Bacterial natural product biosynthetic genes, canonically clustered, have been increasingly found to rely on hidden enzymes encoded elsewhere in the genome for completion of biosynthesis. The study and application of lanthipeptides are frequently hindered by unclustered protease genes required for final maturation. Here, we establish a global correlation network bridging the gap between lanthipeptide precursors and hidden proteases. Applying our analysis to 161,954 bacterial genomes, we establish 5209 correlations between precursors and hidden proteases, with 91 prioritized. We use network predictions and co-expression analysis to reveal a previously missing protease for the maturation of class I lanthipeptide paenilan. We further …

Faculty Publications

Depression is one of the biggest health issues among men who have sex with men (MSM) living with HIV, where sexual identity might play an intricate role. Yet, findings of the relationship between sexual identity and depression were mixed and few studies explored its underlying mechanisms. This study aimed to examine the association between sexual identity and depression, and the potential mediating role of HIV-related stigma and moderating role of age. A cross-sectional survey was conducted among 203 MSM living with HIV in Guangxi, China. Participants provided information on sexual identity, depression, HIV-related stigma, and background information. Descriptive statistics, bivariate …

Faculty, Staff and Student Publications

Wwox-deficient human cells show elevated homologous recombination, leading to resistance to killing by double-strand break-inducing agents. Human Wwox binds to the Brca1 981-PPLF-984 Wwox-binding motif, likely blocking the pChk2 phosphorylation site at Brca1-S988. This phosphorylation site is conserved across mammalian species; the PPLF motif is conserved in primates but not in rodents. We now show that murine Wwox does not bind Brca1 near the conserved mouse Brca1 phospho-S971 site, leaving it open for Chk2 phosphorylation and Brca1 activation. Instead, murine Wwox binds to Brca1 through its BRCT domain, where pAbraxas, pBrip1, and pCtIP, of the A, B, and C binding …

A Mixed Methods Analysis Of Maternal-Infant Feeding Interactions Between Dyads With Opioid Exposure, Kailey T. Rinaldi Mar 2022

A Mixed Methods Analysis Of Maternal-Infant Feeding Interactions Between Dyads With Opioid Exposure, Kailey T. Rinaldi

USF Tampa Graduate Theses and Dissertations

Infants diagnosed with neonatal opioid withdrawal syndrome (NOWS) exhibit neurobehavioral dysregulation that may make them challenging to feed, potentially impeding positive early maternal-infant interactions. The goal of this study was to investigate dyadic behavior during the maternal-infant feeding interaction, specifically infant dysregulation and maternal sensitivity, between dyads with opioid exposure in the neonatal intensive care unit (NICU). This was a secondary data analysis using a mixed-methods design to analyze videorecorded feeding interactions of mothers with opioid use disorder (OUD) and their infants with NOWS. An integrative literature review investigating maternal sensitivity in mothers with OUD was conducted. Verbalizations expressed by …

Research outputs 2022 to 2026

Previous studies have indicated that physical activity may be beneficial in reducing the risk for Alzheimer's disease (AD), although the underlying mechanisms are not fully understood. The goal of this study was to evaluate the relationship between habitual physical activity levels and brain amyloid deposition and AD-related blood biomarkers (i.e., measured using a novel high-performance mass spectrometry-based assay), in apolipoprotein E (APOE) ε4 carriers and noncarriers. We evaluated 143 cognitively normal older adults, all of whom had brain amyloid deposition assessed using positron emission tomography and had their physical activity levels measured using the International Physical Activity Questionnaire (IPAQ). We …

Genetic Connectivity Of Trypanosomes Between Tsetse-Infested And Tsetse-Free Areas Of Kenya, Naomi N. Kimenyi, Kelvin M. Kimenyi, Nelson O. Amugune, Merid N. Getahun

All Peer-Reviewed Publications

The prevalence rates of trypanosomes, including those that require cyclical transmission by tsetse flies, are widely distributed in Africa. Trypanosoma brucei and Trypanosoma congolense are actively maintained in regions where there are no tsetse flies although at low frequencies. Whether this could be due to an independent evolutionary origin or multiple introduction of trypanosomes due to continuous movement of livestock between tsetse-free and -infested areas is not known. Thus, the aim of the study was to carry out microsatellite genotyping to explore intra-specific genetic diversity between T. (Trypanozoon), T. congolense and Trypanosoma vivax from the two regions: tsetse infested and …
