Overexpression Of Sox11 Promotes Corticospinal Tract Regeneration After Spinal Injury While Interfering With Functional Recovery, Zimei Wang, Ashley Reynolds, Adam Kirry, Christopher Nienhaus, Murray G. Blackmore

Biomedical Sciences Faculty Research and Publications

Embryonic neurons, peripheral neurons, and CNS neurons in zebrafish respond to axon injury by initiating pro-regenerative transcriptional programs that enable axons to extend, locate appropriate targets, and ultimately contribute to behavioral recovery. In contrast, many long-distance projection neurons in the adult mammalian CNS, notably corticospinal tract (CST) neurons, display a much lower regenerative capacity. To promote CNS repair, a long-standing goal has been to activate pro-regenerative mechanisms that are normally missing from injured CNS neurons. Sox11 is a transcription factor whose expression is common to a many types of regenerating neurons, but it is unknown whether suboptimal Sox11 expression contributes …

The Development Of Listen: A Novel Intervention For Loneliness, Laurie A. Theeke Phd, Jennifer A. Mallow Phd

Faculty & Staff Scholarship

Purpose: The purpose of this paper is to present the development of LISTEN (Loneliness Intervention

using Story Theory to Enhance Nursing-sensitive outcomes), a new intervention for loneliness.

Methods: LISTEN was developed using the Medical Research Council (MRC) framework for intervention

development. Extensive literature review revealed that belonging, relating, place in community,

challenges, and meanings of coping were concepts significant to loneliness. Past interventions

were limited but it was determined from a recent meta-analysis that enhanced effectiveness

may result from interventions that targeted the poorly adapted cognitive processes of loneliness.

These processes include social undesirability, stigma, and negative thoughts about self …

Proteomic Signature Of The Murine Intervertebral Disc, Matthew R. Mccann, Priya Patel, Agya Frimpong, Yizhi Xiao, Walter L. Siqueira, Cheryle A. Séguin

Bone and Joint Institute

© 2015 McCann et al. Low back pain is the most common musculoskeletal problem and the single most common cause of disability, often attributed to degeneration of the intervertebral disc. Lack of effective treatment is directly related to our limited understanding of the pathways responsible for maintaining disc health. While transcriptional analysis has permitted initial insights into the biology of the intervertebral disc, complete proteomic characterization is required. We therefore employed liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) protein/peptide separation and mass spectrometric analyses to characterize the protein content of intervertebral discs from skeletally mature wild-type mice. A total …

Directed Evolution Of A Yeast-Displayed Hiv-1 Sosip Gp140 Spike Protein Toward Improved Expression And Affinity For Conformational Antibodies, Sebastian K. Grimm, Michael B. Battles, Margaret E. Ackerman

Dartmouth Scholarship

Design of an envelope-based immunogen capable of inducing a broadly neutralizing anti- body response is thought to be key to the development of a protective HIV-1 vaccine. How- ever, the broad diversity of viral variants and a limited ability to produce native envelope have hampered such design efforts. Here we describe adaptation of the yeast display sys- tem and use of a combinatorial protein engineering approach to permit directed evolution of HIV envelope variants. Because the intrinsic instability and complexity of this trimeric glyco- protein has greatly impeded the development of immunogens that properly represent the structure of native envelope, …

Mdr1 Sirna Loaded Hyaluronic Acid-Based Cd44 Targeted Nanoparticle Systems Circumvent Paclitaxel Resistance In Ovarian Cancer, Xiaoqian Yang, Arun K. Iyer, Amit Singh, Edwin Choy, Francis J. Hornicek, Mansoor M. Amiji, Zhenfeng Duan

Pharmaceutical Sciences Faculty Publications

Development of multidrug resistance (MDR) is an almost universal phenomenon in patients with ovarian cancer, and this severely limits the ultimate success of chemotherapy in the clinic. Overexpression of the MDR1 gene and corresponding P-glycoprotein (Pgp) is one of the best known MDR mechanisms. MDR1 siRNA based strategies were proposed to circumvent MDR, however, systemic, safe, and effective targeted delivery is still a major challenge. Cluster of differentiation 44 (CD44) targeted hyaluronic acid (HA) based nanoparticle has been shown to successfully deliver chemotherapy agents or siRNAs into tumor cells. The goal of this study is to evaluate the ability of …

Posterior Approach To Kidney Dissection: An Old Surgical Approach For Integrated Medical Curricula, Frank J. Daly, David L. Bolender, Deepali Jain, Sheryl Uyeda, Todd M. Hoagland

Biomedical Sciences Faculty Publications

Integrated medical curricular changes are altering the historical regional anatomy approach to abdominal dissection. The renal system is linked physiologically and biochemically to the cardiovascular and respiratory systems, yet anatomist often approach the urinary system as part of the abdomen and pelvic regions. As part of an integrated curriculum, the renal system must be covered relatively quickly after the thorax in the cadaver lab, often without the opportunity to fully appreciate the rest of the abdominal contents. This paper provides dissection instructions that follow the one of the historical surgical approaches for nephrectomy, including preservation of the posterior abdominal wall …

Molecular Docking And Inhibition Of Matrix Metalloproteinase-2 By Novel Difluorinatedbenzylidene Curcumin Analog, Aamir Ahmad, Afreen Sayed, Kevin R. Ginnebaugh, Vivek Sharma, Anita Suri, Arundhati Saraph, Subhash Padhye, Fazlul H. Sarkar

Department of Pathology

We recently described the synthesis and characterization of a novel difluorinatedbenzylidene analog of curcumin, commonly referred as CDF, which demonstrated significantly enhanced bioavailability and in vivo anticancer activity. CDF targets many factors similar to curcumin, albeit with more potency, as reported previously. To further highlight this differential behavior of CDF, we chose matrix metalloproteinase protein MMP-2 which is involved in the processes of invasion and metastasis of human tumors. Both curcumin and CDF were characterized for their binding characteristics using in silico docking studies; they were also evaluated via biological assays involving gelatin zymography, miRNA analysis, invasion assays and ELISA. …

Interleukin-8 (IL-8) has been implicated in the pathogenesis of several human respiratory diseases, including tuberculosis (TB). Importantly and in direct relevance to the objectives of this report quite a few findings suggest that the presence of IL-8 may be beneficial for the host. IL-8 may aid with mounting an adequate response during infection with Mycobacterium tuberculosis (M. tb); however, the underlying mechanism remains largely unknown. The major goal of our study was to investigate the contribution of IL-8 to the inflammatory processes that are typically elicited in patients with TB. We have shown for the first time that IL-8 can …

Anti-Tumoral Effects Of Mir-3189-3p In Glioblastoma, Duane Jeansonne, Mariacristina Deluca, Luis Marrero, Adam Lassak, Marco Pacifici, Dorota Wyczechowska, Anna Wilk, Krzysztof Reiss, Francesca Peruzzi

School of Medicine Faculty Publications

Glioblastoma is one of the most aggressive brain tumors. We have previously found up-regulation of growth differentiation factor 15 (GDF15) in glioblastoma cells treated with the anticancer agent fenofibrate. Sequence analysis of GDF15 revealed the presence of a microRNA, miR-3189, in the single intron. We then asked whether miR-3189 was expressed in clinical samples and whether it was functional in glioblastoma cells. We found that expression of miR-3189-3p was down-regulated in astrocytoma and glioblastoma clinical samples compared with control brain tissue. In vitro, the functionality of miR-3189-3p was tested by RNA-binding protein immunoprecipitation, and miR-3189-3p coimmunoprecipitated with Argonaute 2 together …

Diversification Of Importin-Α Isoforms In Cellular Trafficking And Disease States., Ruth A. Pumroy, Gino Cingolani Feb 2015

Diversification Of Importin-Α Isoforms In Cellular Trafficking And Disease States., Ruth A. Pumroy, Gino Cingolani

Department of Biochemistry and Molecular Biology Faculty Papers

The human genome encodes seven isoforms of importin α which are grouped into three subfamilies known as α1, α2 and α3. All isoforms share a fundamentally conserved architecture that consists of an N-terminal, autoinhibitory, importin-β-binding (IBB) domain and a C-terminal Arm (Armadillo)-core that associates with nuclear localization signal (NLS) cargoes. Despite striking similarity in amino acid sequence and 3D structure, importin-α isoforms display remarkable substrate specificity in vivo. In the present review, we look at key differences among importin-α isoforms and provide a comprehensive inventory of known viral and cellular cargoes that have been shown to associate preferentially with specific …

Postcolonial Disability In Mohesen Makhmalbaf’S Kandahar, Sukshma Vedere Feb 2015

Postcolonial Disability In Mohesen Makhmalbaf’S Kandahar, Sukshma Vedere

Ray Browne Conference on Cultural and Critical Studies (2015-2021)

Kandahar (2001), an Iranian film directed by Mohsen Makhmalbaf, details the journey of the protagonist, Nafas, to Kandahar to save her sister from committing suicide on the day of the solar eclipse. The film has gained recent attention by disability studies scholars for the representation of disability in Afghanistan; scholars have discussed the significance of prosthetics and international aid for the disabled in post-war zones of the Third World, but little has been said about disability as a postcolonial embodiment. I argue that Kandahar represents the postcolonial state as a disabled space both literally and metaphorically. It projects the veil …

Measuring Organisational Readiness For Patient Engagement (More): An International Online Delphi Consensus Study, Linda J. M. Oostendorp, Marie-Anne Durand, Amy Lloyd, Glyn Elwyn

Dartmouth Scholarship

Widespread implementation of patient engagement by organisations and clinical teams is not a reality yet. The aim of this study is to develop a measure of organisational readiness for patient engagement designed to monitor and facilitate a healthcare organisation’s willingness and ability to effectively implement patient engagement in healthcare. The development of the MORE (Measuring Organisational Readiness for patient Engagement) scale was guided by Weiner’s theory of organisational readiness for change. Weiner postulates that an organisation’s readiness is determined by both the willingness and ability to implement the change (i.e. in this context: patient engagement). A first version of the …
