Cancer Incidence In First And Second Degree Relatives Of Brca1 And Brca2 Mutation Carriers, Haley Streff May 2015

Cancer Incidence In First And Second Degree Relatives Of Brca1 And Brca2 Mutation Carriers, Haley Streff

Dissertations and Theses (Open Access)

Mutations in the BRCA1 or BRCA2 genes are associated with increased risks for breast, ovarian, and several other cancers. The purpose of this study was to evaluate the incidence of cancers in first and second degree relatives of BRCA mutation carriers compared to the general population. A total of 1086 pedigrees of BRCA mutation carriers were obtained from a prospectively maintained, internal review board approved study of persons referred for clinical genetic counseling at The University of Texas MD Anderson Cancer Center. We identified 9032 first and second degree relatives from 784 pedigrees which demonstrated a clear indication of parental …

Obstetrician And Gynecologist Utilization Of The Nipt Expanded Testing Option, Sarah Mayes Ba May 2015

Obstetrician And Gynecologist Utilization Of The Nipt Expanded Testing Option, Sarah Mayes Ba

Dissertations and Theses (Open Access)

Noninvasive prenatal testing (NIPT) enables the detection of common fetal aneuploidies such as trisomy 21, trisomy 18, trisomy 13, and sex chromosome abnormalities via analysis of cell-free fetal DNA circulating in maternal serum. Although the accuracy of NIPT for fetal aneuploidy is expected to be higher than that of currently available alternative maternal serum screening options, the implications of results are not straight forward. In October 2013, the option to screen for additional trisomies and select microdeletion syndromes, such as 22q11.2 deletion syndrome and 5 p- syndrome, became clinically available. Due to this rapidly evolving prenatal screening technology, clinicians must …

Two-Color Fluorescent Analysis Of Connexin 36 Turnover And Trafficking – Relationship To Functional Plasticity, Yanran Wang May 2015

Two-Color Fluorescent Analysis Of Connexin 36 Turnover And Trafficking – Relationship To Functional Plasticity, Yanran Wang

Dissertations and Theses (Open Access)

Gap junctions (GJ) formed of Cx36 show tremendous functional plasticity on several time scales. Changes in connexin phosphorylation modify coupling in minutes through an order of magnitude, but recent studies also imply involvement of connexin turnover in regulating cell-cell communication. We utilized Cx36 with an internal HaloTag to study Cx36 turnover and trafficking in cultured cells to discriminate newly formed and pre-existing Cx36. New Cx36 in cargo vesicles was added directly to existing gap junctions and newly made Cx36 was not confined to points of addition, but diffused throughout existing gap junctions. Existing connexins also diffused into photobleached areas with …

Management Of Complications Of Wrist Arthroplasty And Wrist Fusion., Michael P. Gaspar, Patrick M Kane, Eon K. Shin May 2015

Management Of Complications Of Wrist Arthroplasty And Wrist Fusion., Michael P. Gaspar, Patrick M Kane, Eon K. Shin

Department of Orthopaedic Surgery Faculty Papers

The human wrist joint is unique from functional and anatomic standpoints. Numerous articulations exist within the wrist that allow for many options for partial wrist fusion and arthroplasty. In cases of pancarpal disease, fusion or arthroplasty of the entire wrist joint can be performed. Because of the high functional demand of the wrist, many of these surgical options can fail, leading to devastating complications. This article addresses the types of fusions and arthroplasties available for the wrist and discusses the potential complications associated with each. Methods to prevent these complications are presented and those to treat them once they have …

The Paleoepidemiology Of Malaria In The Ancient Near East, Nicole Elizabeth Smith May 2015

The Paleoepidemiology Of Malaria In The Ancient Near East, Nicole Elizabeth Smith

Graduate Theses and Dissertations

The end of the Late Bronze Age in the Near East (1300 - 1200 BCE) saw the widespread collapse of several large cultural centers, the reasons for which are a subject of continued debate. Evidence from events leading up to this cultural collapse suggest epidemic disease may have factored into the eventual downfall of these early civilizations. Recent DNA analysis from Egyptian mummies who lived during the period leading up to the Late Bronze Age collapse identified malaria in several elite individuals, suggesting the widespread prevalence of this infectious disease in Egypt. However, the exact prevalence, antiquity, and dynamics of …

Barnstable Brown Diabetes Center Faculty Publications

Lipid accumulation in liver and skeletal muscle contributes to co-morbidities associated with diabetes and obesity. We made a transgenic mouse in which the adiponectin (Adipoq) promoter drives expression of lipoprotein lipase (LPL) in adipocytes to potentially increase adipose tissue lipid storage. These mice (Adipoq-LPL) have improved glucose and insulin tolerance as well as increased energy expenditure when challenged with a high fat diet (HFD). To identify the mechanism(s) involved, we determined whether the Adipoq-LPL mice diverted dietary lipid to adipose tissue to reduce peripheral lipotoxicity, but we found no evidence for this. Instead, characterization …

Coaching And Concussions: Prevention, Education, And Perspective, Colby J. Felts May 2015

Coaching And Concussions: Prevention, Education, And Perspective, Colby J. Felts

Honors Thesis

The large participation of players and coaches in football has made it one of the most popular sports in America. Concussions have specifically gained national attention in football due to frequent, high impact collisions. A concussion injury poses a unique threat in high school football because of the developing adolescent brain and high participation numbers. Additionally, lack of medical personnel to diagnose concussions at the high school level imposes a legal duty on the coaches. The thesis develops a recommended plan of action for directors of Iowa high school football to assist in preserving the future of the sport. A …

Accidental Phosphine Gas Poisoning With Fatal Myocardial Dysfunction In Two Families, Saleem Akhtar, Arshalooz Rehman, Surraya Bano, Anwarul Haque

Department of Emergency Medicine

ABSTRACT Aluminum phosphide is commonly used as a rodenticide and insecticide and is one of the most fatal poisons. The active ingredient is Phosphine gas which inhibits cytochrome oxidase and cellular oxygen utilization. The clinical symptoms are due to multiorgan involvement including cardiac toxicity which is the most common cause of mortality. Severity of clinical manifestations depends upon the amount of the gas to which a person is exposed. There is no specific antidote available. High index of suspicion and early aggressive treatment is the key to success. We report 2 cases of aluminum phosphide toxicity in 2 families due …

Introduction: Cancer metastasis to mediastinal and hilar lymph nodes is indicative of a poor prognosis. It is thus important to understand the pathogenesis of cancer metastasis on a cellular and molecular level with a goal to develop novel therapies directed towards prevention and treatment. In this study we compared cytokine expression in benign versus malignant mediastinal and hilar lymph nodes using EBUS-FNA biopsy specimen. Methods: In this prospective, single center study, we collected EBUS-FNA biopsies from patients recruited using a convenience sampling approach. Cytokines were assayed using Bio-Plex Pro human cancer biomarker panels 1 and 2 in a Bio-Rad 200 …

Yeast V-Atpase Regulation By Phosphofructokinase-1, Chun-Yuan Chan May 2015

Yeast V-Atpase Regulation By Phosphofructokinase-1, Chun-Yuan Chan

Biomedical Sciences ETDs

V-ATPase is a vacuolar (lysosome-like) ATPase-dependent proton pump necessary for maintaining pH homeostasis in the organelles of the endomembrane system. It also contributes to regulation of the cytosol pH and the extracellular pH. In specialized cells (renal intercalated cells, epididymis clear cells, and osteoclasts), V-ATPase proton transport supports urinary acidification, sperm maturation, and bone resorption. Genetic mutations of V-ATPase expressed in those tissue-specific cells cause distal renal tubular acidosis, infertility, and osteopetrosis. V-ATPases are composed of a peripheral domain (V1), which hydrolyzes ATP, and a membrane-bound domain (Vo), which transports proton. V-ATPase activity is tightly regulated in vivo by numbers …

Obesity among children and adolescents is increasingly becoming a major public health problem. Poverty and lower socio-economic status as well as lack of parental awareness regarding a balanced diet and adequate exercise all contribute towards childhood obesity. Unfortunately minority children have been especially vulnerable, among them inner-city Latino children continue to have the highest incidence rates of overweight and obesity; hence targeted intervention for this population group is of paramount importance. Access and affordability of smartphones and mobile internet devices provide an opportunity to create nutrition education interventions that can significantly impact and change attitude and knowledge regarding healthy diet …

Use Of Dietary And Herbal Supplements In Older Adults With Osteoarthritis, Sharon Denise Mcdonald May 2015

Use Of Dietary And Herbal Supplements In Older Adults With Osteoarthritis, Sharon Denise Mcdonald

Dissertations

Over the past several decades, older adults with Osteoarthritis (OA) have increasingly used more natural products in an effort to manage chronic pain. The theoretical framework for this study was the Lazarus and Folkman transactional theory of stress and coping. This framework was chosen based on the belief that older adults with OA are taking natural products as an active problem solving coping strategy to manage the pain, disability, and alterations they experience in their health related quality life including functional status, physical symptoms, emotional health, and social functioning. Older adults also take prescription medications for Osteoarthritis and a variety …

A Measure Of Soft Skill Gains Acquisition With Engagement In Baccalaureate Nursing Programs, Wendy J. Williams-Buenzli

Dissertations

This dissertation examined soft skill acquisition from engagement in baccalaureate nursing programs. Baccalaureate nursing programs prepare nurses to be the future leaders of nursing practice. Soft skills include critical thinking, interpersonal communications, and ethical decision-making. To explore the acquisition of soft skills, this research study analyzed data from the National Survey of Student Engagement to explore for gains in critical thinking, ethical decision-making, and critical thinking. After gains scores were identified, then correlations were run to identify key engagement indicators. The findings of the study found gains in soft skill behaviors from freshman year to senior year of baccalaureate education. …

Precoding And The Accuracy Of Automated Analysis Of Child Language Samples, Rachel Christine Winiecke May 2015

Precoding And The Accuracy Of Automated Analysis Of Child Language Samples, Rachel Christine Winiecke

Theses and Dissertations

Language sample analysis is accepted as the gold standard in child language assessment. Unfortunately it is often viewed as too time consuming for the practicing clinician. Over the last 15 years a great deal of research has been invested in the automated analysis of child language samples to make the process more time efficient. One step in the analysis process may be precoding the sample, as is used in the Systematic Analysis of Language Transcripts (SALT) software. However, a claim has been made (MacWhinney, 2008) that such precoding in fact leads to lower accuracy because of manual coding errors. No …

Computer Science Faculty publications

Comparative co-localization analysis of transcription factors (TFs) and epigenetic marks (EMs) in specific biological contexts is one of the most critical areas of ChIP-Seq data analysis beyond peak calling. Yet there is a significant lack of user-friendly and powerful tools geared towards co-localization analysis based exploratory research. Most tools currently used for co-localization analysis are command line only and require extensive installation procedures and Linux expertise. Online tools partially address the usability issues of command line tools, but slow response times and few customization features make them unsuitable for rapid data-driven interactive exploratory research. We have developed PAPST: Peak Assignment …

A Cohort Study Investigating Autoantibody Levels During And After Infection With Influenza A Virus, Michelle Collins

All NMU Master's Theses

Antinuclear autoantibodies (ANAs) are present in all individuals. In those with autoimmune diseases they are routinely present in elevated levels. Although the nature and development of autoimmune diseases are not fully understood there are many hypotheses as to possible causes of an autoimmune disorder. One possible cause is viral infections. The scope of this thesis study was to examine if autoantibodies levels in individuals without autoimmune disorders increase during or after infection with influenza A virus. Blood was collected from volunteers (n=11) at time intervals of 0, 7, 42 and 63 days, respectively. Antibody levels were measured using ELISA assays …
