Posttranslational modification of macromolecules by ubiquitin-like proteins (UBLs) such as ubiquitin, Sumo and NEDD8 regulate a vast array of processes in the cell. Transfer of UBLs to their target generally occurs by a series of molecular handoffs down an E1‑E2‑E3 cascade. We are interested in understanding how E1‑E2 pairs interact and mediate UBL transfer. To this effect, we studied two E1‑E2 pairs: the Sumo pathway (Sumo utilizes a canonical E1 and E2) and the Atg8 pathway (Atg8 is a UBL involved in autophagy that utilizes a non-canonical E1‑E2 pair).

A biomechanical study was performed to compare range of motion of the multi-body lumbar spine using three different protocols: pure moment, eccentric loading, and a new method called combined loading moment. PTEN (Phosphatase and Tensin homolog deleted on chromosome 10) is a tumor suppressor gene that is commonly mutated in multiple types of human cancers including endometrial, prostate, breast cancers and glioblastoma multiforme (GBM). PTEN functions as a phospholipid phosphatase antagonizing phosphorylation by phosphatidylinositol‑3‑OH kinases (PI3Ks) and thus serves as a major negative regulator of the PI3K pathway. Homozygous Pten knock-out mice die during embryogenesis. Neuroprotection is a strategy to prevent or reduce neuronal cell death regardless of the underlying cause. Erythropoietin (EPO) exhibits neuroprotective effects in a variety of in vitro and in vivo models of neuronal cell death although its classical function is the regulation of red blood cell production. EPO crosses the blood brain barrier and therefore can be delivered systemically. However, long term treatment with exogenous EPO causes over stimulation of the erythropoietic pathway which can be lethal. Localization acuity of a given listener is dependent upon the ability discriminate between interaural time and level disparities. Interaural time differences are encoded by low frequency information whereas interaural level differences are encoded by high frequency information. Much research has examined effects of hearing aid microphone technologies and occlusion separately and prior studies have not evaluated age as a factor in localization acuity. Open-fit hearing instruments provide new earmold technologies and varying microphone capabilities; however, these instruments have yet to be evaluated with regard to horizontal localization acuity.

Platelet activation initiates an upsurge in 18:2 and 20:4 lysophosphatidic acid (LPA) production. The biochemical pathway responsible for LPA production during blood clotting is not fully understood. We have purified a phospholipase A1 (PLA1) from thrombin-activated human platelets using sequential chromatographic steps followed by fluorophosphonate‑biotin affinity labeling and proteomics. We identified acyl‑protein thioesterase 1 (aka. lysophospholipase A1, accession code O75608) as a novel PLA1. Addition of this recombinant PLA1 significantly increased the production of sn‑2‑esterified polyunsaturated LPCs and the corresponding LPAs in plasma. Purpose. The purpose of this study was to examine the influence of patient‑perceived provider availability on nonurgent emergency department use (NUEDU). The study population consisted of a nationally‑representative population of non‑elderly adults who were continuously privately insured (CPI) for at least a year before an emergency department visit that was determined to be nonurgent, and who reported having a regular source of care (RSC).

Background: There are few studies that examine patients' costs of oral oncology drugs from Medicare Part D plan providers. These drugs can impose a financial burden to beneficiaries, and, due to their cost, place beneficiaries in the coverage gap in a short period of time. In addition to examining costs, there is little published concerning the potential impact of drug utilization management techniques on access to these drugs in Medicare Part D plans. This study examined the roles of prior authorization, step-therapy, and placement of drugs in higher formulary tiers on patients' out-of-pocket costs, and access to therapy.

Nursing presence is conceptualized as occurring within the nurse-patient relationship when the nurse acknowledges the uniqueness of the patient, within his or her context of being and chooses to intervene on the patient's behalf with a patient who allows the nurse into a reciprocal relationship. Blast-induced neurotrauma (BINT) is a newly emerging re-occurrence of an old combat-related injury in U.S. military troops returning from deployment in Afghanistan and Iraq. This dissertation study examined the phenomenon of emergence delirium in U.S. military combat veterans. Emergence delirium is a post-anesthetic phenomenon that occurs immediately following emergence from general anesthesia and is characterized by agitation, confusion, and violent physical and/or verbal behavior. Clinical evidence suggests that emergence delirium is occurring with greater frequency among military personnel returning from conflict in the Middle East. This body of work is a culmination of three distinctive phases and is presented in three individual manuscripts. Purpose The purpose of this research study was to describe the nutritional status (well nourished, at risk for malnutrition, or malnourished) of residents admitted to nursing homes and adverse clinical outcomes in those residents after four weeks. Methodology A descriptive, correlational design utilizing medical record data was used to examine the relationship between nutritional status and adverse clinical outcomes of weight loss and pressure ulcers. Results The sample (n=69) was predominately Caucasian (90%) with 38% having one or more pressure ulcers on admission. A series of experiments were conducted to determine the effects of on-arrival vs. delayed respiratory vaccination and exposure to persistently infected bovine viral diarrhea virus challenge on health, gain performance, and physiological and immunological measurements in newly received stocker cattle. Two experiments evaluated timing (d 0 vs. 14) of respiratory, clostridial, or both vaccinations in newly received stocker calves during stress-induced immunosuppression. Female athletes have a four-to-six times greater risk of sustaining a lower extremity injury than their male counterparts (Hewett, 2000). One explanation for the discrepancy in injury rates is increased dynamic knee valgus angles caused by neuromuscular imbalances of the lumbopelvic hip complex (LPHC). The purpose of this study was to determine if implementing a LPHC conditioning program improved neuromuscular deficiencies. Seven female collegiate tennis players competitive at the NCAA Division I level volunteered for the study (19.7 + .95 years, 167.3 + 7.4 cm, and 60.72 kg + 6.71 kg). The lesser mealworm, Alphitobius diaperinus (Panzer) (Coleoptera: Tenebrionidae), is a serious, cosmopolitan pest present in poultry production facilities, where it slows down weight gain in chicks, carries disease–causing organisms, and causes damage to poultry insulation. The first study estimated that the lesser mealworm densities in poultry litter applied to pastures were 3.5, 2.2 and 0.4 million larvae, adults and pupae, respectively, per hectare in Savoy, Arkansas. The repercussions of these adult beetles are that they often disperse to and reinfest poultry houses with the potential to vector disease to poultry, and disperse to invade human dwellings resulting in litigation. Neurofibromatosis type 1 (NF-1) is a multisystem neurocutaneous disorder resulting from mutations in the NF-1 gene on chromosome 17 (q11.2). Mutations in NF-1 result in deficient activity of the tumor suppresor protein neurofibromin, allowing for uncontrolled constitutive activity of the proliferative Raspathway in several neural cell types, including neurons, oligodendrocytes, astrocytes and Schawnn cells. Half of NF-1 cases are inherited in autosomal dominant fashion, while half develop sporadically. The syndrome has 100% penetrance but highly variable expressivity. This study was designed to determine whether adult mouse induced pluripotent stem cells (iPSCs), could be used to produce retinal precursors and subsequently photoreceptor cells for retinal transplantation to restore retinal function in degenerative hosts. iPSCs were generated using adult dsRed mouse dermal fibroblasts via retroviral induction of the transcription factors Oct4, Sox2, KLF4 and c-Myc. As with normal mouse ES cells, adult dsRed iPSCs expressed the pluripotency genes SSEA1, Oct4, Sox2, KLF4, c-Myc and Nanog. Following transplantation into the eye of immunecompromised retinal degenerative mice these cells proceeded to form teratomas containing tissue comprising all three germ layers. At …
