The field of radiation oncology seeks novel techniques to increase the therapeutic ratio to treat malignancies while minimizing damage and toxicities in healthy tissues. Spatial proteomics can visualize and quantify protein expression profiles within tissues at single-cell resolution. Although spatial proteomics can only detect a limited number of proteins compared to spatial transcriptomics, it provides comprehensive spatial information with single-cell resolution. By studying the spatial distribution of cells, we can clearly obtain the spatial context within tissues at multiple scales. Spatial context includes the spatial composition of cell types, the distribution of functional structures, and the spatial communication between functional regions, all of which are crucial for the patterns of cellular distribution. Alternative splicing is a crucial cellular process in eukaryotes, enabling the generation of multiple protein isoforms with diverse functions from a single gene. To better understand the impact of alternative splicing on protein structures, protein-protein interaction and human diseases, we developed ASpdb (https://biodataai.uth.edu/ASpdb/), a comprehensive database integrating experimentally determined structures and AlphaFold 2-predicted models for human protein isoforms. ASpdb includes over 3400 canonical isoforms, each represented by both experimentally resolved and predicted structures, and >7200 alternative isoforms with AlphaFold 2 predictions. Cancer metastasis, the process by which tumour cells migrate and colonize distant organs from a primary site, is responsible for the majority of cancer-related deaths. Understanding the cellular and molecular mechanisms underlying this complex process is essential for developing effective metastasis prevention and therapy strategies. To this end, we systematically analysed 1786 bulk tissue samples from 13 cancer types, 988 463 single cells from 17 cancer types, and 40 252 spots from 45 spatial slides across 10 cancer types. The CRISPR (clustered regularly interspaced short palindromic repeats)/Cas (CRISPR-associated protein) programmable nuclease system continues to evolve, with in vivo therapeutic gene editing increasingly applied in clinical settings. However, off-target effects remain a significant challenge, hindering its broader clinical application. To enhance the development of gene-editing therapies and the accuracy of prediction algorithms, we developed CRISPRoffT (https://ccsm.uth.edu/CRISPRoffT/). Users can access a comprehensive repository of off-target regions predicted and validated by a diverse range of technologies across various cell lines, Cas enzyme variants, engineered sgRNAs (single guide RNAs) and CRISPR editing systems. Pediatric low-grade gliomas (pLGGs) exhibit heterogeneous prognoses and variable responses to treatment, leading to tumor progression and adverse outcomes in cases where complete resection is unachievable. Early prediction of treatment responsiveness and suitability for immunotherapy has the potential to improve clinical management and outcomes. Here, we present a radiogenomic analysis of pLGGs, integrating MRI and RNA sequencing data. We identify three immunologically distinct clusters, with one group characterized by increased immune activity and poorer prognosis, indicating potential benefit from immunotherapies. We develop a radiomic signature that predicts these immune profiles with over 80% accuracy. Furthermore, our clinicoradiomic model predicts progression-free …

The Optimal electrode configuration of Electroencephalograms (EEG) systems for mild cognitive impairment (MCI) detection and monitoring in non-clinical settings, i.e. number of electrodes and the positions of the electrodes, remains to be explored. In the current study, we explored the optimization of electrode configuration for MCI detection. We used a 32-channel EEG device to record the data of 21 MCI patients and 20 cognitively normal elderly (NC) undergoing working memory (WM) tasks. Integrating artificial intelligence (AI) into nursing education presented significant opportunities yet posed challenges due to varied faculty readiness. This Doctor of Nursing Practice (DNP) quality improvement (QI) project evaluated an educational intervention aimed at enhancing nursing faculty's AI proficiency and confidence at Regis University’s Rueckert-Hartman College for Health Professions. Using a mixed-methods, pre- and post-intervention design, validated surveys assessed changes in faculty perceptions, knowledge, and skills related to AI. This study investigates the prevalence and underlying causes of work-related stress and burnout among cybersecurity professionals using a quantitative survey approach guided by the Job Demands-Resources model. Analysis of responses from 50 cybersecurity practitioners reveals an alarming reality: 44% report experiencing severe work-related stress and burnout, while an additional 28% are uncertain about their condition. The demanding nature of cybersecurity roles, unrealistic expectations, and unsupportive organizational cultures emerge as primary factors fueling this crisis. Notably, 66% of respondents perceive cybersecurity jobs as more stressful than other IT positions, with 84% facing additional challenges due to the pandemic and recent high-profile …

Per- and polyfluoroalkyl substances (PFAS) contamination has posed a significant environmental and public health challenge due to their ubiquitous nature. Adsorption has emerged as a promising remediation technique, yet optimizing adsorption efficiency remains complex due to the diverse physicochemical properties of PFAS and the wide range of adsorbent materials. Traditional modeling approaches, such as response surface methodology (RSM), struggled to capture nonlinear interactions, while standalone machine learning (ML) models required extensive datasets. As artificial intelligence (AI) transforms drug development, regulatory frameworks are evolving to oversee its implementation, particularly at the US Food and Drug Administration (FDA) and the European Medicines Agency (EMA). This paper makes three contributions to understanding emerging regulatory approaches. First, we offer a comparative analysis of how these agencies have responded to AI-driven advances, incorporating new US executive orders and the European Union (EU)’s AI Act. Second, we propose a novel analytical framework to understand regulatory divergence: the FDA’s flexible, dialog-driven model contrasts with the EMA’s structured, risk-tiered approach, reflecting broader institutional and political-economic differences. In this study, we prospectively and retrospectively evaluated the occurrence of errors in the management of cutaneous disorders from patient visits and medical records in a single dermatology practice in southeast Virginia over a 3-year period (June 2020-July 2023). The complex immune interactions produced by the tetravalent dengue vaccine Dengvaxia have foregrounded the important role of antibody-dependent enhancement (ADE) in dengue infection. Some evidence exists that ADE may extend beyond the four dengue serotypes to Zika, a closely related flavivirus transmitted by the same mosquito species as dengue, and may also account for the increased severity of some cases. Estimates of the public health impact of dengue vaccination may then need to include its effects on the transmission of Zika in addition to dengue. In eukaryotic cells such as Saccharomyces cerevisiae, protein synthesis begins with the transport of mRNA from the nucleus to the cytoplasm. In order for mRNA to leave the nucleus, it must first bind to specific transport proteins which can then facilitate transport via the nuclear pore complex (NPC), a selectively permeable membrane within the cell's nuclear envelope. Under environmental stress, such as heat shock, S. cerevisiae cells modulate transport rates. In this case, in heat stress or “heat shock” most mRNA transcripts will remain in the nucleus. This paper presents a novel Stackelberg-game theoretic multilayer-online learning framework for cooperative control of nonlinear Physical Human-Robot Interaction (pHRI), where the human is modeled as the leader guiding a robot follower. This hierarchical interaction is captured as a dynamic Stackelberg game, with the human's intention estimated in real-time through online multilayer neural networks (MNNs). This paper addresses the infinite horizon optimal tracking control problem for partially uncertain control-affine nonlinear discrete-time (DT) systems, where the control input dynamics are known. Multi-layer critic and actor neural networks (MNNs) are utilized for online estimation of the infinite horizon value function and optimal control input. The NN weights are tuned online using a direct temporal difference error (TDE)-driven learning approach, which modifies the singular values of the gradient with respect to the NN weights to accelerate their convergence. This paper presents a safety-aware deep reinforcement learning (DRL)-based trajectory tracking control of autonomous surface vessels (ASVs). A multilayer neural network (MNN) observer estimates the ASV's state and uncertain dynamics. By utilizing the estimate state vector from the observer, a safety-aware DRL-based optimal policy is formulated using control barrier function (CBF) and Karush-Kuhn-Tucker (KKT) conditions. An actor-critic MNN with singular value decomposition (SVD)-based update mitigates vanishing gradients. This article considers the infinite time horizon optimal tracking control problem for discrete time (DT) partially uncertain strict feedback systems with application to quadrotor UAVs. First, the strict feedback DT system is transformed into an equivalent affine nonlinear DT system in terms of tracking error dynamics. This paper presents a partially model-free adaptive optimal tracking control method for power systems, specifically targeting a synchronous generator connected through a reactive transmission line. By integrating the tracking error dynamics with reference trajectory dynamics, an augmented system is created. A discounted performance function is introduced to address the nonlinear tracking problem optimally. Unlike traditional methods that compute feedforward and feedback terms separately, the proposed approach calculates both simultaneously by minimizing the discounted performance function. This paper presents a unified framework for the safe and optimal control of heterogeneous quadrotor unmanned aerial vehicles (QUAVs) in formation, enabling multitask missions without requiring precise system dynamics. To address partial state observability, a multilayer neural network (MNN) observer is designed to estimate unmeasured states. Reinforcement learning (RL) is employed for optimal control utilizing an MNN ensuring adaptability. Barrier Lyapunov Functions (BLFs) are integrated into the RL framework to enforce safety by maintaining QUAVs within predefined constraints. This study presents an original interdisciplinary investigation into how reinforcement learning (RL) can model motor and cognitive defects and potentially improve motor and cognitive functions in individuals with cerebral palsy (CP), a non-progressive neurological disorder that impairs movement and adaptability. Integrating computational neuroscience and machine learning, the research applies policy gradient methods and Markov Decision Processes (MDPs) to simulate adaptive learning in agents with and without CP-related constraints.

This thesis develops the mathematical foundations of computed tomography (CT) reconstruction through the lens of harmonic analysis. Beginning with the Schwartz class, we introduce the Fourier transform and its role in expressing the Radon transform and its inversion via a fractional Laplacian. After constructing the Radon transform in general dimension R^d, we specialize to the cases d = 2 and d = 3, demonstrating explicit inversion formulas and the associated instability in lower dimensions.
