Triterpenes are thirty-carbon compounds derived from the universal five-carbon prenyl precursors isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP). Normally, triterpenes are synthesized via the mevalonate (MVA) pathway operating in the cytoplasm of eukaryotes where DMAPP is condensed with two IPPs to yield farnesyl diphosphate (FPP), catalyzed by FPP synthase (FPS). Squalene synthase (SQS) condenses two molecules of FPP to generate the symmetrical product squalene, the first committed precursor to sterols and most other triterpenes. In the green algae Botryococcus braunii, two FPP molecules can also be condensed in an asymmetric manner yielding the more highly branched triterpene, botryococcene. Botryococcene …

Genomic prediction (GP) is now routinely performed in crop plants to predict unobserved phenotypes. The use of predicted phenotypes to make selections is an active area of research. Here, we evaluate GP for predicting grain yield and compare genomic and phenotypic selection by tracking lines advanced. We examined four independent nurseries of F3:6 and F3:7 lines trialed at 6 to 10 locations each year. Yield was analyzed using mixed models that accounted for experimental design and spatial variations. Genotype-by-sequencing provided nearly 27,000 high-quality SNPs. Rice yield is highly sensitive to increased temperature. Given the trend of increasing global temperatures, this sensitivity to higher temperatures poses a challenge for achieving global food security. Early seed development in rice is highly sensitive to unfavorable environmental conditions. Heat stress (HS) during this stage decreases seed size and fertility, thus reducing yield. Here, we explore the transgenerational phenotypic consequences of HS during early seed development on seed viability, germination, and establishment. Approximately two thirds of St. John is National Park territory. However, the land has been threatened with tourism and development, greatly impacting island biodiversity. One species that may become extinct due to this degradation is Solanum conocarpum. S. conocarpum is a rare shrub, endemic to the dry tropical forests of St. John, USVI. This plant is a species of conservation concern and is one of very few native and endemic plants on this island. Very little is known about the ecology and reproduction of S. conocarpum. PREMISE OF THE STUDY: Few genetic markers provide phylogenetic information in closely related species of Isoëtes (Isoëtaceae). We describe the development of primers for several putative low- copy nuclear markers to resolve the phylogeny of Isoëtes, particularly in the southeastern United States.
METHODS AND RESULTS: We identified regions of interest in Isoëtes transcriptomes based on low- copy genes in other plants. Primers were designed for these regions and tested with 16 taxa of Isoëtes and one species of Lycopodium. An abrupt transition in the fossil record between Early Devonian plants with simple structure and structurally-complex later Devonian plants, has frustrated efforts to understand patterns of phylogeny across the Early/Middle Devonian boundary and the evolution of complex forms. Both these aspects have important implications for lignophyte and seed plant evolution. In the first chapter, I evaluate phylogenetic relationships between the earliest seed plants, Aneurophytales, and Stenokoleales, using comprehensive taxon sampling (28 species, including all relevant permineralized species) and a set of 40 discrete and nine continuous morpho-anatomical characters. The interactive effect of exogenous application of salicylic acid (SA) and sodium nitroprusside (SNP), a donor of nitric oxide, on zinc (Zn) toxicity was assessed in Zn-stressed safflower (Carthamus tinctorius L.) seedlings. Exposure to 500 μM ZnSO4.7H2O for 10 days caused an increment in the levels of malondialdehyde (MDA), H2O2, α-tocopherol, phytochelatins (PCs), and proline. Application of SA or SNP, and especially their combination, resulted in remarkable decrease in MDA, H2O2, and proline content while chlorophyll content and the activity of catalase, ascorbate peroxidase, and guaiacol peroxidase exhibited a significant induction in comparison with plants subjected to Zn treatment alone. …

Flow cytometry measurement confirmed the genetic stability of Primula veris L. (cowslip) plants obtained during the micropropagation process. The mean 2C DNA content was 0.97 pg/2C and P. veris can be classified in the group of plants of very small genomes. Comparative morphological analysis of cowslip plants regenerated in vitro and derived from seeds was carried out in three subsequent years under common garden conditions, which limited the effects of environmental factors. The origin of specimens significantly influenced most of the morphological traits examined. This study describes the production of calli from Globularia trichosantha subsp. trichosantha and the quantitative determination of catalpol, aucubin, and verbascoside in the calli and the plant by HPLC. The seeds of the plant were sterilized and germinated in Murashige and Skoog (MS) medium without in vitro plant growth regulator. Hypocotyl, cotyledon, first leaf, epicotyl, apical meristem, and root explants were taken from the 30-day-old aseptic seedlings germinated in vitro. Explants were then transferred to MS media for callus production together with varying concentrations of plant growth regulators. Genomic selection (GS) by selecting lines prior to field phenotyping using genotyping data has the potential to enhance the rate of genetic gains. Genotype × environment (G × E) interaction inclusion in GS models can improve prediction accuracy hence aid in selection of lines across target environments. Phenotypic data on 320 chickpea breeding lines for eight traits for three seasons at two locations were recorded. These lines were genotyped using DArTseq (1.6 K SNPs) and Genotyping-by-Sequencing (GBS; 89 K SNPs). Alkaline soils comprise 30% of the earth and have low plant-available iron (Fe) concentration, and can cause iron deficiency chlorosis (IDC). IDC causes soybean yield losses of $260 million annually. However, it is not known whether molecular responses to IDC are equivalent to responses to low iron supply. The unique morphology of quillworts has received limited attention, and such studies have been limited to only a few species. Our extensive field work during the past two decades has revealed variation in the structures of the plant, including the rhizomorph, scales, phyllopodia, and abscission caps. Polarity of the axes of southeastern quillwort rhizomorphs varies from discoid (most species), to elongate, to upright with axis branching in several species. In species of intermittent streams, these branched rhizomorphs produce plantlets that break off in running water. Historically, disturbances shaped prairie ecosystems both directly through events like fire and soil moving by animals, as well as indirectly through dynamic rainfall patterns and periodic flux in seed availabilities. We hypothesized that the implementation of several distinct disturbance events in Nebraska Sandhills upland prairie would have measurable effects on plant community composition. Oxytropis glandulosa Turcz. (Fabaceae) is a rare perennial plant endemic to Buryatia (Russia). We sequenced three intergenic spacers (the psbA−trnH, trnL−trnF, and trnS−trnG) of chloroplast DNA (cpDNA) in specimens of four populations from two geographic regions (Barguzin and Yeravna depressions) within this species range. The levels of haplotype and nucleotide diversity varied in the range of 0.133−0.911 and 0.0002−0.0059, respectively. The highest values of these parameters are characteristic of populations located in the Barguzin depression. In this study, we aimed to investigate the potential distribution area of Solidago xniederederi, a natural hybrid between North American S. canadensis and European S. virgaurea, in Central and East Europe using the MAXENT modeling approach. The final MAXENT model was constructed based on 83 occurrence records from Austria, Poland, Lithuania, and Latvia and six abiotic environmental variables. Xanthagaricus pakistanicus is described as a new species from lowland northern Pakistan, based on morphological and molecular data. It is characterized by a yellowish pileus, covered with dark brown squamules, a stipe with yellowish fibrils, globose basidiospores, and pileal squamules made up of pseudoparenchymatous epithelium with encrusted walls. Molecular phylogenetic trees were inferred based on nucleotide sequences of the internal transcribed spacer (ITS1-5.8S-ITS2) region and 28S nuclear ribosomal DNA. In phylogenetic analyses the genus Hymenagaricus sensu lato (s. l.) was inferred as a nonmonophyletic group and recovered in two monophyletic clades, consisting of species of the genera Xanthagaricus and Hymenagaricus …

The cuticular wax layer covers the aerial surface of plants and acts as a barrier between plants and the environment. The cuticle plays a key role in the protection of plants from pathogens, UV light, and transpiration. Variation in the wax quality and quantity is influenced by factors like the solvent used for extraction, species, ontogeny, and season. In recent years, transgenic approaches have played a significant role in improving traits that help plants overcome abiotic stresses. Combined gene discovery and functional genomics have helped identify diversified mechanisms and gene families, which improved productivity under various abiotic stresses. We report on the genetic stability and persistent morphological features of transgenic pea (Pisum sativum L.) plants harboring the dicistronic vector construct pG0229MASnhx1/luc over five generations. In addition to salt stress tolerance, the transgenic plants showed frost tolerance compared to wild-type (WT) plants. We monitored the only remaining population of Lotus kunkelii on a monthly basis over 3 years to reveal important characteristics of the population that could help protect it. Studies on interactions between Orobanche species and their hosts are mainly focused on resistance, radical interactions, and haustorium development. The influence of the host plant on morphology of the parasite under environmental controlled conditions is not reported in the literature. With the aim of filling this gap, the results of cultivation experiments of Orobanche crenata on Vicia faba, V. sativa, Pisum sativum, Cicer arietinum, and Lathyrus clymenum are presented. Ekimia ozcan-secmenii Şenol & Eroğlu sp. nov. (Apiaceae) is described from SW Turkey. E. ozcan-secmenii differs from its close affinity, E. bornmuelleri, by life form (monocarpic), petal and bracteole morphology, and mericarp features. These data were more supported by the results of nrDNA ITS sequences. The ML tree depicted based on phylogenetic studies aimed to reveal the closeness of the Laserpitium and Prangos species as well as the location the affinity of species in the genus Ekimia. The taxonomic composition of diatom flora of the Balikhli River (Balikhli Chay), Northwest Iran, was studied from October 2013 through September 2014. In total 109 taxa belonging to 50 genera were identified. Nitzschia (18 spp.), Navicula (9 spp.), and Gomphonema (7 spp.) were the most abundant genera. Twenty-five diatom species were found in more than half of the samples; among them, Nitzschia inconspicua, Planothidium lanceolatum, Amphora pediculus, Navicula trivialis, Gomphonema olivaceum, Nitzschia palea, and Rhoicosphenia abbreviata were the most abundant taxa, which are characteristic for alkaline and eutrophic waters. Recently, the number of algological studies in Turkish inland waters has increased remarkably. However, taxonomic and floristic studies on algae in the Euphrates basin are still scarce. This study contributes new information to the knowledge of the Turkish freshwater algal flora. Phytoplankton samples were collected from the Tatar Dam Reservoir in the Euphrates Basin between January 2016 and December 2016. Here we describe two new species of genus Piper L. from Madagascar: Piper malgassicum and Piper tsarasotrae, the species names referring to the currently known distribution areas. These two species contribute, at least in part, to the production of local voatsiperifery pepper, probably mixed together with P. borbonense, and are therefore economically relevant as spices. We used a selected set of characters (those more easily observable on herbarium samples) for principal component analysis to assess the relative distance between the species, including the analysis of the autochthonous species of Piper known from Africa and Madagascar. Nine discomycete and one sordariomycete (Ascomycota) species are reported for the first time from Turkey. The genera Coccomyces, Kompsoscypha, Pseudopithyella, Strobiloscypha, and Lasiosphaeris have not been reported before in the country. Anthracobia, Plicaria, Sclerotinia, and Pithya species are new records added to the previous knowledge. Macro- and micromorphological descriptions and illustrations for each new taxon are provided.

In this study, the macro- and micromorphological seed characteristics of the genus Crocus L. series Crocus taxa, including C. pallasii , C. turcicus , C. dispathaceus , C. asumaniae , C. mathewii , C. thomasii , C. oreocreticus , and C. cartwrightianus , were observed. Morphological characteristics, such as the size, shape, color, and seed surface structures, were analyzed using stereo and scanning electron microscopes.
