Autism Spectrum Disorder (ASD) is a neurodevelopmental disorder that is characterized by abnormal social behavior, deficits in communication, and motor stereotypy. The SHANK3 gene, responsible for the generation of a scaffolding protein that is integral for the development of synapses, has been identified as one of the primary candidate genes implicated in the disorder. Shank3B is the rodent homolog for this gene. Research has shown that when this gene is disrupted in rodent models (e.g., via knock-out (KO)), ASD-like behaviors result. At least five beta-type connexins are expressed in various layers of the skin (Cx26, Cx30, Cx30.3, Cx31, and Cx32) and all include a glycine residue at position 12. Glycine12 (G12) is located about halfway through the cytoplasmic amino terminus and has been the focus of several studies related to connexin diseases and gap junction channel structure. The importance of this residue is evident in the severity and diversity of diseases associated with amino acid substitutions at G12 including hereditary forms of skin disease, deafness and neuropathy. C1q TNF Related Protein 3 (CTRP3), is a cytokine that is primarily secreted from adipose tissue, which classifies it as an adipokine. Our previous research has shown that CTRP3 prevents alcoholic fatty liver disease (ALD) in male mice. However, even when accounting for confounding factors such as absolute and relative alcohol intake, females are more sensitive to the effects of consumption compared to male mice. Idiopathic pulmonary fibrosis (IPF) is a chronic lung disease which affects about 5 to 8 million individuals in the world. Despite the high prevalence, there is currently no cure for IPF, and the cause of this disease is still unclear. Our laboratory and collaborators have shown that nudix hydrolase 21 (NUDT21, which is also known as cleavage factor 25, CFIm25) is a key regulator of alternative polyadenylation (APA). NUDT21 depletion causes 3’UTR shortening via APA leading to enhanced mRNA stability and protein translation. Anaerobic exercise causes metabolic acidosis to occur in the muscle, which is unfavorable for sustaining high intensity activity, as fatigue starts to set in. In order to compensate for the onset of fatigue produced by an acidic environment, respiratory alkalosis, induced by hyperventilation, can be performed to mitigate these effects. In contrast, research has shown that slowing down the breath rate, such as a two second inhale and three second exhale, has led to an increase in performance outcomes. The Gibbs lab has maintained starvation-selected Drosophila melanogaster for >130 generations. These starvation-selected flies evolved an obese phenotype with a suite of physiological differences compared to control-fed flies. Previous studies have shown that long-term starvation-selected Drosophila contain more lipids, have lower metabolic rates and develop more slowly than controls. This dissertation encompasses 1) Examining the molecular mechanisms contributing to starvation resistance, 2) Functional validation of the candidate plin1 allele, and 3) Rapid physiological and genomic evolution in starvation-selected Drosophila.

Māori (the indigenous peoples of Aotearoa, New Zealand) are intimately connected to wai (i.e., water) yet are overrepresented in New Zealand’s drowning statistics each year. On average Māori account for 20-24% of all preventable and non-preventable drowning fatalities, despite comprising only 15 percent of New Zealand’s population. Drowning remains a significant issue posing a threat to whānau (i.e., families) through premature death being imminent and whakapapa (i.e., genealogy) being interrupted. Fitness tracking apps are popular. There is little data validating their use during and after exercise. PURPOSE: Validate an iOS-based pulse oximeter against a fingertip pulse oximeter and a Polar® heart rate monitor during moderate intensity exercise and recovery. METHODS: Age Estimated Maximal Heart Rate (AEMHR) was calculated for fifteen college-aged students. Participants completed a 30-minute running trial divided into three 10-minute segments intended to elicit heart rate responses of 60, 70, and 80% of AEMHR. Ocean lifeguards have several responsibilities that require precise training. California State Parks is one of the largest providers of open water lifeguard services in the United States, and trains approximately 200 new lifeguards per year. As part of our lifeguard training section’s quality improvement efforts, we conducted a training needs assessment to help determine how well our introductory ocean lifeguard training curriculum prepared lifeguards to perform job related tasks upon successful completion of the training program. The aminoacyl-tRNA synthetases are an essential and universally distributed family of enzymes that plays a critical role in protein synthesis, pairing tRNAs with their cognate amino acids for decoding mRNAs according to the genetic code. Synthetases help to ensure accurate translation of the genetic code by using both highly accurate cognate substrate recognition and stringent proofreading of noncognate products. While alterations in the quality control mechanisms of synthetases are generally detrimental to cellular viability, recent studies suggest that in some instances such changes facilitate adaption to stress conditions. Alcohol and drugs have been identified as key risk factors for youth (aged 15-24 years) and adult drownings in high-income countries (HIC). Whilst alcohol specific drowning prevention education programs have been developed and implemented, youth continue to be over-represented in drowning statistics, including those linked with alcohol. Therefore, this project aimed to: (i) review and assess all alcohol themed drowning prevention campaigns within HICs; (ii) determine whether the campaign had undergone evaluation for effectiveness; and (iii) provide recommendations to improve the effectiveness of future interventions. Most deaths by drowning (91%) have occurred in low- and middle-income countries (LMICs), particularly in Southeast Asia (35%) and Africa (20%), in proportion to total drowning deaths worldwide. Poor data collection in LMICs hinders the planning, implementation, and evaluation of prevention strategies. The objective of this study was to review the rates and risk factors of unintentional drowning in LMICs and to identify drowning prevention strategies within a socio-ecological health promotion framework. Background: This study examined the motor unit (MU) control strategies for non-fatiguing isometric elbow flexion tasks at 40% and 70% maximal voluntary isometric contraction. Methods: Nineteen healthy individuals performed two submaximal tasks with similar torque levels: contracting against an immovable object (force task), and maintaining the elbow joint angle against an external load (position task). Surface electromyographic (EMG) signals were collected from the agonist and antagonist muscles. The signals from the agonist were decomposed into individual action potential trains. Diagnosing concussions provides challenges for healthcare professionals due to current diagnostic protocols utilizing subjective input from patients. Recent studies have shown relationships between specific salivary microRNA levels and concussions, but it is unknown if this is due to concussive forces or physical exertion. Analysis of this distinction may contribute to further confirming the relationship of concussions and microRNA, improving techniques for objective assessments of concussion. Training exercises to develop muscular strength and power, such as the weighted jump squat, commonly utilize barbells and dumbbells to increase resistance. Strength coaches often measure improvements in peak force, peak power, and rate of force development to determine effectiveness of a training program. The purpose of this study was to determine if equipment selection (barbells vs. dumbbells) affects peak force, peak power, and/or rate of force development in weighted jump squats. Though there are several to choose from, the Bruce protocol is the most commonly utilized treadmill protocol when assessing maximal oxygen consumption (VO2max). Originally developed for cardiac patients, the Bruce protocol may not be an appropriate treadmill protocol for young or trained individuals with higher levels of cardiorespiratory fitness. The steep grade used often leads localized muscular fatigue, which may cause participants to end the test prior to reaching their true VO2max. Another popular protocol, the Astrand, also utilizes a steep grade, but the speed remains constant. Similar to the Bruce, the Astrand protocol may not prompt a maximal cardiovascular …

Prediction of energy expenditure allows for calculation of appropriate energy requirements, which is especially important for athletes. Resting metabolic rate (RMR) is the greatest contributor to total daily energy expenditure (TDEE) and is typically measured via indirect calorimetry. Indirect calorimetry is not always available, which results in the need for predictive equations. Most predictive equations have been developed with participants resembling the general population and have not been found to be appropriate for athletes, as they may incorrectly predict RMR due to the unique differences of body composition between athletes and the general population. Adverse pregnancy outcomes (APOs) are believed to be caused by poor placental formation. APOs can be characterized by elevated levels of a circulating Fms-like tyrosine kinase biomarker called Flt-1, produced by the placenta when its blood vessels are shallow or inadequately formed. Flt-1 acts directly on the maternal endothelium to impair vascular function during pregnancy and contributes to maternal features of APOs. I will be exploring the impact of the large protein Titin on heart failure with a preserved ejection fraction (HFpEF). It was hypothesized that myocardial Titin plays a significant role in the progression of HFpEF through isoform, N2BA and N2B, changes. It was hypothesized that an increase in the quantity of N2B Titin, less compliant, and a decrease in N2BA Titin, more compliant, will be observed as HF progresses, contributing to an increase in passive stiffness involved in heart contraction. I will cover a detailed report on the role of Titin in HFpEF and propose research methods to explore further.

Glucose-stimulated insulin secretion (GSIS) from the pancreatic β-cell in response to elevated levels of glucose is controlled by a variety of signals including intracellular calcium and nucleotides such as cAMP, ATP and GTP. These cellular signals are responsible for activation of specific kinases that mediate phosphorylation of key exocytotic proteins that lead to GSIS. In the context of protein kinases, mixed-lineage kinases (MLKs) have been implicated in an assortment of cellular functions, including cell proliferation and apoptosis. Mild cognitive impairment (MCI) is a decline in cognitive function to a greater extent than natural aging. It has been suggested that reduced brain blood flow is strongly associated with reduced cognitive function and neurodegenerative diseases, of which is partially attributed to excessive reactive oxygen species (ROS) production. Mitoquinol mesylate has been shown to scavenge ROS and improve vascular function in healthy older adults. However, the effects of mitoquinol in MCI patients have not been investigated.

The purpose of this study was to examine the relationship between the standard method (i.e., Iowa Oral Performance Instrument) and a practical method in healthy adults. In order to achieve our research purpose, ninety-eight healthy adults (18-40 years) completed lip compression strength measurements (standard method) and lip grasping performance tests using a standard recyclable plastic water bottle (practical method).
