Glioblastoma multiforme (GBM), a high-grade, malignant brain tumor of astrocyte origin, invades normal brain tissue and most often results in death ofthe patient withinayearofdiagnosis. In order to invade the surrounding tissue and thereby metastasize, these tumor cells must migrate through the extracellular matrix. The pmpose ofthis research project is to investigate the family ofmatrix metalloproteinases responsible for extracellular degradation. Specifically, this investigation looks at MMP-2 activation via MT 1-MMP and the possible importance of phosphorylation of the tyrosine residue on the cytoplasmic tail of MTl-MMP. Glial-cell line derived neurotrophic factor (GDNF) is a neurotrophic factor produced by skeletal muscle. GDNF has been shown to be potent survival factor for motor neurons. We have previously shown that GDNF content in skeletal muscle is altered following exercise and GDNF levels 24 hours after the last bout of exercise are significantly different than those measured immediately after exercise. We sought to determine how long after exercise do GDNF protein levels remain altered. We removed extensor digitorum longus (EDL) and Soleus (SOL), 24 hr, 72 hr and 1 week following the last bout of involuntary exercise. Muscles were processed …

Toxic insult by PCBs results in learning and memory deficits in humans. Alterations in expression of neurotrophic factors and/or their receptors have been linked to changes in cognition. How PCBs affect cognition is not known. We propose that PCBs affect cognition by altering neurotrophic factor expression or effects. We exposed cultured C6 glial cells in medium containing PCB (Aroclor 1254 (10ppm)). Control cells were treated with DMSO or regular medium. Cells were incubated at 37o C for up to 72 hours. Medium samples were taken at 6hr, 24hr, 48hr, and 72hr intervals. Enzyme-linked immunosorbent assays (ELISA) were used to determine …

The apolipoprotein E4 (apoE4) genotype is a major risk factor for Alzheimer's disease (AD). Previous studies have shown that apoE4 is less effective than apoE3 in supporting neurite outgrowth in adult mice cortical cultures. However the mechanism underlying this phenomenon is not known. To understand this mechanism and the broader role of apoE isoforms on neuronal biology I examined the rate of apoE isoforms internalization in neurons, effects of apoE isoforms on lipid uptake by neurons, effects of apoE isoforms on neuronal environment - microglia and on neuroregeneration, or neurogenesis.

Neural maps in the vertebrate central nervous system often show discontinuously segregated, domain-to-domain patterns. However, the molecular mechanism that establishes such maps is not well understood. Here we show that in the chicken olivocerebellar system, EphA receptors and ephrin-As are expressed with distinct levels and combinations in mapping domains. When ephrin-A2 is retrovirally overexpressed in the cerebellum, the olivocerebellar map is disrupted, excluding axons with high receptor activity from ectopic expression domains. Conversely, overexpression of a truncated EphA3 receptor in the cerebellum reduces endogenous ligand activity to undetectable levels and causes aberrant mapping, with high receptor axons invading high ligand …

Previous studies have demonstrated that 17β-estradiol can modify apolipoprotein E (apoE) expression. I found that apoE protein varied as a function of the estrous cycle and 17β-estradiol (E2) in a region specific manner in the mouse brain. I also found that apoE concentration was lowest on estrus in the hippocampus, cingulate cortex and frontal cortex; apoE concentration was highest on estrus in the olfactory bulb and cerebellum. No changes in apoE were found in the striatum throughout the estrous cycle. Exogenous E2 significantly raised tissue levels in the olfactory bulb and cerebellum, but did not increase apoE in cortical samples …

Estrogen replacement therapy appears to delay the onset of Alzheimer's disease (AD). The mechanism whereby estrogen prevents the pathogenesis of AD is unknown. In the present study, I examined the effects of 17-β-estradiol (E2) on neurite outgrowth from adult mice cortical neurons (AMC) in culture. I found that E2 increases apoE secretion and neurite outgrowth in AMC neurons from wild type mice in a dose dependent fashion. The neurite outgrowth promoting effect of E2 was not observed in AMC neurons derived from age-, sex-, and stain-matched apoE deficient/apoE gene knockout (apoE KO) mice.
