ปริมาณโลหิตและส่วนประกอบโลหิตที่เพียงพอในธนาคารเลือดนั้นมีความสำคัญในการรักษาผู้ป่วย ดังนั้นธนาคารเลือดทุกแห่งต้องส่งเสริมให้ผู้บริจาคโลหิตมีสุขภาพดีและมีคุณสมบัติเหมาะสมในการบริจาคโลหิตอย่างสม่ำเสมอจากข้อมูลของศูนย์บริการโลหิตแห่งชาติ สภากาชาดไทยพบผู้ถูกปฏิเสธการบริจาคโลหิตประมาณร้อยละ 15-20 จากจำนวนผู้ประสงค์จะบริจาคโลหิตทั้งหมด ส่วนใหญ่มีสาเหตุจากค่าฮีโมโกลบิน (hemoglobin; Hb) ไม่ผ่านเกณฑ์ นำไปสู่ความผิดหวังความไม่พอใจเพราะผู้บริจาคโลหิตต้องเสียเวลา ค่าใช้จ่ายในการเดินทางแต่ไม่ได้บริจาคโลหิตและส่งผลกระทบต่อการจัดหาโลหิตโดยตรง หากสามารถพยากรณ์ผลตรวจ Hb ได้ล่วงหน้าจะช่วยลดผลกระทบปัญหา เป็นประโยชน์ทั้งต่อผู้บริจาคโลหิต ธนาคารเลือดและผู้ป่วย การศึกษานี้มีวัตถุประสงค์เพื่อพัฒนาและเปรียบเทียบประสิทธิภาพเทคนิคการเรียนรู้ของคอมพิวเตอร์ในการพยากรณ์จำแนกกลุ่มผลตรวจ Hb ของผู้บริจาคโลหิต โดยเก็บข้อมูล 44 ตัวแปรของผู้บริจาคโลหิตจำนวน 2,180 รายจากภาคบริการโลหิตแห่งชาติ 12 แห่งและสถานีกาชาดหัวหินเฉลิมพระเกียรติตั้งแต่ 1 ต.ค. 2561 ถึง 31 พ.ค. 2562 นำมากลั่นกรองข้อมูล คัดเลือกตัวแปร พัฒนาตัวแบบทางเหมืองข้อมูลได้แก่ ต้นไม้ตัดสินใจ ซัพพอร์ตเวกเตอร์แมชชีน การจำแนกแบบเบส์อย่างง่ายและโครงข่ายประสาทเทียม จากนั้นเปรียบเทียบประสิทธิภาพการจำแนกกลุ่มของตัวแบบพยากรณ์ พบว่าต้นไม้ตัดสินใจเป็นตัวแบบการจำแนกกลุ่มที่เหมาะสมที่สุดโดยให้ค่าความถูกต้อง ค่าความไว ค่าความจำเพาะ ค่าการพยากรณ์ผลบวก ค่าการพยากรณ์ผลลบสูงที่สุดและค่า AUC เท่ากับร้อยละ 92.20, 82.98, 94.74, 81.25, 95.29 และ 0.943 ตามลำดับ ซึ่งต้นไม้ตัดสินใจที่ได้จากศึกษานี้อาจนำไปพัฒนาต่อเป็นระบบประเมินออนไลน์ก่อนเดินทางมาบริจาคโลหิตได้

โรคติดเชื้อไวรัสมะเร็งเม็ดเลือดขาวในแมว (Feline Leukemia Virus : FeLV) เป็นโรคติดเชื้อไวรัสในแมวที่สำคัญที่เป็นสาเหตุของโรคมะเร็งเม็ดเลือดขาว และ มะเร็งต่อมน้ำเหลือง การตรวจหา FeLV RNA และโปรไวรัส ด้วยวิธีทางอณูชีววิทยามีความสำคัญในการแยกระยะต่างๆของโรค ซึ่งวิธี Rapid immunochromatographic assay ที่นิยมใช้ตรวจแอนติเจนนั้นสามารถตรวจได้เพียงการติดเชื้อระยะ Progressive ที่มี Viremia เท่านั้น งานวิจัยนี้จึงได้พัฒนาเทคนิค Recombinase polymerase amplification (RPA) และ Nested RPA เพื่อตรวจ exogenous FeLV Provirus DNA และ RT-RPA เพื่อตรวจ FeLV RNA และทดสอบตัวอย่างเลือดแมวจำนวน 122 ตัวอย่าง ผลการศึกษาพบว่าเทคนิค RPA สามารถตรวจหา FeLV Provirus ได้ที่ขีดจำกัด 1 ng/µl และไม่เกิดปฏิกิริยาข้ามกลุ่มกับเชื้อก่อโรคในแมว มีความไว ความจำเพาะ เทียบกับเทคนิค PCR ที่ร้อยละ 95.89 และ 100 ตามลำดับ ขณะที่ Nested RPA เทียบกับเทคนิค Nested PCR มีความไว ความจำเพาะ ที่ร้อยละ 98.89 และ 96.88 ตามลำดับ การตรวจหา FeLV RNA ด้วยเทคนิค RT-RPA มีขีดจำกัดที่ 0.1 ng/µl เมื่อเทียบกับเทคนิค RT-PCR และ Rapid Immunochromatographic assay มีความไว ร้อยละ 93.75 และ 95.24 ตามลำดับ และ ความจำเพาะร้อยละ 100 ค่าความสอดคล้องของเทคนิค RPA …

เชื้อ Klebsiella pneumoniae ที่สร้างเอนไซม์ carbapenemases (KPCs) เป็นปัญหาสำคัญของการติดเชื้อในโรงพยาบาลโดยเฉพาะผู้ป่วยเด็ก เนื่องจากมักดื้อยาหลายขนาน ทำให้มียาที่ใช้รักษาได้จำกัด เป็นสาเหตุทำให้มีอัตราการเสียชีวิตสูงขึ้น งานวิจัยนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิค multiplex Recombinase Polymerase Amplification (multiplex RPA) สำหรับตรวจหายีนที่สร้างเอนไซม์ carbapenemase ของเชื้อ K. pneumoniae และเปรียบเทียบความไวและความจำเพาะกับ วิธี Modified Carbapenem Inactivation Method Test (mCIM) และ วิธี nucleotide sequencing กับตัวอย่างเชื้อ K. pneumoniae ในสถาบันสุขภาพเด็กแห่งชาติมหาราชินี ประเทศไทย ตั้งแต่เดือนมิถุนายน ถึง ธันวาคม 2018 โดยเก็บเชื้อที่ผลการคัดกรองพบดื้อยา carbapenem แบ่งเป็นจาก perianal ในงานเฝ้าระวังเชื้อดื้อยา จำนวน 129 ตัวอย่าง และจากสิ่งส่งตรวจของผู้ป่วยเด็ก จำนวน 21 ตัวอย่าง พบให้ผลบวกกับวิธี mCIM จำนวน 149 ตัวอย่าง (99.3%) และพบเป็นยีน blaNDM-1 สูงสุดถึงร้อยละ 89.3 รองลงมาคือยีน blaOXA-232 ร้อยละ 8 และยีน blaNDM-1 ร่วมกับ blaOXA-232 ร้อยละ 2 ตามลำดับ เชื้อที่แยกได้จากสิ่งตรวจพบมีการดื้อยาหลายขนาน โดยดื้อยา carbapenem ร่วมกับ gentamicin, trimethoprim/sulfamethoxazole, ciprofloxacin ขณะที่เชื้อที่แยกจากแผนกผู้ป่วยต่าง ๆ ในงานเฝ้าระวังเชื้อดื้อยา พบการแพร่กระจายของยีน blaNDM-1 สูงสุดใน 4 แผนก จาก 9 แผนก การพัฒนาเทคนิค multiplex RPA พบว่าสามารถเพิ่มปริมาณยีน blaKPC, blaNDM, …

Parkinson’s Disease (PD) is an idiopathic disorder with no known cure. With number of cases steadily rising around the world, it is imperative to turn to the underlying cellular and molecular mechanisms of the disease manifestation and neurodegeneration to craft novel modes of therapy. VPS35 is one of the few genes that have identified and definitively linked to familial PD. The Chrysoperla carnea-group of green lacewings is a cryptic species complex. Species within the group are morphologically similar, yet isolated from one another via reproductive mating song. Chrysoperla zastrowi, a species within the carnea-group, is currently described with a distribution ranging from South Africa to the Middle East and India. However, recent collections of carnea-group lacewings from Guatemala and California were preliminarily identified as Chrysoperla zastrowi based upon similarities in their vibrational courtship songs. The N-terminal domain (NTD) of nuclear human uracil DNA glycosylase (hUNG2) assists in targeting hUNG2 to replication forks through specific interactions with replication protein A (RPA). Here, we explored hUNG2 activity in the presence and absence of RPA using substrates with ssDNA-dsDNA junctions that mimic structural features of the replication fork and transcriptional R-loops. We find that when RPA is tightly bound to the ssDNA overhang of junction DNA substrates, base excision by hUNG2 is strongly biased toward uracils located 21 bp or less from the ssDNA-dsDNA junction. The Bermuda fireworm Odontosyllis enopla exhibits an extremely tight circalunar circadian behavior that results in an impressive bioluminescent mating swarm, thought to be due to a conventional luciferase-mediated oxidation of a light-emitting luciferin. In addition, the four eyes become hypertrophied and heavily pigmented, and the nephridial system is modified to store and release gametes and associated secretions. In an effort to elucidate transcripts related to bioluminescence, circadian or circalunar periodicity, as well as epitoky-related changes of the eyes and nephridial system, we examined the transcriptomic profile of three female O. enopladuring a bioluminescent swarm in Ferry Reach, Bermuda. …

Recombination and mutagenesis are elevated by high levels of transcription. The correlation between transcription and genome instability is largely explained by the topological and structural changes in DNA and the associated physical obstacles generated by the transcription machinery. However, such explanation does not directly account for the unique types of mutations originating from the non-canonical residues such as uracil, which are also elevated at highly transcribed regions. Sperm activation is a fascinating example of cell differentiation, in which immotile spermatids undergo a rapid and dramatic transition to become mature, motile sperm. Because the sperm nucleus is transcriptionally silent, this transition does not involve transcriptional changes. Although Caenorhabditis elegans is a leading model for studies of sperm activation, the mechanisms by which signaling pathways induce this transformation remain poorly characterized. Here we show that a conserved transmembrane zinc transporter, ZIPT-7.1, regulates the induction of sperm activation in Caenorhabditis nematodes. Diabetes mellitus and Parkinson’s Disease (PD) are debilitating diseases that are increasing in prevalence worldwide. One potential cause of these diseases is the accumulation of advanced glycation end products (AGEs), which are macromolecules that cause irreversible damages. AGEs are a diverse group of highly oxidative byproducts produced from α-dicarbonyl compounds (α-dcs), which are highly reactive molecules that bind indiscriminately to protein and DNA and, are regulated by a conserved glyoxalase system (GLO1 and DJ-1) in humans. This project is part of the Meiothermus ruber genome analysis project, which uses the bioinformatics tools associated with the Guiding Education through Novel Investigation –Annotation Collaboration Toolkit (GENI-ACT) to predict gene function. We investigated the biological function of Escherichia coli and Meiothermus ruber proC genes using the complementation assay. In this research project, mutants of varying severity to the functional state of the protein were developed. The results showed that two or more amino acid deletions reduced or eliminated ProC function. Amino acid substitutions, on the other hand, were not severe enough to impact ProC function. Vascular type of Ehlers-Danlos Syndrome (vEDS) is an inherited cardiovascular disease affecting the middle to large sized arteries, with an incidence rate of 1/5000. vEDS patients also show a significant phenotype of easily bruised skin, indicating aberrant wound healing and injury repair ability. Over 70% of the patients carry a glycine mutation located in their COL3A1 gene, which encodes the propeptide of type III collagen. Mutations in glycine residues lead to a disruption in the assembly and maturation of type III collagen. The ability to express a gene of interest in a spatio-temporal manner using Gal4-UAS system has allowed the use of Drosophila model to study various biological phenomenon. During Drosophila eye development, a synchronous wave of differentiation called Morphogenetic furrow (MF) initiates at the posterior margin resulting in differentiation of retinal neurons. Copy number variations (CNVs) are large insertions, deletions, and duplications in the genome that vary between individuals in a species. Animal behavioural traits often covary with gene expression, pointing towards a genomic constraint on organismal responses to environmental cues. This pattern highlights a gap in our understanding of the time course of environmentally responsive gene expression, and moreover, how these dynamics are regulated. Advances in behavioural genomics explore how gene expression dynamics are correlated with behavioural traits that range from stable to highly labile. We consider the idea that certain genomic regulatory mechanisms may predict the timescale of an environmental effect on behaviour. The Colorado potato beetle is one of the most challenging agricultural pests to manage. It has shown a spectacular ability to adapt to a variety of solanaceaeous plants and variable climates during its global invasion, and, notably, to rapidly evolve insecticide resistance. To examine evidence of rapid evolutionary change, and to understand the genetic basis of herbivory and insecticide resistance, we tested for structural and functional genomic changes relative to other arthropod species using genome sequencing, transcriptomics, and community annotation. Background

Transient gene expression is a powerful tool to study gene function in plants. In citrus, Agrobacterium transformation is the method of choice for transient expression studies, but this method does not work efficiently with many gene constructs, and there is a need for a more robust transient expression system in citrus leaves. Biolistic particle delivery is an alternative to Agrobacterium transformation, and in some plants, such as Arabidopsis, gives higher transformation rates in leaf tissues than Agrobacterium.

Generally, ABC transporters are multi-subunit; contain essential cytoplasmic factors which are critical to ATP hydrolysis activity. In this paper, we would like to take a closer look to Mrub_1283, Mrub_1284 and Mrub_1285, three consecutive genes in Meiothermus ruber genome.
