Cellular homeostasis is achieved by the dynamic flux in gene expression. Post-transcriptional regulation of coding and non-coding RNA offers a fast method of adapting to a changing cellular environment, including deadenylation, microRNA (miRNA) pathway, and alternative polyadenylation (APA). In this dissertation, I explored some of the mechanisms involved in the post-transcriptional regulation of gene expression. The main hypothesis in these studies is that a single APA event after DNA damage is governed by specific conditions and factors outside of current known regulators of APA, and that the resultant transcript has a role in the DNA damage response (DDR). Surveillance of circulating microbial populations is critical for monitoring the performance of a molecular diagnostic test. In this study, we characterized 31 isolates of Streptococcus agalactiae (group B Streptococcus [GBS]) from several geographic locations in the United States and Ireland that contain deletions in or adjacent to the region of the chromosome that encodes the hemolysin gene cfb, the region targeted by the Xpert GBS and GBS LB assays. PCR-negative, culture-positive isolates were recognized during verification studies of the Xpert GBS assay in 12 laboratories between 2012 and 2018. One of the major goals in large-scale genomic studies is to identify genes with a prognostic impact on time-to-event outcomes which provide insight into the disease's process. With rapid developments in high-throughput genomic technologies in the past two decades, the scientific community is able to monitor the expression levels of tens of thousands of genes and proteins resulting in enormous data sets where the number of genomic features is far greater than the number of subjects. The past two decades have witnessed significant advances in high-throughput ``omics" technologies such as genomics, proteomics, metabolomics, transcriptomics and radiomics. These technologies have enabled simultaneous measurement of the expression levels of tens of thousands of features from individual patient samples and have generated enormous amounts of data that require analysis and interpretation. One specific area of interest has been in studying the relationship between these features and patient outcomes, such as overall and recurrence-free survival, with the goal of developing a predictive ``omics" profile. The mechanism of resistance in carbapenem-resistant Enterobacteriaceae (CRE) has therapeutic implications. We comprehensively characterized emerging mechanisms of resistance in CRE between 2013 and 2016 at a health system in Northern California. A total of 38.7% (24/62) of CRE isolates were carbapenemase gene-positive, comprising 25.0% (6/24) blaOXA-48 like, 20.8% (5/24) blaKPC, 20.8% (5/24) blaNDM, 20.8% (5/24) blaSME, 8.3% (2/24) blaIMP, and 4.2% (1/24) blaVIM. Between carbapenemases and porin loss, the resistance mechanism was identified in 95.2% (59/62) of CRE isolates. The transcriptional anti-silencing and DNA-binding protein, VirB, is essential for the virulence of Shigella species and, yet, sequences required for VirB-DNA binding are poorly understood. While a 7-8 bp VirB-binding site has been proposed, it was derived from studies at a single VirB-dependent promoter, icsB. Our previous in vivo studies at a different VirB-dependent promoter, icsP, found that the proposed VirB-binding site was insufficient for regulation. Instead, the required site was found to be organized as a near-perfect inverted repeat separated by a single nucleotide spacer. Thus, the proposed 7-8 bp VirB-binding site needed to be re-evaluated. This research is part of the Meiothermus ruber genome annotation project which aims to predict gene function with various bioinformatics tools. We investigated the function of Mrub_3019, which encodes the CasA protein involved in the multi-subunit effector complex for the CRISPR-Cas immunity system and predicted it to be an ortholog of E. coli K12 MG1655 b2760 (casA). We predicted that Mrub_3019 encodes the protein CasA, which is involved in PAM recognition of CRISPR interference pathway. Foreign DNA will bind to CasA, which signals Cas3 for helicase-mediated DNA degradation. Our hypothesis is supported by low E-values for pairwise alignment in NCBI …

This study phylogenetically analyzed 30 AMY-related genes from 11 primates. The results show the gradual expansion of the AMY gene family which could have allowed primates to adapt to various ecological landscapes and maximize energy intake from starch-rich foods in periods of food scarcity.

We have identified an albino mutant in the plant Brassica rapa. This recessive mutation could provide insights into the photosynthetic pathways of all plants. Previously the mutation was mapped to chromosome 10 of Brassica rapa. To narrow down the chromosomal region containing the albino mutation, we grew a segregating population of plants. DNA was extracted from albino and wildtype (green) plants and genotyped using PCR markers on chromosome 10. The genotype detected using the INDEL2 marker perfectly predicted whether a plant was albino or WT. This indicates that the INDEL2 marker is tightly linked to the albino mutation. INDEL2 is …

Most of today’s maize is cultivated outside its original climate zone, where yields are constrained by the changes in climate. Maize is especially vulnerable to high temperatures and drought stress, both of which negatively affect corn yields. An important strategy to combat this is early sowing, which avoids the effects of summer droughts and high temperatures in many places around the globe. Genomic instability, in the form of gene mutations, insertions/deletions, and gene amplifications, is one of the hallmarks in many types of cancers and other inheritable genetic disorders. Trinucleotide repeat (TNR) disorders, such as Huntington’s disease (HD) and Myotonic dystrophy (DM) can be inherited and repeats may be extended through subsequent generations. However, it is not clear how the CAG repeats expand through generations in HD. Two possible repeat expansion mechanisms include: 1) polymerase mediated repeat extension; 2) persistent TNR hairpin structure formation persisting in the genome resulting in expansion after subsequent cell division. In the past 20 years, the number of subterranean taxa discovered in Australia, especially in the Pilbara bioregion, has considerably increased due to incidental environmental surveys often associated with mining development. Bathynellidae are an important component of stygofauna and they occur in most Australian aquifers, but their collection and identification are difficult due to their habitat, and small and fragile bodies with conservative morphology. The study of Pilbaranella ethelensis in the upper Fortescue catchment contributed to a better understanding of the group at local scale, but knowledge at larger catchment scale is still limited. Widely effective treatment for alcohol use disorder is not yet available, because the exact biological mechanisms that underlie this disorder are not completely understood. One way to gain a better understanding of these mechanisms is to examine the genetic frameworks that contribute to the risk for developing this disorder. This dissertation examines genetic association data in combination with gene expression networks in the brain to identify functional groups of genes associated with alcohol consumption and dependence.

PNPase, polynucleotide phosphorylase, is a multifunctional exoribonuclease protein with 3` terminal oligonucleotide polymerase activity. Coded by the PNPT1 gene, the protein is associated with mitochondrial homeostasis and functions as a possible target for cancer therapy. In this study, C. elegans was used to investigate the effect of mutation and overexpression of pnpt-1, the gene that encodes PNPase. It was determined that two specific mutations in pnpt-1 did not affect PNPase expression nor did they produce deleterious phenotypes that affected polycistronic transcript accumulation or ROS production. Emplectonema gracile (Johnston 1837) is a hoplonemertean of marine intertidal hard-bottom communities and is distributed throughout the Northern Hemisphere. Although possessing a planktonic larval stage in its life history, the range of such cosmopolitan marine invertebrate species is often explained by cryptic speciation and anthropogenic transport. The purpose of this study is to test for possible cryptic species using mtDNA markers (COI and 16S rDNA) and to investigate population structure in E. gracile over a portion of its geographic range using mtDNA markers and ddRADseq nuclear SNP data. Alcohol use disorder is a global health issue that affects a significant portion of the population, with affects including both negative mental and physical consequences. Currently, there are few treatment options available to those who suffer from alcohol use disorder, alcohol abuse, or alcohol dependence. Identifying candidate genes or environmental influences would therefore improve the means for possible treatments or identification of those people at risk for alcohol use disorder. Glyceollin phytoalexins are the pathogen-elicited defense metabolites that belong to the isoflavonoid family of molecules of soybean (Glycine max L. Merr). Phytophthora root and stem rot of soybean caused by Phytophthora sojae is a destructive disease throughout the soybean-growing regions worldwide causing devastating economic damages (globally $1-2 billion and over $250 million in the USA) every year. Alzheimer’s disease (hereafter AD) is a progressive neurodegenerative disorder that affects the central nervous system. There are multiple factors that cause AD, viz., accumulation of extracellular Amyloid-beta 42 plaques, intracellular hyper-phosphorylated Tau tangles, generation of reactive oxygen species due to mitochondrial dysfunction and genetic mutations. The plaques and tau tangles trigger aberrant signaling, which eventually cause cell death of the neurons. As a result, there is shrinkage of brain, cognitive defects, behavioral and psychological problems. To date, there is no direct cure for AD. The intestinal microbiome of mammals plays a significant role in host health and response to environmental stimuli and can include both beneficial native bacteria as well as parasitic worms. In this study, I examined the intestinal cestode and bacterial communities of two closely related species of shrew, Sorex monticola and Sorex cinereus, over a six month period in 2016. Specimens were collected approximately every three weeks from May to October from the Sangre de Cristo Mountains in Cowles, New Mexico. Natural selection alters genetic pathways over evolutionary time. However, different pathways have been shown to maintain mutations at varying rates, leading to different levels of conservation across developmental pathways. Sex determination pathways, in particular, show vast diversity across animals despite the fundamental conservation of biological sex itself. Here, I investigated the sex determination pathway of the hemimetabolous insect, Oncopeltus fasciatus. Biocides are antimicrobial compounds that are designed to kill diverse groups of microbes in an untargeted fashion. Glutaraldehyde and DBNPA are commonly used to manage microbial growth in different industries. It is possible that biocide use may result in the development of bacterial resistance. However, resistance to DBNPA is believed to be limited under normal use conditions. We isolated a number of bacteria from produced water from a hydraulically fractured site in West Texas. Birdstrikes to aircrafts are increasing on an annual basis and pose significant aviation safety risks. Identification of the birds involved is key to developing mitigation strategies. Often the only information available to make identifications are feather and/or tissue samples. Relying on feathers alone to identify the bird species requires special expertise and access to museum collections for specimen comparisons. In 2017, feathers and tissue samples were recovered from the engine cowling of an airplane that had just landed at the Oliver Reginald (O. R.) โรคติดเชื้อที่มีสาเหตุมาจาก Mycobacterium avium complex (MAC) มีอุบัติการณ์เพิ่มสูงขึ้นทั่วโลกรวมทั้งในประเทศไทย เนื่องมาจากการเพิ่มขึ้นของจำนวนผู้ป่วยที่มีภาวะภูมิคุ้มกันของร่ายกายต่ำ การวินิจฉัยเชื้อ MAC ในปัจจุบันที่อาศัยการเพาะเลี้ยงเชื้อและทดสอบปฏิกิริยาชีวเคมี นอกจากใช้ระยะเวลานานแล้ว ยังไม่สามารถจำแนกเชื้อได้ถึงระดับสปีชี่ส์ โดยเฉพาะอย่างยิ่งเชื้อ Mycobacterium avium และเชื้อ Mycobacterium intracellulare ซึ่งเป็นเชื้อ MAC ที่สำคัญและเพาะแยกได้บ่อยจากสิ่งส่งตรวจ ดังนั้นวัตถุประสงค์ของการศึกษาในครั้งนี้ คือการพัฒนาเทคนิค Recombinase Polymerase Amplification ร่วมกับการอ่านผลด้วยสี SYBR Green I (RPA/SYBR) ในการตรวจวินิจฉัยเชื้อ MAC ซึ่งอาศัยการเพิ่มปริมาณสารพันธุกรรมภายใต้อุณหภูมิคงที่ ร่วมกับไพรเมอร์ที่จำเพาะต่อเชื้อกลุ่ม MAC เชื้อ M. avium และเชื้อ M. intracellulare และอ่านผลที่เกิดขึ้นด้วยตาเปล่าได้ในทันทีภายหลังการเติมสี SYBR Green I ผลจากการทดสอบกับตัวอย่าง DNA ของเชื้อ MAC จำนวน 120 ตัวอย่าง พบว่าเทคนิค RPA/SYBR สามารถตรวจวินิจฉัยจำแนกสปีชี่ส์ของเชื้อ M. avium และเชื้อ M. intracellulare ได้อย่างมีประสิทธิภาพ โดยมีความไว 100% และความจำเพาะตั้งแต่ 80.7% ขึ้นไป ซึ่งจัดว่ามีความสอดคล้องในเกณฑ์ดีมากเมื่อเปรียบเทียบกับวิธีหาลำดับเบส (สถิติ Kappa มีค่าตั้งแต่ 0.801 ขึ้นไป) ในขณะที่เมื่อเปรียบเทียบกับเทคนิค PCR มีความไวตั้งแต่ 89.2% ขึ้นไปและความจำเพาะตั้งแต่ 97.5% ขึ้นไป และมีความสอดคล้องในเกณฑ์ดีมากเช่นเดียวกัน (สถิติ Kappa มีค่าตั้งแต่ 0.863 ขึ้นไป) นอกจากนี้เทคนิค RPA/SYBR ยังมีค่าความเข้มข้นของ DNA ต้นแบบน้อยที่สุดที่สามารถตรวจได้ เท่ากับ 1 ng/µl และไม่เกิดปฏิกิริยาข้ามกลุ่มกับเชื้อส่วนใหญ่ของ Mycobacterium แม้ว่าเทคนิค RPA/SYBR ที่ได้พัฒนาขึ้น มีขั้นตอนการทดสอบที่ทำง่าย …

เชื้อ Escherichia coli ที่สร้างเอนไซม์ Extended-spectrum β-lactamase (ESBLs) เป็นปัญหาสำคัญที่พบในโรงพยาบาลทั่วโลก การตรวจจีโนไทป์ของเอนไซม์ ESBLs เป็นวิธีที่สำคัญและมีประโยชน์ในการควบคุมการแพร่กระจายของเชื้อดื้อยาและการเลือกใช้ยาต้านจุลชีพอย่างเหมาะสม การศึกษานี้มุ่งหวังจะพัฒนาเทคนิค Multiplex Recombinase polymerase amplification (Multiplex RPA) และ Helicase dependent amplification (HDA) สำหรับตรวจหายีนดื้อยา blaCTX-M, blaOXA, blaSHV และ blaTEM ในเชื้อ E. coli ในสถาบันมะเร็งแห่งชาติ ประเทศไทย เชื้อ E. coli จำนวน 144 ตัวอย่างที่แยกได้จากสิ่งส่งตรวจภายในงานจุลชีววิทยา สถาบันมะเร็งแห่งชาติ ช่วงระหว่างเดือนกุมภาพันธ์ 2017 ถึงเดือนกันยายน 2018 ถูกนำมาทดสอบความไวต่อยาต้านจุลชีพและตรวจคัดกรองการสร้างเอนไซม์ ESBLs ด้วยวิธี Disk diffusion ตามวิธีมาตรฐานของ CLSI ปี ค.ศ. 2017 เชื้อที่สร้างเอนไซม์ ESBLs จำนวน 95 ตัวอย่าง พบดื้อยาสูงสุดในยา Ampicillin (100%), Cefotaxime (100%), Cefdinir (100%) และ Ceftriaxone (99.9%) และเชื้อ E. coli จำนวน 144 ตัวอย่าง มียีน blaCTX-M (100%) รองลงมาคือ blaTEM (42.1%), blaOXA (30.5%) และ blaSHV (2.1%) ตามลำดับ โดยเป็นยีน ESBLs ชนิด CTX-M-15 สูงสุด (51.1%) รองลงมาคือ CTX-M-55 (18.1%), CTX-M-27 (17.0%), CTX-M-14 …

อะไมลอยด์เบต้า 42 (Aβ42) เป็นของเสียที่เกิดจากการย่อยอะไมลอยด์เบต้าพรีเคอเซอร์โปรตีน อาจเสี่ยงต่อการเกิดโรคอัลไซเมอร์เมื่อสะสมที่สมอง โดยมียีนบีเอซีอีวัน (BACE1) ควบคุมการสร้างเอนไซม์เบต้าซีครีเตส เพื่อย่อยอะไมลอยด์เบต้าพรีเคอเซอร์โปรตีน และมียีนเอโปอี (ApoE) เป็นยีนที่ถอดรหัสเป็น Apolipoprotein E ทำหน้าที่ขนส่งอะไมลอยด์เบต้าไปกำจัดที่ตับ คาดว่าการติดเชื้อไวรัสตับอักเสบเรื้อรังอาจส่งผลให้การกำจัดของเสียลดลงด้วย งานวิจัยนี้จึงมีวัตถุประสงค์เพื่อศึกษาระดับอะไมลอยด์เบต้า 42 ในเลือดด้วยหลักการ ELISA และศึกษาความหลากหลายทางพันธุกรรมของยีน BACE1 (rs638405) ด้วยหลักการ PCR-RFLP และยีน ApoE (rs429358 และ rs7412) ด้วยหลักการ real time PCR ในผู้ติดเชื้อไวรัสตับอักเสบบีเรื้อรัง (CHB, N=149) ซีเรื้อรัง (CHC, N=31) และคนปกติ (N=164) โดยเป็นผู้ติดเชื้อไวรัสตับอักเสบบีเรื้อรัง ซีเรื้อรังที่รักษา ณ โรงพยาบาลพระมงกุฎเกล้า และผู้บริจาคโลหิต ณ สถาบันพยาธิวิทยา ในช่วงเดือน สิงหาคม 2561 - มิถุนายน 2562 ผลการศึกษาพบว่าระดับอะไมลอยด์เบต้า 42 ในเลือดของผู้ป่วย CHB มีค่าสูงกว่าคนปกติ (CHB=46.86 pg/ml (N=22), CHC=43.27 pg/ml (N=15) และ คนปกติ =33.72 pg/ml (N=36), p-value=0.002) ข้อมูลจากกลุ่มตัวอย่างที่ตรวจวัดนี้มีจำนวนจำกัด จึงยังไม่สามารถนำไปอ้างอิงได้ นอกจากนี้ผู้วิจัยพบสัดส่วนของจีโนไทป์ของยีน BACE1 ในผู้ป่วย CHB (N=149), CHC (N=31) มีจีโนไทป์ C/G สูงกว่าในคนปกติ (N=164) (p-value = 0.044) และยีน ApoE ในผู้ป่วย CHB (N=149), CHC (N=31) มีจีโนไทป์ ε3ε4 สูงกว่าในคนปกติ (N=148) (p-value=0.001) …

สถานการณ์การดื้อยาปฏิชีวนะของแบคทีเรียเป็นปัญหาใหญ่ที่ส่งผลกระทบไปทั่วโลก จึงจำเป็นต้องศึกษาเพื่อพัฒนาสารทดแทนยาปฏิชีวนะ ตัวเงินตัวทอง (Varanus salvator) เป็นสัตว์เลื้อยคลานมีความสามารถในการอยู่รอดจากสภาวะที่เสี่ยงต่อการติดเชื้อ อาจเนื่องจากสัตว์เหล่านี้มีระบบภูมิคุ้มกันโดยกำเนิดซึ่งมีวิวัฒนาการมาแต่โบราณจึงเป็นจุดเด่นที่น่าศึกษาวิจัย งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาฤทธิ์ต้านเชื้อแบคทีเรียขององค์ประกอบเลือดจากตัวเงินตัวทองและโปรตีนหรือเปปไทด์ที่มีคุณสมบัติต้านจุลชีพ ซึ่งข้อมูลของลำดับกรดอะมิโนของตัวเงินตัวทองที่ได้ในการศึกษาวิจัยครั้งนี้จะเป็นพื้นฐานที่สำคัญใน โดยนำพลาสมาและซีรัมจากตัวเงินตัวทองมาทดสอบคุณสมบัติการยับยั้งเชื้อแบคทีเรียที่ทำให้เกิดโรคด้วยเทคนิค agar diffusion พบว่าพลาสมาสามารถยับยั้งการเจริญของเชื้อแบคทีเรียได้ดีที่สุดมีขนาดเส้นผ่านศูนย์กลางของวงใสยับยั้งเชื้อ (inhibition zone) เท่ากับ 17-20 มิลลิเมตร เมื่อทำการทดสอบหาค่าความเข้มข้นต่ำสุดของพลาสมาและซีรัมที่สามารถยับยั้งเชื้อแบคทีเรีย (minimal inhibitory concentration, MIC) ด้วยวิธี broth dilution พบว่าสารสกัดหยาบพลาสมาให้ผลการทดสอบกับเชื้อได้ดีที่สุดโดยมีค่าความเข้มข้นต่ำสุดที่สามารถยับยั้งเชื้อแบคทีเรีย (MIC) เท่ากับ 125 ไมโครกรัมต่อมิลลิลิตร จากผลของการแยกบริสุทธิ์บางส่วนของพลาสมาและซีรัมโดยเทคนิค ion exchange chromatography ด้วย Q - sepharose คอลัมน์ พบว่าทั้งพลาสมาและซีรัมสามารถแยกโปรตีนแฟรคชันได้ 5 แฟรคชันเท่ากัน เมื่อวิเคราะห์ด้วยเทคนิค sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) พบขนาดโมเลกุลอยู่ระหว่าง 16 – 200 กิโลดัลตัน แต่เมื่อทดสอบคุณสมบัติการต้านเชื้อ พบบางแฟรกชันจากพลาสมา (P) และซีรั่ม (S) ซึ่งกำหนดเป็น P1 / P2 / P5 และ S1 / S2 / S4 ตามลำดับ ที่แสดงฤทธิ์ต้านเชื้อแบคทีเรีย เมื่อวิเคราะห์องค์ประกอบของสารออกฤทธิ์ทางชีวภาพด้วยเทคนิค liquid chromatography mass spectrometry (LC-MS) สามารถระบุชนิดโปรตีนได้ทั้งหมด 33 ชนิดและพบโปรตีนที่มีศักยภาพเป็นเปปไทด์ต้านจุลชีพ 15 ชนิด มีเพียง 3 ชนิด จากส่วนของพลาสมา (P1) 2 ชนิด และ (P2) 1 ชนิด ที่มีโครงสร้างเป็น α-helical peptide …

งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาองค์ประกอบทางพฤกษเคมีและฤทธิ์ต้านเชื้อรา Candida albicans ของน้ำมันเสม็ดขาวจากใบและกิ่งอ่อนของ Melaleuca cajuputi Powell ที่พบในพื้นที่ภาคตะวันออกของประเทศไทย รวมทั้งประเมินผลกระทบของน้ำมันเสม็ดขาวต่อการแสดงออกของยีนที่เกี่ยวข้องกับกลไกการดื้อยาฟลูโคนาโซลในเชื้อราทดสอบ การวิเคราะห์องค์ประกอบทางพฤกษเคมีด้วยวิธีแก๊สโครมาโทกราฟี-แมสสเปกโตรมิทรี (gas chromatography-mass spectrometry, GC-MS) พบว่าน้ำมันเสม็ดขาวมีองค์ประกอบหลัก 6 ชนิด ได้แก่ 1,8-Naphthyridine derivatives (ร้อยละ 10.46), alpha-Pyrone (ร้อยละ 10.11), Terpinolene (ร้อยละ 9.26), gamma-Terpinene (ร้อยละ 8.00), trans-Caryophyllene (ร้อยละ 6.36) และ beta-Elemene (ร้อยละ 5.09) จากการทดสอบฤทธิ์เบื้องต้นของน้ำมันเสม็ดขาวในการยับยั้ง C. albicans สายพันธุ์คลินิกที่ดื้อต่อยาฟลูโคนาโซล จำนวน 16 ไอโซเลท ด้วยวิธี Disc diffusion พบว่าดิสก์บรรจุน้ำมันเสม็ดขาว ขนาด 1 ไมโครลิตรต่อดิสก์ แสดงฤทธิ์ยับยั้งการเจริญของเชื้อทดสอบทุกไอโซเลท โดยมีขนาดเส้นผ่านศูนย์กลางบริเวณยับยั้งเชื้อเฉลี่ยอยู่ระหว่าง 6.67 ± 0.58 ถึง 10.00 ± 0.00 มิลลิเมตร เมื่อทำการทดสอบหาค่าความเข้มข้นต่ำสุดของน้ำมันเสม็ดขาวที่สามารถยับยั้งเชื้อทดสอบ (minimal inhibitory concentration, MIC) และฆ่าเชื้อทดสอบ (minimum fungicidal concentration, MFC) ด้วยวิธี Broth macrodilution พบว่าน้ำมันเสม็ดขาวแสดงฤทธิ์ฆ่าเชื้อราทดสอบทุกไอโซเลท โดยมีค่า MIC และ MFC อยู่ในช่วง 0.31-1.25 และ 0.63-2.5 ไมโครลิตรต่อมิลลิลิตร ตามลำดับ การทดสอบการเสริมฤทธิ์กันของสารผสมน้ำมันเสม็ดขาวและยาฟลูโคนาโซลในการต้านเชื้อราทดสอบด้วยวิธี Checkerboard microdilution พบว่าสารผสมดังกล่าวมีการเสริมฤทธิ์กันในการยับยั้งเชื้อราทดสอบส่วนใหญ่ (ร้อยละ 60) โดยค่า MIC ของยาฟลูโคนาโซลและน้ำมันเสม็ดขาวจะลดลงประมาณ 8-64 และ 8-16 เท่า ตามลำดับ …
การพัฒนาเทคนิค Helicase Dependent Amplification ควบคู่กับการอ่านผลด้วยตาเปล่าโดยใช้สี Sybr Green I (Hda/Sybr Green I) สำหรับการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิต, วรางคณา แย้มเกต

ผลิตภัณฑ์เกล็ดโลหิตที่มีการปนเปื้อนของเชื้อแบคทีเรีย อาจก่อให้การติดเชื้อในกระแสโลหิตที่รุนแรงและนำไปสู่การเสียชีวิตได้ ดังนั้นจึงมีการกำหนดมาตรฐานให้งานธนาคารโลหิต ทำการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตก่อนนำไปใช้กับผู้ป่วย เพื่อลดอาการไม่พึงประสงค์ที่อาจเกิดขึ้น วิธีการตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตในปัจจุบันยังมีข้อจำกัดที่พบได้บางประการ อาทิเช่น การเพาะเลี้ยงเชื้อแบคทีเรียที่ไม่สามารถปฏิบัติได้ในห้องปฏิบัติการทางธนาคารโลหิต การทดสอบที่ใช้ระยะเวลานาน ซึ่งไม่เหมาะสมกับอายุการเก็บรักษาผลิตภัณฑ์เกล็ดโลหิตที่สั้นประมาณ 5 วัน และต้นทุนของการทดสอบที่มีราคาแพง วัตถุประสงค์ของการศึกษาครั้งนี้เพื่อพัฒนาเทคนิค Helicase dependent amplification ควบคู่กับการอ่านผลด้วยตาเปล่าโดยใช้สี SYBR Green I (HDA/SYBR Green I) สำหรับตรวจหาการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิต โดยใช้ Primer ที่ถูกออกแบบให้จำเพาะต่อยีน 16s rRNA ซึ่งเป็น Universal gene ของเชื้อแบคทีเรีย ผลการทดสอบกับตัวอย่างผลิตภัณฑ์เกล็ดโลหิตที่ปนเปื้อนเชื้อแบคทีเรียแบบจำลอง ณ วันต่าง ๆ จำนวน 96 ตัวอย่าง พบว่าเทคนิค HDA/SYBR Green I มีความไวและความจำเพาะสูง เท่ากับ 96% และ 100% ตามลำดับ และมีความสอดคล้องกันในระดับดีมาก เมื่อเปรียบเทียบกับเทคนิค HDA/AGE เมื่อเปรียบเทียบกับเทคนิค PCR การนับจำนวนโคโลนี และเครื่องอัตโนมัติ BacT/Alert System พบว่าเทคนิค HDA/SYBR Green I มีความจำเพาะสูงเท่ากับ 100% แต่มีความไวอยู่ในช่วงระหว่าง 63-76% จึงทำให้มีความสอดคล้องกันในระดับปานกลางถึงดี อย่างไรก็ดี เมื่อนำเทคนิค HDA/SYBR Green I ตรวจการปนเปื้อนเชื้อแบคทีเรียในผลิตภัณฑ์เกล็ดโลหิตเฉพาะวันที่ 2 ถึง 5 ของการเก็บรักษา พบว่ามีความไวที่สูงขึ้น เท่ากับ 88% และมีความสอดคล้องกันในระดับดีมาก เทคนิค HDA/SYBR Green I มีค่าความเข้มข้นน้อยที่สุดของ DNA ต้นแบบที่สามารถตรวจได้ เท่ากับ 1 ng และไม่เกิดปฏิกิริยาข้ามกลุ่มกับสิ่งมีชีวิตอื่น ๆ ที่มีโอกาสพบการปนเปื้อนได้ในผลิตภัณฑ์เกล็ดโลหิต เทคนิค HDA/SYBR Green I …

การทดสอบความไวต่อยา Pyrazinamide (PZA) ด้วยวิธีมาตรฐาน (culture-based susceptibility tests) นั้นทำได้ยาก เนื่องจากสภาวะเป็นกรดในอาหารเลี้ยงเชื้อสามารถทำให้เกิดผลลบและผลบวกลวง การศึกษานี้มุ่งเน้นศึกษาและพัฒนาชุดทดสอบสำหรับใช้วินิจฉัยเชื้อดื้อต่อยา PZA ในเชื้อ Mycobacterium tuberculosis ด้วยชุดทดสอบ colorimetric biphasic medium assay ที่ผสม Nicotinamide (NIC-CBMA) ความเข้มข้น 250, 500, 1000 และ 2000 µg/mL และวิเคราะห์การกลายพันธุ์ที่สัมพันธ์กับการดื้อยา PZA ในยีน pncA, rpsA และ panD ผลการทดสอบในเชื้อวัณโรคดื้อยาทั้งหมด 150 สายพันธุ์ แบ่งเป็นเชื้อดื้อต่อยา PZA 40 สายพันธุ์ และเชื้อไวต่อยา PZA 110 สายพันธุ์ เมื่อทดสอบด้วยวิธี BACTEC MGIT 960 พบว่าความเข้มข้นของ NIC ≥ 1000 µg/mL เหมาะสำหรับใช้วินิจฉัยเชื้อดื้อยา PZA ด้วยวิธี NIC-CBMA ซึ่งมีความไวของชุดทดสอบเท่ากับ 100% ความจำเพาะเท่ากับ 96.36% เมื่อเทียบกับวิธีมาตรฐาน BACTEC MGIT 960 นอกจากนี้พบการกลายพันธุ์ในยีน pncA ที่สัมพันธ์กับการดื้อยา PZA จำนวน 28 แบบ ในเชื้อจำนวน 37 จาก 40 สายพันธุ์ที่ดื้อยา PZA เป็นการกลายพันธุ์แบบ Nonsynonymous 57.89% การกลายพันธุ์แบบ Indels 21.05% การกลายพันธุ์บริเวณ Promoter 13.16% การกลายพันธุ์แบบ Nonsense 5.26% และการกลายพันธุ์แบบ Double mutation 2.63% นอกจากนี้พบการกลายพันธุ์ใหม่จำนวน 4 แบบ …
