The dimethyl sulfoxide reductase (or MopB) family is a diverse assemblage of enzymes found throughout Bacteria and Archaea. Many of these enzymes are believed to have been present in the last universal common ancestor (LUCA) of all cellular lineages. However, gaps in knowledge remain about how MopB enzymes evolved and how this diversification of functions impacted global biogeochemical cycles through geologic time. In this study, we perform maximum likelihood phylogenetic analyses on manually curated comparative genomic and metagenomic data sets containing over 47,000 distinct MopB homologs. Cardiovascular diseases are a leading cause of mortality and morbidity worldwide. Aberrant thrombosis is a common feature of systemic conditions like diabetes and obesity, and chronic inflammatory diseases like atherosclerosis, cancer, and autoimmune diseases. Upon vascular injury, usually the coagulation system, platelets, and endothelium act in an orchestrated manner to prevent bleeding by forming a clot at the site of the injury. Abnormalities in this process lead to either excessive bleeding or uncontrolled thrombosis/insufficient antithrombotic activity, which translates into vessel occlusion and its sequelae. Salicylic acid (SA) is a central plant hormone mediating immunity, growth, and development. Recently, studies have highlighted the sensitivity of the SA pathway to changing climatic factors and the plant microbiome. Here we summarize organizing principles and themes in the regulation of SA biosynthesis, signaling, and metabolism by changing abiotic/biotic environments, focusing on molecular nodes governing SA pathway vulnerability or resilience. We especially highlight advances in the thermosensitive mechanisms underpinning SA-mediated immunity, including differential regulation of key transcription factors (e.g., CAMTAs, CBP60g, SARD1, bHLH059), selective protein–protein interactions of the SA receptor NPR1, and dynamic phase separation of the recently identified …

Toxoplasma gondii is a prevalent pathogenic parasite that infects approximately 25 percent of the US population. For the parasite to successfully establish and maintain infection in its host, properly controlled regulation of gene expression is critical. One way Toxoplasma regulates gene expression is through modification of histone proteins that bind to DNA and can control gene accessibility. Acetylation is a modification that is added to histones that changes chromatin structure to enhance gene activation. Epigenetics is the study of molecular modification of a genome without changing its base pairs. The most studied type of epigenetic mechanism is DNA methylation, which is capable of turning a gene “on” or “off.” Epigenetic potential is the capacity to which an individual can have methylation on its genome. การดื้อยา colistin มีแนวโน้มเพิ่มสูงขึ้นในเชื้อแบคทีเรียกลุ่ม Enterobacterales เนื่องมาจากการนำยา colistin กลับมาใช้ในการรักษาเชื้อดื้อยาหลายขนาน โดยเฉพาะอย่างยิ่งเชื้อแบคทีเรียแกรมลบที่ดื้อยา carbapenem ประกอบกับการถ่ายทอดยีน mobile colistin-resistant (mcr) ผ่านทาง plasmid ซึ่งส่งผลให้การแพร่ระบาดของเชื้อดื้อยา colistin เกิดขึ้นในวงกว้างอย่างรวดเร็ว สถานการณ์เชื้อกลุ่ม Enterobacterales ที่ดื้อยา colistin ในประเทศไทยกำลังเป็นที่สนใจ เนื่องมาจากเริ่มพบอัตราเชื้อดื้อยาที่สูงขึ้นอย่างต่อเนื่องทั้งในภาคการปศุสัตว์และในโรงพยาบาล การศึกษาครั้งนี้ได้ศึกษาลักษณะจีโนมของเชื้อ E. coli และ K. pneumoniae ที่ดื้อยา colistin ที่เพาะแยกได้จากผู้ป่วยของโรงพยาบาลเลิดสิน กรุงเทพมหานคร ในช่วงระหว่างเดือนตุลาคม พ.ศ. 2564 ถึงเดือนกันยายน พ.ศ. 2565 และศึกษาความสัมพันธ์และการกระจายตัวของเชื้อในแผนกต่างๆ ของโรงพยาบาล เชื้อที่เก็บได้ทั้งหมดจำนวน 54 ตัวอย่าง ประกอบด้วยเชื้อ E. coli และ K. pneumoniae จำนวน 14 และ 40 ตัวอย่าง ตามลำดับ ซึ่งให้ผลดื้อยา colistin เมื่อทดสอบด้วยเครื่องอัตโนมัติ Sensititre™ Gram Negative MIC Plate เชื้อส่วนใหญ่เพาะแยกจากปัสสาวะ (46.30%, p-value=0.38) และเสมหะ (29.63%, p-value=0.63) โดยเพาะแยกเชื้อได้จากผู้ป่วยใน (81.48%) มากกว่าผู้ป่วยนอก (18.52%) อย่างมีนัยสำคัญ (p-value=0.000) ภายหลังนำเชื้อทั้งหมดไปวิเคราะห์ลำดับเบสด้วยเทคนิค Illumina whole genome sequencing (WGS) สามารถจำแนกสายพันธุ์ของเชื้อได้อย่างมีประสิทธิภาพ โดยเชื้อ E. coli และ K. pneumoniae มีรูปแบบยีนก่อโรคที่แตกต่างกันอย่างเห็นได้ชัด และพบยีน blaCTX-M, blaTEM, blaOXA ,blaSHV ,ยีน aac, ยีน qnr …

การพัฒนาวิธีการตรวจออกซิไดซ์ไลโพโปรตีนชนิดความหนาแน่นต่ำ โดยใช้เทคนิคการตรวจวัดบนกระดาษร่วมกับ Immunogold Silver Enhancement, สุจิตรา ต๊ะยามัน

โรคหลอดเลือดแดงโคโรนารี (Coronary Artery Disease, CAD) เป็นโรคไม่ติดต่อชนิดเรื้อรังที่เกิดจากหลอดเลือดแดงที่กล้ามเนื้อหัวใจตีบหรือตัน สาเหตุหลักเกิดจากไขมันและเนื้อเยื่อสะสม มีผลทำให้เยื่อบุผนังหลอดเลือดชั้นในหนาตัวขึ้นและเกิดการอุดตัน จากการศึกษาที่ผ่านมาพบว่า ox-LDL เป็นจุดเริ่มต้นของโรคดังกล่าว เนื่องจากมีคุณสมบัติต่างจาก normal LDL โดยไม่สามารถจับได้กับ LDL receptor ได้ แต่สามารถจับได้กับ scavenger receptor ที่ผิวเซลล์แมคโคฟาจจึงทำให้ ox-LDL สามารถเข้าเซลล์ได้อย่างไม่มีสิ้นสุด เกิดเป็นโฟมเซลล์ (foam cells) สะสมอยู่ในผนังหลอดเลือดเกิดเป็นรอยขีดไขมัน (fatty streak) ประกอบกับการกระตุ้นเกล็ดเลือดและเซลล์อิมมูน ทำให้เกิดเป็นลิ่มเลือดอุดตันทางเดินของเส้นเลือด แม้ว่า ox-LDL จะเป็นสาเหตุสำคัญของการเกิดโรคแต่การตรวจวัดระดับไขมัน ox-LDL ยังไม่แพร่หลายในห้องปฏิบัติการเทคนิคการแพทย์ เนื่องจากชุดตรวจมีราคาแพง ใช้เวลาในการวิเคราะห์นาน และยังต้องใช้เครื่องมือที่มีความจำเพาะ ดังนั้นการศึกษาในครั้งนี้มีวัตถุประสงค์เพื่อพัฒนาเทคนิคการตรวจวัดระดับ ox-LDL ด้วยเทคนิคการตรวจวัดบนฐานกระดาษร่วมกับ immunogold silver enhancement โดยออกแบบกระดาษกรองให้มีรูปแบบเป็น microtiter plate อาศัยเทคนิค wax printing ในการสร้างขอบเขต หาสภาวะที่เหมาะสมสำหรับออกซิไดซ์กระดาษ เพื่อให้มีคุณสมบัติเหมาะสมกับการตรึงแอนติบอดี โดยเปลี่ยนเซลลูโลสเป็นหมู่แอลดีไฮด์ งานวิจัยนี้ได้นำสารละลายลิเธียมคลอไรด์มาช่วยเพิ่มประสิทธิภาพการออกซิไดซ์ร่วมกับสารละลายโซเดียมเพอร์ริออเดท พบว่ากระดาษที่ออกซิไดซ์แล้วสามารถคงสภาพนาน 72 ชั่วโมง และแอนติบอดีที่ตรึงไว้คงสภาพนานที่สุด 4 ชั่วโมง ทดลองวิเคราะห์วัดปริมาณ std. ox-LDL และ ox-LDL ที่เตรียมเองในห้องปฏิบัติการด้วยสภาวะเทคนิคดังกล่าว พบว่าสีปฏิกิริยาที่เกิดขึ้นยังไม่ชัดเจน อันเนื่องมากจากข้อจำกัดของความจำเพาะของแอนติบอดี การทำให้เกิดสีด้วย silver enhancement ซึ่งยังคงต้องมีการพัฒนาต่อไปเพื่อให้ผลการวิเคราะห์มีความชัดเจน นอกจากนี้ได้พัฒนากล่องถ่ายภาพเพื่อใช้เก็บผลการวิเคราะห์บนฐานกระดาษ ที่สามารถถ่ายภาพด้วยแสงและระยะที่คงที่ สามารถเคลื่อนย้ายได้ง่ายในภาคสนาม อย่างไรก็ตามแม้การพัฒนาวิธีการวิเคราะห์ ox-LDL ยังไม่ประสบผลสำเร็จ แต่ยังพอได้แนวทางและต้นแบบในการศึกษาต่อไปในอนาคต

Aspergillosis เป็นโรคติดเชื้ออันตรายถึงแก่ชีวิต มีสาเหตุมาจากเชื้อราในจีนัส Aspergillus การวินิจฉัยโรคในปัจจุบัน การเพาะแยกเชื้อและจัดกลุ่มตามลักษณะทางสัณฐานวิทยายังคงเป็นวิธีมาตรฐาน การตรวจหา galactomannan มักใช้ตรวจวินิจฉัยโรคในผู้ป่วยกลุ่มเสี่ยงเท่านั้น ในขณะที่เทคนิคทางอณูชีววิทยาอาศัยข้อมูลสารพันธุกรรมของ “its region” ในการจำแนกกลุ่มเชื้อระดับสปีชีส์ แต่อย่างไรก็ตามข้อมูลดังกล่าวถูกพิสูจน์ว่าสามารถจำแนกกลุ่มเชื้อราก่อโรคในคนในระดับสปีชีส์ได้ต่ำ ในด้านการรักษาสปีชีส์ของเชื้อ Aspergillus มักไม่ถูกนำมาพิจารณาร่วมเพื่อการเลือกกลุ่มยาแม้ว่ายังคงมีผู้ป่วยบางรายที่ตอบสนองต่อการรักษาไม่ดีเนื่องจากเชื้อสาเหตุดื้อต่อยาต้านเชื้อรา ด้วยเหตุดังกล่าวเพื่อศึกษาความสัมพันธ์ระหว่างสปีชีส์และรูปแบบความไวรับต่อยา การค้นหายีนที่สามารถจำแนกเชื้อ Aspergillus ในระดับสปีชีส์ได้อย่างแม่นยำจึงเป็นสิ่งจำเป็นยีน “tef-1α” เป็นสารพันธุกรรมที่ได้รับการพิสูจน์แล้วว่ามีคุณสมบัติสามารถจำแนกเชื้อราเส้นสายในระดับสปีชีส์ได้สูง ในงานวิจัยนี้ ผู้วิจัยได้เปรียบเทียบผลการจำแนกกลุ่มเชื้อ Aspergillus ที่แยกได้จากผู้ป่วยจำนวน 227 ตัวอย่าง โดยใช้ข้อมูลสารพันธุกรรม “its region” และ“tef-1α” จากผลการศึกษาพบว่าสารพันธุกรรมจากยีน“tef-1α” (D=0.8461) มีความสามารถในการจำแนกเชื้อในระดับสปีส์สูงกว่าบริเวณ “its” (D=0.796) โดยเฉพาะอย่างยิ่งใน section Fumigati ข้อมูลสารพันธุกรรมจาก “its region” สามารถแบ่งเชื้อกลุ่มดังกล่าวออกได้เพียง 1 สปีชีส์ คือ A. fumigatus ในขณะที่ยีน“tef-1α” สามารถแบ่งออกเป็น A. fumigatus และอีก 2 สปีชีส์ คือ A. lentulus และ A. udagawa ซึ่งเป็นเชื้อที่ดื้อต่อยา amphotericin B โดยกำเนิด โดยสรุป ผลจากงานวิจัยนี้ชี้ให้เห็นว่าการจำแนกสปีชีส์ของเชื้อ Aspergillus โดยใช้ยีนทุติยภูมิ “tef-1α” สามารถบ่งชี้กลุ่มยาที่เหมาะสมต่อการรักษาได้

This dissertation explores the identity and role of immunoglobulin-like (Ig-like) receptors in chickens, with focus on their implications in disease and disease progression. These receptors, wisely expressed across immune cells, interact with the major histocompatibility complex (MHC) class I molecules to modulate immune responses in mammals. Due to the insufficient representation of chicken Ig-like receptors in online databases, this study systematically annotates the chicken Cluster Homolog of Immunoglobulin-like Receptors (CHIR) genes using advanced bioinformatic techniques, aligning with the release of the 7th edition of the chicken genome assembly that comprises builds for a broiler and layer chicken. Schistosoma haematobium, the agent of urogenital schistosomiasis, and related schistosomes are transmitted by members of the genus Bulinus. Each of the 38 Bulinus species vary in their ability to vector schistosome species and non-schistosome trematodes resulting in a patchwork of snail-parasite compatibilities. Accurately identifying snail intermediate hosts and the disease-causing parasites they transmit is critical for snail control strategies and the management of human schistosomiasis. Defenses against pathogens can take on many forms. For instance, behavioral avoidance of diseased conspecifics is widely documented. Interactions with these infectious conspecifics can also, however, lead to physiological changes in uninfected animals, an effect that is much less well understood. These changes in behavior and physiology are particularly important to study in a reproductive context, where they can impact reproductive decisions and offspring quality. The western honey bee, Apis mellifera L., is a globally important pollinator plagued by several harmful stressors impacting colony health and survival. At least eight A. mellifera subspecies were imported and continue to be the genetic ancestors of U.S. honey bee populations today. Successive genetic bottle-neck events have led to reduced genetic diversity in U.S. honey bees. Schistosomiasis, a commonly neglected tropical disease, is a waterborne parasitic worm infection able to infect through direct skin penetration. This disease affects approximately 270 million people worldwide and ranks only second to malaria as a leading infectious disease. Although some possible alternatives are emerging, currently, the most effective drug treatment is praziquantel (PZQ). Pancreatic ductal adenocarcinoma (PDAC) is a devastating disease with dismal prognosis. The only curative option for patients is surgery, but over 80% of patients are not surgical candidates. Unfortunately, PDAC is resistant to the three remaining options. PDAC is characterized by a profoundly hypoxic and immunosuppressive stroma, which contributes to its therapeutic recalcitrance. Alpha-smooth muscle actin+ (αSMA+) cancer-associated fibroblasts (CAFs) are the most abundant stromal component, as well as mediators of stromal deposition. Peanut allergies can have severe and life-threatening complications. By understanding the biology of individuals struggling with this disease, better treatments can be developed. This project explores the possibility of two phenotypically distinct cell types within CD154+ cell types in patients with peanut allergy. Previous studies show that there is a Th2 cell subpopulation associated with allergies, and within effector CD4+ memory populations two distinct phenotypes have been found. Given the diversity and complexity within immunoglobulin (IG) loci, effective mouse models first require characterization of intra-strain differences and construction of high-quality reference assemblies for IG loci in several representative strains. To understand light chain germline diversity across biomedically significant mouse strains, we profiled the expressed IGK and IGL repertoires of 18 commonly used laboratory mouse strains using AIRR-seq. Across strains, we observed germline IGKV sequences shared by three different IGK haplotypes and a more conserved IGLV germline repertoire among common laboratory strains. สุกรเป็นอีกหนึ่งแหล่งกักเก็บเชื้อแบคทีเรียที่มียีนดื้อยาสร้างเอนไซม์ Extended-Spectrum β-Lactamases (ESBLs) ซึ่งสามารถแพร่กระจายยีนดื้อยามาสู่มนุษย์ สัตว์ และสิ่งแวดล้อมได้ ในการศึกษานี้จึงได้พัฒนาการตรวจหายีนสร้างเอนไซม์ ESBLs จากมูลสุกรโดยตรง ด้วยเทคนิค Multiplex polymerase chain reaction (mPCR), mPCR-Lateral flow assay (mPCR-LFA) และเทคนิค Nested PCR (nPCR) เปรียบเทียบกับการตรวจจากเชื้อ ESBL-producing Escherichia coli ซึ่งเพาะแยกจากมูลสุกรตัวอย่างเดียวกัน ด้วยเทคนิค PCR-sequencing เชื้อ ESBL-producing E. coli จำนวน 149 isolates ที่เพาะแยกจากมูลสุกรทั้งหมด 139 ตัวอย่าง พบความชุกของยีนชนิด blaCTX-M-15 (34.2%) มากที่สุด รองลงมาคือ blaCTX-M-15+TEM-1 (17.5%) และ blaCTX-M-55+TEM-1 (13.4%) โดยเชื้อ ESBL-producing E. coli ส่วนใหญ่มีการดื้อยาหลายขนาน โดยดื้อต่อยา Ceftriaxone (100%), Cefotaxime (100%), Ciprofloxacin (31.5%) และ Trimethoprim/Sulfamethoxazole (30.9%) ความไว ความจำเพาะ และขีดจำกัดของการตรวจจากเชื้อ ESBL-producing E. coli ด้วยเทคนิค mPCR-LFA เท่ากับ 100% และ 0.1-1 ng ตามลำดับ เทคนิค mPCR ที่พัฒนาขึ้น สำหรับตรวจจากมูลสุกรโดยตรง พบว่าต้องใช้ปริมาณเชื้อมากถึง 107 CFU จึงจะตรวจพบ โดยมีความไวในการตรวจยีน blaCTX-M และ blaTEM เท่ากับ 30.43% และ 87.65% ตามลำดับ ขีดจำกัดและความไวของการตรวจพบยีน blaCTX-M โดยตรง …

Ixodes scapularis is a medically important tick that transmits several microbes to humans, including rickettsial pathogen Anaplasma phagocytophilum. In nature, these ticks encounter several abiotic factors including changes in temperature, humidity, and light. Many organisms use endogenously generated circadian pathways to encounter abiotic factors. In this study, we provide evidence for the first time to show that A. phagocytophilum modulates the arthropod circadian gene for its transmission to the vertebrate host. มะเร็งช่องปากชนิดสความัสเซลล์คาร์ซิโนมา (Oral squamous cell carcinoma, OSCC) จัดเป็นมะเร็งที่พบได้บ่อยที่สุดของ มะเร็งศรีษะและลำคอ เคอคิวมินเป็นสารออกฤทธิ์หลักซึ่งพบได้ในขมิ้นชันและถูกใช้เป็นสมุนไพรสำหรับการรักษาโรคต่าง ๆ หลายชนิด รวมทั้ง ได้รับการพิสูจน์ในห้องปฏิบัติการแล้วว่ามีคุณสมบัติในการยับยั้งเซลล์มะเร็งได้หลากหลายประเภท ในขณะที่โพรไบโอติกแบคทีเรีย Lactobacillus rhamnosus GG (LGG) ได้รับการพิสูจน์ในห้องปฏิบัติการถึงคุณสมบัติในการยับยั้งเซลล์มะเร็งได้หลากหลายชนิดเช่นกัน และ มีรายงานถึงการนำ LGG ไปใช้ร่วมกับสารผลิตภัณฑ์ธรรมชาติชนิดอื่น ๆ เพื่อเพิ่มประสิทธิภาพของสารธรรมชาตินั้นอีกด้วย ดังนั้นวัตถุประสงค์ ของการศึกษาในครั้งนี้คือ เพื่อศึกษาผลของการใช้เคอคิวมินร่วมกับน้ำเลี้ยงส่วนใสของแบคทีเรีย LGG (LGG cell-free supernatant, LGG CFS) ในการยับยั้งการมีชีวิตและกระตุ้นให้เซลล์มะเร็งช่องปากชนิด OSCC (SCC-9 cells) เกิดการตายแบบอะพอพโตซิส ผลการศึกษาพบว่า เคอคิวมินและ LGG CFS มีคุณสมบัติในการยับยั้งการมีชีวิตของเซลล์มะเร็งช่องปาก SCC-9 cells โดยขึ้นอยู่กับความเข้มข้นที่ใช้ การใช้เคอคิว มินและ LGG CFS ที่ความเข้มข้นสูง (Curcumin 40 µg/ml และ 25% (volume/volume) LGG CFS ที่ความเข้มข้น 108 CFU/ml หรือความ เข้มเข้นเริ่มต้น (undiluted condition)) ในรูปแบบการใช้สารเดี่ยวหรือสารผสมร่วมกัน ส่งผลยับยั้งการมีชีวิตของทั้งเซลล์มะเร็งช่องปาก SCC9 cells และเซลล์เนื้อเยื่อเหงือก (human gingival fibroblast, HGF cells) เมื่อเปรียบเทียบกับสภาวะควบคุม (p<0.0001 สำหรับการใช้สาร เดี่ยวในเซลล์มะเร็งช่องปาก SCC-9 และการใช้สารเดี ่ยวและสารผสมในเซลล์เนื ้อเยื ่อเหงือก HGF และ p=0.02 สำหรับการใช้สารผสมใน เซลล์มะเร็งช่องปาก SCC-9) ในขณะที ่การใช้เคอคิวมินและ LGG CFS ในรูปแบบของสารผสมร่วมกันที ่ระดับความเข้มข้นต่ำ (Curcumin 5 µg/ml และ 25% (v/v) LGG CFS ความเข้มข้น 1:100 เท่าจากระดับความเข้มข้นเริ ่มต้น) สามารถยับยั ้งการมีชีวิตของเซลล์มะเร็งช่องปาก SCC-9 ได้อย่างมีประสิทธิภาพเมื่อเปรียบเทียบกับสภาวะการใช้สารเดี่ยวของเคอคิวมินและ LGG CFS ที่ระดับความเข้มข้นต่ำ และสภาวะควบคุม (p<0.0001, p=0.002 และ p=0.02 ตามลำดับ) และไม่ส่งผลต่อการมีชีวิตของเซลล์เนื้อเยื่อเหงือก HGF เมื่อเทียบกับกลุ่มควบคุม (p=0.14) ผลการวิเคราะห์ Flow analysis พบว่าสภาวะการใช้เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ สามารถเหนี่ยวนำให้ เซลล์มะเร็ง SCC-9 เกิดการตายแบบอะพอพโตซิสได้มากขึ้นอย่างมีนัยสำคัญทางสถิติเมื่อเปรียบเทียบกับการใช้สารเดี่ยวและสภาวะควบคุม (p<0.0001 ในทุกสภาวะ) นอกจากนั้นสภาวะการใช้เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ สามารถเหนี่ยวนำให้เซลล์มะเร็ง SCC-9 มีระดับการแสดงออกของ Bax/Bcl-2 ratio ที่สูงขึ้น ซึ่งมีผลให้ให้เซลล์มะเร็งเกิดการตายแบบอะพอพโตซิสต่อไป อย่างไรก็ตามสภาวะ เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ ส่งผลต่อระดับการแสดงออกของโปรตีน caspase-3 ที ่สูงขึ้นอย่างไม่มีนัยสำคัญ (p=0.12) แต่ไม่มีผลต่อระอัตราส่วนการแสดงออกของโปรตีน Bcl-xL/Bak และโปรตีน Mcl-1/Bak เมื่อเทียบกับกลุ่มควบคุม (p=0.53 และ p=0.34 ตามลำดับ) การวิเคราะห์ LGG CFS ที่ความเข้มข้น 1:100 เท่าจากระดับความเข้มข้นเริ่มต้น ด้วยเทคนิค LC-MS/MS พบสารประเภท โปรตีนทั้งหมด 30 ชนิด ได้แก่ metabolic enzyme and protein จำนวน 26 ชนิด และ surface layer protein จำนวน 4 ชนิด การใช้สาร เคอคิวมินและ LGG CFS ร่วมกันที่ระดับความเข้มข้นต่ำ น่าจะมีผลในการยับยั้งเซลล์มะเร็งช่องปากชนิด SCC-9 แต่ไม่มีผลกระทบต่อเซลล์ เหงือก HGF ผลการศึกษาในครั้งนี้อาจนำไปสู่การพัฒนาวิธีป้องกันหรือรักษามะเร็งช่องปากชนิด OSCC รูปแบบใหม่ได้ในอนาคต

Increasing prevalence of nosocomial infections by antimicrobial resistant pathogens resulting in higher mortality rates and financial burden is of great concern. Pseudomonas aeruginosa represents one of six highly virulent “ESKAPE” pathogens that exhibit considerable intrinsic drug resistance as well as mechanisms for acquiring further resistance. As many of these mechanisms are regulated through gene expression, we sought to identify regulatory strategies and patterns at play in 23 clinical isolates collected from Baku, Azerbaijan and Tyler, Texas, USA. Ependymoma is a primary solid tumor of the central nervous system. Supratentorial ependymoma (ST-EPN), a subtype of ependymomas, is driven by an oncogenic fusion between the ZFTA and RELA genes in 70% of cases. We introduced this fusion into neural progenitor cells of mice embryos via in utero electroporation of a non-viral binary piggyBac transposon system containing ZFTA-RELA. From preliminary data in the LoTurco lab, inducing the expression of ZFTA-RELA into different neural progenitor cells produces tumors of varying lethality and cellular composition.
