Platelets from 24 patients with immune thrombocytopenia resistant to standard therapy (refractory ITP), 35 patients with nonimmune thrombocytopenia (non-ITP), and 32 normal donors were studied in regard to platelet surface-bound IgG (PBIgG) and the ability of these platelets to be bound by human monocytes in vitro (monocyte-platelet rosette assay). Fourteen (58%) of the platelet samples from refractory ITP patients but none (0%) from the non-ITP or control donors had PBIgG greater than 800 molecules IgG/platelet. Seventeen of 24 (71%) of the ITP patients had platelets which demonstrated increased monocyte- platelet rosette formation [rosette index (RI) greater than 2], whereas only …

Ten patients with idiopathic thrombocytopenic purpura (ITP) were studied before and following a rise in circulating platelets subsequent to infusions of intravenous gammaglobulin (400 mg/kg/day x 5 days). We quantitated the amount of circulating IgG capable of binding to normal donor platelets in vitro using an 125I-monoclonal anti-human IgG assay, as well as the amount of IgG associated with the patients' platelets before and following therapy. We found no evidence for a decrease in platelet-specific IgG antibodies in these patients undergoing an acute response to therapy. These data suggest that the short-term efficacy of intravenous gammaglobulin is due to effects …

The effects of zinc on embryos and larvae of the fathead minnow (Pimephales promelas) were studied using standard 8-day embryo-larval bioassay techniques. The objective was to determine if there was a period in embryonic development which would be the most sensitive to the toxic and teratogenic effects of zinc. Five developmental stages were exposed to 0.5, 1.0 and 3.0 mg/L. After 96 hr, all animals exposed to 3.0 mg Zn/L were dead. The hatching stage was the most affected by 1.0 mg/L, with only 59% surviving after 96 hr, while the tailbud stage showed essentially control-level survival. However, virtually all …

Despite the extensive literature on salamander cranial morphology, few authors mention the structure or development of the mandibular symphysis. Adult specimens representing eight caudate families and larval Ambystoma opacum, representing nine developmental stages, were evaluated histologically to determine the structure and development of the mandibular symphysis.

From this analysis it was determined that early in the ontogeny of the mandibular symphysis Meckel's cartilages are separated by a band of undifferentiated cartilage. As development progresses, this band of undifferentiated cartilage gradually transforms into a wedge of fibrocartilage which fuses to Meckel's cartilages.

Piglets from 42 litters of Yorkshire sows were weighed 0, 1, 2, 3, 4, 5, 7, 10, and 14 d after farrowing in order to explore the highly variable weight gains during the first 2 wk after birth. All litters were adjusted to 10-12 piglets by fostering. Litters differed greatly in average gain per piglet during the first 3 d after farrowing, with litter means ranging from 5 to 227 g d‒1 per piglet for the 3-d period. Use of drinking water was determined for 51 litters of piglets by weighing and refilling a water dispenser each day for the first 4 d after farrowing. Measures of spillage, evaporation, and piglet drinking behavior indicated that most disappearance of water represented actual drinking by the piglets. Water use varied greatly among litters with an average of 46 g d‒1 per piglet over the 4 d, and a range of negligible use to about 200 g d‒1. Water use, especially on day 2, was correlated negatively with the average weight gain of the litter during the first …

A cytoarchitectural analysis of the rat olfactory tubercle using Nissl-stained coronal sections revealed that the dense cell layer (DCL) consisted of medium sized striatal cells in the cortical regions and small sized "granule" cells in the cap regions. Also delineated from this experiment was a rim of neuropil, nearly devoid of neurons as well as neuronal processes, outlining the islands of Calleja. Acetylcholinesterase (AChE) localization utilizing light microscopy revealed that the olfactory tubercles (OT) contained AChE-positive fibers that were orientated dorsoventrally in the molecular and multiform layers. Queuosine, a nucleoside found exclusively in the first position of the anticodon of transfer RNA (tRNA), is generated post-transcriptionally in an exchange of guanine for queuine by the modifying enzyme tRNA-guanine ribosyltransferase. Phorbol ester tumor promoters were shown to induce queuine hypomodified tRNA, and that phorbol ester action was due, in part, to inhibition of queuine transport across the cell membrane. An unidentified protein designated conditioned medium factor (CMF) that potentiated a phorbol-induced inhibition of queuine uptake was also documented. We suggest here that phorbol ester-induced inhibition of queuine uptake is not a significant factor in inducing queuine-deficient tRNA although …

Chromium-52 has recently been suggested for use as a new agent to label red cells for the in vivo quantitation of red cell volume (1). In this paper, the development and validation of a routine 52cr labeling procedure is described.

The accuracy, precision, and detection limits of chromium analysis by Zeeman effect atomic absorption spectroscopy was evaluated in the concentration range of 1 - 10 ug Cr/L.

Laminin, a basement membrane glycoprotein promotes both cell attachment and neurite outgrowth. Separate domains on laminin elicit these responses, suggesting that distinct receptors occur on the surface of cells. NG108-15 neuroblastoma-glioma cells rapidly extend long processes in the presence of laminin. We report here that 125I-labeled laminin specifically binds to these cells and to three membrane proteins of 67, 110, and 180 kDa. These proteins were isolated by affinity chromatography on laminin-Sepharose. The 67-kDa protein reacted with antibody to the previously characterized receptor for cell attachment to laminin. An agar medium with improved selection for Treponema hyodysenteriae was developed. A method was developed for extraction of plasmid DNA from bacterial cells embedded in agarose blocks. Cell - containing blocks were treated with various lysing reagents and inserted into the wells of an agarose gel. Upon electrophoresis the plasmid DNA migrated out of the embedding block and into the gel leaving intact chromosomal DNA in the well. This method was tested with various organisms and found to be effective for plasmid isolation. In comparison to "traditional" procedures, this new method is less tedious since chemical separation of plasmids is not required prior to electrophoresis. Also, a higher yield of plasmid …

We studied the efficiency of a standard-kit preparation using 1 mg 111In-labeled 96.5 monoclonal antibody in combination with 19 mg of unlabeled antibody in the diagnostic imaging of 27 patients with documented metastatic melanoma. Twenty-three of 26 patients (88%) demonstrated immunoscintigraphic localization of tumor. Of 104 metastatic sites previously documented by conventional studies, 62 (60%) were identified by immunoscintigraphy. A total of 77 sites demonstrated localization of radiolabeled antibody. Fifty-four (70%) corresponded to known sites of disease; eight sites (10%) were "discovered" by immunoscintigraphy and subsequently confirmed by conventional studies; 15 imaged sites (20%) could not be confirmed by conventional …

The cytoarchitecture of the substantia nigra pars lateralis (SNl) of the opossum was examined by light and electron microscopic procedures. Nissl-stained, Golgi-impregnated, HRP-reacted and electron microscopic preparations were utilized in this study. SNl neurons were divided into three groups on the basis of cell size; small, medium-sized, and large neurons. Small cells had a nucleus with some clumped heterochromatin, a relatively organelle-free cytoplasm, and long, thin, sparcely branching dendrites. Electron microscopic examination of small cells confirmed the light microscopic observations. A recent study showed that cAMP analogs lowered cAMP levels in rat hepatocytes. The present work demonstrates that cAMP analogs also lowered cAMP in a rapid, concentration-dependent manner in heart and fat cells. In order to determine if the cAMP-dependent protein kinase mediated this effect, techniques were developed to assay the protein kinase activity ratio in hepatocytes treated with cAMP analogs. The activation of protein kinase and phosphorylase in hepatocytes by 8-pClΦS-cAMP (where 8-pClΦS- indicates 8-parachlorothiophenyl-) was concentration-dependent and occurred in parallel to proportionate decreases in cAMP. Microsurgical fertilization is the technique in which a spermatozoon or sperm nucleus is injected into the cytoplasm of an egg. To establish a foundation for the use of microsurgical fertilization as a means of treating infertility, this study evaluated the procedure in hamster and human eggs. Hamster sperm nuclei were microinjected into hamster eggs to determine the rate of abnormal fertilization and to ultrastructurally assess cellular damage by transmission electron microscopy. In mammalian cell-virus systems, it has been observed that damage caused by exposure of the cell to ultraviolet radiation (UV) will result in an increase in viral plaque development rate. This phenomenon is termed the Large Plaque Effect (LPE). Apparently, viral plaque development increases at a faster rate for Herpes Simplex Virus (HSV) when it is assayed on certain UV-irradiated mammalian cells. The consequence of this increase in plaque development rate is that viral plaques appear larger on irradiated monolayers of cells when compared to plaques that developed on unirradiated cellular monolayers.

Nuclear protein extracts from day 12.5 mouse embryos were used to study protein binding to DNA sequences 5' of the Hox 1.5 homeo box. Embryos of this developmental stage are known to express this gene. DNA binding protein blotting and retardation gel techniques show that murine embryonic nuclear proteins specifically bind a 753-base pair (bp) DNA fragment from the region upstream of the Hox 1.5 homeo box. A fusion protein containing the Hox 1.5 homeo domain constructed in lambda gt11 also binds the same 753-bp DNA fragment. Macroscopically, the testes of Chthonerpeton indistinctum consist of a series of oval lobes. The number and size of lobes is variable in Chthonerpeton indistinctum. Histologically, the testes are divided into structural units, locules separated by septa of connective tissue. Inside of each locule spermatogenesis takes place in cysts of germinal cells that divide synchronically. Chthonerpeton indistinctum is divided chronologically into four stages of development based on the shape of the nuclei of the spermatids and their arrangement in the cyst: (1) round, (2) kidney shaped, (3) attenuate, and (4) bouquet arrangement.

Effects of pituitary stalk-transection on plasma concentrations of luteinizing hormone (LH), follicle stimulating hormone (FSH) prolactin (PRL) and progesterone were investigated during the estrous cycle of ewes. Pituitary stalk (SS) or sham (SH) transection was performed on day 1 (estrus = day 0) of the estrous cycle. A Teflon or Silastic barrier was placed between the cut ends of the stalk to prevent reorganization of the portal vasculature. Immediately following surgery, pulsatile administration of gonadotropin releasing hormone (GnRH, 200 ng/hr) or .9% NaCl was initiated and continued for the duration of the experiment. Acrosin is an acrosomal enzyme which is reported to be involved in sperm penetration through cumulus, zona, and ooplasma. This enzyme has been reported to be significantly lower in sperm of infertile men compared to sperm of fertile men. The acrosin assay which has been developed can be used for studies in patients with suspected infertility and may be used on a fraction of most semen specimens being evaluated in a clinical andrology laboratory.

Piglets from 51 litters were weighed weekly over a 5-wk lactation in an experiment that studied the effects of farrowing crate design and other factors on variation in piglet weight. Of two farrowing crates used, one had low horizontal bars that tended to impede access to the upper row of teats, while the other had angled vertical bars which permitted freer access. Fourteen-day weights were more uniform in the vertical-bar crates than in the horizontal-bar crates (P < 0.025), but the difference waned by day 35 (P > 0.05). There were no significant differences in mean body weight owing to crate type (P < 0.05) for any of the weekly measurements but, by day 14, there was a significant linear trend (P < 0.05) at decreasing main body weight with increasing litter size. The interaction between crate type and litter size was significant at the later ages (P < 0.01 at day 35) because the linear trend was more pronounced for horizontal-bar crates than for vertical. Litters differed greatly in the uniformity of their body weights. Much of the variation became established in the first and, to a lesser extent, the second week after birth. In those weeks, weight gains were highly variable and were not closely related to weight at the beginning of the week. Thereafter, the established differences tended to be perpetuated to 35 d, because weight gains were largely proportional to body weight. Within-litter competition appeared to influence weight gain considerably.

Luteinizing hormone (LH) increases intracellular concentrations of adenosine 3', 5 '-cyclic monophosphate and the phosphoinositides, phosphatidylinositol (PI), PI 4'-phosphate (PIP) and PI 4' .5 1 - bispbosphate (PIP2). It is believed that cAMP and the phosphoinositides act concertedly to regulate mitochondrial conversion of cholesterol to pregnenolone. The mammalian protooncogene homologue of the avian v-ets sequence from the E26 retrovirus consists of two sequentially distinct domains located on different chromosomes. Using somatic cell hybrid panels, we have mapped the mammalian homologue of the 5' v-ets-domain to chromosome 11 (ETS1) in man, to chromosome 9 (Ets-1) in mouse, and to chromosome D1 (ETS1) in the domestic cat. The mammalian homologue of the 3' v-ets domain was similarly mapped to human chromosome 21 (ETS2), to mouse chromosome 16 (Ets-2), and to feline chromosome C2 (ETS2). …

Suckling behavior and weight change of piglets were studied during the first 2 wk after birth. Thirty-six litters comprised a balanced comparison of parity number (first or second), litter size (8, 10 or 12), and two farrowing crate designs, one of which had a low horizontal bar that tended to impede the piglets' access to the udder. Piglets showed the expected preference for the anterior teats although the preference was more apparent in the second parity than in the first. Teat fidelity was most pronounced at the two ends of the udder; piglets occupying the middle positions did more fighting …
