We previously showed that the hypoxia-activated prodrug TH-302 (Evofosfamide) reduces intratumoral hypoxia through a tissue remodeling process, initiates tumor vasculature reorganization, and sensitizes aggressive, spontaneous murine models of prostate cancer to immune checkpoint blockade (ICB). In a clinical trial testing the combination of TH-302 with cytotoxic T-lymphocyte-associated protein (CTLA-4) blockade (NCT03098160) a subset of metastatic, ICB refractory patients showed prolonged progression free survival. While these studies highlight hypoxia as therapeutically tractable, we lack a complete understanding of the contribution of the tumor vasculature to hypoxia reduction therapy, as well as the downstream consequences of hypoxia reduction on the cellular composition …

Mammals, including humans, develop progenitor tissues for both male and female reproductive tract organs before they fully differentiate into a male or female tract. The progenitor tissue for the male reproductive tract is known as the Wolffian duct (WD), and the progenitor tissue for the female reproductive tract is the Müllerian duct (MD). The WD further differentiates into the vas deferens, epididymis, and seminal vesicle, while the MD differentiates into the oviduct, uterus and upper vagina. Lung cancer metastasis is leading the causes of cancer-related mortality in the United States and worldwide. Epithelial-to-mesenchymal transition (EMT) is a model for metastasis that results in loss of specialized epithelial cell contacts and acquisition of mesenchymal invasive capacity. Zinc finger E-box-binding homeobox 1 (ZEB1) recognizes and binds to E-boxes of epithelial gene promoters to repress its transcription. ZEB1 has inconsistent molecular weights, which have been attributed to post-translational modifications (PTMs). Human papillomavirus (HPV) is the causative agent of cervical cancer and some cancers of the penis, vulva, vagina, anus, and oropharynx. Current therapies for these cancers include a combination of surgery, radiotherapy, and chemotherapy that often results in permanent, life altering adverse effects. Immunotherapy is partially effective, but with significant recurrence and lower long-term survival. Importantly, there are no few biomarker-selective targeted therapies for these cancers. The CDH1 gene codes for the epithelial-cadherin (E-cad) protein, which is embedded in the plasma membrane of epithelial cells to form adherens junctions. E-cad is known to be essential for maintaining the integrity of epithelial tissues, and the loss of E-cad has been widely considered a hallmark of metastatic cancers enabling carcinoma cells to acquire the ability to migrate and invade nearby tissues. In C. elegans, the gene glp-1 encodes for a Notch receptor called GLP-1, one of two found in C. elegans’ genome. The gene has been previously implicated in the development of the hermaphroditic germline as well as playing a role in the mitosis/meiosis decision. Genetic screening has further identified it as potentially playing a role in the development of the male somatic gonad, making it an ideal candidate for a reverse genetic. We did this by silencing glp-1 and observing if any alterations to the gonad’s phenotype occur.

Background: Natural killer (NK) cells originate from the differentiation of hematopoietic stem cells (HSCs) in the common lymphoid progenitor pathway, and HSCs can be obtained from umbilical cord blood (UCB). Comparative studies of NK cell differentiation between cultured and freshly isolated HSCs are important in the development of NK cell therapy for cancer. The Caudal Type Homeobox transcription factors cdx are a family of genes found in vertebrates that regulates body regionalization and anterior-posterior patterning. They are also responsible for regulating axial elongation, but the mechanisms behind this behavior are not known. Previous studies in mouse embryonic stem cells have shown that the cdx genes are necessary for upregulating the gene sp5 which may be linked to axial elongation. Invasive plant species’ success may be a result of allelopathy, or the release of secondary metabolites that are harmful for surrounding plant species. Allelopathy can be mediated through the abiotic environment by chemical sorption or transformation, so the substrate on which interactions occur can lead to differential outcomes in allelopathic potential. Understanding the effects of anthropogenic factors, such as oxybenzone, on sea urchins is extremely important to protect the growth of coral reefs- the ocean’s hotspot for biodiversity. Spawning of sea urchins, Lytechinus variegatus, was induced and eggs were fertilized in different concentrations of oxybenzone contaminated water. 48 hours later, data was collected underneath a light microscope. Apoptosis, commonly known as programmed cell death, constantly occurs in humans. As a cancer cell increases in acidity, apoptosis is induced. In healthy cells, proton pump proteins allow for H+ ions to permeate cellular membranes, regulating pH. However, proton pump inhibitors (PPIs), such as omeprazole, prevent proton movement. In previous studies, omeprazole induced cell death in Jurkat T lymphocytes; however, there was no confirmation of whether the cells died through apoptosis, or through necrosis, where the cell bursts. Infection in immunocompromised individuals can cause neurological damage and infection during pregnancy can lead to fetal death. The target of rapamycin complex 1 (TORC1) is a master regulator of cell growth as well as a negative regulator of longevity. A recent study in yeast showed that the vacuolar TORC1 stimulates protein synthesis and cell growth while the endosomal TORC1 inhibits autophagy, a pro-longevity process. Since late endosome frequently exchanges membranes with vacuoles, the spatial dissection of TORC1 raises an interesting question as to how to inhibit the endosomal TORC1 and boosts the vacuolar TORC1 to extend the health span. During an mRNA’s lifecycle, RNA-binding proteins (RBP) are added and removed in order to facilitate export from the nucleus to the cytoplasm through the nuclear pore complex (NPC). During the RNA export process, a class of proteins termed Dead-box proteins (Dbps) are strategically located at the cytoplasmic face of the NPC to remove mRNPs from the mRNA-protein complex, providing one way directionality for export in the process termed mRNA-protein (mRNP) remodeling. Transcriptional Silencing Of Cdk18 And Its Role In Lung Carcinogenesis Of Brg1-Mutant Lung Cancers, Loryn M. Phillips

BRG1 is mutated in 10% of lung cancers, making this mutation clinically relevant. The downstream effects of BRG1 included significantly affecting the cell cycle control and chromosomal replication pathway. CDK18, a cyclin-dependent kinase, was determined to be the gene with significantly decreased expression (p < 0.0001) in the cell cycle control and chromosomal replication pathway. CDK18 is active during the S-phase of the cell cycle and required for genomic stability. Studies were conducted to determine the epigenetic mechanisms that contributed to the repression of CDK18. Histone methylation contributes to the repression of CDK18, and histone deacetylation globally regulates expression of CDK18, while cytosine methylation was found to not contribute to CDK18 expression. Studies were performed to elucidate CDK18 functional role lung carcinogenesis and the genome-wide impact of CDK18 repression. Repression of CDK18 affects cell migration, proliferation, cell-cell signaling, and anchorage-independent growth that contribute to cancer initiation and progression.

Aging is associated with metabolic rewiring. Most lifespan-extending longevity interventions target metabolic pathways. Genetic or pharmacological metabolic approaches that mediate longer lifespan and healthspan, are often linked to detrimental effects on human health. Here, I show that supplementation of botanical extracts from Artemisia scoparia (SCO), dramatically increases animal lifespan. Mechanistically, this lifespan extension is dependent on AMPK-DAF-16 signaling via inhibition of the insulin pathway. In SCO-fed worms, longer lifespan was accompanied by higher fat storage and improved resiliency to age-related physiological stress. Notably, DAF-16, the master regulator of longevity, activated the fatty acid desaturation processes, which promoted the accumulation of …

Currently, there are no approved treatment options for ZIKV infection. Herein, we describe the development of a bacterial ferritin-based nanoparticle vaccine candidate for ZIKV. Hepatocellular carcinoma (HCC) is a leading cause of cancer deaths globally and is a growing clinical problem with poor survival outcomes beyond early-stage disease. Surveillance for HCC has primarily relied on ultrasound and serum α-fetoprotein (AFP), but combined they only have a sensitivity of 63% for early-stage HCC tumors, suggesting a need for improved diagnostic strategies. Alterations to N-glycan expression are relevant to the progression of cancer, and there a multitude of N-glycan-based cancer biomarkers that have been identified with sensitivity for various cancer types including HCC. The treatment of glioblastoma (GBM) using low intensity electric fields (∼1 V cm-1) is being investigated using multiple implanted bioelectrodes, which was termed intratumoral modulation therapy (IMT). Previous IMT studies theoretically optimized treatment parameters to maximize coverage with rotating fields, which required experimental investigation.
