Mango (Mangifera indicia L.) is a popular tropical fruit which has a great value in both domestic and international markets. Mango is a climacteric fruit which ethylene production increases dramatically, thus leading to a quick ripening process and a short postharvest life. Polyamines (PAs) have been reported to be involved in fruit ripening. Changes in endogenous free polyamines accumulation were measured in two mango cultivars; ‘Nam Dok Mai No.4’ and ‘Nam Dok Mai Si Tong’. Mangoes were allowed to ripen at 25±1 ºC after harvest. Acute hepatopancreatic necrosis disease (AHPND) caused by the bacterium Vibrio parahaemolyticus carrying toxin produced plasmid (VPAHPND), has led to severe mortalities in farmed penaeid shrimp throughout Asia. Our studies demonstrated that a chronic non-lethal heat shock (chronic-NLHS) could enhance resistance of the Pacific white shrimp Penaeus vannamei to VPAHPND infection. Shrimp exposed to chronic-NLHS (28 °C to 38 °C, 5 min for 7 days) had higher survival rate (>50%) than that of the control non-heated shrimp (20%) when they were challenged with VPAHPND. L-Phenylalanine (L-Phe) is one of the most important amino acids in food and pharmaceutical industries. It is widely used as a nutritional supplement and a precursor for the synthesis of food additives. The market of L-Phe has been stimulated by increasing demand for the low-calorie sweetener, aspartame. In Escherichia coli, the key enzyme which catalyzes the first committed step of aromatic amino acid biosynthesis pathway is 3-deoxy-D-arabino-heptulosonate-7- phosphate synthase (DAHP-synthase). In the RNA interference (RNAi) process, small RNAs called microRNAs (miRNAs) modulate gene expression post-transcriptionally by acting as guides to direct Argonaute (Ago)-containing RNA-induced silencing complexes (RISCs) to targeted mRNAs. The Penaeus monodon miR-750 was previously reported as the highly upregulated miRNA during white spot syndrome virus (WSSV) infection. In this study, the computational program and 2D gel electrophoresis technique were used to identify miR-750 target genes. Produced by levansucrase that uses sucrose as a substrate, levan and levan oligosaccharides (GFn) have potential applications in food and pharmaceutical industries such as prebiotics, anti-obesity and anti-tumor agents. Previous study reported that N251A and N251Y mutants of levansucrase from Bacillus licheniformis RN-01 could effectively produce short-chain oligosaccharides upto GF3, instead of long-chain levan synthesis. This study hypothesized that these mutations probably reduced GF3 binding affinity in the active site of levansucrase that contains fructosyl-Asp93 intermediate and caused GF3 to be in an unfavorable orientation for transfructosylation; therefore, levansucrase could not effectively extend GF3 to GF4. The previous study of OsCam1-1 showed that the transgenic KDML105 rice over-expressing OsCam1-1 was more tolerant to salt stress than the wild type, so to investigate the role of calmodulin encoded by OsCam1-1 in the salt-responsive mechanism, the transcriptome approach using RNA-seq technique was applied. Transcriptome profile of the transgenic KDML105 rice over-expressing OsCam1-1 under 4 hr of salt stresss (150 mM NaCl) revealed that several cell processes were affected; signaling, hormone-mediated regulation, transcription, secondary metabolism, stress responses, lipid metabolism, glycolysis, TCA cycle, glyoxylate cycle, photosynthesis, and carbohydrate metabolism. The IKK-NF-κB signaling cascade is one of the crucial responsive mechanisms in inflammatory and immune responses. The key kinase proteins called inhibitor of kappa B kinases (IKKs) serve as the core elements involved in cascade activation. Here, the complete open reading frames of IKK homologs including PmIKKβ, PmIKKε1 and PmIKKε2, from the black tiger shrimp Penaeus monodon were identified and characterized for their functions in shrimp antiviral responses. Human immunodeficiency virus type-1 (HIV-1) can cause acquired immunodeficiency syndrome (AIDS), a world serious public health problem. HIV-1 protease (HIV-1 PR) and reverse transcriptase (HIV-1 RT) are necessary target enzymes which have been used for antiretroviral drugs development. Herein, Lignosus rhinocerus (LR), Auricularia polytricha (AP) and Astraeus spp. (AH) crude extracts were screened for inhibitory activities on both enzymes. Crude hexane extract of AP (APH) exhibited significant inhibition on both HIV-1 PR and RT activities not only in an in vitro non-cell based but also cell based assays. วิทยานิพนธ์นี้เป็นการนำเสนองานวิจัย การพัฒนาวิธีการใหม่เพื่อควบคุมและเฝ้าระวังโรคติดเชื้อ โดยประกอบด้วยเนื้อหาสำคัญสองส่วน ได้แก่ (i) การพัฒนาแบบจำลองเซลล์ลำไส้ของมนุษย์ที่เพาะเลี้ยงบนกระดาษที่มีการดัดแปรพื้นผิว เพื่อใช้ประโยชน์ในการศึกษากลไกการบุกรุกของเชื้อจุลชีพก่อโรคในระบบทางเดินอาหาร และ (ii) การพัฒนาต้นแบบไบโอเซนเซอร์แบบไร้สายเพื่อการตรวจติดตามการเจริญของเชื้อจุลชีพก่อโรค โดยในส่วนแรก เซลล์ลำไส้ของมนุษย์ HT-29 จำนวน 200,000 เซลล์ ถูกเพาะเลี้ยงในบริเวณชอบน้ำของกระดาษกรอง Whatman No.1 โดยกระดาษถูกดัดแปรพื้นผิวด้วยสารแมทริกซ์นอกเซลล์ (Extracellular matrix: ECM) ชนิดต่าง ๆ ได้แก่ Matrigel, Collagen-1 และ Laminin พื้นที่เพาะเลี้ยงเซลล์มีขนาด 12.56 ตารางมิลลิเมตร ภายใต้สภาวะที่เหมาะสม เซลล์ HT-29 ที่ถูกเพาะเลี้ยงบนกระดาษ มีชีวิตรอดได้ถึง 28 วัน และแสดงลักษณะของเซลล์ที่เจริญเต็มที่ โดยมีการแสดงออกของวิลไล (villi) ที่ผิวเซลล์ด้านบน (apical) และมีการแสดงออกของโปรตีน ZO-1 ซึ่งแสดงถึงเซลล์มีการสร้างไทต์จังก์ชันเกิดขึ้น จากการใช้สารเชื่อมพันธะสำหรับเชื่อมพันธะ ECM แต่ละชนิด พบว่าเมื่อใช้สารเจนิพินในการเชื่อมพันธะ เซลล์มีชีวิตรอดทั้งหมด (100 -112.8%) ในขณะที่มีเพียง 1.1 - 67.3 % ของเซลล์รอดชีวิต หากใช้สารกลูตารัลดีไฮด์ในการเชื่อมพันธะ แต่การใช้สารเชื่อมพันธะทั้งสองชนิด ทำให้เซลล์ไม่สามารถแบ่งตัวมีรูปร่างแบบสามมิติได้ เมื่อสังเกตด้วย SEM เซลล์มีลักษณะแบน การดัดแปรพื้นผิวกระดาษด้วยสาร ECM ชนิด Matrigel ส่งผลให้เซลล์มีชีวิตรอดมากที่สุดและสาร ECM ชนิด Laminin ทำให้เซลล์เกิดการแปรสภาพ (cell differentiation) ได้ดีที่สุด งานวิจัยส่วนที่ 2 เป็นการพัฒนาไบโอเซนเซอร์ไร้สายสำหรับติดตามการเจริญของเชื้อจุลชีพก่อโรค ได้แก่ เชื้อ Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Streptococcus gordonii และ Candida albicans พบว่าเชื้อดังกล่าวสามารถสร้างไบโอฟิล์มบนผิวของขั้วไฟฟ้าคาร์บอนอสัณฐาน (glassy carbon electrode) และสามารถให้ค่าศักย์ไฟฟ้าที่เป็นลบได้ …

Grus antigone sharpii จัดเป็นนกกระเรียนที่เคยสูญพันธุ์ไปจากธรรมชาติของประเทศไทย การเพาะเลี้ยงและขยายพันธุ์นกกระเรียนในกรงเลี้ยงเป็นขั้นตอนสำคัญที่สามารถเพิ่มจำนวนประชากรเพื่อนำไปปล่อยคืนสู่ธรรมชาติในอนาคตได้ โดยสิ่งสำคัญที่ต้องคำนึงถึงคือความหลากหลายทางพันธุกรรม โดยเฉพาะอย่างยิ่งในกลุ่มของพ่อแม่พันธุ์ตั้งต้น ในการศึกษาครั้งนี้มีวัตถุประสงค์เพื่อคัดกรองเครื่องหมายไมโครแซทเทลไลท์จำนวน 28 ตำแหน่ง ซึ่งเป็นไพรเมอร์ที่เคยใช้ศึกษาในนกกระเรียน G. americana และนกกระเรียน Anthropoides paradisea เพื่อใช้วิเคราะห์ความหลากหลายทางพันธุกรรมของนกกระเรียนพันธุ์ไทยจากสวนสัตว์เปิดเขาเขียว (KKOZ, n = 11) และสถานีวิจัยเพาะเลี้ยงนกน้ำบางพระ (BB, n = 17) จังหวัดชลบุรี ผลการศึกษาพบว่าไพรเมอร์ทั้ง 28 คู่สามารถเพิ่มปริมาณไมโครแซทเทลไลท์ดีเอ็นเอของนกกระเรียนพันธุ์ไทยได้ แต่มีเพียง 22 คู่ที่แสดงความเป็น polymorphism และมีตำแหน่งไมโครแซทเทลไลท์ที่ปรากฏค่า linkage disequilibrium 8 ตำแหน่ง การเบี่ยงเบนออกจากสมดุล Hardy-Weinberg พบที่โลคัส Gram8 และ Gpa38 ซึ่งเป็นผลมาจาก null allele และการเพิ่มขึ้นของอัลลีลแบบ heterozygous ตามลำดับ จากการวิเคราะห์ค่าความหลากหลายทางพันธุกรรมพบว่า ค่าเฉลี่ยของ expected และ observed heterozygosity มีค่าเท่ากับ 0.63 และ 0.69 ค่า inbreeding coefficient มีค่าเท่ากับ -0.100 ซึ่งแสดงว่านกกระเรียนมีความหลากหลายทางพันธุกรรมค่อนข้างสูง และเกิด inbreeding น้อย นอกจากนี้ยังพบว่านกกระเรียนจาก KKOZ และ BB มีความแตกต่างทางพันธุกรรมต่ำ (FST = 0.035) และไม่มีการแยกออกเป็นกลุ่มประชากรย่อย (K = 1) จากผลการศึกษาครั้งนี้บ่งบอกได้ว่านกกระเรียนพันธุ์ไทยจาก 2 พื้นที่การศึกษาน่าจะเหมาะสมที่จะใช้เป็นพ่อแม่พันธุ์เพื่อเพิ่มจำนวนประชากรในกรงเลี้ยงให้มากขึ้น และสามารถนำไปปล่อยคืนสู่พื้นที่ชุ่มน้ำตามธรรมชาติในอนาคตได้

Bacterial isolate HM7 was isolated from Dynastes hercules larvae excrement. It was identified as Bacillus sp. by 16S rRNA gene analysis. When the bacteria were cultured for β-mannanase production in medium containing konjac flour it had the highest specific activity of 138 U/mg at 36 h, and when cultured in medium containing coconut meal it had the highest specific of 114 U/mg at 48 h. The 1089 base pairs long of a β-mannanase encoding gene (Man26HM7) was successfully cloned and expressed in Escherichia coli BL21 (DE3), using pET21-b expression system. หลังคาเขียวเพิ่มพื้นที่สีเขียวในเมืองและอาจรองรับสังคมนกโดยให้ที่พักพิง อาหารและแหล่งน้ำ งานวิจัยนี้ศึกษาความหลากชนิดของนกและรูปแบบการใช้ประโยชน์บนหลังคาเขียว 7 แห่งในกรุงเทพมหานครที่มีขนาดพื้นที่และสัดส่วนพื้นที่สีเขียวแตกต่างกัน โดยสำรวจนกบนหลังคาเขียวทุกเดือนระหว่างเมษายน 2559-พฤษภาคม 2560 พบนกรวม 26 ชนิดและนกส่วนใหญ่เป็นนกประจำถิ่น พบนก 7-18 ชนิดบนหลังคาเขียว นกกินปลีอกเหลือง Cinnyris jugularis เป็นนกที่พบบนทุกหลังคาเขียว นกกระจอกบ้าน Passer montanus เป็นนกที่มีความชุกชุมสูงสุดโดยมักปรากฏเป็นฝูงใหญ่ พบนกอพยพบางชนิด เช่น นกอัญชันป่าขาเทา Rallina eurizonoides นกแซงแซวสีเทา Edolius leucophaeus นกขมิ้นท้ายทอยดำ Oriolus chinensis นกกระเบื้องผา Monticola solitarius และนกอีเสือสีน้ำตาล Lanius cristatus การหาอาหารและการเกาะพักเป็นรูปแบบการใช้ประโยชน์ที่มีความถี่สูงสุด (ร้อยละ 47 และ 31 ตามลำดับ) นกใช้ไม้ยืนต้นสำหรับการเกาะพักและการเคลื่อนที่ ลักษณะทางกายภาพและชีวภาพบางประการส่งผลต่อสังคมนกที่ใช้ประโยชน์บนหลังคาเขียว ดังนั้นหลังคาเขียวจึงมีศักยภาพในการอนุรักษ์ความหลากหลายทางชีวภาพในระบบนิเวศเมือง

The alternansucrase (ALT, EC 2.4.1.140) catalyses transferring of glucose from sucrose to produce a polymer with α-1,6 and α- 1,3 linkages, so-called "alternan". The ALT-encoding gene from Leuconostoc citreum ABK-1 (WTalt) was successfully cloned and expressed in Escherichia coli BL21 (DE3). The two versions of truncated SH3-like domains, ∆3SHALT and ∆7SHALT, were successfully constructed. The WTALT, ∆3SHALT and ∆7SHALT possessed optimum temperature of 40 °C, while optimum pH of WTALT was at 5.0 but ∆3SHALT's and ∆7SHALT's were shifted to pH 4.0. ในปัจจุบัน ออทิซึมสเปกตรัม (Autism spectrum disorder, ASD) ยังคงเป็นโรคที่ยังคงไม่ทราบกลไกระดับโมเลกุล แน่ชัด ยีนและโปรตีนที่เคยมีการรายงานการแสดงออกผิดปกติไปในผู้ป่วยออทิซึมสเปกตรัมมีความสามารถในการทำซ้ำ (Reproducibility) ต่ำหรืออาจไม่มีเลย ซึ่งน่าจะมาจากความหลากหลายของอาการแสดงของผู้ป่วย จากการศึกษาล่าสุด แสดงให้เห็นว่าการจัดกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกสามารถช่วยในการระบุยีนและกลไกที่ผิดปกติไปในกลุ่มย่อยของผู้ป่วย อย่างไรก็ตาม การแบ่งกลุ่มผู้ป่วยเป็นกลุ่มย่อยตามความรุนแรงของลักษณะทางคลินิกใน ออทิซึมสเปกตรัมยังไม่มีการใช้กันอย่างแพร่หลายในการค้นหาโปรตีน Biomarker ที่เกี่ยวข้องกับออทิซึมสเปกตรัม ในการศึกษาครั้งนี้ ผู้วิจัยทำการวิเคราะห์โดยการแบ่งกลุ่มผู้ป่วยจำนวน 85 รายออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R และทำการวิเคราะห์รูปแบบการแสดงออกของยีนในออทิซึมสเปกตรัมในแต่ละกลุ่มย่อยเปรียบเทียบกับกลุ่มควบคุมคนปกติที่อายุใกล้เคียงกันและเพศเดียวกันกับผู้ป่วย (Sex/age-matched control) ซึ่งสามารถระบุยีนที่มีการแสดงออกผิดปกติ (Differentially expressed genes, DEGs) ในออทิซึมสเปกตรัม นอกจากนี้ ผู้วิจัยได้ทำการวิเคราะห์รูปแบบการแสดงออกของโปรตีโอมจากเซลล์ไลน์เม็ดเลือดขาว (Lymphoblastoid cell lines,LCLs) ด้วยเทคนิค Two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) ร่วมกับ Nano-liquid chromatography-tandem mass spectrometry (Nano-LC-MS/MS) รายชื่อของโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญที่ได้จากการวิเคราะห์ถูกนำมาเปรียบเทียบกับ DEGs และผลการวิเคราะห์ Meta-analysis ของโปรตีนและโปรตีโอมในการศึกษาก่อนหน้า หน้าที่ทางพยาธิสภาพที่มีความเกี่ยวข้องกับโปรตีนถูกวิเคราะห์โดยโปรแกรม Ingenuity Pathway Analysis (IPA) และได้ทำการคัดเลือกโปรตีนที่น่าสนใจมาทำการตรวจสอบโดยใช้ Western blot analysis จากการศึกษาครั้งนี้พบว่าผลการวิเคราะห์การแบ่งกลุ่มผู้ป่วยออกเป็นกลุ่มย่อยทางคลินิกตามคะแนน ADI-R สามารถแบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อย ซึ่งพบว่าหนึ่งในกลุ่มนั้น เป็นกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง จากผลวิเคราะห์รูปแบบ ทรานสคริปโตมของเซลล์ไลน์เม็ดเลือดขาว พบว่ามีจุดโปรตีนจำนวน 82 จุดที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษารุนแรง และได้ทำการคัดเลือก 18 จุดโปรตีนจากทั้งหมดมาวิเคราะห์ชนิดของโปรตีนโดยเทคนิค Nano-LC-MS/MS ซึ่งพบว่ามีความเกี่ยวข้องกับการทำงานของระบบประสาทและกระบวนการอักเสบ จากการศึกษานี้พบว่าจากโปรตีนทั้งหมด การแสดงออกของ Isocitrate dehydrogenase (IDH2) และ Diazepam binding inhibitor (DBI) ลดลงในออทิซึมสเปกตรัม ซึ่งโปรตีนเหล่านี้ยังพบการแสดงออกผิดปกติในการวิเคราะห์ Meta-analysis ของทรานสคริปโตมและโปรตีโอมร่วมด้วย การแบ่งกลุ่มออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยทางคลินิกตามด้วยการวิเคราะห์ทรานสคริปโตมและโปรตีโอมแบบบูรณาการร่วมกัน ทำให้การศึกษานี้สามารถระบุโปรตีนที่มีการแสดงออกผิดปกติไปอย่างมีนัยสำคัญในกลุ่มย่อยของออทิซึมสเปกตรัมที่มีความบกพร่องทางภาษาอย่างรุนแรง ซึ่งโปรตีนเหล่านี้อาจทำหน้าที่เป็นตัวแทนในการช่วยระบุกลไกทางโมเลกุลของออทิซึมสเปกตรัมในอนาคต ตลอดจน อาจพัฒนาไปเป็น …

ออทิซึมสเปกตรัมเป็นความผิดปกติที่เกี่ยวกับการพัฒนาของระบบประสาทและสมองที่มีความชุก 1 ใน 68 คนของเด็กในสหรัฐอเมริกา โดยจะมีระดับความรุนแรงและอาการแสดงทางคลินิกแตกต่างกันออกไปในแต่ละบุคคล ออทิซึมสเปกตรัมจัดเป็นโรคที่ไม่ทราบสาเหตุการเกิดที่แน่ชัด แต่เป็นที่เชื่อกันว่าสาเหตุของการเกิดโรคนั้นอาศัยปัจจัยร่วมหลายปัจจัย ได้แก่ ทั้งปัจจัยทางพันธุกรรมและปัจจัยทางสิ่งแวดล้อม การวินิจฉัยออทิซึมสเปกตรัมในปัจจุบัน ใช้วิธีอิงจากการสังเกตพฤติกรรมผู้ป่วย วินิจฉัยโรคและความรุนแรงตามลักษณะอาการทางคลินิกเท่านั้น ยังไม่มีสารบ่งชี้ทางห้องปฏิบัติการทางการแพทย์ที่สามารถใช้ในการวินิจฉัย หรือติดตามความรุนแรงของโรคอย่างจำเพาะ โดยผลการศึกษาก่อนหน้านี้ แสดงให้เห็นว่าผู้ป่วยโรคออทิซึมสเปกตรัมมีความผิดปกติในระดับอณูชีวโมเลกุลมากมายทั้งในสมองและเลือด ซึ่งมีหลายประการที่สอดคล้องกับพยาธิสภาพระดับชีวโมเลกุลที่เกิดขึ้นในสมอง อย่างไรก็ตาม ยังไม่มียีนหรือโปรตีนชนิดใดที่สามารถใช้ในการอธิบายโรคออทิซึมสเปกตรัมได้ทั้งหมดในปัจจุบัน แสดงให้เห็นว่าอาจมีกลไกอื่น ๆ ที่สามารถทำให้เกิดพยาธิสภาพได้ ยิ่งไปกว่านั้น แม้โรคออทิซึมสเปกตรัมจะเป็นโรคที่มีอัตราการถ่ายทอดทางพันธุกรรมสูง แต่มีฝาแฝดร่วมไข่จำนวนไม่น้อยที่คนหนึ่งเป็นโรคแต่อีกคนไม่เป็น หรือมีระดับความรุนแรงของโรคไม่เท่ากัน เนื่องจากฝาแฝดร่วมไข่มีลำดับเบสของดีเอ็นเอที่เหมือนกันทุกประการเมื่อแรกเกิด แสดงให้เห็นว่า กลไกเหนือพันธุกรรมหรือเอพิเจเนติกส์อาจเป็นสาเหตุหรือมีความเกี่ยวข้องกับการเกิดโรคออทิซึมสเปกตรัม รีโทรทรานสโพซอนไลน์วัน (retrotransposon LINE-1) เป็นส่วนของจีโนมที่สามารถเคลื่อนที่ได้ และมีอยู่ถึงประมาณร้อยละ 20 และในจีโนมมนุษย์ อย่างไรก็ตามบทบาทของ LINE-1 ในผู้ที่เป็นออทิซึมสเปกตรัมยังคงไม่ทราบแน่ชัด แต่มีรายงานว่าพบการควบคุมที่ผิดปกติของ LINE-1 ในสมองของหนูที่ขาด MECP2 และมีพฤติกรรมคล้ายออทิซึมสเปกตรัม ดังนั้นคณะผู้วิจัยจึงทำการวิเคราะห์ระดับเมธิเลชันของ LINE-1 เพื่อตรวจสอบการเปลี่ยนแปลงระดับเมธิเลชันของ LINE-1 ในเซลล์ของผู้ที่เป็นออทิซึมสเปกตรัม และผลต่อการแสดงออกของยีน รวมไปถึงบทบาทหน้าที่ทางชีวภาพของยีนนั้น ๆ โดยทำการศึกษาในเซลล์ไลน์เม็ดเลือดขาว (lymphoblastoid cell lines; LCLs) ของผู้ที่เป็นออทิซึมสเปกตรัมและคนปกติ นอกจากนี้ยังได้ทำการวิเคราะห์ทางชีวสารสนเทศศาสตร์ร่วมด้วย โดยได้ทำการวิเคราะห์ร่วมระหว่างรายชื่อยีนที่มี LINE-1 แทรกตัวอยู่และมีการแสดงออกที่ผิดปกติในผู้ที่เป็นโรคออทิซึมสเปกตรัม และได้คัดเลือกรายชื่อยีนมาทำการศึกษาบาทหน้าที่ทางชีวภาพ และความสัมพันธ์ของเครือข่ายการควบคุมยีนของยีนเหล่านั้นด้วยโปรแกรม Ingenuity Pathway Analysis (IPA) ซึ่งพบว่ายีนเหล่านั้นอาจจะมีความสัมพันธ์กับโรคทางระบบประสาทและสมอง ในส่วนของการวิเคราะห์ระดับเมธิเลชันของ LINE-1 ในดีเอ็นเอซึ่งสกัดมาจาก LCLs ของผู้ที่เป็นโรคออทิซึมสเปกตรัมและคนปกติ และดำเนินการโดยใช้เทคนิคการวิเคราะห์ Combined Bisulfite Restriction Analysis (COBRA) ซึ่งพบว่าใน LCLs ของกลุ่มตัวอย่างที่เป็นออทิซึมสเปกตรัมมีระดับเมธิเลชันของ LINE-1 ที่เปลี่ยนแปลงไปเมื่อเทียบกับคนปกติ และเมื่อทำการวิเคราะห์ระดับเมธิเลชันของ LINE-1 ในแต่ละกลุ่มของ LCLs ที่เป็นออทิซึมสเปกตรัม ซึ่งถูกจัดแบ่งกลุ่มตามหลักเกณฑ์ของ ADI-R เนื่องจากออทิซึมสเปกตรัมเป็นโรคที่มีลักษณะอาการและความรุนแรงที่หลากหลายมาก คณะผู้วิจัยพบว่าแต่ละกลุ่มมีรูปแบบการเปลี่ยนแปลงของระดับเมธิเลชันของ LINE-1 ที่จำเพาะ และในบางกลุ่มไม่พบการเปลี่ยนแปลงของระดับเมธิเลชันของ LINE-1 นอกจากนี้ยังพบว่าการเมธิเลชันของ …

โรคออทิซึมสเปกตรัมเป็นกลุ่มโรคของความผิดปกติของพัฒนาการทางระบบประสาทที่มีสาเหตุมาจากหลายปัจจัย การศึกษาก่อนหน้าพบว่าในเลือดของผู้ป่วยออทิซึมสเปกตรัมจะรูปแบบการแสดงออกของยีนแตกต่างจากคนปกติ และยังพบว่ามีการเปลี่ยนแปลงของปฏิกิริยาเมธิเลชันบนดีเอ็นเอในเลือดและสมองของผู้ป่วยออทิซึมสเปกตรัมซึ่งปฏิกิริยาดีเอ็นเอเมธิเลชันเป็นกลไกที่ควบคุมการแสดงออกของยีน และยังเป็นกลไกที่ตอบสนองต่อสิ่งแวดล้อมที่เปลี่ยนแปลงไป ดีเอ็นเอเคลื่อนที่ Alu หรือ Alu elements เป็นดีเอ็นเอส่วนที่มีจำนวนซ้ำ ๆ ในจีโนม และสามารถเหนี่ยวนำการแสดงออกของยีนได้ผ่านกลไกเมธิเลชันของดีเอ็นเอ ซึ่งเป็นกลไกหนึ่งที่ควบคุมอัตราการเพิ่มจำนวนตนเองในจีโนม โดย Alu elements มีสัดส่วนการเกิดปฏิกิริยาเมธิเลชันสูงถึงร้อยละ 23 ของปฏิกิริยาเมธิเลชันที่เกิดขึ้นทั้งหมดบนจีโนมของมนุษย์ อย่างไรก็ตามยังไม่เคยมีผู้ทำการศึกษาว่ากลุ่มยีนที่แสดงออกผิดปกติในผู้ป่วยโรคออทิซึมสเปกตรัมนั้นมีความเกี่ยวข้องกับ Alu elements หรือไม่ ในงานวิจัยนี้ผู้ทำวิจัยได้ทำการค้นพบว่ากลุ่มยีนที่มี Alu elements แทรกตัวอยู่มีระดับแสดงออกของยีนที่ผิดปกติในเลือด หรือเซลล์ไลน์ที่พัฒนามาจากเลือดของผู้ป่วยออทิซึมสเปกตรัมผ่านการวิเคราะห์แบบ meta-analysis และโดยผู้ทำวิจัยพบว่ามียีนจำนวน 423 ยีนที่มีการแสดงออกผิดปกติซึ่งเมื่อนำมาวิเคราะห์หาบทบาทหน้าที่ทางชีวภาพของกลุ่มยีนเหล่านี้ด้วยเครื่องมือชีวสารสนเทศ พบว่ามีความเกี่ยวข้องกับกระบวนการพัฒนาการของระบบประสาท และมีความเกี่ยวกับโรคที่มีความผิดปกติคล้ายกับโรคออทิซึมสเปกตรัม และนอกจากนี้ผลการศึกษาในเซลล์โมเดลจากผู้ป่วยออทิซึมสเปกตรัมที่ใช้ในการศึกษาครั้งนี้ พบว่ามีการเปลี่ยนแปลงของระดับและรูปแบบการเกิดปฏิกิริยาเมธิเลชันของ Alu elements ในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่ผ่านการจัดจำแนกกลุ่มผู้ป่วยด้วยข้อมูลแบบสอบถามทางพฤติกรรมสำหรับการวินิจฉัยผู้ป่วยออทิซึมสเปกตรัม สอดคล้องกับผลการวิเคราะห์รูปแบบการแสดงออกของยีนในผู้ป่วยกลุ่มที่มีระดับเมธิเลชันผิดปกติจะมีรูปแบบการแสดงออกของยีนที่มี Alu elements แทรกตัวอยู่ผิดปกติด้วย ซึ่งข้อมูลการค้นพบใหม่นี้ได้แสดงให้เห็นความถึงสัมพันธ์ระหว่างระดับปฏิกิริยาเมธิเลชันของ Alu elements กับความผิดปกติของระดับการแสดงออกของยีนในผู้ป่วยออทิซึมสเปกตรัมเมื่อมีการจัดจำแนกผู้ป่วยตามลักษณะอาการพฤติกรรมทางคลินิก ซึ่งอาจใช้อธิบายความผิดปกติทางชีวโมเลกุลที่จำเพาะในผู้ป่วยแต่ละกลุ่มได้ และอาจพัฒนาเป็นตัวบ่งชี้ทางชีวภาพในเลือดของโรคออทิซึมสเปกตรัมในอนาคตได้

กระบวนการสร้างนิวไรท์ เป็นกระบวนการที่สำคัญในการเจริญพัฒนา การซ่อมแซมส่วนที่เสียหาย รวมถึงการสื่อสารระหว่างเซลล์ของเซลล์ประสาท ซึ่งหากกระบวนการการสร้างนิวไรท์เกิดความผิดปกติสามารถก่อให้เกิดโรคความผิดปกติของระบบประสาท เช่น โรคอัลไซเมอร์ โรคพาร์กินสัน และโรคฮันติงตัน เป็นต้น ในการศึกษาวิจัยนี้ศึกษาฤทธิ์การต้านสารอนุมูลอิสระ และการกระตุ้นการสร้างนิวไรท์จากสารสกัดเมล็ดของหมามุ่ยไทย และหมามุ่ยอินเดียในเซลล์ประสาทเพาะเลี้ยงชนิด Neuro2a การศึกษาคุณสมบัติในการต้านสารอนุมูลอิสระโดยวิธี DPPH และ ABTS assays นอกจากนั้นทำการศึกษาอัตราการรอดชีวิตของเซลล์ประสาทโดยวิธี MTT พร้อมกับศึกษาการแสดงออกของยีนที่เกี่ยวข้องกับการกระตุ้นการงอกของนิวไรท์ ประกอบด้วย Ten-4 และ GAP-43 โดยวิธี Real-time PCR ซึ่งจากการทดสอบพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียด้วยตัวทำละลายเอทานอลมีความสามารถในการต้านสารอนุมูลอิสระ และพบว่าสารสกัดจากเมล็ดหมามุ่ยไทย และเมล็ดหมามุ่ยอินเดียไม่เป็นพิษต่อเซลล์ประสาท จากนั้นทำการศึกษาฤทธิ์ และกลไกของสารสกัดจากสมุนไพรต่อการกระตุ้นการงอกของนิวไรท์ โดยพบว่าสารสกัดจากเมล็ดหมามุ่ยไทยด้วยตัวทำละลายเอทานอล สามารถกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 และ GAP-43 ในขณะที่สารสกัดจากเมล็ดของหมามุ่ยอินเดียด้วยตัวทำละลายเฮกเซนกระตุ้นการสร้างนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน Ten-4 อีกทั้งศึกษาผลการยับยั้งการแสดงออกของยีน Ten-4 ต่อการกระตุ้นการสร้างนิวไรท์ของสารสกัดสมุนไพร ซึ่งพบว่ามีการสร้างนิวไรท์ และความยาวของนิวไรท์ลดลง จากการศึกษาสามารถบ่งชี้ได้ว่าสารสกัดจากเมล็ดหมามุ่ยไทย และหมามุ่ยอินเดียสามารถปกป้องเซลล์ประสาทจากสารอนุมูลอิสระได้ และสามารถกระตุ้นการงอกของนิวไรท์โดยผ่านการเพิ่มการแสดงออกของยีน GAP-43 และ Ten-4 ซึ่งเป็นอีกทางเลือกหนึ่งในการพัฒนายาการกระตุ้นการเจริญพัฒนาของเซลล์ประสาทสำหรับป้องกัน หรือรักษาโรคความผิดปกติของระบบประสาท
