Vibrio parahaemolyticus causing Acute hepatopancreatic necrosis disease (VPAHPND) has been a major cause of serious shrimp aquaculture economic losses. Moreover, many isolates have exhibited the resistance of various antibiotic drugs. Most recently, bacteriophage or phage, the killer of bacteria, has been gaining interest as an alternative tool to prevent and control pathogens. Dextransucrase is an enzyme in the glycoside hydrolase 70 family that catalyze the hydrolysis of sucrose to fructose and glucose, the glucose residues are transferred to synthesize dextran via transglycosylation reaction. Dextransucrase from Leuconostoc citreum ABK-1 (Lcdextransucrase ), which has 99.5% homology with dextransucrase in the NCBI database, was cloned, expressed, and purified. The size of the Lcdextransucrase was predicted by ExPASy and the result showed that its size was 168 kDa with a pI of 5.15. Protein expression was confirmed by western blot analysis. Epidermal growth factor receptor (EGFR) has been identified as a promising target in the treatment of non-small cell lung cancer (NSCLC). Mutations, including L858R/T790M double and L858R/T790M/C797S triple mutations, in the TK domain of EGFR have been linked to the severity of several cancers, especially NSCLC. Alzheimer’s disease (AD) is a common and devastating disease characterized by pathological aggregations of beta-amyloid (Aβ) plaques extracellularly, and Tau tangles intracellularly. While our understandings of the aetiologies of AD have greatly expanded over the decades, there is no drug available to stop disease progression. This study involves two parts. First, we demonstrate the potential of P. edulis pericarp (PP) extract in protecting against Aβ-mediated neurotoxicity in mammalian cells and Caenorhabditis elegans models of AD. Molecular dynamics studies have been providing a great number of atomistic details of biological systems to understand enzymatic activity and underlying reaction mechanisms. Herein, molecular dynamic (MD) simulations were employed to study the binding mechanisms of organophosphate pesticides towards two metallohydrolases, i.e., glycerophosphodiesterase (GpdQ) and methyl parathion hydrolase (MPH). OPH are the class of enzymes which breaks down harmful organophosphates, including the pesticides and warfare agents into less harmful byproducts. In this study, we investigated the cytotoxicity of 6 anthraquinones (ventilanone K, emodin, chrysophanol, aurantio-obtusin, 1-O-methyl-2-methyoxychrysophanol, and questin) toward A549 lung cancer cells and emodin (ED) exerted the most potent cytotoxicity with IC50 value of 34.27 ± 1.27 µM. The anti-cancer activity of ED has been well reported but its usage in practical applications has been restricted due to its poor solubility. โรคออทิซึมสเปกตรัมเป็นความผิดปกติเกี่ยวกับพัฒนาการในช่วงเริ่มแรกของระบบประสาทและสมองซึ่งมีอาการทางคลินิกที่หลากหลายมาก ปัจจุบันยังไม่มีสารบ่งชี้เพื่อช่วยในการวินิจฉัยและแบ่งผู้ป่วยเป็นกลุ่มย่อย แต่การวินิจฉัยที่รวดเร็วเป็นสิ่งสำคัญที่จะช่วยให้ผู้ป่วยออทิซึมสเปกตรัมสามารถได้รับการเรียนรู้และพัฒนาพฤติกรรมที่จำเป็นในการดำรงชีวิตได้ การวินิจฉัยโรคออทิซึมสเปกตรัมตามมาตรฐานสากลใช้แบบสอบถามที่มีความซับซ้อนและใช้เวลามาก ซึ่งในปัจจุบันมีการนำเอาเทคโนโลยีปัญญาประดิษฐ์มาประยุกต์ใช้ในการวิเคราะห์ข้อมูลขนาดใหญ่ที่มีความซับซ้อน อย่างไรก็ตามการนำเอาปัญญาประดิษฐ์มาใช้วิเคราะห์อาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมยังไม่แพร่หลายนัก ในการศึกษานี้ผู้วิจัยต้องการนำเอาปัญญาประดิษฐ์มาใช้แบ่งผู้ป่วยออทิซึมสเปกตรัมจำนวน 85 ราย ออกเป็นกลุ่มย่อยตามอาการทางคลินิกจากข้อมูลแบบทดสอบในการวินิจฉัยผู้ป่วยออทิซึมสเปกตรัม ADI-R จำนวน 123 ข้อ จากนั้นได้ทำการสร้างโมเดลในการจัดกลุ่มและเปรียบเทียบประสิทธิภาพรวมถึงจำนวนข้อคำถามที่ใช้ในแต่ละโมเดล นอกจากนี้ผู้วิจัยได้ศึกษาหาสารบ่งชี้ทางชีวภาพที่เป็นตัวแทนของโรคออทิซึมสเปกตรัมกลุ่มย่อยจากลักษณะอาการทางคลินิกโดยอาศัยการบูรณาการโอมิกส์หลายระดับ จึงได้ดำเนินการวิเคราะห์รูปแบบทรานสคริปโตม และวิเคราะห์รูปแบบโปรตีโอมของผู้ป่วยออทิซึมสเปกตรัมโดยใช้เซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์เป็นต้นแบบ โดยใช้เทคนิคการแยกโปรตีนด้วยกระแสไฟฟ้า 2 มิติ (2D-GE) จากนั้นจะทำการระบุโปรตีนด้วยเทคนิค LC-MS/MS เพื่อหาโปรตีนที่ถูกรบกวนการทำงานอย่างมีนัยสำคัญ และบูรณาการข้อมูลเพื่อศึกษาความสอดคล้องและสัมพันธ์กันระหว่างการแสดงออกของโปรตีนและยีน และทำการยืนยันระดับการแสดงออกที่ผิดปกติไปของยีนด้วยเทคนิค Real-time qPCR และโปรตีนด้วยเทคนิค Western blot ในการศึกษาครั้งนี้จากผลการวิเคราะห์ข้อมูลลักษณะอาการทางคลินิกของผู้ป่วยออทิซึมสเปกตรัมจากแบบทดสอบ ADI-R 123 ข้อ พบว่าวิธี K-means clustering ที่แบ่งผู้ป่วยออกเป็น 4 กลุ่มย่อยนั้นให้ประสิทธิภาพในการแบ่งดีที่สุด และเมื่อทำการสร้างโมเดลในการจัดกลุ่มผู้ป่วยตามผลการแบ่งกลุ่มดังกล่าว พบว่าอัลกอริทึม Random forest ที่มีจำนวนต้นไม้ 10 ต้น โดยกระบวนการคัดเลือกข้อมูลแบบ Forward selection ให้ผลการทำนายถูกต้องมากที่สุด (100%) โดยใช้ข้อคำถาม ADI-R เพียง 5 ข้อ ในการแบ่งผู้ป่วยออกเป็นกลุ่มย่อย และจากการวิเคราะห์รูปแบบโปรตีโอมจากเซลล์เพาะเลี้ยงชนิดลิมโฟบลาสต์ของผู้ป่วยออทิซึมสเปกตรัมและออทิซึมสเปกตรัมกลุ่มย่อยด้วยเทคนิค 2D-GE ผู้วิจัยได้ทำการคัดเลือกจุดโปรตีนที่มีการแสดงออกผิดปกติอย่างมีนัยสำคัญและมีค่าสัดส่วนความเข้มแตกต่างกันอย่างน้อยสองเท่ามา 19 จุด เพื่อทำการระบุชนิดของโปรตีนด้วยเทคนิค LC-MS/MS โดยพบว่าจากโปรตีนทั้งหมด HNRNPA2B1 มีระดับการแสดงออกเพิ่มขึ้นในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่ไม่มีความผิดปกติด้านภาษารุนแรง โดยที่โปรตีน HNRNPA2B1 MDH2 DLD และ MSN มีระดับการแสดงออกลดลงในผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อยที่มีความผิดปกติด้านภาษารุนแรง ดังนั้นจากการศึกษาวิจัยในครั้งนี้ได้แสดงให้เห็นว่าการใช้เทคโนโลยีปัญญาประดิษฐ์ในการแบ่งผู้ป่วยออทิซึมสเปกตรัมออกเป็นกลุ่มย่อยตามลักษณะอาการทางคลินิกนั้น สามารถลดความหลากหลายตามลักษณะอาการที่แสดงออกของโรคออทิซึมสเปกตรัมในการวิเคราะห์รูปแบบทรานสคริปโตมและโปรตีโอม รวมถึงการค้นหาโปรตีนที่จำเพาะต่อผู้ป่วยออทิซึมสเปกตรัมกลุ่มย่อย ได้อย่างมีประสิทธิภาพมากขึ้น ซึ่งโปรตีนเหล่านี้อาจใช้เป็นโปรตีนบ่งชี้ในการศึกษาเชิงลึกในประชากรผู้ป่วย เพื่อพัฒนาเป็นสารบ่งชีทางชีวภาพ (Biomarker) หรือศึกษากลไกพยาธิสภาพในระดับชีวโมเลกุลของโรคออทิซึมสเปกตรัมได้ต่อไปในอนาคต

DEAD (Asp-Glu-Ala-Asp) box polypeptide 41 (DDX41), a receptor belonging to the DExD family, was recently identified as an intracellular DNA sensor in vertebrates. Here, we investigated the presence of cytosolic DNA sensing, DDX41 in shrimp Penaeus monodon. By searching for gene involved in DNA sensing cascade within the shrimp P. monodon EST database, 3 cDNA fragments exhibited similarity to DDX41 in various species were identified. Sequences assembly resulted in a complete ORF of Penaeus monodon DDX41 (PmDDX41) which has 1868-bp encoding a putative protein of 620 amino acid residues. Inulin-type fructooligosaccharides (IFOS) are well-known prebiotics which can be produced from sucrose using bacterial inulosucrase (E.C. 2.4.1.9). However, the use of inulosucrase for IFOS synthesis has some limitations because it also synthesizes some polysaccharides and exhibits lower stability than the fungal enzymes. To overcome these limitations, improvement of inulosucrase was made by using enzyme immobilization and enzyme engineering. First, inulosucrase from Lactobacillus reuteri 121 was immobilized on a novel, high capacity core-shell chitosan beads using glutaraldehyde as a cross-linker. To study melanoma mechanism, screening the amount of melanin production and invasion study are major keys for drug development. In this work, we developed paper-based devices for culturing melanoma cells. World’s population of older people is rising, dietary supplements promoting healthy lifespan are needed. Moreover, Alzheimer’s disease (AD), an age-related degenerative disease, becomes a public health problem in aging society. Recent reports using cell culture models of AD suggest that amyloid precursor protein (APP), a protein causally related to AD, plays an important role as an inhibitor of neurite outgrowth. Medicinal herbs with neurite outgrowth stimulatory effect may help to prevent and cure AD. Growth factors and hormones often mediate their biological effects through series of protein-protein interaction between cytoplasmic signaling molecules or transcription factors. Interfering of these protein-protein interactions has been shown to reduce or inhibit biological and growth responses to various growth factors and hormones. We previously demonstrated that progesterone receptor (PR) contains a polyproline domain (PPD) which directly interact to Src homology 3 (SH3) domain-containing molecules and expression of PR-PPD inhibits EGFR-mediated NSCLC cell proliferation. หนึ่งในสาเหตุสำคัญของการเกิดโรคความเสื่อมของระบบประสาทนั้นเกิดได้จากภาวะเครียดออกซิเดชันภายในเซลล์ประสาท โดยพบว่าสารสื่อประสาทชนิดกลูตาเมตเมื่อมีระดับสูงผิดปกติสามารถเหนี่ยวนำให้เกิดอนุมูลอิสระและภาวะเครียดออกซิเดชันนำไปสู่การสูญเสียและการตายของเซลล์ประสาทได้ โดยเฉพาะการตายของเซลล์จากการเกิดกระบวนการออโตฟาจีมากเกินไป ซึ่งเป็นกระบวนการที่มีบทบาทสำคัญในการควบคุมการตายของเซลล์ผ่านวิถีอื่นๆ รางจืด (Thunbergia laurifolia) เป็นพืชสมุนไพรไทยมีสรรพคุณในการถอนพิษ ต้านการอักเสบ และ ต้านอนุมูลอิสระได้ดี จุดประสงค์ในการวิจัยครั้งนี้ คือ ศึกษาคุณสมบัติการต้านอนุมูลอิสระของสารสกัดหยาบจากใบรางจืด และการปกป้องการตายของเซลล์ประสาทผ่านวิถีออโตฟาจี จากการเหนี่ยวนำภาวะเครียดออกซิเดชันด้วยกลูตาเมตโดยทดสอบในเซลล์ประสาทส่วนฮิปโปแคมปัสของหนูชนิด HT-22 ใบรางจืดถูกนำมาสกัดด้วยเอทานอล จากผลการทดสอบพบว่าสารสกัดหยาบจากใบรางจืดมีความสามารถในการต้านอนุมูลอิสระ และมีปริมาณสารประกอบ ฟลาโวนอยด์และฟีนอลิกสูง โดยจากการวิเคราะห์ด้วยเครื่อง LC-MS และเปรียบเทียบผลที่ได้กับฐานข้อมูล METLIN-(CA, USA) พบสารออกฤทธิ์สำคัญที่ยังไม่มีรายงานในสารสกัดเอทานอลของใบรางจืด ได้แก่ Apigenin 7-O-glucoside และ 7-Hydroxycoumarin ยิ่งไปกว่านั้นสารสกัดหยาบจากใบรางจืดยังสามารถลดการตายของเซลล์ประสาทจากการเหนี่ยวนำความเป็นพิษด้วยกลูตาเมต โดยสามารถยับยั้งการเกิดอนุมูลอิสระภายในเซลล์ และเพิ่มการแสดงออกของยีนต้านอนุมูลอิสระที่ทำหน้าที่สำคัญในกระบวนการต้านอนุมูลอิสระในร่างกาย ได้แก่ superoxide dismutase 1 และ 2 (SOD1 and SOD2) catalase (CAT) และ glutathione peroxidase (GPx) ซึ่งสัมพันธ์กับการลดการเกิดกระบวนการออโตฟาจีภายในเซลล์และเพิ่มปริมาณ ไมโตคอนเดรียโปรตีน โดยพบว่าสารสกัดหยาบจากใบรางจืดสามารถลดอัตราส่วนการแสดงออกของโปรตีน LC3B-II/LC3B-I และเพิ่มการแสดงออกของไมโตคอนเดรียโปรตีน TOM20 เมื่อทำการตรวจหาตำแหน่งที่จำเพาะต่อการแสดงออกของโปรตีนในกระบวนการออโตฟาจี (LC3B) และไมโตคอนเดรียด้วยกล้องจุลทรรศน์ชนิดคอนโฟคอล พบว่า สารสกัดหยาบจากใบรางจืดสามารถลดการเกิด LC3-puncta ซึ่งเป็นลักษณะเด่นของการเกิดกระบวนการออโตฟาจี นอกจากนี้สารสกัดหยาบจากใบรางจืดยังลดการ colocalization ของ LC3B และไมโตคอนเดรีย จากผลการศึกษานี้เราพบสารออกฤทธิ์สำคัญใหม่ที่ยังไม่มีรายงานมาก่อนของใบรางจืด โดยสารออกฤทธิ์นี้มีความสามารถในการปกป้องเซลล์ประสาทชนิด HT-22 จากภาวะเครียดออกซิเดชันและเป็นปัจจัยสำคัญทำให้สารสกัดจากใบรางจืดมีความสามารถลดการตายของเซลล์ประสาทผ่านวิถีออโตฟาจีได้ ซึ่งอาจสามารถนำไปพัฒนาเป็นสมุนไพรทางเลือกหรือยาเพื่อใช้ในการป้องกันหรือรักษาการเกิดโรคความเสื่อมของระบบประสาทได้ในอนาคต

Cholangiocarcinoma (CCA), a malignancy transformed from cholangiocytes in the bile ducts is more common in Asia and has the highest incidence rate in Thailand. CCA is an aggressive malignancy which has high mortality, high recurrence rate and poor prognosis due to late diagnosis and lack of effective treatment, therefore identification of novel therapeutic targets could lead to the development of more efficient therapy. Levansucrase (LS) is an exocellular fructosyltransferase catalyzing the hydrolysis of sucrose and polymerization of levan and levan-type fructo-oligosaccharides (LFOs) that have various potentials in pharmaceutical and biological applications such as prebiotics and anticancer agents. LFOs with the chain length of 5 (LFO5 or GF4) showed inhibition activity against the SW480 colorectal cell lines up to about 83%. การปลอมปนของสารสเตียรอยด์ชนิดเดกซาเมทาโซนและเพรดนิโซโลนในผลิตภัณฑ์สมุนไพรเป็นปัญหาสำคัญที่ก่อผลเสียร้ายแรงต่อสุขภาพของผู้ใช้ ในหลาย ๆ ประเทศทั่วโลก โดยเฉพาะอย่างยิ่งในประเทศกำลังพัฒนา งานวิจัยนี้ ประสบความสำเร็จในการพัฒนาอุปกรณ์สำหรับแยกและตรวจวัดปริมาณการปลอมปนของสารสเตียรอยด์ดังกล่าว โดยใช้อุปกรณ์ตรวจวิเคราะห์ฐานกระดาษและใช้หลักการทางเคมีไฟฟ้า (electrochemical paper-based analytical device หรือ ePAD) ซึ่งประกอบด้วยกระดาษ Whatman SG81 ตลับพลาสติกจากการพิมพ์สามมิติ และขั้วไฟฟ้าชนิด screen printed electrode โดยกระดาษ Whatman SG81 ใช้สำหรับแยกสารสเตียรอยด์ชนิดเดกซาเมทาโซนและเพรดนิโซโลน โดยใช้วัฏภาคเคลื่อนที่เป็นสารละลาย 60% ethyl acetate ใน cyclohexane ซึ่งผลการศึกษาพบว่าสามารถแยกสารสเตียรอยด์ดังกล่าวออกจากสิ่งเจือปนต่าง ๆ ได้ภายใน 7 นาที สารสเตียรอยด์ที่แยกออกจากกันบนกระดาษ นำมาวัดปริมาณด้วยเทคนิคเคมีไฟฟ้าชนิด differential pulse voltammetry โดยใช้ตลับพลาสติกจากการพิมพ์สามมิติที่ถูกออกแบบทำให้สารละลายอิเล็กโทรไลต์ถูกขังอยู่บนกระดาษ ทำให้สามารถตรวจวัดสัญญาณทางเคมีไฟฟ้าได้คงที่ ซึ่งอุปกรณ์ที่พัฒนาขึ้นสามารถตรวจวัดปริมาณปลอมปนของสารสเตียรอยด์ทั้งชนิดเดกซาเมทาโซนและ เพรดนิโซโลนได้ในช่วงความเข้มข้นระหว่าง 10 ถึง 500 µg/mL โดยมีค่าสัมประสิทธิ์สหสัมพันธ์เท่ากับ 0.988 และ 0.994 ตามลำดับ มีขีดจำกัดในการตรวจวิเคราะห์เชิงคุณภาพเท่ากับ 3.59 µg/mL และ 6.00 µg/mL ตามลำดับ และขีดจำกัดในการตรวจวิเคราะห์เชิงปริมาณมีค่าเท่ากับ 11.98 µg/mL และ 20.02 µg/mL ตามลำดับ การตรวจวัดปริมาณปลอมปนของสารสเตียรอยด์ทั้งสองชนิดในสิ่งตัวอย่างผลิตภัณฑ์สมุนไพรจริงด้วยวิธีที่พัฒนาขึ้นมีค่าสอดคล้องกับผลที่ตรวจด้วยวิธี HPLC และเป็นวิธีที่ใช้งานง่าย มีต้นทุนต่ำ รวดเร็ว และมีศักยภาพนำไปใช้ตรวจหาสิ่งตัวอย่างผลิตภัณฑ์สมุนไพรที่สงสัยว่ามีการปลอมปนของสารสเตียรอยด์ ในระดับภาคสนาม

Durian (Durio zibethinus M.), a major economic fruit, actively produces infamous sulfur-containing compounds during ripening. However, sulfur metabolism which is important in this fruit is rarely investigated. In this study, the integration of metabolomics using CE-TOF/MS and transcriptomics using de novo RNA-seq assembly was performed to reveal metabolic changes involved in sulfur metabolism and chemical composition related to fruit quality. During ripening, the expression level of genes involved in sulfur metabolism is concurred with the reduced sulfate ion content in placenta and the up-accumulation of sulfur metabolism-related metabolites in pulps. A paper-based analytical devices (PADs) provides a great benefit in medical diagnostics, especially in the developing world. The first part of this dissertation describes the effect of sample viscosity on PADs for the measurement of pH and nitrite concentration in human saliva. Preliminary study found that viscosity had a significant effect on the calibration curve of nitrite and a consistent curve could be generated after added washing buffer. Nitrite measurement in artificial saliva with viscosity 1.87-5.10 mPa-s was tested on the proposed device and also showed a good correlation with the standard spectrophotometry method (p=0.8484, paired sample t-test, n=20).
