Many species of Neotropical migrant songbirds have been reported to be declining in the last few decades, and the loss and fragmentation of stopover habitat has been implicated as a potential cause of these population declines. To manage properly a fragmented forest landscape for migrating songbirds information is needed on how the size and the habitat of forest fragments influence their use by migrants during stopover. Broxton Rocks Nature Preserve, a 1,233-hectare preserve containing late Pleistocene sandstone (Altamaha grit) outcrops of various sizes and topographies in a conifer-dominated loblolly pine - hardwood forest, represents a unique habitat to the coastal plain of Georgia. Using modified pitfall traps, I sampled arthropods in four subhabitats (bare rock, lichens, ecotone, wiregrass) at Broxton Rocks between January and November 1999. A total of 312,006 arthropods belonging to four classes (Arachnida, Diplopoda, Chilopoda, Insecta) represented by 24 orders was trapped. Nest predation is an important source of mortality in songbirds and may contribute to declines in Neotropical migrants. I used artificial nests baited with fresh Japanese Quail and Zebra Finch eggs in conjunction with observations of natural nests to quantity the ettects ol nest height and vegetation on nest predation on St. Catherine's Island, a barrier island in southeast Georgia. Because of intense browsing by white-tailed deer. I predicted that lower, more exposed nests would be least successful. During postembryonic development of Caenorhabditis elegans, the heterochronic gene lin-14 controls the timing of developmental events in diverse cell types. Three alternativelin-14 transcripts are predicted to encode isoforms of a novel nuclear protein that differ in their amino-terminal domains. In this paper, we report that the alternative amino-terminal domains of LIN-14 are dispensable and that a carboxy-terminal region within exons 9 to 13 is necessary and sufficient for in vivo LIN-14 function. A transgene capable of expressing only one of the three alternativelin-14 gene products rescues a lin-14 null mutation and is developmentally regulated by lin-4. …

Arrestins are regulatory proteins that participate in the termination of G protein-mediated signal transduction. The major arrestin in the Drosophila visual system, Arrestin 2 (Arr2), is phosphorylated in a light-dependent manner by a Ca2+/calmodulin-dependent protein kinase and has been shown to be essential for the termination of the visual signaling cascade in vivo. Here, we report the isolation of nine alleles of the Drosophila photoreceptor cell-specific arr2 gene. Flies carrying each of these alleles underwent light-dependent retinal degeneration and displayed electrophysiological defects typical of previously identified arrestin mutants, including an allele encoding a protein that lacks the major Ca2+/calmodulin-dependent protein …

Introns have typically been discovered in an ad hoc fashion: introns are found as a gene is characterized for other reasons. As complete eukaryotic genome sequences become available, better methods for predicting RNA processing signals in raw sequence will be necessary in order to discover genes and predict their expression. Here we present a catalog of 228 yeast introns, arrived at through a combination of bioinformatic and molecular analysis. Introns annotated in the Saccharomyces Genome Database (SGD) were evaluated, questionable introns were removed after failing a test for splicing in vivo, and known introns absent from the SGD annotation were …

The structural and functional changes in the arterial wall of humans and animals accompany the development of hypertension and atherosclerosis. Hence, great interest exists in the mechanisms that govern vascular smooth muscle cell contraction and proliferation. Rat aortic rings maintained in organ culture show a significant loss of contractility; whereas, vascular smooth muscle cells in culture quickly dedifferentiate into a secretory phenotype with a marked capacity for proliferation. The addition of all-trans retinoic acid (RA) and retinol to the plasma-containing medium preserved the contractile response of cultured vessels. Removal of endothelium prior to culture abolished these effects. All-trans retinoic acid …

A medicinal herb survey was conducted in ten major marketplaces in Andean and coastal communities of Ecuador. Collections were made during the summer of 1997. Four plant collections from each market were pressed, dried, identified, and the medicinal information for each plant was recorded. Photographs of fresh plants, market vendors and the marketplaces were taken.
Fifty three families were represented in the 112 species collected. Forty-two species (37%) were introduced to Ecuador while seventy species (63%) were native to Ecuador. Plant taxa were grouped into 22 categories of medicinal usage. The most common 3 uses were: 1. General tonic and …

South Fork of Sand Lick Creek, Logan County, West Virginia, drains an abandoned coal strip mine which had exploited Pottsville series coalbeds (Pennsylvania strata). These strata outcrop throughout southwestern West Virginia. North Fork watershed is relatively unchanged, save a small roadcut throughout. South Fork benthic community had not recovered although mining activity had ceased about 20 years earlier. Benthic communities were analyzed with detrended correspondence analysis (DCA). Family Chironomidae predominated South Fork benthic community throughout the study. North Fork’s benthic community had as major contributors acid resistant caddisfly family Hydropsychidae, mayfly family Baetidae, and stonefly families Perlodidae and Nemouridae. Family …

The origin of Carex x deamii Herm. has been in question ever since it was named to science in 1938. Collections of the hybrid have always been associated with C. shortiana Dewey and either C. typhina Michx. or C. squarrosa L.. C. typhina and C. squarrosa are closely related taxa (Section Squarrosae) and often are morphologically similar. Because of this similarity, determining the correct parental species to the hybrid has been extremely difficult. It is known that the hybrid is sterile and only reproduces asexually. Migratory birds spend a significant part of each year in transition between their breeding and resting grounds. During this transitional period migrants rely on stopover sites to provide them with the necessary habitat to replenish lost energy stores, rest, and wait for favorable weather conditions before continuing migration. The majority of stopover work related to passerines in North America has occurred along coastal migration routes. Meanwhile, relatively little work has been conducted at inland stopover sites, areas which may be equally important to migrating birds. Human monocytic ehrlichiosis (HME) is an emerging, zoonosis transmitted to mammals by ixodid ticks. Prevalence of infected ticks and distribution of infection foci indicate relative risk of human exposure to ehrlichiosis and may be influenced by factors such as geographic isolation and human disturbance. To test this, individual and pooled lone star ticks, Amblyomma americanum (L.), collected from 3 populations from the coast and barrier islands of Georgia were screened for Ehrlichia chaffeensis Anderson, Dawson, Jone, and Wilson, the agent of HME. Intestinal bile acid 7α-dehydroxylating bacteria have recently been implicated in cholesterol allstone disease. Eubacterium sp. V.P.I. 12708, a bile acid 7α-dehydroxylating bacterium, contains multiple bile acid inducible (bai) genes which encode the enzymes responsible for bile acid 7a-dehydroxylation. The baiE gene encodes a bile acid dehydratase activity in Eubacterium sp.V.P.I. 12708. Using the polymerase chain reaction assay we determined the presence or absence of baiE-like genes in five clostridial bile-acid 7α-dehydroxylating strains: Clostridium sp. TO-931, Clostridium sp. HD-17, Clostridium sp. TN-271, Clostridium bifermentans 1-55, and Clostridium sordellii ATCC 9714. Results from all the strains tested showed amplification at the predicted …

Intricate interplay may exist between pre-mRNA splicing and the cell division cycle, and fission yeast Dsk1 appears to play a role in such a connection. Previous genetic analyses have implicated Dsk1 in the regulation of chromosome segregation at the metaphase/anaphase transition. Yet, its protein sequence suggests that Dsk1 may function as a kinase specific for SR proteins, a family of pre-mRNA splicing factors containing arginine-serine repeats. Using an in vitro system with purified components, we showed that Dsk1 phosphorylated human and yeast SR proteins with high specificity. Lakes are natural resources, which have many effects on the land surrounding them, They support adjacent communities by providing water and regulating temperatures, helping to define the surrounding ecosystem, and serving as sources of drinking water as well as recreation. The prolonged presence of human activity in a watershed can disturb the physical and chemical cycles of the lake and its surrounding ecosystems (Henderson-Sellers and Markland 1987). Plethodon nettinqi was listed as a threatened species in 1989 by the United States Fish and Wildlife Service (Pauley, 1991). Its total range is within 5 counties in eastern West Virginia. There are fewer then 60 disjunct populations known and most populations are above 3,000 ft. and are associated with emergent rocks or narrow ravines with Rhododendron. It is hypothesized the P. netting! survived lumbering practices at the turn of the century by taking refuge beneath large emergent rocks and narrow ravines with Rhododendron. The Environmental Protection Agency is currently developing sediment quality criteria in order to protect the nation's water sources. In order to assist in the development of standards for the disposal of heavy metals, our laboratory is undergoing sediment toxicity studies on Chironomus riparius. It is an EPA approved test organism that spends most of its life in the sediment. Following a previous study (Cochrane et al., 1994), the organism's response to heavy metals will be monitored on the genetic level. Salamanders are important components of many forested communities. However, their fossorial habits and seasonality often make them difficult to sample. The use of artificial coverboards (coverboards) is a relatively new and little-studied technique for monitoring the terrestrial activity of salamanders Coverboards are designed to simulate fallen tree limbs and logs, and to provide a moist refuge for forest salamanders. Despite several studies, questions remain concerning the value of coverboards. This study employed DNA Polymerase Chain Reaction (PGR) and Single-Stranded Conformation Polymorphisms (SSCP) on a mitochondrial control region target to assess population structure and possible gene flow in the Florida Burrowing Owl (Athene cunicularia floridana), a Florida Species of Special Concern. Although widespread in Florida, Burrowing Owls occur at low densities in semi-isolated populations that are susceptible to extirpation from human development and the demographic consequences of small population size. Chrysops vittatus, a small deer fly, and Tabanus atratus, a horse fly, were captured concurrently in Bulloch County, Georgia during June through September of 1989-1997 Cultures were made from adult females, and the frequency and diversity of spiroplasma carriage were determined by deformation and endpoint testing.
Of the 90 Chrysops vittatus processed, 23 (25 .6%) carried spiroplasmas. The most prevalent spiroplasmas were representatives of group Vlll (20/23): strain B1357 (7/23); Spiroplasma syrphidicola (5/23); S. chrysopicola (4/23); strain TAAS-1 (4/23). Groups XVIII and XXXV were represented by S. Htorale (2/23) and strain B2649 (1/23), respectively.

A tripartite domain of the murine immunoglobulin μ heavy-chain enhancer contains the μA and μB elements that bind ETS proteins and the μE3 element that binds leucine zipper-containing basic helix-loop-helix (bHLH-zip) factors. Analysis of the corresponding region of the human μ enhancer revealed high conservation of the μA and μB motifs but a striking absence of the μE3 element. Instead of bHLH-zip proteins, we found that the human enhancer bound core binding factor (CBF) between the μA and μB elements; CBF binding was shown to be a common feature of both murine and human enhancers. The coat proteins of the RNA phages MS2 and Qβ are structurally homologous, yet they specifically bind different RNA structures. In an effort to identify the basis of RNA binding specificity we sought to isolate mutants that convert MS2 coat protein to the RNA binding specificity of Qβ. A library of mutations was created which selectively substitutes amino acids within the RNA binding site. Genetic selection for the ability to repress translation from the Qβ translational operator led to the isolation of several MS2 mutants that acquired binding activity for Qβ RNA. Las Vegas Wash, a natural wash east of the city of Las Vegas, Nevada, carries stormwater, groundwater drainage, and sewage effluent from three sewage treatment plants to Lake Mead. The Wash provides nearly the only surface water outlet for the entire 2,193 mi2 of Las Vegas Valley. A drainage area of 1,586 mi2 contributes directly to the Wash through surface flow which is channeled to Las Vegas Bay of Lake Mead, while drainage of the remaining 607 mi2 is presumably subsurface and may drain toward Las Vegas Wash.

Mutations in the S. cerevisiae VPS29 and VPS30 genes lead to a selective protein sorting defect in which the vacuolar protein carboxypeptidase Y (CPY) is missorted and secreted from the cell, while other soluble vacuolar hydrolases like proteinase A (PrA) are delivered to the vacuole. This phenotype is similar to that seen in cells with mutations in the previously characterized VPS10 and VPS35 genes. Vps10p is a late Golgi transmembrane protein that acts as the sorting receptor for soluble vacuolar hydrolases like CPY and PrA, while Vps35p is a peripheral membrane protein which cofractionates with membranes enriched in Vps10p. The purpose of this study was to determine the effects of creatine supplementation on strength and power performance in well-trained football players. Twenty-one subjects were randomly assigned to three treatment groups: 1) creatine monohydrate, 2) creatine monohydrate+dextrose, or 3) placebo utilizing a double-blind design. Prior to assignment, subjects were matched according to lean body mass. After a five day loading dose, a maintenance dose was administered for 12 days. Participation by children in organized sport continues to increase. A 1994 survey of 10,000 households by the National Sporting Goods Association (1994) found that approximately 40,000 children between the ages of 7-17 years participate in an organized sport or recreational activity annually. In the realm of organized sports, especially endurance activities, there continues to be interest in how cardiovascular fitness is attained and how it can be maximized. Measurement of cardiovascular fitness is an important piece to that puzzle.
