The Role Of Zinc In Dihydroorotase, Pamela S. Gilchrist

Dihydroorotase (4,4—dihydroorotic acid amidolyase, EC 3.5.2.3.) which catalyzes the reversible cyclization of N-carbamyl-L-aspartate to L-dihydroorotate has been purified from orotate-grown Clostridium oroticum. The enzyme is stable in 0.3 M sodium chloride and 10 µ ZnSO4. Sodium dodecyl sulfate gel electrophoresis indicates the enzyme to be composed of two identical subunits each with a molecular weight of 58,000 + 6000. Dihydroorotase is shown to be a zinc-containing metalloenzyme with 2 g atoms of zinc per 58,000 g of protein. The role of zinc in dihydroorotase is discussed.

Incorporation Trends Of (I-C14) Acetate In Pregnancy Lipids Of Various Organs, D.P. Sharma, T. A. Venkitasubramanian

Comparative evaluation of (I-C14) acetate incorporation was studied in ovary, adrenal, blood plasma and placenta in guinea pigs in mid pregnancy. The respective level of incorporation within the sample for total lipids did not differ in pregnancy and non-pregnant states. Placental lipids in general, and phospholipids in particular were highly incorporated in contrast to other organs/tissues. Amongst non-pregnancy total lipids, the blood plasma showed an elevated incorporation over ovarian and adrenal lipids. The ovary in pregnancy revealed depressed incorporation in general, except for esterified cholesterol which contained higher incorporation. The 14C light and dark bottle technique for measurement of primary production was utilized as a means of assessing the amount of inorganic carbon being converted Into organic form by the photosynthesis of phytoplankton populations In the Boulder Basin of Lake Mead.
Spatial and time series changes of productivity levels observed at eight sampling locations within Boulder Basin Indicate that the Influence of treated municipal arts' industrial effluent flowing into Les Vegas Bay is contributing high levels of available nutrients at Las Vegas Wash Inlet to cause productivity to approximate those levels associated with polluted waters.

Accurate quantum mechanical close coupling (CC) integral cross sections are reported for rotationally inelastic Ar-N2 collisions at room temperature using an empirical intermolecular potential. These cross sections are used to test the infinite order sudden (IOS) approximation and the results of several methods [classical trajectories (CT), full sudden (SA), generalized phase shift (GPS), and classical limit generalized phase shift (CGPS)] as reported in the preceding paper by Pattengill. The IOS approximation works very well for cross sections involving low rotational states. The CT method works well for all the cross sections examined, justifying the popular belief that the rotational motion …

A detailed time study of the incorporation of label from sodium-[1-14C]acetate, [1-14C]ethanol, and [2-14C]glucose into the aspartyl moiety of N-acetylaspartic acid (NAA) was conducted. As expected the specific activity of aspartate increased rapidly with time and peaked within 15-20 min after which it fell sharply: but significantly that of the aspartyl moiety of NAA rose very slowly even after the specific activity of aspartate had fallen to less than 1 per cent of the peak values. A study was conducted to find the effect of magnesium, calcium, manganese and zinc ions on the infection of Psudomonas aeruginosa strain 1C by RNA bacteriophage 7s. When an 18 hour progeny experiment was performed, it was found that magnesium, calcium and manganese had different effects on bacteriophage production and was dependent on the bacterial growth conditions. RNA bacteriophage 7s progeny production was significantly enhanced by the addition of magnesium to cultures of Psudomonas aeruginosa 1C grown in a magnesium deficient medium. Under these environmental conditions there was a slight increase in progeny in the presence of calcium. When Psudomonas …

Cells of Clostridium oroticum, an anaerobic bacterium, were grown on orotate as a carbon and energy source. Ureidosuccinase, an inducible enzyme in the pathway for pyrimidine degradation has been shown to convert ureidosuccinate to aspartate, CO2 and NH3 as reported by Liebenmm and Kornberg (7). Aspartate and CO2 were formed in approximately a 1:1 ratio from ureidosuccinase activity.
Ureidosuccinase was found to be a Mn+2 requiring enzyme with a pH optimum of approximately pH 6.5. Enzyme activity is labile to O2, temperature, pH, dilution and high ionic strength. The optimum conditions for storage …

Intestines of fifty-six albino rats of the Holtzman strain were removed and perfused with Krebs bicarbonate buffered solutions containing caffeine. After one hour of absorbtion, caffeine was extracted from the serosal fluids in chloroform and concentrations were determined spectrophotometrically. It was found that all portions of the small intestine transported caffeine against a concentration gradient. This transport mechanism demonstrated saturation kinetics and was inhibited by ouabain. It was concluded that the movement of caffeine from the mucosa to the serosa of the small intestine cannot be the result of simple diffusion. The combination of L-DOPA and pargyline caused a decrease in level of aspartate and an increase in that of glutamine in vivo in cerebral cortex, cerebellum, brain stem, hypothalamus, neostriatum and cervical cord of rat. There was also a decreased incorporation of radioactivity from [1-14C]acetate into amino acids in rico, most notably in cerebellum and brain stem. The labelling of glutamine was especially affected. In addition, cortical slices were prepared from guinea pigs which had been pretreated with pargyline. These slices were incubated with and without 1 mM L-DOPA in media containing [l- 14C]acetate. Pooled urine samples from patients receiving 100-200 mg of naltrexone per day orally were extracted; the basic (alkaloid) compounds derived were isolated by preparative thin-layer chromatography. The major metabolite of naltrexone was found to be an epimer of N-cyclopropylmethyl- 14-hydroxy-7,8-dihydronormorphine wherein the 6-keto group of naltrexone had been reduced to yield the 6β-hydroxy epimer (an isomorphine). This conclusion was based on infrared, mass, and nuclear magnetic resonance spectra studies. Furthermore, the reduction product formed in vitro in a soluble chicken liver enzyme system from naltrexone and an in vivo metabolite of naloxone derived from the chicken were found to have …

In these exciting times when elementary and high schools teach modern biology, including many of the intricacies of biochemical genetics, the long slow process by which our present knowledge in this area was gained is not often fully appreciated. A third of a century elapsed before Mendel’s work was "rediscovered" and properly appreciated. Archibald E. Garrod’s (1) prophetic appreciation of the relation genetics and biochemistry, beginning soon after the so-called rediscovery of Mendel, lay fallow for more than forty years despite the fact that he published widely and relatively voluminously. The diurnal vertical movement of zooplankton was first recorded in freshwater lakes by Weismann (1877) in Lake Constance (Bodensee), although Cuvier was credited with observing the migration of Daphniae in 1817 (Gushing, 1955). The migration is best observed in deep oligotrophic lakes and migrations of 50 m per day are described (Worthington, 1931). In addition, two species of chaetognaths are reported to migrate 400 m a day in marine waters off of Lisbon (Waterman and Berry, 1967) while Birge (1895) found little evidence of the phenomenon at all in Lake Mendota, Wisconsin. Phytoplankton samples were collected in Lake Mend 6 times from September 1910 to June 1971 for 8 stations at depths of 0. 3, 5, 10, 20, and 30 m. These samples were processed through a Millipore filter apparatus and 79 planktonic algae were identified. Algal divisions represented were Bacillariophyta, 42 species; Chlorophyta, 18 ; Cyanophyta, 9; Chrysophyta, 3; Cryptophyta, 3; Pyrrophyta, 2; and Euglenophyta, 2. Blue-green algae were dominant in late summer and fall; green algae, diatoms, and, cryptomonads in winter; and green algae in spring. The early summer flora was best represented by the Chlorophyta, Cryptophyta, and Chrysophyta. Palmer's …

Ornithine plays an important role in mammalian intermediary metabolism. Ornithine can be (1) utilized as a component of the urea cycle, (2) decarboxylated to form putrescine, a precursor of the polyamines, spermidine and spermine, and (3) converted metabolically to arginine, proline and glutamate. In prior investigations conducted in our laboratories and others, free endogenous hepatic pools of ornithine were found to be elevated following hydrazine administration. Consequently, time-course alterations of arginase, ornithine δ-transaminase, ornithine transcarbamylase, and ornithine decarboxylase activities were investigated regarding the effect of hydrazine treatment on hepatic ornithine metabolism in the rat. Partial cross sections (opacity functions) for rotational transitions in atom-diatom collisions are computed in the infinite-order sudden (IOS) approximation and compared with accurate close-coupling (CC) calculations. Agreement is good in the dominant coupling (small total angular momentum J) region. Simple methods for calculating integral inelastic cross sections are discussed, and it is found that accurate cross sections can often be computed very simply, even when large numbers of channels are coupled together, by using IOS or first-order sudden (FOS) approximations for small J and CC or exponential Born (EBDW) methods for large J.

I. An isothermal titration microcalorimeter having a colume of 4 ml and capable of temerature control to ±2 x 10-5°C is described. Major components include a constant temperature water bath controlled to ±3 x 10-4°C, a platinum reation vessel, and an isothermal control circuit consisting of constant Peltier thermoelectric cooling and variable Joule heating controlled by a thermistor in an AC Wheatstone bridge circuit. The calorimeter was tested by measuring the heat of ionization of water and was found to produce data accurate to ± 0.1% where small samples are used such as in the investigation of many biological systems.

Barley plants grown in Mg, Mn, Zn and Cu deficient media were analyzed for specific activity, total activity and amount of RuDPCase present. Percent dry weight, as well as chlorophyll and soluble protein content were determined. All deficiencies caused lower chlorophyll and RuDPCase concentrations than occurred in the control. Soluble protein content, however, was higher in all deficient plants than in the control, either on a dry or a fresh weight basis. The addition of 18 μM Mn^2+ increased the activity of Mg^2+ containing RuDPCase preparations as much as 133% above normal. The specific activity of RuDPCase from Mg, Mn, …

The virulence of animal-passaged Mycoplasma arthritidis (158) was demonstrated to be enhanced in rats by growth in oleic acid-enriched broth. The virulence of a laboratory-maintained culture of the same strain was unaffected by similar growth conditions. No differences either in the immune response as measured by complement-fixing (CF) antibodies or in the histogenesis of the disease were detected when rats were injected with animal-passaged M. arthritidis grown either in the presence or in the absence of oleic acid. All tests for metabolism-inhibiting (MI) antibodies in the rat were negative.

Purified Bacillus subtilis DNA taken up by one-day-old barley embryos and permanently fixed within the barley cell is shown to have maintained its biological activity in B. subtilis transformation. Cesuim chloride density gradient centrifugation of DNA isolated from the roots of barley embryos after the uptake and incorporation of radioactive, bromouracil-labelled B. subtilis DNA indicates the presence of the bacterial DNA in several discreet bands of well-defined density, whose density is changed by sonication. These preliminary findings concerning the fate of bacterial DNA taken up by barley roots are discussed in terms of the recombination model of Ledoux and Huart …

now being packed and protected with "grape guards"—gas releasing generators consisting of chemically treated paper which releases small quantities of sulphur dioxide. Grape guards inserted in unventilated storage boxes are activated by the high humidity in the box to release their sulphur dioxide and prevent decay of the packaged fruit. The guards were developed in the United States two years ago.

A fatty acid synthetase from Corynebacterium diphtheriae has been purified to a specific activity of 450 nmoles of malonyl coenzyme A incorporated per min per mg. The enzyme is optimally active in 0.5 M phosphate buffer. C. diphtheriae appears to be the most primitive organism having a multienzyme complex for fatty acid synthesis.

The effects of acyl-CoA derivatives (C8 to C20) on the activity
of the fatty acid synthetases from yeast and Corynebacterium
diphtheriae have been examined. Both enzyme
systems are inhibited by the longer chain acyl thioesters
(C16 to C20) and protected against this inhibition by bovine
serum albumin (BSA). Identical relief from acyl-CoA inhibition
is provided by the 6-0-methylglucose-containing
lipopolysaccharide (MGLP), from Mycobacterium phlei. It
is shown that MGLP forms a stable complex with palmitylCoA.
This interaction accounts for the BSA-like effects of
the polysaccharide. BSA and MGLP have two further effects

This program was a status study of the interaction between Las Vegas Wash, an enriched stream, and Las Vegas Bay, a wedge shaped arm of one of the world's deeper reservoirs. The program centered primarily on identification and counting of planktonic algae from several points in Las Vegas Bay. Additional work on nutrient enrichment of water samples was conducted to aid in interpretation of algal distribution related to nutrient input. Examination of a variety of physical, chemical, and biological parameters, both at many surface points in the bay, as well as in vertical profile, was also accomplished and further aided …

On Improved Wkb (Uniform Asymptotic) Quantum Conditions, Dunham Corrections, The Langer Modification, And Rkr Potentials, Russell T. Pack

An improvement to the WKB quantum condition for the rotation-vibration levels of diatomic molecules, known empirically to give RKR potentials of increased accuracy and to account for the Dunham correction Y00, is derived using uniform asymptotic methods. The role of the Langer transformation in this problem is also discussed.

Phytoplankton successions, applications of the general growth equation, and physical measurements have been employed to investigate events occurring at the interface between industrial and sewage effluent contained in Las Vegas Wash and the waters of Lake Mead, Nevada. The data indicate that the entering waters tend to form a density current interrupted at intervals by dynamic effects generated in the lake. The dynamic relationships described here for the spring months suggest that a much more thorough understanding of physical, chemical and biological interactions is necessary to permit solution of the numerous problems of Las Vegas Bay discussed or alluded to …
