The purpose of beef cattle and beef product research at UNL is to provide reference information that represents the various populations (cows, calves, heifers, feeders, carcasses, retail products, etc.) of beef production. Obviously, researchers cannot apply treatments to every member of a population; therefore, they must sample the population. The use of statistics allows researchers and readers of the Nebraska Beef Report the opportunity to evaluate separation of random (chance) occurrences and real biological effects of a treatment. Evaluation Of Single Nucleotide Polymorphisms Associated With Fertility And Production Traits In Holstein And Multi-Generational Angus Females, Rebecca Ann Hill

The objective of this study was to test the association of single nucleotide polymorphisms (SNPs) with fertility in two populations consisting of Holstein cows and multi-generational Angus cows. The candidate gene approach was utilized and previously described SNPs were tested for possible associations with fertility. Single nucleotide polymorphisms on three genes were evaluated including leptin receptor LEPR, calpastatin CAST, and DGAT1. Fertility traits were evaluated in conjunction with production traits for Holstein females and growth traits for Angus females. One SNP was significantly associated with birth weight (P < 0.05) in Angus females while a trend (P < 0.10) was observed for two markers influencing birth weight performance and three markers influencing weaning weight performance. An association of two SNP for birth weight and back fat thickness in Angus females was identified.!A trend (P < 0.10) was observed for one marker within LEPR influencing average services to conception, two markers within CAST influencing average days open, two markers within CAST and one marker within DGAT1 influencing average protein production, and one marker within CAST and one marker within DGAT1 influencing average milk production. One SNP within LEPR was significantly associated with average milk production (P < 0.05) in Holstein females. An association of one SNP within CAST and one SNP within DGAT1 for average protein production and average milk production in Holstein females was identified. An association of one SNP within CAST for average days open and average protein production in Holstein females was also identified. The association of these markers indicates that the evaluated quantitative trait loci (QTL) region may harbor causative mutations responsible for the variation observed in fertility and production traits. Further evaluation of SNP in these regions is necessary in order to identify mutations accounting for the largest degree of variation for fertility and production traits.

Two experiments assessed the potential of altrenogest, an artificial progestogen, and deslorelin acetate, a gonadotropin-releasing hormone (GnRH) agonist, for suppressing estrus and ovulation in cyclic mares. In the first experiment, mares were administered a luteolytic dose of prostaglandin-F2á on d 6 of diestrus, and were then treated with 1 of 3 formulations of altrenogest in slow-release vehicle (6 mares/group): 1) Biorelease altrenogest LA150 (225 mg total as a 1.5 mL injection); 2) Biorelease altrenogest LA225 (225 mg total as a 1.0 mL injection); or 3) Biorelease altrenogest LA225 (450 mg total as a 2.0 mL injection). The exotic Eurasian Collared-Dove (Streptopelia decaocto) was first sighted in Arkansas at Harrison (Boone Co.) on 25 June 1989. Since this initial sighting the species has grown in numbers and is now present in 42 of 75 counties across the state. In the spring and summer of 2009 and 2010, 20 nests were observed in the urban areas of Fort Smith (Sebastian County). Fifteen of the 20 nests (75%) were located on human-made structures of which 13 (65%) were on an electrical substation and two (10%) were on utility poles. The remaining 5 nests (25%) were in trees. The spothanded crayfish, Orconectes (Procericambarus) punctimanus (Creaser), is an endemic crayfish of the Interior Highlands (Ozark Plateau) of Arkansas and Missouri. Fieldwork within the Arkansas portion of its overall distribution from 1999–2012 and a search of museum and literature records revealed a state distribution of 11 counties in northern Arkansas. Eighty-two collections of this crayfish were made from the White River system in 9 counties, including Baxter, Fulton, Independence, Izard, Lawrence, Marion, Randolph, Sharp, and Stone. Two additional counties, Clay and Searcy, are also added to the distribution of O. punctimanus from museum and literature records. This crayfish inhabits areas …

Military installations present unique challenges to natural resource managers managing wildlife populations. For those species that are hunted or trapped, it is important to provide data to these installations for achieving sustainable harvests. Pine Bluff Arsenal (PBA), a military installation in southeastern Arkansas, manages for a host of wildlife species including white-tailed deer (Odocoileus virginianus). However, baseline data regarding population size for deer are lacking. We used infrared technology and distance sampling to estimate the size of the winter, post-harvest deer population on PBA. The least darter, Etheostoma microperca, has a limited distribution in Arkansas and is designated as a species of greatest conservation need by the Arkansas Game and Fish Commission. The fish was first documented in the state in 1938 in Wildcat Creek west of Springdale with additional discoveries in 1960, 1973, 1981, and 1982. A 1997 study documented the persistence of the species in Healing Spring Run and a spring run tributary of Osage Creek, 2 of the historic streams. Studies of locomotor performance often link variation in morphology with ecology. While maximum sprint speed is a commonly used performance variable, the absolute limits for this performance trait are not completely understood. Absolute maximal speed has often been shown to increase linearly with body size, but several comparative studies covering a large range of body sizes suggest that maximal speed does not increase indefinitely with body mass but rather reaches an optimum after which speed declines. Because of the comparative nature of these studies, it is difficult to determine whether this decrease is due to biomechanical constraints on maximal speed …

Many studies suggest that zoo visitors are a cause of stress among animals; among primates, visitor presence can lead to an increase in aggressive displays, time spent non-visible to the public, and a decrease in overall activity. This study tested the effectiveness of using species-specific behaviors among a group of captive chimpanzees. This study describes the anatomical, light microscopic, and ultrastructural characterization of the immune organs in ostrich chicks. In this study 6 healthy 8-week-old ostrich chicks were processed for light and transmission electron microscopy. Hematoxylin and eosin as well as improved Weigert staining were used for light microscopy. The small thymus, which clustered caudally along both sides of the neck, contained Hassall's corpuscles in the medulla. The cloacal bursa, which formed the hanging and lateral walls of the cloaca, enveloped the cloaca but did not form a truly independent bursa. In the present study, antisense primers were designed from 7 differentially displayed expressed sequence tags (ESTs) previously identified at our laboratory. Total RNA was isolated from the mammary gland tissue of lactating buffalo to carry out 5' rapid amplification cDNA ends (RACE) PCR. 5' RACE PCR amplicons, ranging from 161 bp to 336 bp, were cloned and sequenced. These 5' RACE sequences were analyzed using the blastn (Megablast) bioinformatics tool available from NCBI. To determine the seroprevalence of Anaplasma in Minnesota cattle, the database of the Veterinary Diagnostic Laboratory (VDL), University of Minnesota was searched over a 10 year period (2001 to July 2010). A total of 438,407 bovine serum samples were tested by complement fixation test (CFT) and competitive enzyme linked immunosorbent test (cELISA). The positive rate of seroprevalence by CFT was 2.4% out of 211,484 samples, with a 0.13% anticomplementary result. The VDL at the University of Minnesota started using cELISA after 2002, and CFT application was suspended as diagnostic test for anaplasmosis after 2005 due to its complexity. With cELISA, …

In order to determine the availability of fermented protein feedstuffs (FPFS)-such as cottonseed, blood, and feather meals—with Aspergillus oryzae, 2 phases of feeding experiments were adopted for 7 weeks. Adverse environmental conditions (contamination of water with nitrite, pesticides, or other pollutants), nutritional deficiencies, bacterial and viral infections, and parasitic invasions may induce anemia in fish, in both natural and aquaculture conditions. Changes in hematological values and cellular composition of peripheral blood and head kidney hematopoietic tissue of bled common carp (Cyprinus carpio L.) were evaluated over a 4 week period post-bleeding (30% blood loss). This study was conducted to evaluate the effects on sperm motility and abnormality in ram semen extended with skimmed milk (M), sodium citrate (SC), Tris (T), and Bioxcell® (B) after storage in liquid form at 4 °C and in frozen form. Ejaculates were collected from 3 Akkaraman rams by artificial vagina twice a week during the non-breeding season. After pooling, each pooled ejaculate was split into 4 equal aliquots and diluted with skimmed milk (M), sodium citrate (SC), Tris (T), and Bioxcell® (B) extenders. Sperm motility was significantly higher (P < 0.05) in M compared with B, SC, and T on the first and second days (48 h) of storage. With regard to the percentage of total abnormal spermatozoa, the results in M were different from (P < 0.05) from those of SC and T on the first day (24 h) of storage but no different from B. Differences among extenders were found to be significant post-thawing for spermatozoa motility and the percentage of total abnormal spermatozoa. The total abnormality of semen diluted with M was significantly lower than that observed in the other extenders. Consequently, it was found that skimmed milk was better than the other extenders in terms of the sperm parameters evaluated during liquid storage and post-thaw.

The objective of this study was to evaluate the reproductive performance of primiparous and multiparous Saanen goats after intrauterine laparoscopic artificial insemination with frozen semen. Twenty-four Saanen goats, divided in 2 groups: group 1 consisted of 11 primiparous goats and group 2 consisted of 13 multiparous goats. Estrus was synchronized by 20 mg fluorogestone acetate (FGA)-impregnated intravaginal sponges and the IM administration of 125 mg of cloprostenol (PGF2\alpha) and eCG (400 IU), 48 h before sponge removal. A simple and rapid diagnostic method of Anaplasma phagocytophilum infection was established, employing the loop-mediated isothermal amplification (LAMP) reaction. With 1 set of 4 specific primers targeting the citrate synthase (gltA) gene of A. phagocytophilum, the LAMP assay rapidly amplified the target DNA sequence in a single tube in 1 h, requiring only a water bath for reaction. When visualized by gel electrophoresis, the LAMP products appeared as a ladder-like pattern, with many bands of different sizes from 180 base pairs up to the loading well. The tiger population that once inhabited the Korean peninsula was initially considered a unique subspecies (Panthera tigris coreensis), distinct from the Amur tiger of the Russian Far East (P. t. altaica). However, in the following decades, the population of P. t. coreensis was classified as P. t. altaica and hence forth the two populations have been considered the same subspecies. From an ecological point of view, the classification of the Korean tiger population as P. t. altaica is a plausible conclusion. A unique community-funded project in Puerto Rico has launched whole-genome sequencing of the critically endangered Puerto Rican Parrot (Amazona vittata), with interpretation by genome bioinformaticians and students, and deposition into public online databases. This is the first article that focuses on the whole genome of a parrot species, one endemic to the USA and recently threatened with extinction. It provides invaluable conservation tools and a vivid example of hopeful prospects for future genome assessment of so many new species. Consumer interest in forage-fed products has grown and little research has been done comparing performance of forage-fed animals and beef finished on different forages. Kefir is a fermented milk traditionally made from a unique starter culture, which consists of numerous bacteria and yeast species bound together in an exopolysaccharide matrix produced by certain lactic acid bacteria. Many health benefits are associated with traditionally produced kefir; however, bulging and leaking packaging, caused by secondary yeast fermentation during storage, has limited large scale manufacture traditionally produced kefir. Different antimicrobials are added in the manufacturing of dairy products such as flavored yogurts and processed cheese. Potassium Metabisulfite (PM) and Potassium Nitrite (PN) have been reported to have antimicrobial effect on pathogenic microorganisms such as Clostridium botulinum and Listeria monocytogenes. Yogurt is known for its health benefits, due to the presence of cultured bacteria. PM and PN are not commonly used in the dairy industry hence it would be interesting to study their influence in yogurt culture bacteria. The Spiny Lobster (Panulirus argus) fishery in Florida is closed during the spawning season (March-July) except for a two-day recreational `miniseason' for sport divers in July, several days prior to the opening of the commercial fishing season. In Monroe County, recreational fishers, who possess a valid Saltwater Fishing License with crawfish stamp, are allowed to harvest six lobsters per day, each with a minimum carapace length of 76.2 mm (3.0 inches). During these two days, approximately 50,000 people attempt to catch lobster, and the number of boats visiting the reef has been estimated to be up to 900 times higher …

Brief description of an encounter with a Snowy Plover discovered at a nest site near the mouth of Tahkenitch Creek in Douglas County, Oregon. The plover was buried up to its neck in sand, presumably caused by high winds. The article describes the authors' efforts to resuscitate the plover.

Insulin Dose Response Curves And Factors Affecting Insulin Sensitivity In Horses, Jeanne D. Lestelle

Three experiments were conducted to further study the use of intravenous insulin injection to measure insulin sensitivity in horses. In the first experiment, glucose responses to multiple doses of recombinant human insulin were compared in insulin sensitive and insensitive mares. Regression lines describing the responses in insensitive mares had less (P = 0.003) steep slopes and greater (P = 0.006) effective doses at 50% response (ED50) compared to sensitive mares. Curvilinear regression models provided a good fit (R2 = 0.95) for the prediction of ED50 from the individual responses to 50 mU/kg BW insulin. The second experiment estimated the repeatability …

Small pumpkinseed sunfish ( Lepomis gibbosus), were found to be capable of removing the spine of Bythotrephes longimanus, an invasive cladoceran. Because fish consumption may be important in the dispersal or control of Bythotrephes, aquarium feeding experiments were conducted to 1) establish if the spine removal behavior of the pumpkinseeds was locally unique; 2) quantify how frequently pumpkinseeds exhibit the behavior; 3) determine if pumpkinseed handle Bythotrephes more quickly than other species of fish; and 4) verify if Bythotrephes' resting eggs pass through the digestive systems of pumpkinseeds in viable condition.
