The purpose of this study was to evaluate genetic similarities and distances among pure lines of poultry using the RAPD technique. In this study four layer and three meat type pure lines were used. Venous blood samples from five males and five females of each line were collected from the venae cutenea ulnaris. The RAPD technique was used to generate a DNA fingerprint of lines. Male Sprague-Dawley rats (SD) were kept in medium between the postnatal 80^{th} and 130^{th} day while environmental temperature (from 20 to 5 ºC) and photoperiod were decreased gradually. The weight of the animals adapted to the cold and the weight and length of the external digitorum longus (EDL) muscle were measured. When compared with controls, in a cold environment the weight and fiber length of the muscle was reduced significantly. A total of 1000 beef cattle were examined after being slaughtered, of which 373 were Holstein and 627 were Brown Swiss. The high incidence of lens abnormalities was observed to be as follows: coloboma (20.4%), microphakia (6%) and cataract (3.9%) in Holsteins for a total incidence rate of 30.3%, and coloboma (5.6%), microphakia (2.6%) and cataract (4.2%) in Brown Swiss for a total incidence rate of 12.4 %. Holsteins were more frequently affected than Brown Swiss with coloboma and microphakia, but not with unilateral cataract formation. This experiment was conducted to compare the kid growth and doe milk production under kid suckling and milk replacer (MR) regimes. Thirty does with single kids were randomly assigned into two equal groups (15 kids per group): the control group, where kids suckled freely, and the MR group, where kids were fed with MR containing 21% fat and 22% protein. Effects Of Progesterone And Testosterone On The Hyaluronidase Activities And Sperm Characteristics In Sheep, Sadetti̇n Tanyildizi, Tanzer Bozkurt

The aim of this study was to determine the effects of progesterone and testosterone hormones on the hyaluronidase activities of serum and semen and on sperm characteristics in sheep. Thirty healthy Akkaraman sheep were used. The progesterone and testosterone were injected intramuscularly at the rate of 0.5 mg/kg and 5 mg/kg, respectively. Then the blood and semen samples were obtained from the sheep at 1, 2, 4, 24, 48, 72 and 168 h. The serum hyaluronidase activities of ewes and rams in progesterone and testosterone groups increased significantly (p < 0.01, p < 0.001) when compared with the control groups at all times. Additionally, there were significant (p < 0.001) increases in the levels of semen hyaluronidase activity in the progesterone group except at 4 h. Furthermore, semen hyaluronidase activities in the testosterone group were detected to decrease significantly (p < 0.01) in comparison with the control rams except at 4 h. The use of progesterone and testosterone caused a significant (p < 0.05, p < 0.01) increase in spermatozoa motility, respectively. However, sperm concentrations were determined to decrease significantly (p < 0.01, p < 0.001) in both groups. Although the values for semen volume decreased significantly (p < 0.05, p < 0.01) in the progesterone group, the same values were observed to increase significantly (p < 0.01) in the testosterone group except at 48 h when compared with the control groups. The results of this study showed that there were no significant correlations between semen hyaluronidase activities and sperm characteristics. In conclusion, the use of both hormones did not have any deleterious effects on hyaluronidase activities but for the semen samples in the testosterone groups. However, progesterone and testosterone hormones cause harmful effects on the fertilization ability of rams due to decreased sperm concentration values.

In this study, the capture efficiencies and catch rates of three different traps were compared for the blue crab Callinectes sapidus in Beymelek Lagoon, Antalya, Turkey. All gear was used simultaneously and in the same habitats. The mean catch per unit effort (CPUE) was determined to be significantly higher for hoop nets than for traps. Gear effectiveness was examined by comparing CPUE (CPUE as number of crab per trap per haul, and g per trap haul), mean weight of individual crabs, catch composition and width frequency distributions of the blue crab for each gear.

Characterisation Of An Exopolysaccharide Preventing Phage Adsorption In Lactococcus Lactis Subsp. Cremoris Ma39, Mustafa Akçeli̇k, Pinar Şanlibaba

Lactococcus lactis subsp. cremoris strain MA39 produces an extracellular polysaccharide containing rhamnose, glucose and galactose. A 16.5 kb plasmid encoding exopolysaccharide production in MA39 was determined by plasmid curing experiments. Phage adsorption assays showed that four different lactococcal phages were adsorbed to MA39-40; only the 16.5 kb plasmid cured an exopolysaccharide non-producing mutant of strain MA39, with high efficiency (93.2-98.5%) while adsorption of these phages was completely inhibited in the wild type strain MA39. These results suggest that exopolysaccharide prevents phage adsorption by masking phage receptor sites.

Fish were exposed to three different sublethal doses in order to determine the effects of Cypermethrin on some biochemical parameters (Ca-Calcium, P-Phosphorus, Na-Sodium and TP-Total Protein) of rainbow trout (Oncorhynchus mykiss). After 15 days exposure, Ca and P decreased, while TP and Na took various values depending on the doses of synthetic pyrethroid.

Male Wistar albino rats, bred and fed under standard laboratory conditions, were administered for 20 days two dosages [Group I: 750 mg/kg body weight (n = 15) and Group II: 1500 mg/kg body weight (n = 15)] of oral hexachlorobenzene resolved in corn oil. Control group animals (n = 15) received only corn oil for 20 days. Twenty-four hours after the administration of the last dose, renal tissue samples, obtained by laparotomy, were fixed in 3% glutaraldehyde in phosphate buffer and 1% aqueous osmium tetroxide. Thymic nurse cells are isolated thymic epithelial/thymocyte complexes described in vitro. Their in vivo existence is controversial. We examined rat thymi in order to distinguish thymic nurse cells in vivo using the zinc iodide-osmium tetroxide (ZIO) fixation/staining technique. There were mainly two types of strongly ZIO reactive cells in the thymic cortex: macrophages and small clusters of thymic epithelial cells. The latter group was composed of thymic epithelial cells and intact thymocytes forming complexes. Thus we suggest that these structures are in situ equivalents of thymic nurse cells.

A specimen of Squatina aculeata (sawback angelshark) was caught in a trawl on 15 May 1997 in Iskenderun Bay at about 120-200 m depth and recorded for the first time from the eastern Mediterranean coast of Turkey. Thus, a new species of fish was recently added to the chondrictyes fish fauna in the eastern Mediterranean Sea off Turkey.

In this study a series of weight-length relationships are presented for 13 fish species caught between September and December 2000 off the south coast of İskenderun Bay. The samples were collected with longlines. The values of the exponent b in the weight-length relationship, W = aL^b, ranged from 2.391 to 3.675 and the median value was 2.714.

A New Fish Record From Turkish Seas: The Blackfin Sorcerer Nettastoma Melanurum Rafinesque, 1810 (Nettastomidae), Nuri̇ Başusta, Cemal Turan, Mevlüt Gürlek

Two and five specimens of the blackfin sorcerer, Nettastoma melanurum, were trawled on 25 and 30 April 2001 respectively at Samandağ, Hatay (36º10'N, 35º20'E; 36º05'N, 35º40'E) at about 80-200 m depth. It is recorded for the first time from the eastern Mediterranean coast of Turkey. Thus, a new species and family have been added to the fish fauna in Turkish seas.

A female specimen of Dioctophyma renale was recovered from the abdominal cavity of a stray dog necropsied in Istanbul, Turkey on 8 June, 1999. The nematode was 55.5 cm in lenght, with a diameter of 0.7 (0.6-0.8) cm. This is the first case report of D. renale from Turkey.

An adult, male common buzzard (Buteo buteo) with a leg fracture and multiple proliferative cutaneous nodules on its feet was brought to Akdeniz University Burdur Veterinary Faculty by a farmer who found the bird at his farm. There were similar small nodules on the eyelids and beak. The fractured bone ends had become necrotized. The buzzard was euthanized. On histopathological examination of nodules, large, eosinophilic intracytoplasmic inclusion bodies (Bollinger bodies) were seen. Avipox was diagnosed on the basis of the typical gross and microscopic features.

A case of neonatal renal oxalosis in a 7-day-old calf is described. Clinical biochemistry revealed increased blood urea nitrogen. Gross findings included thin renal medulla and dilated renal calyces containing numerous, small, pale yellow and granular calculi. Histologically, most cortical and medullary tubules were dilated with birefringent crystals, whereas the glomeruli appeared unaffected. The crystals were morphologically consistent with calcium oxalate by the Pizzolato technique. Diffuse interstitial fibrosis and focal accumulations of lymphocytes were also noted.

In this study, the detection of canine parvovirus (CPV) in a fecal sample from a dog with enteritis was performed for the first time using the polymerase chain reaction (PCR) in Turkey. The final PCR product was analyzed using the restriction fragment length polymorphysm (RFLP) technique. RFLP analysis using Apa LI and Eco RV restriction endonucleases revealed homology in the nucleotide sequence in at least the VP2 coding region of the virus DNAs detected in the fecal specimen and prepared from attenuated vaccine virus as a positive control.

In this study, the histopathology of enzootic epidermal papillomas in cage cultured mirror carp from the Liman region of Eğirdir Lake was investigated. Macroscopically, the lesions were creamy-white, had a villus-like structure and were localised in the operculum, lateral line and fins. Histopathological studies showed that lesions were the result of hyperplasia of fibrous connective tissue and epidermal tissues. The cells of both tissues were faintly stained by HxE and PAS. Under light microscopy, cytoplasmic vacuolation in epidermal cells in the lesions was prominent. Mucous cells were relatively common and larger than normal. Chemotherapy effectively cured all lesions.

A trivalent Escherichia coli vaccine (TECA) was prepared from serotypes O1, O2 and O78 and one series of TECA was combined with an inactivated Newcastle Disease vaccine. TECA and combined vaccine were experimentally investigated for immunogenic effects in pullets and hens. This study was carried out at two stages consisting of rearing and laying periods: (1) A number of fourty chicks aged 15 days were subcutaneously vaccinated with 0.2 ml TECA . The effects of 90 Hz and 5 mT EMF on blood Ca^{++}, P^{+++}, Na^{+}, K^{+} and Cl^{-} levels were investigated. For this purpose, 72 Swiss-Albino mice were used. On days 15, 30 and 45, blood was taken from both the experimental and control groups. A statistically significant decrease in the calcium level was only observed on day 30. On days 15 and 45, a statistically significant increase in the phosphorus level was determined. In this study, 120 British male mice, weighing 35-40 g, and 5 months old were used. Three groups, one control (Group I) and two experimental (Groups II and III), were formed. One experimental group (Group II) received 60 Hz and 5 mT, while the other (Group III) received 90 Hz frequency and 5 mT magnetic flux density efficacy electromagnetic field (EMF), between 900 and 1700. As regards fiber diameter quality and yields, Turkish Merinos have proven successful. They have better fertility and meat yields than native species bred in Turkey. It is of significance to know the similarity degree between Turkish Merinos and Deutsches Merinofleischschaf. Various methods of statistical analysis may be employed to determine the similarity degree between related species. One of the methods to be employed is cluster analysis. This study was performed to establish techniques used in the isolation and identification of Vibrio parahaemolyticus and investigate the presence of V. parahaemolyticus in fish and clams. Between June 1998 and June 2000, a total of 46 fish samples including 14 anchovies, 12 whiting, 10 horse mackerel and 10 sardines collected from fish markets in Bursa and a total of 70 clam samples collected from shellfish stations in Sakarya and Kocaeli districts were tested for V. parahaemolyticus. This study was conducted with the aim of identifying the tick species infesting cattle, sheep and goats and their seasonal activities in the Burdur area of Turkey. The study was carried out between 1 September 1999 and 31 August 2000 on animals owned by local farmers in Burdur and 14 surrounding villages. The study centres were visited twice a month. This study was conducted to determine the presence and contamination levels of Listeria spp. in 100 samples of chicken meat, including 40 mince meat samples, 30 meatball samples and 30 burger samples which were retailed fresh in the markets of Ankara. USDA/FSIS suggested methods were used for the isolation and identification of Listeria spp., while the number of Listeria spp. was detected by the Most Probable Number (MPN) technique. In this study, the oviducts of 10 laying hens aged 8 months and 10 oviducts of pullets aged 2 months were comparatively examined. The oviducts of hens consisted of five regions: infundibulum, magnum, isthmus, uterus and vagina. Tissue samples were taken from the five oviduct regions. The samples were fixed in 10% neutral formalin and embedded in paraffin and then cut into 6 micron sections. Tissue sections were stained with triple, PAS, PAS/Ab, Ab, performic acid/Ab and examined with a light microscope. The epithelium of the oviduct consisted of ciliated and secretory cells.
