Functional Characterization And Expression Of A Cytosolic Iron-Superoxide Dismutase From Cowpea Root Nodules, Jose F. Moran, Euan K. James, Mario C. Rubio, Gautam Sarath, Robert V. Klucas, Manuel Becana

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

An iron-superoxide dismutase (FeSOD) with an unusual subcellular localization, VuFeSOD, has been purified from cowpea (Vigna unguiculata) nodules and leaves. The enzyme has two identical subunits of 27 kD that are not covalently bound. Comparison of its N-terminal sequence (NVAGINLL) with the cDNA-derived amino acid sequence showed that VuFeSOD is synthesized as a precursor with seven additional amino acids. The mature protein was overexpressed in Escherichia coli, and the recombinant enzyme was used to generate a polyclonal monospecific antibody. Phylogenetic and immunological data demonstrate that there are at least two types of FeSODs in plants. An enzyme …

John Owens: Speeches and Appearances

We begin our Master Conservationist recognition in Production Agriculture with the Buskirk Family of Hemingford in the Panhandle. Will David and Toni Buskirk, Dean and Sonya Buskirk, Chad Buskirk, and Dustin and Shelly McConville, please come forward? They started with a half-section which has grown to 2,855 acres, as well as a tree-planting business. The family formed a partnership in 1995 and named it “Trees are Us” in 1998. They have used no-till farming on 1,000 acres of cropland since 1980 and planted over 165,000 trees. A low-pressure irrigation system saves about 400 acre inches of water every growing season. …

Research On The Genetics Of Screwworms In Lincoln, Nebraska, Steven Skoda, Dennis Berkebile, Margaret Allen

Department of Entomology: Faculty Publications

The success of the screwworm eradication program - these pests have been eliminated from the North American continent to Panama - has been facilitated by insightful research. The recent research on screwworms by ARS scientists at Lincoln, NE has concentrated on 1) discovering molecular markers useful in population genetics and population identification; 2) genetic research useful in developing a genetic sexing strain (males-only) in screwworms. Amplified fragment length polymorphisms (AFLP) have been studied in screwworms; this technique shows the potential for use in population identification and population genetics. AFLP's will be further explored to determine their utility: a grant proposal …

Integrated Weed Management Plan For The Lower Las Vegas Wash, Elizabeth Bickmore

Publications (WR)

The Las Vegas Wash (Wash) is the primary drainage for the metropolitan Las Vegas Valley, eventually finding its way into the Colorado River watershed system. Pursuant to the recommendations of the Water Quality Citizens Advisory Committee, the Las Vegas Wash Coordination Committee (LVWCC) was formed in 1998 to develop a comprehensive plan for the long-term stabilization and management of the Wash. Consisting of representatives from 28 government agencies, businesses, environmental groups and citizens, the LVWCC formulated the Las Vegas Wash Comprehensive Adaptive Management Plan (CAMP) and designated the Southern Nevada Water Authority (SNWA) as the lead agency for the implementation …

Effect Of Sucrose Ester Addition On Nucleation And Growth Behavior Of Milk Fat-Sunflower Oil Blends, Marina Cerdeira, Silvana Martini, Richard W. Hartel, Maria Lidia Herrera Sep 2003

Effect Of Sucrose Ester Addition On Nucleation And Growth Behavior Of Milk Fat-Sunflower Oil Blends, Marina Cerdeira, Silvana Martini, Richard W. Hartel, Maria Lidia Herrera

Nutrition, Dietetics, and Food Sciences Faculty Publications

The effects of addition of the sucrose esters (SE) P-1670, P-170, and S-170 to a high-melting fraction of milk fat (HMF) and its blends with sunflower oil (SFO) on nucleation and growth were studied by laser polarized light turbidimetry and polarized light microscopy (PLM). The three SE delayed nucleation of HMF at the temperatures selected. P-1670 did not modify average crystal size after 3 h at crystallization temperature (Tc) or crystal size distribution and modified crystallization kinetics only slightly. P-170 and S-170, however, markedly diminished crystal size and narrowed crystal size distribution. Activation free energies of nucleation at equivalent supercooling, …

Chromosome-Wide Control Of Meiotic Crossing Over In C. Elegans, Kenneth J. Hillers, Anne M. Villeneuve Sep 2003

Chromosome-Wide Control Of Meiotic Crossing Over In C. Elegans, Kenneth J. Hillers, Anne M. Villeneuve

Biological Sciences

A central event in sexual reproduction is the reduction in chromosome number that occurs at the meiosis I division. Most eukaryotes rely on crossing over between homologs, and the resulting chiasmata, to direct meiosis I chromosome segregation, yet make very few crossovers per chromosome pair [1] and [2]. This indicates that meiotic recombination must be tightly regulated to ensure that each chromosome pair enjoys the crossover necessary to ensure correct segregation. Here, we investigate control of meiotic crossing over in Caenorhabditis elegans, which averages only one crossover per chromosome pair per meiosis [3] and [4], by constructing genetic maps …

Cultivation Of Tropheryma Whipplei From Cerebrospinal Fluid, Matthias Maiwald, Axel Von Herbay, David N. Fredricks, Cleber C. Ouverney, Jon C. Kosek, David A. Relman

Faculty Publications, Biological Sciences

Whipple disease (WD) is a systemic disorder caused by the bacterium Tropheryma whipplei. Since the recognition of a bacterial etiology in 1961, many attempts have been made to cultivate this bacterium in vitro. It was eventually isolated, in 2000, from an infected heart valve, in coculture with human fibroblasts. Here we report the isolation of 2 new strains of T. whipplei from cerebrospinal fluid (CSF) of 2 patients with intestinal WD but no neurological signs or symptoms. One culture-positive specimen was obtained before treatment; the other was obtained 12 months after discontinuation of therapy, at a time of intestinal remission. …

Parameter Estimation For Distributed Delay Based Population Models From Laboratory Data: Egg Hatching Of Oulema Duftschmidi Redthenbacher (Coleoptera, Chrysomelidae) As An Example, Maurizio Severini, Johann Baumgärtner, Lidia Limonta

All Peer-Reviewed Publications

The cohort development of poikilotherms under favorable temperature conditions may be described by the time-invariant distributed delay models that require three parameters, i.e. the thermal threshold T0, the thermal constant W and the variability parameter H representing distributed maturation times, also known as time distributed or stochastic development. Here, a parsimonious method is developed that uses stage-frequency matrices obtained under controlled conditions. The analysis of these matrices permits the estimation of the three parameters, while the incorporation of both developmental threshold and thermal constant into the time-invariant distributed delay model permits the representation of stochastic cohort development under constant temperatures. …

A tobacco mosaic virus (TMV)-based vector was utilized for expression of a cytosolic form of the bovine herpesvirus type 1 (BHV-1) protein glycoprotein D (gDc). Nicotiana benthamiana plants were harvested 7 days after inoculation with RNA transcripts derived from the TMV-gDc recombinant virus. Recombinant gDc protein of expected electrophoretic mobility accumulated in inoculated leaves to a concentration of about 20 μg/g of fresh leaf tissue. Oil-based vaccines were formulated with crude foliar extracts to immunize mice parentally. After a single injection, animals developed a sustained and specific response to both the isolated gD and native virus particles. Cattle vaccinated with …

Book Review: Insecta Coleoptra Chironidae (Huchet, J.-B. 2003), Andrew B. T. Smith

University of Nebraska State Museum: Entomology Papers

The Fauna of Madagascar series has been around for almost 50 years. In that time authors have published on a variety of taxa that occur on this remarkable island. Throughout the history of the series there has been a heavy emphasis on beetles. The latest volume by Jean-Bernard Huchet is no exception. Huchet is an expert on Chironidae (or Chironinae depending on which scarab classification scheme you ascribe to). He is in the process of giving a much-needed taxonomic overhaul to the group. Chironidae are odd-looking scarabs that occur in Africa, Asia (mainly India), and Madagascar and are allied taxonomically …

A Neuronal-Specific Differentiation Protein That Directly Modulates Retinoid Receptor Transcriptional Activation, Kenneth W. Henry Ii, Michael L. Spencer, Maria Theodosiou, Dingyuan Lou, Daniel J. Noonan

Molecular and Cellular Biochemistry Faculty Publications

BACKGROUND: The specificity of a nuclear receptor's ability to modulate gene expression resides in its ability to bind a specific lipophilic ligand, associate with specific dimerization partners and bind specific DNA sequences in the promoter regions of genes. This sequence of events appears to be the basis for targeting an additional regulatory complex composed of a variety of protein and RNA components that deliver signals for facilitation or inhibition of the RNA polymerase complex. Characterization of the tissue and cell-specific components of these coregulatory complexes appear to be integral to our understanding of nuclear receptor regulation of transcription.

Topic: Nu Program Highlights In The Areas Of Food, Agriculture And Natural Resources, And The Future Of The Nebraska Agricultural Industry, John Owens

John Owens: Speeches and Appearances

Tonight I would like very much to talk with you about the good news of the Institute of Agriculture and Natural Resources at the University of Nebraska- Lincoln.

It is a pleasure to talk about good news. For the last two-plus years of my going-on three years in Nebraska, we have had a great deal of painful news to discuss, as Nebraska's budget crisis affected its land-grant university through budget cuts. Throughout the university and certainly in the Institute of Agriculture and Natural Resources, cuts occurred that we did not want to make and never would have considered had they …

On Friction And Surface Cracking During Sliding Of Ice On Ice, Maurine Montagnat, Erland M. Schulson

Dartmouth Scholarship

As a complement to earlier measurements on the friction of both granular fresh-water ice and S2 columnar salt-water ice, new experiments were performed on the friction of S2 columnar fresh-water ice sliding against itself at low velocities (5 × 10−7 to 5 × 10−1 m s−1) and at −10°C, using the same double-shear device as was used earlier. The results showed that under a given set of experimental conditions the kinetic coefficient of friction of S2 fresh-water ice compares favorably with that of the other two variants. The experiments also revealed friction-induced surface cracks and recrystallized grains. These deformation features are explained, …

Tnt Biotransformation And Detoxification By A Pseudomonas Aeruginosa Strain, Byung-Taek Oh, Patrick J. Shea, Rhae A. Drijber, Galina K. Vasilyeva, Gautam Sarath

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Successful microbial-mediated remediation requires transformation pathways that maximize metabolism and minimize the accumulation of toxic products. Pseudomonas aeruginosa strain MX, isolated from munitionscontaminated soil, degraded 100 mg TNT L-1 in culture medium within 10 h under aerobic conditions. The major TNT products were 2-amino-4,6-dinitrotoluene (2ADNT, primarily in the supernatant) and 2,2′- azoxytoluene (2,2′AZT, primarily in the cell fraction), which accumulated as major products via the intermediate 2-hydroxylamino-4,6-dinitrotoluene(2HADNT). The 2HADNT and 2,2′AZT were relatively less toxic to the strain than TNT and 2ADNT. Aminodinitrotoluene (ADNT) production increased when yeast extract was added to the medium. While TNT transformation rate was …

The Evidence For Pain In Fish: The Use Of Morphine As An Analgesic, Lynne U. Sneddon

Veterinary Science and Medicine Collection

This paper discusses the evidence for pain perception in fish and presents new data on morphine analgesia in fish. Recent anatomical and electrophysiological studies have demonstrated that fish are capable of nociception, the simple detection of a noxious, potentially painful stimulus and the reflex response to this. To prove pain perception, it must be demonstrated that an animal’s behaviour is adversely affected by a potentially painful event and this must not be a reflex response. The present study examined the acute effects of administering a noxious chemical to the lips of rainbow trout (Oncorhynchus mykiss) to assess what changes occurred …
