Generation Of High Efficiency Ssdna Hybridization Probes By Linear Polymerase Chain Reaction (Lpcr), Gregory W. Konat

Scanning Microscopy

The polymerase chain reaction (PCR) methodology can be employed to produce DNA hybridization probes. The major advantages of this paradigm over other techniques include superior specific activity of the probes, the versatility of sequence selection, the ability to produce short probes, and the simplicity of the procedure. We have further improved the efficiency of PCR probes by generating single stranded (ssDNA) probes that do not reanneal with themselves in solution, and hence, their availability for the interaction with the complementary sequences of the target is profoundly increased. Protocols for 32P-dCTP labeled and digoxigenin-dUTP labeled probes have been elaborated to …

Feeding Habits Of Juvenile Largemouth Bass Micropterus Salmoides (Lacepede), Thomas R. Goodwill Jul 1996

Feeding Habits Of Juvenile Largemouth Bass Micropterus Salmoides (Lacepede), Thomas R. Goodwill

Honors Theses

The stomachs of 110 juvenile largemouth bass, Micropterus salmoides (Lacepede) were examined to learn the early feeding habits of bass in a brackish environment. Bass were found to have five primary food sources: Taphromysis bowmani, Cloeon sp., Gammarus mucronatus, Micropterus salmoides, and Lepidophthalmus louisianum. Three feeding trends were observed during the course of the study. First, bass seemed to display prey selection. Second, bass length increased as prey size increased. Third, the types of prey eaten by the bass changed as the bass increased in size.

Dairy Science

Transgenic mice were produced by microinjection of a DNA construct composed of the bovine κ-casein (κ-CN) cDNA under the control of the goat β-CN 5' promoter elements and 3' flanking regions into pronuclear-stage embryos. The gene construct targeted the expression of bovine κ-CN RNA to the mammary gland and secretion of bovine κ-CN in the milk. In the three lines studied (BC-7, BC-31 and BC-67) the transgene was stably integrated and propagated as a Mendelian locus. Expression of the bovine protein in lactating mice from the three transgenic lines was demonstrated by northern and western blots. In ten different tissues …

Habitat Sampling And Habitat Selection By Female Wild Turkeys: Ecological Correlates And Reproductive Consequences, A. V. Badyaev, Thomas E. Martin, W. J. Etges Jul 1996

Habitat Sampling And Habitat Selection By Female Wild Turkeys: Ecological Correlates And Reproductive Consequences, A. V. Badyaev, Thomas E. Martin, W. J. Etges

Wildlife Biology Faculty Publications

Habitat sampling can allow much more effective habitat selection for longterm activities such as nesting and may be directly linked to fitness. We studied the process of habitat sampling and selection in female Wild Turkeys (Meleagris gallopavo) in the Arkansas Ozarks. In particular, we tested the prediction that movements prior to selecting nesting habitat correlate with the quality of selected habitat. Our results supported the prediction that greater habitat sampling (as reflected by greater area covered prior to nesting) allows acquisition of better nesting habitat; greater movements were correlated with choice of better nesting sites with more cover that allow …

Biological Sciences Faculty Publications

We characterized the gene required to initiate the degradation of 2,4-dichlorophenoxyacetic acid (2,4-D) by the soil bacterium Burkholderia sp. strain TFD6, which hybridized to the tfdA gene of the canonical 2,4-D catabolic plasmid pJP4 under low-stringency conditions. Cleavage of the ether bond of 2,4-D by cell extracts of TFD6 proceeded by an (alpha)-ketoglutarate-dependent reaction, characteristic of TfdA (F. Fukumori and R. P. Hausinger, J. Bacteriol. 175:2083-2086, 1993). The TFD6 tfdA gene was identified in a recombinant plasmid which complemented a tfdA transposon mutant of TFD6 created by chromosomal insertion of Tn5. The plasmid also expressed TfdA activity in Escherichia coli …

Characterization Of A Chromosomally Encoded 2,4-Dichlorophenoxyacetic Acid/A-Ketoglutarate Dioxygenase From Burkholderia Sp. Strain Rasc, Yuichi Suwa, Alice D. Wright, Fumitasu Fukimori, Kathy A. Nummy, Robert P. Hausinger, William E. Holben, Larry J. Forney Jul 1996

Characterization Of A Chromosomally Encoded 2,4-Dichlorophenoxyacetic Acid/A-Ketoglutarate Dioxygenase From Burkholderia Sp. Strain Rasc, Yuichi Suwa, Alice D. Wright, Fumitasu Fukimori, Kathy A. Nummy, Robert P. Hausinger, William E. Holben, Larry J. Forney

Biological Sciences Faculty Publications

The findings of previous studies indicate that the genes required for metabolism of the pesticide 2,4-dichlorophenoxyacetic acid (2,4-D) are typically encoded on broad-host-range plasmids. However, characterization of plasmid-cured strains of Burkholderia sp. strain RASC, as well as mutants obtained by transposon mutagenesis, suggested that the 2,4-D catabolic genes were located on the chromosome of this strain. Mutants of Burkholderia strain RASC unable to degrade 2,4-D (2,4-D- strains) were obtained by insertional inactivation with Tn5. One such mutant (d1) was shown to have Tn5 inserted in tfdARASC, which encodes 2,4-D/alpha-ketoglutarate dioxygenase. This is the first reported example of a …

Genomics: Past & Future

AT THE END OF THE ROAD in Little Cottonwood Canyon, near Salt Lake City, Alta is a place of near-mythic renown among skiers. In time it may well assume similar status among molecular geneticists. In December 1984, a conference there, co-sponsored by the U.S. Department of Energy, pondered a single question: Does modern DNA research offer a way of detecting tiny genetic mutations—and, in particular, of observing any increase in the mutation rate among the survivors of the Hiroshima and Nagasaki bombings and their descendants? In short the answer was, Not yet. But in an atmosphere of rare intellectual fertility, …

Oxidative Stress Detection With Escherichia Coli Harboring A Katg'::Lux Fusion, S. Belkin, D. R. Smulski, Amy Cheng Vollmer, T. K. Van Dyk, R. A. Larossa

Biology Faculty Works

A plasmid containing a transcriptional fusion of the Escherichia coli katG promoter to a truncated Vibrio fischeri lux operon (luxCDABE) was constructed. An E. coli strain bearing this plasmid (strain DPD2511) exhibited low basal levels of luminescence, which increased up to 1,000-fold in the presence of hydrogen peroxide, organic peroxides, redox-cycling agents (methyl viologen and menadione), a hydrogen peroxide-producing enzyme system (xanthine and xanthine oxidase), and cigarette smoke. An oxyR deletion abolished hydrogen peroxide-dependent induction, confirming that oxyR controlled katG'::lux luminescence. Light emission was also induced by ethanol by an unexplained mechanism. A marked synergistic response was observed when cells …

Female Choice In Sage Grouse: The Roles Of Attraction And Active Comparison, Robert M. Gibson Jul 1996

Female Choice In Sage Grouse: The Roles Of Attraction And Active Comparison, Robert M. Gibson

School of Biological Sciences: Faculty Publications

Previous studies of female choice in sage grouse Centrocercus urophasianus have implicated both the acoustic quality and repetition rate of the stereotyped strut display as putative cues for female choice. Stages in the choice process at which specific components of male courtship display influence female decisions were investigated using field observations of female premating behavior. Females visited a subset of territorial males and then actively chose one of these as a mate. The order in which males were visited suggested that females searched until an acceptable mate was found, rather than employing a “best-of-n” tactic. Numbers of females visiting a …

Patch Diseases In Kentucky Bluegrass Lawns, Paul C. Vincelli, A. J. Powell Jr. Jul 1996

Patch Diseases In Kentucky Bluegrass Lawns, Paul C. Vincelli, A. J. Powell Jr.

Agriculture and Natural Resources Publications

“Patch diseases” can be very destructive when Kentucky bluegrass is grown under intensive management. Two patch diseases with similar symptoms can occur. Necrotic ring spot often appears in early summer. Summer patch, the more common disease in Kentucky landscapes, develops in middle to late summer.

These diseases are often a concern in lawns under intensive management for two reasons:

  1. Improper application of certain lawn management practices, such as close mowing, can aggravate disease development.
  2. When a lawn receives significant inputs of fertilizer and irrigation and frequent mowing, the homeowner or landscape manager usually has high aesthetic expectations and tolerates …

Impact Of A Parasitic Plant On The Structure And Dynamics Of Salt Marsh Vegetation, Steven C. Pennings, Ragan M. Callaway Jul 1996

Impact Of A Parasitic Plant On The Structure And Dynamics Of Salt Marsh Vegetation, Steven C. Pennings, Ragan M. Callaway

Biological Sciences Faculty Publications

We investigated the effect of a native parasitic plant, Cuscuta salina, on the structure and dynamics of the plant community in a California salt marsh. Cuscuta was common in the middle marsh zones. The abundance of Cuscuta was positively correlated with the abundance of Limonium californicum at two sampling scales (0.25— and ≈\approx40—m2 quadrats). Sampling at the scale of individual plants indicated that the dominant plant in the marsh, Salicornia virginica, was preferred by Cuscuta as a host over Arthrocnemum subterminale, Limonium californicum, and Frankenia salina. This result was confirmed with host—choice experiments in the field. Based on spatial …

The Real Cost Of Beef: A Monetary Assessment Of The Environmental Degradation Caused By Beef Production, Carolyn Babcock Jul 1996

The Real Cost Of Beef: A Monetary Assessment Of The Environmental Degradation Caused By Beef Production, Carolyn Babcock

UNLV Theses, Dissertations, Professional Papers, and Capstones

The environmental degradation caused by beef production is severe. The current literature assesses the ecological damage, but falls short of assigning a per pound dollar amount to reflect the real cost of beef. In the United States, most of the environmental focus has been centered around the use of public lands for grazing and the grazing fee the government considers appropriate. The fee covers the maintenance of the grazing program, which includes maintenance of the actual land. This paper will concentrate on beef-related environmental degradation and select specific damage for monetary valuation.

Morphometry Of Juvenile And Subadult Loligo Pealei And L. Plei From The Northern Gulf Of Mexico, Guillermo Sánchez, Harriet M. Perry, Christine B. Trigg, Michael Vecchione, Clyde Roper, Patricia M. Biesot

Faculty Publications

Two species of Loligo are abundant in northern Gulf waters: the long-finned squid, Loligo pealei. and the arrow squid, Loligo plei. Variability within species and similarities between the species often hamper accurate identification. The two species more closely resemble each other in areas of sympatry. and there is overlap in almost all of the diagnostic characters. Small specimens of Loligo are not easily identified. and there are few studies detailing their morphometry. Because of the taxonomic uncertainties associated with the identification of juveniles and subadults of L. pealei and L. plei, the species were differentiated by isoelectric …

Characterization Of A Newly Cloned Dna Damage- Inducible Gene From Saccharomyces Cerevisiae Strain D43, June-Home Chen Jul 1996

Characterization Of A Newly Cloned Dna Damage- Inducible Gene From Saccharomyces Cerevisiae Strain D43, June-Home Chen

Master's Theses

The identification and characterization of DNA damage-inducible genes is an important topic in the study of the integrity of the cellular genome. Previously, a TnlO-lacZ derivative was inserted randomly into yeast cells and the resulting mutants were screened for insertions in DNA damage-inducible genes. S. cerevisiae strain D43 was identified as one such TnlO-lacZ insertion mutant. DNA sequencing results show the insertion in D43 is located within the TDH2 gene, which encodes glyceraldehyde-3-phosphate dehydrogenase. Treatment of strain D43 with DNA damaging agents and heat shock suggest TDH2 is responsive to DNA damage, but not to heat shock. Haploid mutants carrying …

Lead Activation Of A Developmentally Regulated Calcium Channel In Rat Hippocampal Nerve Terminals, Troy E. Rhodes Jul 1996

Lead Activation Of A Developmentally Regulated Calcium Channel In Rat Hippocampal Nerve Terminals, Troy E. Rhodes

Theses and Dissertations in Biomedical Sciences

Low level lead (Pb2+) exposure may produce lasting deficits in learning and memory by altering calcium (Ca2+) dependent processes. Isolated presynaptic nerve terminals from rat hippocampus were loaded with the intracellular (Ca2+) indicator Fura-2. The changes in cytoplasmic free calcium ([Ca2+]i) were measured by stopped-flow fluorescence spectroscopy following depolarization with elevated potassium on a millisecond time scale (Lentzner et al., 1992). Depolarization promoted a rapid increase in Ca2+i which occured in two kinetically distinguishable phases: a fast component, representing the activity of rapidly inactivating Ca2+ channels (τ …

Linking Local Species Interactions To Rates Of Speciation In Communities, Mark A. Mcpeek Jul 1996

Linking Local Species Interactions To Rates Of Speciation In Communities, Mark A. Mcpeek

Dartmouth Scholarship

Regional species diversity limits the diversity of local communities by defining the pool of species that are available to colonize sites. Biogeographical processes that influence speciation and extinction rates determine the size and composition of this regional species pool. Community ecologists are beginning to recognize the importance of these biogeographical processes in influencing diversity patterns among local communities, but the potential for local interactions to influence biogeographical processes, especially speciation, has been little considered. In this paper I discuss one mechanism by which variation in the strengths of local interactions can influence the potential for population differentiation and thus for …
