Iron Uptake By Symbiosomes From Soybean Root Nodules., Kristin Levier, David A. Day, Mary Lou Guerinot
Iron Uptake By Symbiosomes From Soybean Root Nodules., Kristin Levier, David A. Day, Mary Lou Guerinot
Dartmouth Scholarship
To identify possible iron sources for bacteroids in planta, soybean (Glycine max L. Merr.) symbiosomes (consisting of the bacteroid-containing peribacteroid space enclosed by the peribacteroid membrane [PBM]) and bacteroids were assayed for the ability to transport iron supplied as various ferric [Fe(III)]-chelates. Iron presented as a number of Fe(III)-chelates was transported at much higher rates across the PBM than across the bacteroid membranes, suggesting the presence of an iron storage pool in the peribacteroid space. Pulse-chase experiments confirmed the presence of such an iron storage pool. Because the PBM is derived from the plant plasma membrane, we reasoned that it …
Spikelet And Floret Naming Scheme For Grasses With A Spike Inflorescence, Wallace Wilhelm, Gregory S. Mcmaster
Spikelet And Floret Naming Scheme For Grasses With A Spike Inflorescence, Wallace Wilhelm, Gregory S. Mcmaster
United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications
Effective schemes to name uniquely and define parts of grass plants allow researchers to communicate results of their experiments accurately and precisely. Naming schemes for vegetative parts of grass plants have been developed; however, no system exists to name uniquely parts of the inflorescence. In this report, we present a method to extend existing systems to name parts of grass spikes. Spikelet positions are denoted by the letter S and numbered acropetally along the rachis. For example, the third spikelet position from the base of the rachis is named S3. Likewise, floret positions along the rachilla are named F (or …

Application Of Monoclonal Antibodies Against Creatine Kinase Mm And Bb Isoenzymes For Creatine Kinase Mb Isoenzyme Detection, Lei-Chieh Chuang
Chemistry & Biochemistry Theses & Dissertations
Nondiagnostic ECGs and unknown characteristic chest pains increase the diIIiculty of Acute Myocardial Infarction (AMI) diagnosis. Biochemical markers, such as creatine kinase MB isoenzymes (CK-MB) and myoglobin, are employed for the early detection of AMI. Compared to myoglobin, CK-MB reveals a higher specific indicator for AMI detection owing to its abundance in cardiac tissue. Hence, a successful CK-MB detection is expected to improve AMI diagnosis and therapy.
A CK-MB detecting ELISA system was set up based on a sandwich assay principle by using specific CK-B and CK-M monoclonal antibodies. The dimeric nature of the CK-MB isoenzyme provides two epitopes on …

Comparison Of Positive Screening And Confimatory Results From Federally Mandated Drug Testing Of Urine, Mary M. Stuck
Community & Environmental Health Theses & Dissertations
The purpose of this study was to analyze the accuracy of FDA approved immunoassays for the detection of drug positive urine samples.
Federal civilian employees are tested under the strict protocol of the Department of Health and Human Services mandatory guidelines for federal workplace drug testing programs. The guidelines provide for a two instrument testing protocol for the analysis of urine samples. The first is an FDA approved immunoassay. Samples which test positive on this screening immunoassay are then submitted for confirmatory testing with the "gold standard" gas chromatography/mass spectrometry.
Agency monthly drug testing summaries, required under the DHHS guidelines …
Neuronal Toxicity Of Humoral Factors From Patients With Diabetic Neuropathy And Mechanism Of Neuronal Death On Cultured Adrenergic Neurons: Studies Of Autoimmune Pathogenesis In Diabetic Neuropathy, Dong Liu
Theses and Dissertations in Biomedical Sciences
Diabetic neuropathy is an extremely common medical problem because of the high frequency of diabetes and its complications. Diabetic neuropathy consists of not one, but many patterns of nerve injury, each potentially with its own discrete pathogenic mechanisms. Evidence is accumulating that indicates autoimmunity plays a role in diabetic peripheral neuropathy. However, the significance of immunopathogenesis of diabetic neuropathy is as yet unclear.
Using the N1E-115 mouse neuroblastoma cell line (NB) as a model of the adrenergic autonomic neuron, we found that sera of IDDM patients with neuropathy inhibit growth and differentiation of adrenergic neurons in culture (Pittenger et al, …
Molecular Phylogeny And Typing Of Blackflies (Diptera: Simuliidae) That Serve As Vectors Of Human Or Bovine Onchocerciasis, Jianming Tang, Kenneth Pruess, Eddie W. Cupp, Thomas R. Unnasch
Molecular Phylogeny And Typing Of Blackflies (Diptera: Simuliidae) That Serve As Vectors Of Human Or Bovine Onchocerciasis, Jianming Tang, Kenneth Pruess, Eddie W. Cupp, Thomas R. Unnasch
Department of Entomology: Faculty Publications
A subregion of the mitochondrial large subunit (16s) rRNA gene was amplified by polymerase chain reaction (PCR) from nine species of blackflies (Diptera: Simuliidae) which serve as natural or experimental vectors of human or bovine Onchocerca parasites. PCR products from each species of blackfly were tested by directed heteroduplex analysis (DHDA), and their genotypes established according to diagnostic banding patterns of the heteroduplex products. Three alleles of mitochondrial 16s rRNA were found to exist in members of the Simulium (Edwardsellum) damnosum sensu lato complex from West Africa, and two alleles were found in the Neotropical Simulium (Psilopelmia …

A Truncated Form Of The Pho80 Cyclin Of Saccharomyces Cerevisiae Induces Expression Of A Small Cytosolic Factor Which Inhibits Vacuole Inheritance., Teresa Nicolson, Barbara Conradt, William Wickner
Dartmouth Scholarship
Vacuoles project streams of vesicles and membranous tubules into the yeast bud where they fuse, founding the daughter cell organelle, vac5-1, which encodes a truncated form of the Pho80 cyclin, inhibits normal vacuole inheritance. An in vitro inheritance assay which measures the fusion of vacuoles serves as a model for several steps of this process. We find that cytosol isolated from the vac5-1 mutant is unable to promote the fusion of wild-type vacuoles in the in vitro assay. Wild-type vacuoles are irreversibly inactivated in a time- and temperature-dependent manner if preincubated with vac5-1 cytosol and ATP, suggesting the presence of …
Marine Pore-Water Sulfate Profiles Indicate In Situ Methane Flux From Underlying Gas Hydrate, Walter S. Borowski, Charles K. Paull, William Ussler Iii
Marine Pore-Water Sulfate Profiles Indicate In Situ Methane Flux From Underlying Gas Hydrate, Walter S. Borowski, Charles K. Paull, William Ussler Iii
EKU Faculty and Staff Scholarship
Marine pore-water sulfate profiles measured in piston cores are used to estimate methane flux toward the sea floor and to detect anomalous methane gradients within sediments overlying a major gas hydrate deposit at the Carolina Rise and Blake Ridge (U.S. Atlantic continental margin). Here, sulfate gradients are linear, implying that sulfate depletion is driven by methane flux from below, rather than by the flux of sedimentary organic matter from above. Thus, these linear sulfate gradients can be used to quantify and assess in situ methane flux, which is a function of the methane inventory below.

An assessment of 10 tetrameric restriction enzymes (TREs) was conducted by using a computer-simulated restriction fragment length polymorphism (RFLP) analysis for over 100 proximally and distally related bacterial small-subunit (SSU) rRNA gene sequences. Screening SSU rDNA clone libraries with TREs has become an effective strategy because of logistic simplicity, commercial availability, and economy. However, the rationale for selecting the type and number of TREs has not been systematically evaluated. Our objective nas to identify the optimal combination of TREs for RnP screening of cloned SSU rRNA genes from undefined bacterial clone libraries. After computer-simulated TRE digestion, the resultant fragments were …
Windbreaks: An Important Component In A Plasticulture System, Laurie Hodges, James R. Brandle
Windbreaks: An Important Component In A Plasticulture System, Laurie Hodges, James R. Brandle
Department of Agronomy and Horticulture: Faculty Publications
Windbreaks reduce wind speed and modify the microclimate in sheltered areas. Many producers use wind barriers in their production systems, but few producers recognize all of the benefits available or understand the principles involved in windbreak function and design. Wind has direct and indirect effects on plant growth and development. Direct effects include soil abrasion, increased transpiration, and lodging. Indirect effects are based on changes in the crop microclimate, which influence plant growth and yield. Windbreaks increase soil and air temperatures and can extend the growing season in sheltered areas, resulting in increased crop development, earlier crop maturity, and market …
Microhardness And Scanning Electron Microscopy Analysis Of Nd:Yag Laser And Acid Treatment Effects In Dentin, J. M. White, G. L. Adams
Microhardness And Scanning Electron Microscopy Analysis Of Nd:Yag Laser And Acid Treatment Effects In Dentin, J. M. White, G. L. Adams
Scanning Microscopy
Laser irradiation at and above parameters producing the modification threshold for dentin causes structural changes to the dentin surface. This study determined the microhardness of dentin before and after laser modification and acid treatment using a repeated-measures de-sign. Seven dentin sections (4 mm in thickness) were cut from freshly extracted non-carious third molars using a diamond saw. The middle occlusal third was used as the test dentin surface. One section served as a control (C); three received laser modification (L) and then acid treatment (L + A); and three received acid (A) and then laser treatment (A + L). Laser …

Scanning Microscopy
Using ethylene glycol (EG) and vitamin D3 as crystal-inducing diet (CID) in rats, we investigated the effect of the dosage of EG on the generation of chronic calcium oxalate (CaOx) nephrolithiasis. We collected weekly 24 hour urines and measured herein the amount of oxalate, calcium, glycosaminoglycans (GAG's), creatinine, protein, alkaline phosphatase (AP), 𝝲-glutamyl transpeptidase (𝝲GT), and N-acetyl-𝝱-glucosaminidase (NAG). The potential of these urines to inhibit crystal growth and agglomeration was also evaluated. After four weeks, the kidneys were screened by histology and radiography for the presence of CaOx crystals and the amount of kidney-associated oxalate was biochemically measured. Using 0.5 …

Abundant Protein-Containing Particles In The Sea, Richard A. Long, Farooq Azam
Faculty Publications
The interaction of bacteria with particulate organic matter has implications for organic matter cycling and bacterial ecology in the ocean. Until recently, the focus has been on 'classical' particles visible by unaided eye (marine snow) or light microscopy. Recent discoveries of several new types of abundant particles, from sub-micrometer to sub-marine snow, are changing our ideas of the physical and chemical nature of the particle field with which pelagic bacteria interact. Previous workers have discovered polysaccharide-containing (Alcian Blue stainable) transparent exopolymer particles (TEP) ranging from 3 to 100s of micrometers. Looking for additional components of the sub-marine snow particle field, …

Settlement And Early Post-Settlement Survival Of Sessile Marine Invertebrates On Topographically Complex Surfaces: The Importance Of Refuge Dimensions And Adult Morphology, Linda J. Walters, David S. Wethey
Faculty Publications
We predicted that both refuge dimension and growth form would influence settlement and short-term post-settlement success (≤7 d) of sessile marine invertebrates that live attached to hard substrata in low energy environments. Individuals with unlimited attachment to the substrata should rapidly be protected by their growth form, thus decreasing their need to settle in refuges and limiting the length of time any locations on heterogeneous substrata act as refuges. Alternatively, organisms with limited attachment to the substrata should remain susceptible to the causes of mortality for a longer time, and as a result should settle in high quality refuges (sites …
Problems In Preparation Of Chromosomes For Scanning Electron Microscopy To Reveal Morphology And To Permit Immunocytochemistry Of Sensitive Antigens, A. T. Sumner
Scanning Microscopy
Although much information about chromosome structure and behaviour has been obtained using light microscopy, greater resolution is needed for a thorough understanding of chromosome organisation. Scanning electron microscopy (SEM) can provide valuable data about these three-dimensional organelles. The introduction of methods using osmium impregnation of methanol-acetic acid-fixed chromosome spreads revolutionised matters, producing life-like images of chromosomes. Nevertheless, it became clear that osmium impregnation introduced various artefacts, although the resulting images were still useful. Methanol-acetic acid-fixed chromosomes are, in fact, flattened on the glass substratum, and the 3-dimensional appearance obtained after osmium impregnation is the result of swelling during this process. …

The Method Of Radicle Emergence In Phytolacca Americana (Pokeweed), Staci A. Van Winkle
The Method Of Radicle Emergence In Phytolacca Americana (Pokeweed), Staci A. Van Winkle
Honors Theses
Germination is a key element in the development of a plant. This process encompasses several stages, one of which is the elongation of the radicle tip. This process has been noted to occur by different methods such as rapid mitotic division or cell elongation. However, such a mechcmism has not been studied in Phytolacca americana (pokeweed). It was the intention of this study to observe at the cellular level the nature of the Phytolacca americana radicle during normal germination. This was done using a routine histological microtechnique which involves embedding in paraffin and sectioning with a microtome. The sections were …
Preparation Of Plasmid Dna In Transfection Complexes For Fluorescence And Electron Spectroscopic Imaging, Marek Malecki
Preparation Of Plasmid Dna In Transfection Complexes For Fluorescence And Electron Spectroscopic Imaging, Marek Malecki
Scanning Microscopy
The aim of this project was to develop procedures necessary to study mechanisms of receptor mediated gene transfer by means of integrated microscopy.
Plasmid DNA was incorporated into a transfection complex consisting of poly(L)lysine and transferrin to which the nuclear localization signal was conjugated. This complex was presented to cultured glioma cells. Preparation of the transfected DNA for imaging was pursued by two methods. In the first method tetramethylrhodamine, nanogold, and ferritin were linked through streptavidin to the biotinylated plasmid DNA. Trafficking of the fluorescent derivatives was studied in living cells with fluorescence microscopy. Then, selected cells were rapidly cryo-immobilized. …

Common Epidermal Lesions Of The Bowhead Whale, Balaena Mysticetus, William G. Henk, Dianna L. Mullan
Scanning Microscopy
Samples of common skin abnormalities from 23 subsistence-harvested bowhead whales (Balaena mysticetus) were examined. Most lesions fell into three broad classes: shallow lacerations, circular depressions, and epidermal sloughing. Both circular depression lesions and epidermal sloughing lesions may be divided into more than one subgroup based on morphological criteria. Examination of each of the subgroups using light and scanning electron microscopy suggests relationships among the subgroups of a class. These proposed relationships are discussed, as are some possible etiologies. Scanning electron microscopy reveals abundant bacteria and diatoms present in association with each lesion class but no evidence of a particular association …
Feasibility Exploration: "Perfectly" Integrated Crop-Livestock Production, Donald Taylor, Diane H. Rickerl
Feasibility Exploration: "Perfectly" Integrated Crop-Livestock Production, Donald Taylor, Diane H. Rickerl
Economics Research Reports (1969 - 2017)
In this research report, the following question is examined. Can individual integrated crop and cow-calf operations be simultaneously "balanced" from the standpoints of (1) amounts of manure produced "matching" (plus or minus 10%) the soil fertility needs of producers' cropland and rangeland and (2) amounts of feedgrains and roughages produced matching (plus or minus 10%) the nutrient needs of producers' livestock? Answers to the question were sought through examination of livestock manure production and utilization and livestock feedstuff production and consumption on eight South Dakota integrated crop-livestock case farms.

Molecular Studies On Biv Infection And Its Interaction With Other Bovine Viruses, Charles Wood, Harish Minocha, Yunqi Geng
Molecular Studies On Biv Infection And Its Interaction With Other Bovine Viruses, Charles Wood, Harish Minocha, Yunqi Geng
Nebraska Center for Virology: Faculty Publications
Bovine immunodeficiency virus (BIV) is a bovine lentivirus that infects animals world-wide. The authors have cloned the BIV env and gag proteins and several sub-unit peptides spanning the entire coding regions of the two proteins as fusions to the bacterial trpE protein and expressed them in Escherichia coli (Spindler et al. 1984). These recombinant proteins can readily be expressed but the expression levels of these peptides varied. When these proteins were tested against a panel of bovine sera, only sera from BIV-infected animals reacted specifically. One major antigenic determinant was identified in the cartoxyl terminus of the p26 gag protein …
