Infection of quiescent cells with the DNA tumor virus simian virus 40 induces expression of the cellular thymidine kinase (TK) gene a minimum of 10- to 20-fold, and this induction depends upon the viral protein large T antigen (T-Ag). To define both human TK promoter elements and T-Ag functional domains required for transcriptional induction, we have established a system in which stable Rat-1 transfectants harboring TK promoter-luciferase hybrid genes are infected with recombinant adenoviruses expressing either wild-type or mutant forms of T-Ag and luciferase expression is measured as an indicator of promoter activity. On the morning of November 10, 1991, a deer hunter in the Pine Ridge area of extreme northwestern Nebraska shot a young female mountain lion (Gertzen, 1991, Omaha World-Herald, November 15, Sunrise ed. p. 17sf; Henion, 1991, Omaha World-Herald, November 15, Metro ed. p. 17). Although mountain lions (Felis concolor) were part of the original mammalian fauna of Nebraska (Jones, 1949, J. Mammal. 30: 313; 1962, Bull Univ. Nebraska State Mus. 4: 87-100; 1964, Univ. Measures of juvenile abundance are presently in wide use as a key element in the management of the Atlantic States' coastal fishery resources. Estimates of the relative interannual abundance of early juveniles (age-0) generated from scientific survey programs have been found to provide a reliable and early estimator of future year class strength (Goodyear 1985, Lipcius and Van Engel 1990). The search for a molecular mechanism of auxin action has rendered the “Auxin-Binding-Protein 1” (ABP-1) the top candidate for a functional auxin receptor (Venis and Napier, 1995), although its status remains disputed (Jones, 1994; Hertel, 1995; Napier, 1995; Venis, 1995). ABP-1 had long been thought to be localized exclusively in epidermal cells in coleoptiles (Löbler and Klämbt, 1985), and thus had fitted nicely the “epidermal-growth-control-hypothesis” (Kutschera, 1987, 1992). Emergence patterns and population levels of Douglas-fir beetles (DFB), Dendroctonus pseudotsugae Hopkins, in the seventh year of infestation following the Clover Mist Fire were evaluated on the Clarks Fork Ranger District of the Shoshone National Forest. Adult emergence in 1995 returned to a single peak beginning in mid-June, about a week later than normal likely due to cold, wet spring weather. Adults of both the '93-'94 and '94-'95 generations emerged, demonstrating for the second year in a row that DFB can extend its life cycle up to two years. In North America, the Colorado potato beetle, Leptinotarsa decemlineata, is often infected with the host-specific, gut-inhabiting Colorado potato beetle spiroplasma (CPBS). CPBS is apparently a commensal, but it may be useful in biocontrol if it can be transformed to express an insect-lethal gene. Difficulty in cultivating the organism, however, has hindered the development of a suitable transformation system. In this study, we eliminated the need for coculturing CPBS with insect cells. CPBS was reliably isolated with the BBL Anaerobic GasPak Jar system (low redox, enhanced CO(inf2)), which was easier to use and less expensive than insect cell coculture methods. A …

Marine phytoplankton fix carbon dioxide primarily through the action of ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO), the first enzyme in the Calvin Cycle. Although the regulation of this enzyme has been studied in algal cultures and higher plants, little is known regarding RubisCO regulation in natural phytoplankton populations. To determine if natural communities of phytoplankton utilize transcriptional regulation to control RubisCO expression, we investigated the diel relationship between 14C carbon fixation and RubisCO large subunit (rbcL) transcript levels, rbcL DNA, chlorophyll a, autofluorescent cell counts and bacterial direct counts for natural communities of the southeastern Gulf of Mexico. We have previously demonstrated that hydrogen atoms on the hydrogen-terminated Si(100)-2xl:H surface can be extracted by applying either positive or negative voltage pulses between an STM tip and the sample surface with a certain tunneling current. This phenomenon suggests that electron excitation and field evaporation both play a role in atom transfer. Under our experimental conditions, hydrogen atoms are extracted in pairs from either a single dimer or from two adjacent dimers in neighboring rows. Hydrogen atoms can also be redeposited onto the Si(100)-2xl:H surface from a tungsten tip, which has picked up hydrogen from the sample surface through the …

The recognition that apoptosis is regulated by an evolutionarily conserved set of polypeptides from the nematode Caenorhabditis elegans to humans suggests that a conserved set of biochemical mechanism(s) may also be involved in the response. Early evidence suggested that the endogenous endonuclease implicated in apoptosis in most model systems is Ca2+ -dependent, and subsequent work from a number of independent laboratories suggests that alterations in cytosolic Ca2+ homeostasis are one of the conserved biochemical pathways regulating the response. The influence of bleeding on post-mortem tenderization of fish muscle during storage was studied. Six fish species were used for the present study. Among them, delay of post-mortem tenderization of muscle by bleeding was shown in the pelagic fishes: yellowtail, horse mackerel, and striped jack. On the other hand, bleeding had no influence on the muscle firmness of the bottom fishes: red sea bream, flatfish, and rudder-fish. According to transmission electron microscopy, degradation of pericellular collagen fibrils was delayed in bled yellowtail and horse mackerel. Laser microprobe mass spectrometry (LMMS) provides spot analysis with a lateral resolution of 1-5 μm. Focused laser ionisation, combined with mass spectrometry, yields information on elements, molecular identification of inorganic substances, and structural characterisation of organic molecules of typically less than 2-3 kDa. Quantification in complex heterogeneous systems, such as biological materials, is not possible at the present state of the art. The strength of the method lies in the qualitative information on the molecular composition of the analyte, not just element detection, with a lateral resolution at the light microscopy level. This paper deals with the ability of scanning force microscopy to determine contact forces of various materials. Indeed, with high spring constants at low relative humidity, the nature of the material can be determined by measurement of the contact force as the tip approaches. Cantilevers with a high spring constant are used to achieve solid-solid contact for the tip-sample system. The capillary force estimation provides information on the development of the height of the water meniscus formed between the tip and different surfaces depending on the relative humidity. The labeling of proteins with fluorescent compounds for microscopy has allowed a greater understanding of biological processes. The preparation of fluorescent proteins is the first step in development of their use in microscopy. Methods are described to label and characterize a protein as an example of the general approach for other proteins. Skeletal muscle alpha-actinin was labeled with either fluorescein-5-maleimide or 5-iodoaceamidofluorescein and the reaction characterized. The maleimide reaction was much more rapid and efficient than the iodoacetamide reaction giving a coupling efficiency of 65% under the given ration conditions. Pelage Coloration In Oldfield Mice (Peromyscus Polionotus): Antipredator Adaptation?, Mark C. Belk, Michael H. Smith

Reflectance spectra (in full light and moonlight) of dorsal pelage of 469 adult oldfield mice (Peromyscus polionotus) comprising nine regional samples were compared to reflectance spectra of corresponding surface and subsurface soil samples to test for a selective effect of predation on color of pelage. Mice were slightly more reddish in hue, generally had more chroma, and were less reflective than soil at all locations. Color of pelage within locations was not significantly correlated with color of soil. Among all locations, brightness of pelage and soil were significantly positively correlated. A significant new development in gold labeling for microscopy has been achieved through the use of gold cluster compounds that are covalently attached to antibodies or other probe molecules. These unique gold probes are smaller than most colloidal gold conjugates and exhibit improved penetration into tissues, higher labeling densities, and allow many new probes to be made with peptides, nucleic acids, lipids, drugs, and other molecules. A new fluorescent-gold conjugate is useful for examining localization by fluorescence microscopy, then visualizing the same label at the ultrastructural level in the electron microscope.

Female wild Japanese monkeys (Macaca fuscata), as with all male cercopithecoids, use the mesiobuccal surfaces or the elongated crests of the mandibular third premolars (P3s), as cutting blocks that wear against edges of maxillary canines during threat manifestation or food-eating. In other words, the crests of their P3s are honed with the maxillary canines. The crests become sloped during growth and more heavily striated with the advance of age. The number, directions, lengths, and widths of these striations have been analyzed quantitatively using scanning electron microscopy (SEM). The ability to deal with a huge number of independent and heterogeneous repositories is the most critical problem in Global Information Systems. One approach to enable efficient query processing is by utilizing semantic descriptions (organized as ontologies) of such repositories whenever available.

In this context semantic relationships among ontologies can be used Query Processors. Three kind of relationships are considered: synonyms and hypernyms. Using synonyms the semantic of the query is preserved; however, when synonyms are not available and hypernyms or hyponyms are used there exists some loss of information that must be measured.

The developmental time scale of the cochlea varies from species to species. We investigate here the condition of the neonatal cochlea in the chinchilla, a species increasingly used in auditory research. We have examined the morphology of cochlear hair cells using scanning microscopy, and the development of auditory function during the first postnatal month by monitoring auditory brainstem evoked responses (ABR). We find that although there were some outer hair cell kinocilia present in middle and apical areas, the hair cells otherwise were mature at 24 hours after birth. Furthermore, cochlear auditory thresholds are adult-like at birth. However, whilst there …

Advances in in vitro fertilization and in vitro culture techniques have allowed considerable progress in identifying physiological requirements of mammalian embryos. Parrish et al. (11) reported a major breakthrough on in vitro fertilization when his group identified heparin as an important factor for the capacitation of spermatozoa. Capacitation is necessary for fertilization of matured oocytes. During the precoculture era of embryos (prior to 1980), the development of early preirnplantation embryos was vary limited regardless of medium, medium supplement, gas atmosphere, osmolarity or pH used (7).
