In Vivo Detection Of Porcine Reproductive And Respiratory Syndrome Virus Rna By In Situ Hybridization At Different Times Post-Infection, Jung-Hyang Sur, Vicki Cooper, Judith A. Galeota, Richard A. Hesse, Alan R. Doster, Fernando A. Osorio

Nebraska Center for Virology: Faculty Publications

We studied the distribution of porcine reproductive and respiratory syndrome virus (PRRSV) RNA in tissues by in situ hybridization at different times postinfection (p.i.). The probe used for in situ hybridization was prepared by reverse transcription of PRRSV RNA, followed by PCR amplification of the cDNA. The sequence amplified corresponded to 433 bp from PRRSV open reading frame 7, which is contained in the nucleocapsid protein gene and which is highly conserved in both European and American strains (H. Mardassi, L. Wilson, S. Mounir, and S. Dea, J. Clin. Microbiol. 32:2197–2203, 1994). An immunohistochemical technique was used to detect PRRSV …

Characterization Of Experimentally Induced, Nonaflatoxigenic Variant Strains Of Aspergillus Parasiticus., Shubha Kale Ireland, J. W. Cary, D. Bhatnagar, J. W. Bennett

Faculty and Staff Publications

Six previously isolated, nonaflatoxigenic variants of Aspergillus parasiticus, designated sec mutants, were characterized morphologically by electron microscopy, biochemically by biotransformation studies with an aflatoxin precursor, and genetically by Northern (RNA) hybridization analysis of aflatoxin biosynthetic gene transcripts. Scanning electron micrographs clearly demonstrated that compared with the parental sec+ forms, the variant sec forms had an abundance of vegetative mycelia, orders of magnitude reduced number of conidiophores and conidia, and abnormal metulae. Conidiospores were detected in sec cultures only at higher magnifications (x500), in contrast to the sec+ (wild-type) strain, in which abundant conidiospores (masking the vegetative mycelia) were observed even …

Pancreatic Expression Of Interleukin-4 Abrogates Insulitis And Autoimmune Diabetes In Nonobese Diabetic (Nod) Mice., Regula Mueller, Troy Krahl, Nora Sarvetnick

Journal Articles: Regenerative Medicine

Diabetes in nonobese diabetic (NOD) mice is a T cell-dependent autoimmune disease. The destructive activities of autoreactive T cells have been shown to be tightly regulated by effector molecules. In particular, T helper (Th) 1 cytokines have been linked to diabetes pathogenesis, whereas Th2 cytokines and the cells that release them have been postulated to be protective from disease. To test this hypothesis, we generated transgenic NOD mice that express interleukin (IL) 4 in their pancreatic beta cells under the control of the human insulin promoter. We found that transgenic NOD-IL-4 mice, both females and males, were completely protected from …

Monoclonal Antibodies For Detection Of The H7 Antigen Of Escherichia Coli, Yongsheng He, James E. Keen, Ralph B. Westerman, E. Travis Littledike, Jimmy Kwang

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Two murine monoclonal antibodies (MAbs) (2B7 and 46E9-9) reactive with the H7 flagellar antigen of Escherichia coli were produced and characterized. A total of 217 E. coli strains (48 O157:H7, 4 O157:NM, 23 O157:non-H7, 22 H7:non-O157, and 120 non-O157:non-H7), 17 Salmonella serovars, and 29 other gramnegative bacteria were used to evaluate the reactivities of the two MAbs by indirect enzyme-linked immunosorbent assay (ELISA). Both MAbs reacted strongly with all E. coli strains possessing the H7 antigen and with H23- and H24-positive E. coli strains. Indirect ELISA MAb specificity was confirmed by inhibition ELISA and by Western blotting (immunoblotting), using partially …

Activation Of The Human Thymidine Kinase (Tk) Promoter By Simian Virus 40 Large T Antigen Requires Both The T Antigen Prb Family-Binding Domain And Tk Promoter Sequences Resembling E2f-Binding Sites., Michelle M. Anderson, Jun Chen, Charles N. Cole, Susan E. Conrad

Dartmouth Scholarship

Infection of quiescent cells with the DNA tumor virus simian virus 40 induces expression of the cellular thymidine kinase (TK) gene a minimum of 10- to 20-fold, and this induction depends upon the viral protein large T antigen (T-Ag). To define both human TK promoter elements and T-Ag functional domains required for transcriptional induction, we have established a system in which stable Rat-1 transfectants harboring TK promoter-luciferase hybrid genes are infected with recombinant adenoviruses expressing either wild-type or mutant forms of T-Ag and luciferase expression is measured as an indicator of promoter activity. The results show that (i) a 135-bp …

A Late Baird's Sandpiper In Keith County, Stephen J. Dinsmore

Nebraska Bird Review

On 23 December 1994, Gordon Brown and I were walking the North Platte River below Keystone Dam as part of the Lake McConaughy Christmas Bird Count (CBC). At approximately 8:30 a.m. MST, about 0.5 mi below the dam, we observed a group of 8 Killdeer and a smaller shorebird, which we immediately recognized as a "peep." We studied the bird for about 30 minutes at distances as close as 40 feet and then returned in the afternoon to photograph it. The size and black legs immediately eliminated Least Sandpiper, and the dark rump eliminated White-rumped Sandpiper. The remaining possibilities were …

Regulation Of Ribulose Bisphosphate Carboxylase Gene Expression In Natural Phytoplankton Communities .1. Diel Rhythms, S. L. Pichard, L. Campbell, J. B. Kang, F. Robert Tabita, John H Paul

Marine Science Faculty Publications

Marine phytoplankton fix carbon dioxide primarily through the action of ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO), the first enzyme in the Calvin Cycle. Although the regulation of this enzyme has been studied in algal cultures and higher plants, little is known regarding RubisCO regulation in natural phytoplankton populations. To determine if natural communities of phytoplankton utilize transcriptional regulation to control RubisCO expression, we investigated the diel relationship between 14C carbon fixation and RubisCO large subunit (rbcL) transcript levels, rbcL DNA, chlorophyll a, autofluorescent cell counts and bacterial direct counts for natural communities of the southeastern Gulf of Mexico. Studies …

Hydrogen Atom Extraction And Redeposition On Hydrogen-Terminated Silicon Surface With Scanning Tunneling Microscope At Room Temperature, D. H. Huang, Y. Yamamoto Aug 1996

Hydrogen Atom Extraction And Redeposition On Hydrogen-Terminated Silicon Surface With Scanning Tunneling Microscope At Room Temperature, D. H. Huang, Y. Yamamoto

Scanning Microscopy

We have previously demonstrated that hydrogen atoms on the hydrogen-terminated Si(100)-2xl:H surface can be extracted by applying either positive or negative voltage pulses between an STM tip and the sample surface with a certain tunneling current. This phenomenon suggests that electron excitation and field evaporation both play a role in atom transfer. Under our experimental conditions, hydrogen atoms are extracted in pairs from either a single dimer or from two adjacent dimers in neighboring rows. Hydrogen atoms can also be redeposited onto the Si(100)-2xl:H surface from a tungsten tip, which has picked up hydrogen from the sample surface through the …

Applications Of Laser Microprobe Mass Spectrometry In Biology And Medicine, Annette Bakker, Luc Van Vaeck, Wim Jacob Aug 1996

Applications Of Laser Microprobe Mass Spectrometry In Biology And Medicine, Annette Bakker, Luc Van Vaeck, Wim Jacob

Scanning Microscopy

Laser microprobe mass spectrometry (LMMS) provides spot analysis with a lateral resolution of 1-5 μm. Focused laser ionisation, combined with mass spectrometry, yields information on elements, molecular identification of inorganic substances, and structural characterisation of organic molecules of typically less than 2-3 kDa. Quantification in complex heterogeneous systems, such as biological materials, is not possible at the present state of the art. The strength of the method lies in the qualitative information on the molecular composition of the analyte, not just element detection, with a lateral resolution at the light microscopy level. The applications of this technique in the field …

Influence Of Bleeding On Post-Mortem Tenderization Of Fish Muscle During Chilled Storage, M. Ando, A. Nishiyabu, T. Nakagawa, Y. Makinodan

Scanning Microscopy

The influence of bleeding on post-mortem tenderization of fish muscle during storage was studied. Six fish species were used for the present study. Among them, delay of post-mortem tenderization of muscle by bleeding was shown in the pelagic fishes: yellowtail, horse mackerel, and striped jack. On the other hand, bleeding had no influence on the muscle firmness of the bottom fishes: red sea bream, flatfish, and rudder-fish. According to transmission electron microscopy, degradation of pericellular collagen fibrils was delayed in bled yellowtail and horse mackerel. In the case of striped jack, though, collagen fibril degradation could not be observed; slower …

The Role Of Calcium In The Regulation Of Apoptosis, David J. Mcconkey Aug 1996

The Role Of Calcium In The Regulation Of Apoptosis, David J. Mcconkey

Scanning Microscopy

The recognition that apoptosis is regulated by an evolutionarily conserved set of polypeptides from the nematode Caenorhabditis elegans to humans suggests that a conserved set of biochemical mechanism(s) may also be involved in the response. Early evidence suggested that the endogenous endonuclease implicated in apoptosis in most model systems is Ca2+ -dependent, and subsequent work from a number of independent laboratories suggests that alterations in cytosolic Ca2+ homeostasis are one of the conserved biochemical pathways regulating the response. Molecular targets for Ca2+ are now being identified and include signal transduction intermediates, endonuclease(s) and proteases, and the enzymes involved in the …

Contact Force Dependence On Relative Humidity: Investigations Using Atomic Force Microscopy, E. Finot, E. Lesniewska, J. -C. Mutin, S. I. Hosain, J. -P. Goudonnet

Scanning Microscopy

This paper deals with the ability of scanning force microscopy to determine contact forces of various materials. Indeed, with high spring constants at low relative humidity, the nature of the material can be determined by measurement of the contact force as the tip approaches. Cantilevers with a high spring constant are used to achieve solid-solid contact for the tip-sample system. The capillary force estimation provides information on the development of the height of the water meniscus formed between the tip and different surfaces depending on the relative humidity. Finally, we focus our attention on measurements of moduli of elasticity which …

Covalent Labeling Of Proteins With Fluorescent Compounds For Imaging Applications, Darl R. Swartz Aug 1996

Covalent Labeling Of Proteins With Fluorescent Compounds For Imaging Applications, Darl R. Swartz

Scanning Microscopy

The labeling of proteins with fluorescent compounds for microscopy has allowed a greater understanding of biological processes. The preparation of fluorescent proteins is the first step in development of their use in microscopy. Methods are described to label and characterize a protein as an example of the general approach for other proteins. Skeletal muscle alpha-actinin was labeled with either fluorescein-5-maleimide or 5-iodoaceamidofluorescein and the reaction characterized. The maleimide reaction was much more rapid and efficient than the iodoacetamide reaction giving a coupling efficiency of 65% under the given ration conditions. The fluorescein-5-maleimide alpha-actinin was functionally characterized and there was essentially …
