Phosphotransferase Activity In Clostridium Acetobutylicum From Acidogenic And Solventogenic Phases Of Growth, Robert W. Hutkins, Eva R. Kashket

Clostridium acetobutylicum cells, when energized with fructose, transported and phosphorylated the glucose analog 2-deoxyglucose by a phosphoenolpyruvate-dependent phosphotransferase (PT) system. Butanol up to 2% did not inhibit PT activity, although its chaotropic effect on the cell membrane caused cellular phosphoenolpyruvate and the 2-deoxyglucose-6-phosphate to leak out. Cells harvested from the solventogenic phase of batch growth had a significantly lower PT activity than did cells from the acidogenic phase.

Galactose-fermenting (galactose-posirive) strains of Streptococcus tbermopbilus, alone and combined with galactosepositive and galactose-negative strains of Lactobacillus bulgaricus, were used as starter cultures in the manufacture of Swiss and Mozzarella cheese and were paired with Streptococcus lactis (also galactose-positive) in short-method Cheddar cheese manufacture. Experimental Swiss cheese made with the galactosepositive Streptococcus tbermopbilus starter alone contained a large amount of galactose (ca. 26 to 28 /amol/g of curd) 28 h after hooping compared with control Swiss (< 2/amol/g) made with a nongalactose fermenting strain of Streptococcus tbermopbilus and a galactosepositive strain of Lactobacillus bulgaricus. The objective of the tall fescue breeding program at the University of Kentucky is the development of varieties characterized by superior nutritive value (including reduced levels of perloline and loline alkaloids), minimal infestation of the fungal endophyte, Acremonium coenophialum), higher palatability, improved disease resistance, and wider adaptation through the derivation of intergeneric and interspecific hybrids of ryegrass and tall fescue species. Kentucky 31, Kenwell, and Kenhy were varieties released from this breeding program. Johnstone tall fescue, developed cooperatively by the Kentucky Agricultural Experiment Station and USDA-ARS, is the most recently released variety. It is characterized as having low levels of …

The species of the Megaluropus group, here placed in three genera, two of which are described as new, are reported from the Caribbean Sea and the eastern Pacific Ocean. The west African Megaluropus longimerusSchellenberg is redescribed and a lectotype chosen. It is placed in the new genus Gibberosus which otherwise has three American species, G. falciformis, G. myersi, and a new species, G. devaneyi. Megaluropus is now confined to the Old World. A second new genus from the New World, Resupinus, is described to include R. visendus and two new species, R. spinicaudatus and R. coloni …

Hornellia (Metaceradocus) atlanticus, a new species from Belize, related to M. occidentalis, an eastern Pacific species, is reported from the Caribbean Sea and a morphologically remote new species, M. tequestae, is described from the Florida Keys.

Three models were used to estimate heritabilities for milk yields at different production levels and for different years as twice the regression of daughter residual effects on dam residual effects. The denominator is the residual mean square for dams. The numerator is the difference between the residual term for sum of dam's and daughter's records and sum of residual terms for records of dams and daughters. Combined effects of levels of vector resistance and insecticide application in control of rice tungro virus (RTV) were determined in three field tests. Cultivar “IR28,” with high levels of resistance to the vector, Nephotettix virescens (Distant), had low RTV infection in all treatments including the untreated check. In moderately resistant “IR36,” RTV decreased with an increase in level of insecticide but did not decrease to a level equaling the untreated “IR28.” The N. virescens-susceptible cultivar “IR22” had extremely high levels of RTV infection at all insecticide levels. An experiment was conducted on a 2-5% sloping Mercer silt loam soil on a farm in Bourbon/Harrison Counties where very poor corn was grown in 1984 because of soil compaction and drought. Soil test levels of the experimental area were: pH 4.8. and 99, 316, 2070. and 120 lbs/A of P, K, Ca, and Mg, respectively. Examination of the field showed that the compacted zone occurred uniformly in the 0-5 inch surface layer of soil. Cause of this compacted layer was thought to be due to excessive disk tillage in prior years when preparing the field for tobacco production and …

The southern Western Australian Shark Working Group was formed to investigate the southern shark fishery and advise the Minister for Fisheries on options for future management. Membership comprised six fishermen from regional professional fishermen's associations and two Fisheries Department officers. The Working Group considered that each shark fisherman obtaining an entitlement should then be restricted in the length of fishing gear he can use and the number of months he can fish. The individual's time and gear entitlement would be approximately that length of gear being used and time spent fishing in a period shortly before the bench mark date. …

Research is also conducted in the Bog, which holds an understandable fascination for researchers and students of natural history because of its size, complexity, diversity and geographical isolation from similar communities. We surveyed the IRRI farm to determine the extent of parasitization of yellow stemborer egg masses. The egg masses were randomly collected at weekly intervals from July to October 1984 from rice fields, 15-20 days after transplanting, and brought to the laboratory for collection of emerging parasites. Three species of hymenopterous parasitoids—Tetrasticus schoenobii, Telenomus rowani, and Trichogramma japonicum—were found from 700 egg masses.
We introduced a method to calculate percent parasitism as based on parasite biology and behaviour. Among the three species, the combination of T. rowani and T. japonicum was the highest multiparasitization of yellow stem borer egg …

On April 23 and 24, 1986 a survey was conducted by the Virginia Institute of Marine Science (VIMS) on the temporary easement (3.248 acres) for the construction of the proposed bridge over the Machipongo River in Accomack County, Virginia (Figure 1). The study was done at the request of the Virginia Department of Highways and Transportation, Suffolk, Virginia for Project 0182-001-102, RW-201.

Visna lentiviruses have a natural tropism for cells of the macrophage lineage of sheep and goats, but virus replication in these cells in vivo is restricted so that only small quantities of virus are produced. One restricting factor suggested in previous studies is that virus replication is dependent on the maturity of the cells: the more mature the cell, the less restrictive the replication of the virus. Since monocytes in peripheral blood are precursors of macrophages, we investigated the effect of cell maturation on virus replication under limited control conditions in vitro by inoculating blood leukocytes with virus and retarding …

Farms in the Albany, Esperance, Jerramungup and Katanning districts were surveyed to determine parasite control strategies currently used, to evaluate the adoption of recommended control practices and to detect areas of ineffective parasite control. For cattle, the number of drenches given in the low rainfall zone was higher than the number given in the high rainfall zone.

Prime farmland disturbed by surface mining must by law be returned to a productivity level equal to that before mining. The coal operator has several test crops that can be selected to determine whether these production standards have been met before final bond release will be made. Grain sorghum and soybeans are two of the crops that may be used. A method is presented that allows rapid and accurate measurement of changes in the spatial pattern of space occupation in encrusting organisms. The boundaries of organisms are represented as polygons, and geometric difference calculations are used to establish the perimeters of the areas gained or vacated between sampling dates. The influence of interspecific interactions on occupation of space is also quantified: geometric intersection calculations are used to define the perimeters of the areas gained or lost in overgrowth interactions. A method is presented for estimating the sampling frequency necessary for detection of significant changes in occupation of space. The method …

The mammalian protooncogene homologue of the avian v-ets sequence from the E26 retrovirus consists of two sequentially distinct domains located on different chromosomes. Using somatic cell hybrid panels, we have mapped the mammalian homologue of the 5' v-ets-domain to chromosome 11 (ETS1) in man, to chromosome 9 (Ets-1) in mouse, and to chromosome D1 (ETS1) in the domestic cat. The mammalian homologue of the 3' v-ets domain was similarly mapped to human chromosome 21 (ETS2), to mouse chromosome 16 (Ets-2), and to feline chromosome C2 (ETS2). …

Four groups of rats were tested on an eight-arm radial maze under a free-choice procedure. The subjects were maintained at either 80% or 100% of their preexperimental free-feeding weights through restricted access to either food or water. Water-deprived subjects received water in the maze; food-deprived subjects received food. Water-deprived subjects learned the task faster than food-deprived subjects. The four groups developed different response patterns. These were measured by the mean transition size, the average angular distance (in 45° units) between consecutively chosen arms. Rats foraging for food and water developed different search strategies, with water-deprived subjects exhibiting lower mean transition …

Those aspects of an organism's biology that influence the number of progeny produced make up the fertility component of its fitness. The fertility of both male and female Drosophila pseudoobscura is influenced by multiple matings. In the former, rates of multiple matings and the genotypes of females' other mates interact to affect male fitness. Female fecundity and productivity increases with multiple matings, while longevity declines. Mating rates could be the result of these conflicting selection pressures. Microtubule proteins isolated from calf brain will undergo gelation-contraction in the presence of ATP. We have now examined this process by video-enhanced contrast microscopy. After ATP addition to steady-state microtubules, slow (1-5 micron/min), linear movements of particles and microtubules toward aggregation centers occur. The resulting structures resemble mitotic spindle asters. During the time when gel contraction occurs, asters move (at 1-5 micron/min) toward other nearby asters. This is accompanied by the apparent shortening of the microtubules running between the asters. This is the first example of isolated microtubules undergoing a process that has similarities to half-spindle shortening during anaphase A. …

The simian virus 40 large tumor antigen (T-ag) is found in both the nuclei (nT-ag) and plasma membranes (mT-ag) of simian virus 40-infected or -transformed cells. It is not known how newly synthesized T-ag molecules are recognized, sorted, and transported to their ultimate subcellular destinations. One possibility is that these events depend upon structural differences between nT-ag and mT-ag. To test this possibility, we compared the structures of nT-ag and mT-ag from simian virus 40-infected cells.
