Browse all Theses and Dissertations

ALS is a neurodegenerative disease characterized by degeneration of upper and lower motor neurons in the brain and spinal cord leading to progressive paralysis and ultimately death. Perturbations in RNA metabolism and RNA binding proteins have emerged as underlying defects in ALS pathogenesis. Matrin-3 is a multifunctional RNA binding protein that has been linked to familial and sporadic ALS. Matrin-3 is normally found in the nucleus, but mutations in the gene cause mislocalization of the protein from the nucleus into the cytoplasm of neuronal cells where it forms protein aggregates. In this study, we show that over-expressing human MATR3 in …

Constitutively active oncogenic mutant K-Ras is one the principal contributors to human cancers including 90% of pancreatic, 50% of colorectal and 32% of non-small cell lung cancers. However, except for K-Ras G12C oncogenic mutant, which only presents in about 13% of non-small cell lung cancer patients, there is no anti-K-Ras therapy for a considerable subset of K-Ras mutations in human tumors, reflecting challenges for targeting oncogenic K-Ras activity. K-Ras is a membrane-bound small GTPase; when active, it triggers multiple signaling pathways regulating a variety of key cellular functions such as cell growth, proliferation and survival. To initiate these signaling cascades, …

Reconstruction Of Gut Microbiome Via Intermittent Feeding, Kourtney Sprague Jan 2022

Reconstruction Of Gut Microbiome Via Intermittent Feeding, Kourtney Sprague

Browse all Theses and Dissertations

The benefits of intermittent fasting have been studied across many facets of health. It is known that physiologically fasting results in a metabolic switch from liver-derived glucose to adipose cell derived ketones to be used as energy and signaling molecules. Fasting down-regulates inflammation, increases expression of antioxidant defenses, and activates pathways for DNA repair and autophagy. While fasting or intermittent feeding effects on host physiology have been identified, the relationship between fasting and gut microbiome is not as well known. In times of gut rest, resident microbes undergo detoxication and motility, and in times of activity, gut microbes undergo DNA …

Sensor-Based Prediction Of Mental Effort During Learning From Physiological Data: A Longitudinal Case Study, Ankita Agarwal, Josephine Graft, Noah Schroeder, William Romine

Biological Sciences Faculty Publications

Trackers for activity and physical fitness have become ubiquitous. Although recent work has demonstrated significant relationships between mental effort and physiological data such as skin temperature, heart rate, and electrodermal activity, we have yet to demonstrate their efficacy for the forecasting of mental effort such that a useful mental effort tracker can be developed. Given prior difficulty in extracting relationships between mental effort and physiological responses that are repeatable across individuals, we make the case that fusing self-report measures with physiological data within an internet or smartphone application may provide an effective method for training a useful mental effort tracking …

Climate Change Is Driving Greening Trend Whereas Tree Cover Is Declining In The Western Himalaya Of Pakistan, Arshad Mahmood Khan, Nadia Khalid

Journal of Bioresource Management

Recent studies suggest that two factors including climate change (especially precipitation) and human land use management (HLUM) are primarily driving greening trend around the world. However, studies targeting the impact of both factors on different vegetation layers (i.e. herbs, shrubs and tree layers) are lacking. This study hypothesize that precipitation relatively influence the under-storey vegetation cover more quickly, and HLUM (say afforestation/reforestation, deforestation) is more important towards top-storey layer. To investigate this, 13 randomly selected permanent stations in the wild natural west Himalayan coniferous forests of Sudhan Gali, Pakistan were explored from 2000-2016 to collect vegetation health data by two …

Enhancing The Fertilizer Value Of Cattle Manure Using Organic Resources For Soil Fertility Improvement: A Review, Benedicta E. Ayamba, Robert C. Abaidoo, Andrews Opoku, Nana Ewusi-Mensah

Journal of Bioresource Management

The poor quality of cow dung in Ghana resulting from the opportunistic feeding mode of cattle impedes its adoption due to high quantities needed for application. This calls for nutrient optimization of cattle manure with inexpensive locally available sources of organic inputs that could enhance its quality. Though large volumes of crop residues and other agro-wastes are produced on-farm, they are underutilized and mostly a nuisance. Major food crops take approximately, 44 %, 42 % and 56 % of the total nitrogen (N), phosphorus (P) and potassium (K), respectively and are found in crop residues. There is therefore the need …

The interface between terrestrial ecosystems and inland waters is an important link in the global carbon cycle. However, the extent to which allochthonous organic matter entering freshwater systems plays a major role in microbial and higher-trophic-level processes is under debate. Human perturbations can alter fluxes of terrestrial carbon to aquatic environments in complex ways. The biomass and production of aquatic microbes are traditionally thought to be resource limited via stoichiometric constraints such as nutrient ratios or the carbon standing stock at a given timepoint. Low concentrations of a particular constituent, however, can be strong evidence of its importance in food …

Lake Tanganyika (LT) is the largest tropical freshwater lake, and the largest body of anoxic freshwater on Earth's surface. LT's mixed oxygenated surface waters float atop a permanently anoxic layer and host rich animal biodiversity. However, little is known about microorganisms inhabiting LT's 1470 meter deep water column and their contributions to nutrient cycling, which affect ecosystem-level function and productivity. Here, we applied genome-resolved metagenomics and environmental analyses to link specific taxa to key biogeochemical processes across a vertical depth gradient in LT. We reconstructed 523 unique metagenome-assembled genomes (MAGs) from 34 bacterial and archaeal phyla, including many rarely observed …
