Contemporary research suggests that shear force is a major contributor to discomfort and pressure injury. However, the variation of shear forces during human-exoskeleton interaction dynamics is a highly unexplored field. The high cost of commercial triaxial force sensors may be a major factor in the notable lack of research in this field. Arteriovenous (AV) grafts are commonly used to provide vascular access for hemodialysis in patients with end-stage renal disease. Despite their widespread use, AV grafts are prone to complications such as stenosis and thrombosis. Early detection of these conditions remains challenging with current monitoring methods too costly or insufficient. This work presents the design, fabrication, and validation of an LC pressure sensor embedded within a model AV graft to enable real-time monitoring. Diabetes mellitus patients must regularly monitor their blood glucose levels to manage glycaemia, typically requiring capillary tests at least three times daily and laboratory tests one to two times per month. These conventional methods involve finger pricking, causing significant discomfort and stress. This study introduces an innovative non-invasive glucose monitoring approach by integrating photoplethysmography (PPG) technology with an artificial intelligence (AI) algorithm, complemented by a mobile application using Flutter, IoT systems, and Firebase cloud for real-time data access. This thesis develops and experimentally validates an online adaptive Linear Quadratic Regulator (LQR) control method for prosthetic joint systems using Recursive Least Squares (RLS)-based real-time parameter estimation on the Quanser QUBE-Servo 2 platform. Traditional LQR controllers assume a fxed system model, which limits adaptability and results in reduced tracking accuracy, poor robustness, and loss of optimal performance when applied to dynamically changing prosthetic joints, infuenced by load variations, user gait changes, and mechanical wear. Limited experimental validation exists for combining RLS with online LQR adaptation in prosthetic-like systems.

Auditory Nerve Interface (ANI) devices, currently in developmental stages, represent an emerging neuroprosthetic approach to restore hearing in severe sensorineural loss patients by directly stimulating the auditory nerve via the Utah Slanted Electrode Array (USEA). Conventional implant leads are comprised of multiple wires made from materials such as platinum-iridium and MP35N stainless steel that are overmolded in an insulative material. The fabrication of microfluidic devices often requires specialized methods. The development of these methods requires careful characterization and understanding of the processes involved. Using primarily PolyJet 3D printing technology, microfluidics offers a wide scope of applications such as microfluidic benthic microbial fuel cells (MBMFCs) and electrostatic artificial muscles. MBMFCs benefit from the confinement of the microbes resulting in close proximity between the electrode and the organisms. Using a modular design called the Sponge, assembly and upscaling is possible. In this work, we developed and investigated the cancer cell killing efficacy of different instances using real-time high-sensitivity measurements facilitated by Guided-Mode Resonance (GMR) sensors. The GMR biosensor technology detects shifts in the spectral peak caused by changes in the refractive index of the surrounding medium, offering a label-free method for monitoring cellular events.

A549 cell line wild type and HER2 knockout (lung cancer cells) were exposed to different concentrations of Triton X-100 and ADC (Herceptin with αHFc-CL-DMDM). This process is an essential non-value-added activity that creates a bottleneck. In this dissertation, streamlined components, aerosol administration synchronization, and/or Excipient Enhanced Growth (EEG) technologies were utilized to develop and test several novel devices and aerosol delivery systems. Accomplishing slow translocation speed with high sensitivity has been the greatest mission for solid-state nanopore (SSN) to electrically detect nucleobases in single-stranded DNA (ssDNA). In this study, a method to detect nucleobases in ssDNA using a SSN is introduced by considerably slowing down the translocation speed and effectively increasing its sensitivity. In the inertial actuation, the robot used the transfer of momentum to navigate, and this momentum could be generated by spinning masses and wheels. In order to obtain a cell-based biosensor, the sensing method needs to be reliable and repeatable in a liquid environment. My research focuses on electrokinetic transport. Particularly, in this dissertation, we focus on fabrication and testing of micro electrodes with nanostructured surfaces to minimize the electrode polarization (EP) effects for biosensor applications. In the first study, electrochemical deposition of gold nanoparticles on to planar gold electrodes was used to generate rough surfaces. Dendritic nanostructures that reduced EP up to two orders of magnitude was obtained by optimizing the deposition conditions. These structures also enhanced dielectrophoresis (DEP) response of our bio-chips, making them usable in physiological buffers. This paper details an investigation into methods and designs of 3D printing a microfluidic system capable of droplet emulsion using NinjaFlex filament. The specific field in which this paper’s experiment is rooted is dubbed “BioMEMS,” short for bio microelectromechanical systems. One prominent research area in BioMEMS is developing a “lab on a chip.” Essentially, the goal is to miniaturize common lab processes to the micro scale, rendering it possible to include these processes in a small chip.
