Fluoroscopy is considered the gold standard for locating catheters during cardiac electrophysiology (EP) procedures. However, fluoroscopy emits ionizing radiation which can lead to adverse health effects when exposed to in high doses (World Health Organization, 2016). Electroanatomic mapping (EAM) systems display the three-dimensional location of EP catheters and measure the local electrical activity of the heart. They can minimize a physician’s reliance on fluoroscopy and can help reduce radiation exposure during a case (Casella, 2011).

Filtration is a technology that is used almost ubiquitously in society from uses raging from filtration of macroparticles from water to pharmaceutical grade filtration products to remove anything larger than a protein. However, with such a wide range of uses, most filtration products have the same issue; membrane clogging (fouling) that prevents continuous use and requires frequent maintenance. This thesis hypothesizes that by applying a direct current (DC) to an insulating array of posts, they will create a foul-less insulative dielectrophoretic filter (iDEP) that does not clog since particles will levitate above the insulating array.

Impedance spectroscopy (IS) is an important tool for cell detection and characterization in medical and food safety applications. In this thesis, the Cal Poly Biofluidics Lab’s impedance spectroscopy system was re-evaluated and optimized for single-cell impedance spectroscopy. To evaluate the IS system, an impedance spectroscopy bioMEMS chip was fabricated in the Cal Poly Microfabcrication lab, software was developed to run IS experiments, and studies were run to validate the system. Atrial fibrillation (AF) is the disordered activation of the atrial myocardium, which is a major cause of stroke. Currently, the most effective, minimally traumatic treatment for AF is percutaneous catheter ablation to isolate arrhythmogenic areas from the rest of the atrium. The standard in vitro evaluation of ablation catheters through lesion studies is a resource intensive effort due to tissue variability and visual measurement methods, necessitating large sample sizes and multiple prototype builds. Cerebral ischemia leading to an ischemic stroke is a possible complication of a transcatheter aortic valve replacement (TAVR) procedure. This is because embolic debris can become dislodged and travel through the aortic arch, where they either continue to the descending aorta and join the systemic circulation or travel into the cerebral vasculature through the three daughter vessels that branch off the top of the aortic arch. The production and approval process for the various labels used in clinical trials wastes significant time and resources through the need to outsource label production or rely on large reams of pre-cut label stock for each revision throughout the process. An in-house, on-demand label printing and cutting system is a potential remedy to this waste. Previous work by Cheadle et al. resulted in a functional electomechanical prototype of the label cutting aspect of this research, capable of rudimentary linear cuts. The purpose of this study was to define a lower standard cuff occlusion time to induce reactive hyperemia in assessing endothelial dysfunction. In this study, strong evidence was found by a novel technique that used oscillometric methods, which supported that 3 minute reactive hyperemia was sufficient to elicit a significant difference in arterial compliance from baseline. Twenty healthy Cal Poly students were assessed, (n=12 female, n=8 male) aged 22 years old with a standard deviation of 2.04 years. Arterial compliance was estimated by measuring the peak-to-peak oscillations for baseline, 3 minute reactive hyperemia, and 5 minute reactive hyperemia tests, with …

The goal of this study is to design and manufacture a microfluidic device capable of measuring changes in impedance valuesof microfluidic cell cultures. Tocharacterize this, an interdigitated array of electrodes was patterned over glass, where it was then bonded to a series of fluidic networks created in PDMS via soft lithography. The device measured ethanol impedance initially to show that values remain consistent over time. Impedance values of water and 1% wt. saltwater were compared to show that the device is able to detect changes in impedance, with up to a 60% reduction in electrical impedance in saltwater. In this thesis, a prototype ECG monitor was developed that is integrated into an elastic shirt and takes a 3-lead ECG for over 5 days. The high-quality measurements can be used to identify markers indicative of various detrimental heart conditions. Measurements recorded by the device are encrypted and stored onto a micro-SD card. Current Holter monitors are expensive and have functional lives less than 48 hours; however, extended duration monitoring has been proven more useful in diagnosis. The device designed demonstrates that ECG measurements can be taken over longer durations without sacrificing quality, comfort, or device cost.

Movement disorders are a group of syndromes that often arise due to neurological abnormalities. Approximately 40 million Americans are affected by some form of movement disorder, significantly impacting patients’ quality of life and their ability to live independently. Deep brain stimulation (DBS) is one treatment that has shown promising results in the past couple decades, however, the currently used open-loop system has several drawbacks. 3-D paper-based microfluidic devices (micoPADs) are small and portable devices made out of paper that offer a promising platform for diagnostic applications outside of a laboratory. These devices are easy to use, low cost, require no power source, and capable of detecting multiple targets simultaneously. The work in this thesis demonstrated the ability of a 3-D paper-microfluidic platform to simultaneously detect 5 targets. Rubber cord stock was used in conjunction with an acrylic housing unit to apply pressure along the edge of the channel. However, this minimally invasive technique has shown to cause ischemic brain lesions, or “silent infarcts”, in 90% of TAVR patients, which can increase the patient’s risk for stroke by two to four times in future years [3]. There are a number of applications, from hearing aids to microfluidic pumps, which utilize micro-scale actuating diaphragms. These MEMS (micro-electromechanical system) devices can be actuated by electrostatic forces, which utilize an induced electric field to pull two charged plates towards one another. Such devices were fabricated and electrostatic actuation of the diaphragms was performed to analyze its viability as a micro-speaker. Since the advent of recommendations for placing infants in the supine position during sleep to reduce the incidence of sudden infant death syndrome, clinicians have noted an increase in the frequency of cranial asymmetry due to deformation of suture sections of the infants’ skulls as a result of constant concentrated stress in one area at the back of their head. This specific form of cranial deformation is known as positional plagiocephaly and its rate of occurrence has increased from 0.3% in 8.2% within the past 30 years.

The multi-transducer sensor array design consists of five standard stethoscope diaphragms mounted to a rigid frame for a-priori knowledge of their relative spatial locations in the x-y plane, with compliant z-direction positioning to ensure good contact and pressure against the subject’s skin for reliable acoustic coupling. When the heart is not able to pump blood to the body efficiently, fluid can back up into the veins that take blood through the lungs to the left atrium. It is a known problem that a Spinal Cord Stimulator (SCS) can interact with an Implantable Cardioverter Defibrillator (ICD) when both devices are implanted in the same patient. Interactions between the SCS and ICD can cause inappropriate therapy which can be harmful to the patient. While ICD devices have a distinct narrowband sensing bandwidth, the pulse configurations that current SCS devices deliver were not designed with this frequency region in mind.
