Personalized Medicine - from a one-size fits all to a tailored approach is the future in pharmaceutical therapeutics and medical applications. The aim of this study is demonstrate additive manufacturing in two personalized medicine applications: 1. Patient- customized solid dose medicines, and 2. Patient-customized wound care device. The aim of the first application is to develop an inexpensive and accurate method to design and fabricate patient-customized tablets. Particularly this paper focuses on fabricating and dose controlling of patient-customized extended release prednisolone-poly(vinyle alcohole) PVA. Microscale propulsion impacts a diverse array of fields, with simplistic microrobots allowing for novel innovations in microscale surgery and drug delivery. Propulsion at the microscale is constrained by physics, with time-reversal and geometric symmetries limiting available propulsion mechanisms. However, certain fluid environments and surface coatings allow for the propulsion of microparticles through externally applied magnetic fields. Presented here is a detailed analysis of microparticles propelling using spontaneous symmetry breaking, flagella surface coatings, and multi-modal actuation mechanisms. Accomplishing slow translocation speed with high sensitivity has been the greatest mission for solid-state nanopore (SSN) to electrically detect nucleobases in single-stranded DNA (ssDNA). In this study, a method to detect nucleobases in ssDNA using a SSN is introduced by considerably slowing down the translocation speed and effectively increasing its sensitivity. The ultra-thin titanium dioxide (TiO2) coated hexagonal boron nitride (h-BN) nanopore was fabricated, along with an ionic-liquid [bmim][PF6]/2.0 M KCl aqueous (cis/trans) interfacial system, to increase both the spatial and the temporal resolutions. Vascular grafting has been an effective remedy to vascular diseases for decades. Autografting by using the veins or arteries in the patient’s body is found to have patency rate for years. However, autografting may not be possible when compatible vessels are not available. In this case, synthetic vascular grafts are found to be effective replacements. Nevertheless, synthetic grafts with diameter less than 4 mm becomes challenging due to thrombogenicity and mismatch of mechanical properties with the native vessels. Blood-contacting cardiovascular stents often induce a secondary clotting event due to unrestricted enzymatic activities. The use of hemocompatible polyurethane coatings on these implantable devices is one of the most promising methods to reduce device rejection. In this study, four commercial polyurethane films of various thicknesses and compositions were evaluated for their anticoagulation properties. Results suggested that these films exhibited excellent thermal and physico-mechanical properties while capable of increasing contact time with blood plasma by over a thousand-fold as compared to a control surface. Bone cancer is an aggressive disease and has peak occurrence during physiological stimulation of growth and aging. Astronauts who undergo long-term space missions also acquire an increased risk of bone tissue degeneration and cancer. Few in-vitro models currently exist capable of reproducing the complex microenvironment of bone tissue to support multicellular activity in a three-dimensional structure. This limits the ability to understand disease progression and develop suitable treatment strategies. Electrospinning is a technology used to generate small fibers down to nano-scale size. This method of fiber creation has been around for many years. However, in recent years electrospinning has found increased applications, especially in the area of tissue engineering due to its ability to create fibers with properties similar to the extracellular matrix in tissue. An electrospinning platform can illustrate concepts of engineering, electro-mechanical system design, manufacturing, and biomedical applications in one single package. As the fabrication of soft actuators with conventional microfabrication methods are tedious, expensive, and time consuming, employment of 3-D printing fabrication methods appears promising as they can simplify fabrication and reduce the production cost. Traditional metals such as stainless steel, titanium and cobalt chrome are used in biomedical applications (implants, scaffolds, etc.) but suffer from issues such as osseointegration and compatibility with existing bone. The development of a medical drug delivery device that allows for the deployment drugs into the adventitial tissue of blood vessels requires the inflation of a silicone elastomer. The inflated silicone must be able to consistently endure multiple loading cycles without failing so that the device can operate reliably. In this work, the approach to the manipulation of alginate artificial cell soft-microrobots, both individually and in swarms is shown. Fabrication of these artificial cells were completed through centrifugation, producing large volumes of artificial cells, encapsulated with superparamagnetic iron oxide nanoparticles; these artificial cells can be then externally stimulated by an applied magnetic field. The construction of a Permeant Magnet Stage (PMS) was produced to manipulate the artificial cells individually and in swarms. The stage functionalizes the permanent magnet in the 2D xy-plane. This thesis is an examination of two material systems derived from polylactic acid (PLA) and polyethylene glycol (PEG). PLA is a polymer commonly sourced from renewable sources such as starches and sugars. It is a relatively strong, biodegradable polymer, making it ideal for use in the body. Even though it has a relative high strength, PLA is also brittle leading to the use of plasticizers to increase flexibility. Selective Laser Melting (SLM) of Additive Manufacturing is an attractive fabrication method that employs CAD data to selectively melt the metal powder layer by layer via a laser beam and produce a 3D part. This method not only opens a new window in overcoming traditional NiTi fabrication problems but also for producing porous or complex shaped structures. The goal of this dissertation was to develop a realistic and patient-specific computational model of the heart that ultimately would help medical scientists to better diagnose and treat heart diseases. In order to achieve this goal, a three dimensional finite element model of the heart was created using magnetic resonance images of the beating pig heart. This model was loaded by the pressure of blood inside the left ventricle which was measured by synchronous catheterization. It is widely accepted that cells behave differently responding to the stiffness of their extracellular matrix (ECM). Such observations were made by culturing cells on hydrogel substrates of tunable stiffness. However, it was recently proposed that cells may sense how strongly they are tethered to ECM, not the local stiffness of ECM. To investigate both hypotheses, we developed a method to fabricate Janus polyacrylamide (PAAM) gels. We squeeze two drops of different concentrations in the Hele-Shaw geometry to generate radial Stokes flow. IPMCs are one of the most widely used types of electro-active polymer actuator, due to their low electric driving potential and large deformation range. Recent advances in patterning techniques and emerging surface microtechnologies have allowed cell micropatterning to control spatial location of the cells on a surface as well as cell shape, attachment area, and number of contacting neighbor cells. These parameters play important roles in cell cellular behaviors. Production of ethyl dodecanoate (biodiesel) using lauric acid and methanol with a solid acid catalyst of sulfated zirconia is studied by using two distillation sequences. In the first sequence, the methanol recovery column follows the reactive distillation column. In the second sequence, the reactive distillation and methanol recovery columns are thermally coupled. Thermally coupled distillation sequences may consume less energy by allowing interconnecting vapor and liquid streams between the two columns to elminate reboiler or condenser or both. Abstract Head injuries can occur in most sporting activities. The seriousness of these injuries can vary greatly depending on the sport. Many sports such as American football, cricket, baseball and hurling try to reduce this risk by designing helmets that are suitable to be worn by the players. The peridynamic method is used here to analyse the effect of van der Waals forces on the mechanical behaviour and strength and toughness properties of three-dimensional nanofibre networks under imposed stretch deformation. The peridynamic formulation allows for a natural inclusion of long-range forces (such as van der Waals forces) by considering all interactions as ‘long-range’. We use van der Waals interactions only between different fibres and do not need to model individual atoms. Fracture is introduced at the microstructural (peridynamic bond) level for the microelastic type bonds, while van der Waals bonds can reform at any time. We conduct statistical …
