Studying Mass And Mechanical Property Changes During The Degradation Of A Bioadhesive With Mass Tracking, Rheology And Magnetoelastic (Me) Sensors, Zhongtian Zhang
Studying Mass And Mechanical Property Changes During The Degradation Of A Bioadhesive With Mass Tracking, Rheology And Magnetoelastic (Me) Sensors, Zhongtian Zhang
Dissertations, Master's Theses and Master's Reports
In this research, the degradable polymer 4-arm poly (ethylene glycol)-glutaric acid-dopamine (PEG-GA-DM4) was synthesized. The degradation behavior of crosslinked PEG-GA-DM4 bioadhesive was studied with mass tracking, oscillatory rheology, and magnetoelastic (ME) sensors. Changes in mechanical properties were correlated with both dry mass and wet mass changes during the degradation. The results indicate that the loss of mechanical property in the bioadhesive can take place without losing the dry mass. The mass loss profile cannot describe the degradation behavior completely. In addition to studying the degradation of PEG-GA-DM4, this research also confirms the application of ME …
Collapsible Silicone Tubes: An In Vitro Model For Tracheal Traction, Kevin D. Garman
Collapsible Silicone Tubes: An In Vitro Model For Tracheal Traction, Kevin D. Garman
Master's Theses (2009 -)
Obstructive sleep apnea (OSA) is characterized by recurrent episodes of airway collapse and airflow limitation during sleep. Fragmented sleep and reductions in blood oxygen saturation lead to several comorbidities, including hypertension, cardiovascular disease, and cerebrovascular disease. Longitudinal forces (tracheal traction) acting on the soft tissues surrounding the upper airway have been proposed to play a significant role in stabilizing the airway and preventing collapse. However, the relative contribution of longitudinal forces as compared to other factors that affect airway stability (airway geometry, tissue properties, muscle activity) remains unclear. This in-vitro study aimed to investigate to what extent longitudinal forces can …

Towards A Bioselective Surface For Treatment Of Sepsis In A Hemoperfusion Device, Karl F. Schilke, Ramya Raman, Bonan Yu, Erik Hahn, Jonathan Su
Biomedical Engineering Western Regional Conference
Sepsis is a dysregulated immune response to circulating bacteria/endotoxin. Hemoperfusion is a promising treatment, but is not universally effective and can unacceptable blood damage. We are developing protein-repellent hydrogel nanofilms that can be grown on a variety of surfaces, and functionalized with “click-ready” pathogen-binding proteins expressed by Genetic Code Expansion (GCE). The extremely rapid, bioorthogonal “click” immobilization chemistry eliminates prior purification, maximizes surface loading, and guarantees spatial orientation of the immobilized proteins. We have demonstrated pathogen binding and blood protein repulsion by these layers, suggesting that they may be part of effective treatments for sepsis and other applications.

Formulation And Testing Of Biodegradable Polymeric Coating On Zinc Wires In Cardiovascular Stent Application, Avishan Arab Shomali
Dissertations, Master's Theses and Master's Reports
Biodegradable and biocompatible poly (L-lactic-acid) (PLLA) coating was applied on a modified zinc (Zn) substrate by dip coating, with the intent to delay the bio-corrosion and slow the degradation rate of zinc substrate. 3-(Trimethoxysilyl) propyl methacrylate (MPS) was used for modification of the zinc substrate for promoting the adhesion between the metallic substrate and the polymer coating. It is hypothesized that the delay in Zn biodegradation could be useful in the first several weeks to prevent the early loss of mechanical integrity of the endovascular stent and to improve the healing process of the diseased vascular site. The PLLA coating …

Development Of An Electrospun And 3d Printed Cellular Delivery Device For Dermal Wound Healing, Ryan M. Clohessy
Theses and Dissertations
The goal of this research was to develop a system of individualized medicine that could be applied to dermal wounds serving as a wound dressing and synthetic extracellular matrix while delivering stem cells to the wound bed. First, fabrication parameters for electrospinning polymer fibers were determined. This involved evaluating fiber morphology with respect to polymer selection and solution concentration. Next, construct fabrication was examined to produce an integrated void space, or cargo area, suitable to maintain stem cells. In vitro studies to ensure stem cell viability and phenotype were conducted, and results supported the notion that cells could be administered …

The World Health Organization estimates that over 30 million people require some sort of prosthetic technology. However, traditional prosthetic fitting practices take a lot of time and cost a lot of money, making them inaccessible to millions of people around the world. StandUP Worldwide is an interdisciplinary project team devoted to creating low-cost prosthetic technologies for use around the world, especially in resource poor areas. They are currently developing a low cost, below-the-knee prosthetic kit that can be easily deployed in a resource-poor area. The following presents their solution for a below-the-knee prosthetic socket, foot, and leg.

Mechanical Characterization Of Carbon Fiber And Thermoplastic Ankle Foot Orthoses, Amanda Wach
Master's Theses (2009 -)
The needs of an increasingly young and active orthotic patient population has led to advancements in ankle foot orthosis (AFO) design and materials to enable higher function. The Intrepid Dynamic Exoskeletal Orthosis (IDEO) is a custom energy-storing carbon fiber AFO that has demonstrated improved clinical function, allowing patients to return to high-intensity activities such as sports and military service. An improved understanding of AFO mechanical function will aid prescription and fitting, as well as assist in design modifications for different patient populations. This study investigated the mechanical properties of AFOs, specifically structural stiffness, rotational motion, and strut deflection, to discern …

The Role Of Biological Fluid And Dynamic Flow In The Behavior And Cellular Interactions Of Gold Nanoparticles, Emily K. Breitner, Saber M. Hussain, Kristen K. Comfort
Chemical and Materials Engineering Faculty Publications
Background: Due to their distinctive physicochemical properties, nanoparticles (NPs) have proven to be extremely advantageous for product and application development, but are also capable of inducing detrimental outcomes in biological systems. Standard in vitro methodologies are currently the primary means for evaluating NP safety, as vast quantities of particles exist that require appraisal. However, cell-based models are plagued by the fact that they are not representative of complex physiological systems. The need for a more accurate exposure model is highlighted by the fact that NP behavior and subsequent bioresponses are highly dependent upon their surroundings. Therefore, standard in vitro models …

Breast Cancer/Stromal Cells Coculture On Polyelectrolyte Films Emulates Tumor Stages And Mirna Profiles Of Clinical Samples, Amita Daverey, Karleen M. Brown, Srivatsan Kidambi
Department of Chemical and Biomolecular Engineering: Faculty Publications
In this study, we demonstrate a method for controlling breast cancer cells adhesion on polyelectrolyte multilayer (PEM) films without the aid of adhesive proteins/ ligands to study the role of tumor and stromal cell interaction on cancer biology. Numerous studies have explored engineering coculture of tumor and stromal cells predominantly using transwell coculture of stromal cells cultured onto coverslips that were subsequently added to tumor cell cultures. However, these systems imposed an artificial boundary that precluded cell−cell interactions. To our knowledge, this is the first demonstration of patterned coculture of tumor cells and stromal cells that captures the temporal changes …

Mechanical Engineering
In today’s world of medical innovation, regulations and requirements set by organizations such as the Food and Drug Administration (FDA), the International Organization for Standardization (ISO), and ASTM International (American Society for Testing and Materials) can inhibit rapid innovation by demanding rigorous testing of new designs. For arterial stents, the standard is that each design must be tested to simulate 10 years of life in an environment congruent to an in vivo environment. Endologix in Irvine, California develops and manufactures minimally invasive treatments for aortic diseases, with a focus on stent grafts for the treatment of abdominal aortic aneurysms (AAA). …

Design And Development Of Two Component Hydrogel Ejector For Three-Dimensional Cell Growth, Thomas Dunkle, Jessica Deschamps, Connie Dam
Honors Scholar Theses
Hydrogels are useful in wound healing, drug delivery, and tissue engineering applications, but the available methods of injecting them quickly and noninvasively are limited. The medical industry does not yet have access to an all-purpose device that can quickly synthesize hydrogels of different shapes and sizes. Many synthesis procedures that have been developed result in the formation of amorphous hydrogels. While generally useful, amorphous hydrogels exhibit limited capability in tissue engineering applications, especially due to their viscous properties. This endeavor aims to modulate the appropriate gelation parameters, optimize the injection process, and create a prototype that allows for the extrusion …
Peracetic Acid: A Practical Agent For Sterilizing Heat-Labile Polymeric Tissue-Engineering Scaffolds, William R. Trahan
Peracetic Acid: A Practical Agent For Sterilizing Heat-Labile Polymeric Tissue-Engineering Scaffolds, William R. Trahan
Theses and Dissertations
Advanced biomaterials and sophisticated processing technologies aim to fabricate tissue-engineering scaffolds that can predictably interact within a biological environment at a cellular level. Sterilization of such scaffolds is at the core of patient safety and is an important regulatory issue that needs to be addressed prior to clinical translation. In addition, it is crucial that meticulously engineered micro- and nano- structures are preserved after sterilization. Conventional sterilization methods involving heat, steam and radiation are not compatible with engineered polymeric systems because of scaffold degradation and loss of architecture. Using electrospun scaffolds made from polycaprolactone (PCL), a low melting polymer, and …
Biocorrosion Rate And Mechanism Of Metallic Magnesium In Model Arterial Environments, Patrick Bowen
Biocorrosion Rate And Mechanism Of Metallic Magnesium In Model Arterial Environments, Patrick Bowen
Dissertations, Master's Theses and Master's Reports
A new paradigm in biomedical engineering calls for biologically active implants that are absorbed by the body over time. One popular application for this concept is in the engineering of endovascular stents that are delivered concurrently with balloon angioplasty. These devices enable the injured vessels to remain patent during healing, but are not needed for more than a few months after the procedure. Early studies of iron- and magnesium-based stents have concluded that magnesium is a potentially suitable base material for such a device; alloys can achieve acceptable mechanical properties and do not seem to harm the artery during degradation. …
Development Of A Novel Device For The Perfusion Driven Decellularization Of Skeletal Muscle, Benjamin Kasukonis
Development Of A Novel Device For The Perfusion Driven Decellularization Of Skeletal Muscle, Benjamin Kasukonis
Graduate Theses and Dissertations
Decellularization of skeletal muscle is a process that removes cellular components of skeletal muscle tissue while leaving behind the intact extracellular matrix (ECM). Skeletal muscle ECM is currently being studied as a biologic scaffold for repairing volumetric muscle loss (VML) because the removal of cells greatly reduces the antigenicity of the donor tissue. Decellularization usually relies on passive diffusion of detergents, surfactants and/or osmotic solutions to strip cells from the ECM. However, passive diffusion alone is usually not sufficient for complete removal of cells from the interior of large pieces of skeletal muscle using detergents, such as sodium dodecyl sulfate …
Improvements To Uhmwpe, Brooke Mckelvogue
Improvements To Uhmwpe, Brooke Mckelvogue
Journal of Undergraduate Research at Minnesota State University, Mankato
Ultra high molecular weight polyethylene (UHMWPE) is a material used in artificial implants for articular joint replacements. However, these implants have a limited lifespan in which the patient will be pain-free due to the wear of the UHMWPE components. Recently crosslinking, or exposing the material to radiation, has been used to extend the wear resistance of UHMWPE. Crosslinking introduces another set of drawbacks; mainly the reduction of the fracture toughness of UHMWPE and the generation of free radicals, which leave the polymer vulnerable to damage from oxidation. Currently, research is being conducted on other methods to increase the wear resistance …

Utilizing Fast Spin Echo Mri To Reduce Image Artifacts And Improve Implant/Tissue Interface Detection In Refractory Parkinson’S Patients With Deep Brain Stimulators, Subhendra N. Sarkar, Pooja R. Sarkar, Efstathios Papavassiliou, Rafael Rojas
Publications and Research
Introduction. In medically refractory Parkinson’s disease (PD) deep-brain stimulation (DBS) is an effective therapeutic tool. Postimplantation MRI is important in assessing tissue damage and DBS lead placement accuracy. We wanted to identify which MRI sequence can detectDBS leads with smallest artifactual signal void, allowing better tissue/electrode edge conspicuity.
Methods. Using an IRB approved protocol 8 advanced PDpatientswere imagedwithinMRconditional safety guidelines at lowRF power (SAR ≤ 0.1 W/kg) in coronal plane at 1.5T by various sequences. The image slices were subjectively evaluated for diagnostic quality and the lead contact diameters were compared to identify a sequence least affected by metallic leads.

Microsphere Spray System For Wound Coverage, Nicholas J. Andersen
Microsphere Spray System For Wound Coverage, Nicholas J. Andersen
Theses and Dissertations--Biomedical Engineering
Spinal fusion is used to treat diseases or disorders of the spine by fusing together two or more vertebrae. Two associated risks with spinal fusion are infection and blood loss. Administration of tranexamic acid is used to prevent blood loss, and transfusions are given following blood loss. Surgical site infections are prevented with vancomycin powder spread into the surgical wound, while established infections are treated by debridement and delivery of antibiotics for 4 to 6 weeks. The present research explored an alternate method to prevent and treat blood loss or infection in spinal fusion. Poly(lactic-co-glycolic acid) (PLGA) microspheres was used …

Modeling The Zimmer Fitmore And Ml Taper Implantation, Tyler Kazuo Franklin
Modeling The Zimmer Fitmore And Ml Taper Implantation, Tyler Kazuo Franklin
Master's Theses
With more young adults requiring total hip
arthroplasties the need for bone saving implants becomes
more important. The Zimmer Fitmore is a new bone saving
implant that utilizes an implantation technique that
reduces the damage to the muscle tissue allowing for
patients to have a short recovery time as well as a new
design that allows it to rest on the medial cortex. There
has been anecdotal evidence that this device leads to early
revision within six months of implantation due to failures
occurring in the medial cortex. The main goal of this
study was to computationally model the Zimmer …

Closed Loop Control Of A Cylindrical Tube Type Ionic Polymer Metal Composite (Ipmc), Benjamin Mead
UNLV Theses, Dissertations, Professional Papers, and Capstones
The goal of this research is to provide a framework for the integration of tube type, cylindrical Ionic Polymer Metal-Composite (IPMC) into conventional devices. IPMCs are one of the most widely used types of electro-active polymer actuator, due to their low electric driving potential and large deformation range. For this research a tube type IPMC was investigated. This IPMC has a circular cross section with four separate electrodes on its surface and a hole through the middle. The four electrodes allow for biaxial bending and accurate control of the tip location. One of the main advantages of using this type …
Effect Of Extracellular Matrix (Ecm) Protein Micropatterns On The Behavior Of Human Neuroblastoma Cells, Ishwari Poudel
Effect Of Extracellular Matrix (Ecm) Protein Micropatterns On The Behavior Of Human Neuroblastoma Cells, Ishwari Poudel
Department of Engineering Mechanics: Dissertations, Theses, and Student Research
Recent advances in patterning techniques and emerging surface microtechnologies have allowed cell micropatterning to control spatial location of the cells on a surface as well as cell shape, attachment area, and number of contacting neighbor cells. These parameters play important roles in cell cellular behaviors. Cell micropatterning has thus become one of the most important strategies for biomedical applications, such as, tissue engineering, diagnostic immunoassays, lab-on-chip devices, bio-sensing, etc., and cell biology studies as well. For neuronal cells, there have been attempts to distribute neuronal cells on specific patterns to control cell-to-cell interaction. However, there have been very limited understanding …

An Engineering Evaluation Of Ankle Prosthetics, Christopher Thian, Stephen Tiernan, Fiona Mcevoy, Robert Flavin
Conference Papers
There are a wide range of different types of ankle replacements on the market today each with a
different mechanical design. Unfortunately the results of ankle replacements are not as good as hip
and knee replacements; this is due to the complexity of the ankle joint. In the early days of ankle
replacements some of the prosthetics only lasted 4 months. Recent developments have improved the
longevity of the replacements although, there are still many complications and failures of the
replacements, these include; the prosthetic components migrating into the bone, the components
failing due to stresses induced by the forces …
