Synthesis Of Thiol-Acrylate Hydrogels For 3d Cell Culture And Microfluidic Applications, Anowar Hossain Khan
Synthesis Of Thiol-Acrylate Hydrogels For 3d Cell Culture And Microfluidic Applications, Anowar Hossain Khan
LSU Doctoral Dissertations
Globally cell culture is an $18.98 billion industry as of 2020, with an 11.6 percent annual growth rate. Drug discovery has an estimated worth of $69.8 billion in 2020 and is predicted to grow to $110.4 billion by 2025. Three-dimensional (3D) cell culture of cancer cells is one of the rapidly growing felids since it better recapitulates in vivo conditions compared to two-dimensional (2D) models. However, it is challenging to grow 3D tumor spheroids outside the body, and some of the existing technology can generate these spheroids outside the human body but poorly mimic in vivo tumor models. Therefore, there …
An Electrochemical, Fluidic, Chip-Based Biosensor For Biomarker Detection, Lauren Bell
An Electrochemical, Fluidic, Chip-Based Biosensor For Biomarker Detection, Lauren Bell
Theses and Dissertations--Biomedical Engineering
Biosensors and their use in both the research and clinical field for the detection and monitoring of critical biomarkers are prevalent and constantly improving. However, continued research needs to be done to address shortcomings, such as low sensitivity, poor specificity, and poor readiness for integration into research use and patient care. The objective of this research was to create a combined fluidic, chip-based biosensor that could detect different biomarkers with high sensitivity and ease of use. For assessing the developed sensor, three separate biomarkers were tested: glucose, cholesterol, and oxygen. Both the glucose biosensor and cholesterol biosensor were combined with …
Impact Of Percutaneous Transluminal Intervention On Vascular Hypoxia And The Role Of Micro-Calcifications On Atherosclerotic Plaque Rupture, Andrea Corti
Dissertations and Theses
Atherosclerotic disease is initiated by cholesterol build-up beneath the endothelium, which evolves into a fibroatheroma, a lipid-rich plaque covered by a fibrous cap. Many of these plaques are considered as vulnerable, as they grow to occlude the luminal section of the artery (chronically stenotic) or become mechanically unstable with a higher chance of rupturing (rupture-prone). On one hand, chronically stenotic vulnerable plaques are symptomatic lesions that are often treated with Percutaneous Transluminal Intervention (PTI). Unfortunately, approximately 10% of PTI procedures are followed by severe Neointima Hyperplasia (NH), which causes a new occlusion and failure of the implant. A significant precursor …

Nano-, Encapsulated Manganese Oxide (Nemo) Particles For Mri Enhancement In Microfluidic Tumor Models, Kasey Alexis Freshwater
Graduate Theses, Dissertations, and Problem Reports (ETD)
Breast cancer is among the deadliest cancers in women, with approximately 2.3 million new cases and 685,000 deaths reported globally in 2020 alone. Magnetic Resonance Imaging (MRI) is commonly used to diagnose, stage, and develop treatment plans for patients predisposed to breast cancer. However, current MRI-based screening of breast cancer has high false-positive rates up to 25%, leading to unnecessary invasive follow-up screening and testing. The gold-standard intravenous gadolinium (Gd)-based contrast agents (GBCAs) account for false-positive rates because they are always “ON”, providing a constant MRI signal that highlights both benign and malignant tumors. The work detailed in this thesis …
Time-Lapse Observation Of Sarcomeric Addition In Hypertrophic Models Based On A Tissue Like Cardiomyocyte Culture, Ailin Wei
All Dissertations
Cardiac hypertrophy is the response of the heart to increased mechanical stress exerted on the heart. In a hypertrophic heart, the muscles elongate or/and thicken with cardiomyocytes growing sizes. It is assumed that cardiomyocytes elongate by adding sarcomeres in series; the cardiomyocytes thicken with the addition of sarcomeres in the parallel position.
In this study, a tissue-like culture model was achieved with step-like ICDs. Uniaxial static stretch has been applied to our unique culture model for studying mechanical-load-induced cardiac hypertrophy. Hypertrophic responses of these cardiomyocytes have been confirmed on transcriptional and translational levels in our tissue-like model. The two-photon excitation …

Development Of Solution Blow Spun Nanofibers As Electrical And Whole Cell Biosensing Interfaces, Craig Levar Miller
All Dissertations
Infectious pathogens place a huge burden on the US economy with more than $120 billion spent annually for direct and indirect costs for the treatment of infectious diseases. Rapid detection schemes continue to evolve in order to meet the demand of early diagnosis. In chronic wound infections, bacterial load is capable of impeding the healing process. Additionally, bacterial virulence production works coherently with bacterial load to produce toxins and molecules that prolongs the healing cycle. This work examines the use of nonwoven polymeric conductive and non-conductive nanofiber mats as synthetic biosensor scaffolds, drug delivery and biosensor interface constructs. A custom-made …

Makara Journal of Science
The emerging technology in biomedical engineering requires biocompatible materials, which are also referred to as biomaterials. For a material to be considered biocompatible, it should not interact with human tissues in a harmful way, and vice versa. Various properties of biocompatible materials, such as mechanical and optical properties, have to be considered for different biomedical applications. One of the most popular applications of biomaterials is for contact lenses. Hydrogels, specifically poly(2-hydroxyethyl methacrylate) (PHEMA) hydrogels, are among the most popular ones in ophthalmologic applications, especially in soft contact lenses. This paper reviews the use of PHEMA hydrogels as one of the …

Promotion Of Human Schwann Cell Proliferation Using Heparin/Collagen Coated Nerve Conduits, John Magness
Chemical Engineering Undergraduate Honors Theses
Often in the aftermath of an injury or surgery, the sense of touch and muscle control is lost in the affected area as nerves are damaged or severed and fail to grow back completely. The regeneration of the nerve cells can be promoted by treating the nerves with nerve conduits. Nerve conduits are hollow cylinders of bio-compatible materials that can be surgically implanted to the disconnected nerve to promote and direct the growth of nerves. The objectives of this research are to investigate the ability of nerve conduits treated with layer-by-layer coatings to promote the growth of Schwann cells, to …

Design And Control Of A Peristaltic Pump To Simulate Left Atrial Pressure In A Conductive Silicone Model, Jeremy Collins
Mechanical Engineering Undergraduate Honors Theses
According to the CDC, atrial fibrillation is responsible for more than 454,000 hospitalizations and approximately 158,000 deaths per year. A common treatment for atrial fibrillation is catheter ablation, a process in which a long flexible tube is guided through the femoral artery and to the source of arrhythmia in the heart, where it measures the electrical potential at various locations and converts problematic heart tissue to scar tissue via ablation. This paper details the design and control of a low-cost ($400) peristaltic pump system using repetitive control to replicate blood pressure in the left atrium in a conductive silicone model …
Surface-Eroding Drug Delivery Films For Sequential And/Or Intermittent Release Of Psychoactive Drugs, Jason Price

Honors Theses
ABSTRACT JASON BLAKE PRICE: Surface-Eroding Drug Delivery Films for Sequential and/or
Intermittent Release of Psychoactive Drugs For patients with psychiatric diseases, adherence to medication schedules, medication
errors, and abuse are common issues. Promising new forms of therapy for these patients, such as micro-dosed lysergic acid diethylamide (LSD), where patients receive 10-20% of a full dose every third day, present further drug delivery challenges. Sequential or intermittent release of drugs from an implanted device could ensure long-term drug compliance, automate drug dosing during the life of the implant, and eliminate potential for abuse and medication errors. To this end, we generated …

Dissertations and Theses
Localized chemical delivery plays an essential role in the fundamental information transfers within biological systems. Thus, the ability to mimic the natural chemical signal modulation would provide significant contributions to understand the functional signaling pathway of biological cells and develop new prosthetic devices for neurological disorders. In this paper, we demonstrate a light-controlled hydrogel platform that can be used for localized chemical delivery in a high spatial resolution. By utilizing the photothermal behavior of graphene-hydrogel composites confined within micron-sized fluidic channels, patterned light illumination creates the parallel and independent actuation of chemical release in a group of fluidic ports. The …

Applied Machine Learning In Extrusion-Based Bioprinting, Shuyu Tian
Theses and Dissertations
Optimization of extrusion-based bioprinting (EBB) parameters have been systematically conducted through experimentation. However, the process is time and resource-intensive and not easily translatable across different laboratories. A machine learning (ML) approach to EBB parameter optimization can accelerate this process for laboratories across the field through training using data collected from published literature. In this work, regression-based and classification-based ML models were investigated for their abilities to predict printing outcomes of cell viability and filament diameter for cell-containing alginate and gelatin composite hydrogels. Regression-based models were investigated for their ability to predict suitable extrusion pressure given desired cell viability when keeping …

Characterization Of Oxone Mediated Tempo-Oxidized Nano Cellulose Mixed-Matrix Membranes During Ultrafiltration And Hemodialysis, Kristyn Robling
Biomedical Engineering Undergraduate Honors Theses
The ninth leading cause of death in the United States is kidney disease, and hemodialysis is the process most commonly prescribed for treatment. It utilizes a selectively permeable membrane filter to remove toxins such as urea from the blood and retain necessary protein levels. However, traditional filters, such as cellulose triacetate, used during dialysis can be inefficient in terms of separation performance and reduction of fouling. Recent exploration of nanoparticles has resulted in the creation of Oxone Mediated TEMPO-Oxidized Nano Cellulose which has properties that are believed to increase hydrophilicity, increase tensile capacity, decrease membrane resistance and lower fouling, making …

Modified Stent Design For A Coronary Bifurcation Lesion, Abigail Nowell
Biological Sciences Undergraduate Honors Theses
Currently, 18.2 million adults aged 20 and older are diagnosed with Coronary Artery Disease (CAD) (Benjamin et al., 2019). Stenosis is the most common intervention. However, when a patient has a bifurcated artery, treatment becomes more difficult and is often unsuccessful. This project created a new stent and balloon complex that was tested in vitro using a gel phantom artery model. Two separate prototypes have been created and tested so far, with improvements made upon each. Testing is still underway with Prototype 2.

3d Printing Of Iron Oxide Incorporated Polydimethylsiloxane Soft Magnetic Actuator, Rasoul Bayaniahangar
Dissertations, Master's Theses and Master's Reports
Soft actuators have grown to be a topic of great scientific interest recently. As the fabrication of soft actuators with conventional microfabrication methods are tedious, expensive, and time consuming, employment of 3-D printing fabrication methods appears promising as they can simplify fabrication and reduce the production cost. Complex structures can be fabricated with 3-D printing such as helical coils can achieve actuation performances that otherwise would not be possible with simpler geometries. In this thesis development of soft magnetic helical coil actuators of iron-oxide embedded polydimethylsiloxane (PDMS) was achieved with embedded 3-D printing techniques. Composites with three different weight ratios …

A Physical And Computational Reverse-Engineering Approach To Determine Dimensional Change And Its Relationship To Oxidation In Retrieved Orthopedic Implants, Josephine Kalshoven
ENGS 88 Honors Thesis (AB Students)
Oxidation of the Ultra-High Molecular Weight Polyethylene (UHMWPE) tibial inserts of total knee arthroplasty devices is a major factor underlying multiple modes of failure for these devices, including delamination, wear, and fracture. Previous research has demonstrated that oxidation of UHMWPE is driven by a high concentration of free radicals in the polyethylene. However, even new devices created with undetectable amounts of free radicals are oxidizing in vivo. One theory is that, in the absence of residual free radicals, oxidation is facilitated by absorbed species (e.g. lipids, ROS) delivered or exacerbated by contact stress. However, no method exists to comprehensively …
Development Of A Sonically Powered Biodegradable Nanogenerator For Bone Regeneration, Avi S. Patel
Development Of A Sonically Powered Biodegradable Nanogenerator For Bone Regeneration, Avi S. Patel
University Scholar Projects
Background: Reconstruction of bone fractures and defects remains a big challenge in orthopedic surgery. While regenerative engineering has advanced the field greatly using a combination of biomaterial scaffolds and stem cells, one matter of difficulty is inducing osteogenesis in these cells. Recent works have shown electricity’s ability to promote osteogenesis in stem cell lines when seeded in bone scaffolds; however, typical electrical stimulators are either (a) externally housed and require overcomplex percutaneous wires be connected to the implanted scaffold or (b) implanted non-degradable devices which contain toxic batteries and require invasive removal surgeries.
Objective: Here, we establish a biodegradable, piezoelectric …
Suture-Less Trocar Site Closure Clip, Hares A. Patel
Suture-Less Trocar Site Closure Clip, Hares A. Patel
Electronic Theses and Dissertations
Introduction: Following laparoscopic surgery, there is a need, in many cases, to close trocar sites to prevent hernias. Currently, devices that exist on the market are suture based, but the lack of standardization in the suturing techniques together with the time-consuming nature of the procedure leads to the need for improvement in trocar site closure products. Trocar closure sites do not need to be fully closed on the fascial layer; rather, sufficiently blocking the hole at the abdominal wall can significantly reduce post-operative herniation. A retrospective study on trocar site herniation (TSH) after laparoscopic surgery indicates a TSH incidence …
An Engineer’S Take On The Bone-Ligament Interface: Utilizing Novel Technology To Improve Clinical Outcomes, Emma Garcia
An Engineer’S Take On The Bone-Ligament Interface: Utilizing Novel Technology To Improve Clinical Outcomes, Emma Garcia
Shared Knowledge Conference
Ligament repair is a common surgical practice with a significant lack of viable replacements. The current gold standard for repair is the use of tendon grafts from cadavers or from another place in the patient’s body; however, these often cause more problems than they solve including immune responses or a lack of mobility in another place in the body. Synthetic replacements are of growing interest, though the ability to mimic the complex structure of the ligament and how it connects to the bone remains an obstacle. Our lab built a 3D bioprinter combined with an electrospinner to address this complicated …

Orientation of plasmonic nanostructures is an important feature in many nanoscale applications such as photovoltaics, catalyst, biosensors DNA interactions, protein detections, hotspot of surface-enhanced Raman spectroscopy (SERS), and fluorescence resonant energy transfer (FRET) experiments. Silver nanocubes with significant spectral signatures between 400-700 nm are observed in this experimental research. Whereas study of single cells will enable the analysis of cell-to-cell variations within a heterogeneous population. These variations are important for further analysis and understanding of disease propagation, drug development, stem cell differentiation, embryos development, and how cells respond to each other and their environment. Adipose-derived mesenchymal stem cells possess the …

Adaptation of cancer cells to changes in the biochemical microenvironment in an expanding tumor mass is a crucial aspect of malignant progression, tumor metabolism, and drug efficacy. In vitro, it is challenging to mimic the evolution of biochemical gradients and the cellular heterogeneity that characterizes cancer tissues found in vivo. It is well accepted that more realistic and controllable in vitro 3D model systems are required to improve the overall cancer research paradigm and thus improve on the translation of results, but multidisciplinary approaches are needed for these advances. This work develops such approaches and demonstrates that new droplet-based cell-encapsulation …
Electrochemical And Spectroscopic Study Of Peptide-Template Melanins, Daniel Courter, Olivia Rossi, Zhen Tian
Electrochemical And Spectroscopic Study Of Peptide-Template Melanins, Daniel Courter, Olivia Rossi, Zhen Tian
Honors Theses and Capstones
Melanins are organic polymers made up of 5,6-dihydroxyindole carboxylic acid (DHICA) and 5,6-dihydroxyindole (DHI). Biochemical systems synthesize melanins through a complex reaction pathway. Previous research exhibited that naturally-occurring melanins (NatMel) and their synthetic analogues (SynMel) can potentially serve as electrode materials for aqueous charge storage devices. Small surface area creates technical challenges for these melanins as charge storage materials. This research herein demonstrates that the structure of melanins can affect the charge storage capacities of monovalent Na+cations and divalent Mg2+ cations. 2-D sheet-like peptide melanins (PepMel) were synthesized into tripeptides consisting of aspartic acid (D), phenylalanine (F), …

We set out in August 2017 to develop an active noise suppressing device that would be usable both in military and civilian activities. Due to constraints in knowledge and time, we decided our best course of action was to divide the project into two equal projects with the hope to be able to combine them into a single project at the end of the allotted time. This consisted of an active noise suppression device and a passive noise suppression device. The passive device would have no electronic components and the active device would be pure circuitry with no housing. This …
