Antimicrobial Properties Of Citric Acid Based Polymers, Lee-Chun Su
Bioengineering Theses - Archive
A series of citric acid derived polymers synthesized in our lab were investigated and compared for their physiochemical and antimicrobial properties. We selected citric acid based polymers since citric acid is a product of natural metabolism. These polymers are not only biocompatible but also possess an antimicrobial effect. In polymer synthesis, citric acid (CA) provides profound functional groups available for binding and crosslinking to allow the controllable crosslinking degrees of polymers. A number of citric acid based polymers has been synthesized and studied for their chemical, mechanical, and biological characteristics for use in various biomedical applications. Since bacterial infections became …
Regenerative Peripheral Nerve Interfacing Of Sensory Modalities, Nilanjana T. Dutta
Regenerative Peripheral Nerve Interfacing Of Sensory Modalities, Nilanjana T. Dutta
Bioengineering Theses - Archive
Peripheral neural interfaces (PNI) has proved potential to record motor commands for the control of these prosthetic limbs, together with stimulation of specific sensory axons, so as to transmit specific sensory sub-modalities. Although PNI is hypothesized to provide adequate sensory feedback, realizing this degree of specificity is not fully established. Recently, a regenerative multi-electrode interface (REMI) was reported to be capable of recording peripheral nerve activity from a transected nerve. This study is aimed at interfacing neural activity specific to sensory sub-modalities, such as thermal pain, mechanoception and limb stretching using the REMI, in a freely moving animal. In the …

Characterization Of Immune Response And Peripheral Nerve Regeneration Of Implanted Regenerative Multi-Electrode Interface Post-Subchronic Amputation, Camilo Andres Sanchez Useche
Bioengineering Theses - Archive
Interfacing high degree-of-freedom robotic prosthetics directly to the peripheral nervous system aims at improving the quotidian life of amputees. Despite advancements in robotics that have lead to human-like upper limb prosthetics, long-term interfacing has not been accomplished, thus voiding them from clinical use. Interfacing electrodes in the central nervous system fail due to the foreign body reaction and glial scar formation at the site of implantation. Concrete mechanisms for the failure of long-term Interfacing at the peripheral nervous level remain unclear. A regenerative multi-electrode interface (REMI) which allows for action potential recording as early as 7 days and provides support …

Development Of A Measurement System For Quantifying Ultrasound Field Distribution Of Different Transducers, Jayanth Kandukuri
Bioengineering Theses - Archive
Ultrasound has been widely used in many fields, such as nondestructive examination and medical imaging. Ultrasound is mostly characterized by its pressure and, dynamic pressure wave can be considered to be the foundation of ultrasound physics and imaging techniques. It is thus important for understanding ultrasound pressure properties to that of ultrasound intensity. Moreover, there are many applications that require mainly characterizing ultrasound pressure field (instead of intensity field alone). Therefore, reconstruction of an ultrasound pressure field is highly desirable not only for education but also for research. There may be some commercially available systems that can be used for …

Fabrication Of Small Diameter Vascular Graft Using Stacked Collagen Films, Sandeep Shah
Bioengineering Theses - Archive
Collagen I have been widely used in the field of vascular tissue engineering. They are characterized better for their interaction at the cellular level but are often limited as vascular graft due to their weak mechanical strength and thrombic property. The crosslinking chemicals or polymers support are used to overcome weak mechanical property. Crosslinking agents tend to have cytotoxic effects while blend with synthetic polymers have mismatch or compliancy issues inside the body. Here in this set of study, we used stacked collagen films and embedded drug delivery system within the film to construct small tubular conduits to meet the …

Rapid, reliable and easy to implement approaches are needed for fabrication of circular microchannels in polymeric microfluidics. PDMS has been a choice of material in microfluidics and it is being used as prototype due to its unique property such as flexibility and transparency. Traditional method of fabrication such as photolithography and soft lithography are complicated and time consuming. In this thesis, a non-lithography and bottom-up self-assembled approach is shown. The resulting devices can have uses in many applications such as assays, biosensing, separation of mixtures, micro-mixing and bioreactors. This thesis presents novel approaches of manufacturing self-assembled microchannels in polydimethyleneoxide (PDMS) …

Effect Of Ascorbic Acid On Cdte Quantum Dots And Investigation Of Different Chemical Sensors On Cdtehg Quantum Dots, Yamini Chitrala
Bioengineering Theses - Archive
The objective of my thesis is to investigate the effect of ascorbic acid on CdTe quantum dots, which is an antioxidant. It was observed that the quantum dots recovered their luminescence intensity after the interaction of the ascorbic acid, where the intensity was quenched by the photosensitizer. The luminescence intensity increased gradually for 0.1mM- 0.5mM concentration of ascorbic acid and remained stable with higher concentrations of the antioxidant. Also, it was observed that under some reaction conditions like pH, temperature and concentration, ascorbic acid had a great influence on its photoluminescence. In the second part of the research project, I …

Nerve conduits have shown to successfully bridge nerve gaps of ≤ 3 cm; however repair of longer, mixed or motor nerve gaps still remains a clinical challenge. In order to study regeneration techniques in clinically relevant long gaps, larger animal models are required. Standardized methods to evaluate functional outcome are necessary to clinically translate experimental treatments. In this study, the capability of the biodegradable and transparent cross-linked doped polyester elastomer (CUPE) to serve as a nerve conduit material in a multiluminal nerve conduit across a 5 cm gap in a rabbit common peroneal nerve model was evaluated. The effect of …

Near Infrared Quantitative Phase Imaging Of Cellular Manipulation Under Different Physio-Chemical Environments, Bipin Joshi
Bioengineering Theses - Archive
Quantitative phase imaging using Digital Holographic Microscopy (DHM) is emerging as a label-free and wide-field method of characterizing cells with high spatio-temporal resolution. In parallel, silicon based micromechanical and electronic devices are allowing both manipulation (e.g. electrical stimulation, mechanical actuation) as well as characterization (electrical and mechanical) of micro and nano-scopic samples. This has revolutionized development of lab-on-a-chip devices for high throughput analysis of cells and molecules for diagnosis of disease and screening of drug-effects. However, very little progress has been made in optical (e.g. fluorescence, Raman etc) characterization of samples on these silicon-based devices. Especially, wide-field high-resolution optical imaging …

Diffusion tensor imaging (DTI), a modality of MRI that measures water diffusion properties noninvasively, is highly sensitive to subtle structural changes of white matter. It provides a unique and noninvasive method for delineating in-vivo architecture of human brain white matter. Four metrics derived from DTI, fractional anisotropy (FA), mean diffusivity (MD), axial diffusivity (AxD) and radial diffusivity (RD), have been widely used to quantitatively characterize the white matter disruption in mental disorders and degenerative diseases. An atlas-based approach incorporating a digital white matter atlas and registration and skeletonization functions of tract-based spatial statistics (TBSS) has been developed recently. This approach …

Abdominal adhesion is a prevalent, troublesome condition occurring after surgical procedures and can cause a variety of side effects such as, abdominal pain with a highly associated cost ($1.3 billion annually for) for expensive surgical removal. Many companies and research groups have studied and developed techniques to prevent adhesion after surgery; however, none have been completely effective or practical for use. Current materials have many drawbacks, such as the inability to degrade, poor surgical handling, and attracting bacteria. Up to 93% of patients receiving abdominal surgeries will develop abdominal adhesions, and a viable treatment option needs to be developed to …

Involvement Of Mixed Lineage Leukemia And Homeobox Proteins In Dna Damage Recognition And Repair, Somdutta Chakraborty
Bioengineering Theses - Archive
Deoxyribonucleic Acid (DNA) molecules are informational molecules, encoding genetic information which is indispensable in the development and functioning of living organisms. DNA encodes for most proteins essential for all bodily functions and a few of which are known to be involved in the DNA Damage Response/ Repair mechanism. DNA damage recognition and repair is crucial and the cell has an innate mechanism of repairing it. Unrepaired DNA damage leads to genetic mutation or even cancers, which are sustained by further cell division and genetic multiplication. Other researchers have already confirmed the involvement of several proteins and histones like PARP and …

Evaluation Of Tissue Response From The Regenerative Multielectrode Array (Remi) Implant Using Atf-3 And Cjun, Sarita Bhetawal
Bioengineering Theses - Archive
Currently available peripheral nerve regenerative interfaces such as the sieve and the micro-channel regenerative electrode, intended to be used for the control and natural feel from advanced robotic prosthetic devices, eventually fail due to nerve damage due to axonopathy. The Regenerative Multi-electrode array (REMI) developed in our lab overcomes this limitation by providing a non-obstructive pathway for regenerating axons. However, it is possible that the mismatch at the electrode-tissue interface may cause axonal injury during limb stretching. In order to determine if micro-motion at the REMI causes neuronal injury I evaluated the nuclear expression of Activating Transcription Factor 3 (ATF-3), …

Cancer has been one of the leading causes of death worldwide that necessitates the development of theranostic systems for effective cancer management. Previously, we developed biodegradable-photoluminescent polymer coated magnetic nanoparticles (BPLP-MNPs) as a theranostic system for cancer management. These BPLP-MNPs were highly stable with excellent biocompatibility and consisted of magnetic targeting and imaging capabilities. However, these particles experienced reduced fluorescence due to the presence of dark/blackness of MNPs, which absorbs excited/emitted light. Therefore, it was hypothesized that surface modification on MNPs can overcome this limitation and enhance fluorescence for BPLP-MNPs.The goal of this research was therefore to employ various surface …

Neuroprosthetics facilitate amputees with motor function restoration by interfacing nervous system with artificial limb and enabling control of the prosthetic device by volitional thought. Though such neurointerfacing provides dexterity nearly mimicking natural limb agility, the clinical viability of such prosthetics is limited by the lack of long-term reliability. In central nervous system, these implants fail due to foreign body reaction and glial scar formation around the implant. The cause of decrease in active sites of interfacing in peripheral nervous system, which is less invasive than central nervous system implantation, has not been established. Recently an open architecture, regenerative peripheral neural …

Bioengineering Theses - Archive
Prostate cancer is the most common male cancer in the United States, and the second leading cause of cancer death in men. The conventional diagnostic procedures use an invasive approach to collect the biopsy tissue samples. Also, treatments currently used include radiotherapy, chemotherapy and surgical removal of the prostate associated with high risks of surgical complications. Among all the imaging modalities, Near Infrared (NIR) optical imaging could be a possible candidate for minimally invasive diagnosis of prostate cancer with avoidance of harmful radiation and relatively low cost. This motivation brings the development of multifunctional polymeric (di- lactide-co-glycolic acid) (PLGA) Nanoparticles …

The mechanical properties of the cellular environment play an important role in the functioning of the cell. Cells attach to the extracellular matrix or other surrounding cells, thus sensing their mechanical properties and tune their own internal mechanical properties accordingly. A number of in vitro studies have been performed to study various cellular mechanical phenomena, but most of them employ a very stiff substrate such as glass or plastic that does not encompass the physiologically relevant range. Also, for tissues of the nervous system, the relationship between the substrate stiffness and the cellular behavior is less understood. In this study, …

Malignant Breast Neoplasm (MBN) or breast cancer is a heterogeneous cancer type originating in the breast tissue. Surgery is considered the primary treatment for breast cancer, with many early stage patients being cured without recurrence. The goals of the surgery include complete resection of the primary tumor, with negative margins to reduce the risk of local recurrences. Further, pathological staging of the tumor and axillary lymph nodes are carried out to extract necessary prognostic information. Surgery works well in case of complete Mastectomies. However, the biggest challenge for this technique is determining adequate margins on the partial mastectomy (lumpectomy) specimen. …

A Viscoelastic Model For Human Prolapsed Vaginal Wall Tissue, Milton Ma
Bioengineering Theses - Archive
Pelvic organ prolapse (POP) affects thousands of elderly women each year. Improvements in surgical repair could benefit from the understanding of the biomechanical properties of the vaginal wall (VW) tissues to better tailor the needed repair. VW tissue property has been measured by uniaxial tensile test on excised fresh human anterior VW tissues. Data available, however, are limited to elastic modulus obtained in vitro. Recently, in vivo biomechanical data of VW from POP patients have been obtained by adapting a cutometer-like device, the BTC-2000TM (SRLI). The Voigt and Standard Linear models in conjunction with the framework of quasi-linear viscoelasticity theory …

The HRT II - Rostock Corneal Module (HRT II-RCM) provides excellent resolution, contrast and optical sectioning capability for cornea imaging. It offers the most detailed views of cornea structure and pathology. However, the major limitation of this microscope is its scanning method. It has a threaded dial on the front side of the microscope which is coupled to the objective, and this dial has to be rotated manually to change the focal plane and to scan through the cornea. The dial is within inches of the cornea. During the scans, manual control causes serious interference and degrades the quality of the …

Pain is a sensory and emotional experience as a result of possible or definite tissue damage. The level of pain is subjective to the individual's consciousness of sensory stimuli. Pain stimuli can be noxious (i.e., potentially damaging) or innocuous (i.e., normally not painful). Therefore, pain perception can be based on the degree of the stimuli. Studies have been done using brain imaging modalities to assess pain. Functional Near-infrared spectroscopy (fNIRS) is a noninvasive imaging tool and has been used to study the hemodynamic variations in the brain induced by a variety of activations. This research was a feasibility study to …

Evaluation Of Spatial Resolution And Localization Accuracy Of Absorbing Heterogenity Embedded In Homogenous And Layered Turbid Media Using Functional Near Infrared Spectroscopy, Niranjana Nandakumar
Bioengineering Theses - Archive
Functional Near-Infrared Spectroscopy (fNIRS) is a non-invasive widely used technique for imaging the brain activation patterns in the cerebral cortex of neonates and adults. For the fNIRS imaging of these brain activation patterns, it is common to assume the head geometry as a homogenous medium due to the resulting computational simplicity of the image reconstruction problem. In reality the head is not a simple homogenous medium and prior research has been conducted to show that light propagates differently in a spatially heterogeneous model of the head compared to a homogenous one, which alters the sensitivity of NIR light detection on …
