Quantitative volumetric assessment of filamentous actin (F-actin) fibers remains challenging due to their interconnected nature, leading researchers to utilize threshold-based or qualitative measurement methods with poor reproducibility. Herein, a novel machine learning-based methodology is introduced for accurate quantification and reconstruction of nuclei-associated F-actin. Utilizing a convolutional neural network (CNN), actin filaments and nuclei from 3D confocal microscopy images are segmented and then each fiber is reconstructed by connecting intersecting contours on cross-sectional slices. In-pile instrumentation is critical for advancing operations and materials discovery in the nuclear industry. Ensuring optimal performance of sensors in high temperatures is the first step in demonstrating their viability in the harsh in-pile environment. This work demonstrates the high temperature capabilities of a line heat source and measurement technique previously shown to extract thermal conductivity of nuclear fuel sized samples within a laboratory environment at room temperature. This method uses a hybrid AC/DC measurement technique to obtain rapid measurements of the temperature dependent voltage change of a heater wire, which also acts as a resistance thermometer. Glioblastoma multiforme (GBM) is a highly aggressive and invasive tumor of the central nervous system (CNS). Survival rates are abysmal, with only 7.2% of patients alive 5-years after diagnosis. Because of this, understanding epigenetic alterations that give GBM tumors their aggressive phenotypes is critical for the development of more targeted and effective therapies. These alterations frequently affect a group of proteins called the Polycomb group proteins, which play important oncogenic and tumor suppressive roles in cancer. One Polycomb protein, the RING1- and YY1-binding protein (RYBP), is downregulated in a majority of GBM patients, suggesting a strong tumor suppressive property. In …

El Nino and La Nina are worldwide environmental phenomena brought about by repetitive changes in the water temperature of the Pacific Ocean. Even though the El-Nino impact focuses on a smaller area in the Pacific Ocean near the Equator, these developments have global repercussions, where temperature and precipitation are influenced across the globe, causing droughts and floods simultaneously. In this dissertation, we first derived a drought vulnerability index for the Nile basin, identifying regions with high and low drought risk under ENSO conditions. This Study Included the Design, Pilot, and Evaluation of a Fourth-Grade Science Unit Entitled "Raising the Green Roof", Exploring These Connections. Five Classrooms in Two Midwestern U.S. Public Elementary Schools Participated, and 4th-Grade Students (N = 73) Drew Systems Models at Three Time Points (212 Models) and Wrote their Ideas. While perfluorosulfonic acid (PFSA) membranes have primarily been used in fuel cells due to their chemical, thermal, and mechanical stability, one PFSA, Nafion, boasts two unique characteristics: a broad glass transition (~55 °C to 130 °C) and a temperature-persistent electrostatic network. The combination of these two characteristics endows Nafion with exceptional shape memory properties – the ability of a material to morph and transform into pre-programmed shapes when exposed to an external stimulus – with enhanced permanent shape memorization, and a potentially near-infinite number of temporary shape memorization. This study introduced a four-way CFD-DEM coupling approach to simulate the shear-induced particle migration (SIPM) mechanism leading to formation of the lubrication layer (LL) during concrete pumping. The CFD-DEM simulations considered the coupled effect of concentration (10 %–40 %) and wide size distribution (1–17 mm) of aggregate and rheology of the mortar for forces between the suspending matrix and the particles (and vice versa), as well as force transmission directly between particles (and the pipe wall). The Icarus project began in 2017 where a team performs high-altitude balloon (HAB) launches to conduct atmospheric and payload design research. The project involves sending a data-collecting payload containing recording equipment to high altitudes (~100,000 feet). This paper outlines the Icarus project as well as the efforts to design a payload that will remain stable throughout flight while also protecting the equipment housed within the payload shell. This report details the complete design of an automated bottling system. Each bottle is 5.97 centimeters tall and has an outer diameter of 3.81 centimeters. The goal of the project is to successfully fill and cap twelve bottles in under three and a half minutes. The system will have two conveyors in series, a water dispenser, an electrolyte dispenser, capping station, Human Machine Interface (HMI) and Allen Bradley programmable logic controller (PLC). In this dissertation, relationships between polyphenylene sulfide (PPS) chain structure and melt processing parameters are linked to thermoplastic composite (TPC) properties and mechanical performance. Deviations from linear PPS structure and the formation of branching/crosslinked moieties upon exposure to TPC manufacturing steps presents an opportunity to systematically uncover the interdependencies of melt-state thermal history on polymer rheology, crystallization, crystal structure, and translate these properties to the continuum level. Current design code equations for estimating the elastic modulus of specialty concretes like ultra-high performance concrete are not all suitable. Developing empirical curves from exhaustive experimental testing is challenging for concretes that may have high levels of cement replacement with mineral admixtures and fibers at varying volume contents. As such, an alternative approach is taken that seeks to understand the constituent microscale stiffness mechanisms and relate the results to the macroscale. Special moment frames (SMFs) having reduced beam section (RBS) connections are commonly used in high seismic areas as they resist lateral forces while preserving critical welded connection regions. RBS SMFs provide brace-free, unobstructed frame options which can be favorable for architectural purposes. However, when building designs require non-rectangular framing, laterally skewed connections may be needed. This project was intended to investigate the feasibility of a proposed modular dairy processing facility collocated with a small dairy on the University of Nebraska–Lincoln’s Innovation Campus. Priority was placed upon cost savings for equipment to be used in processing. Process flow diagrams, floor plans, production schedules, risk assessments, and plans for wastewater treatment were all developed to address the needs of the plant should it be built. The purpose of this research project is to investigate the presence of patented in vitro models for perineural invasion (PNI) and tumor innervation, the two most prominent modes of cancer-nerve crosstalk that play an active role in cancer initiation and progression. PNI occurs through a positive feedback loop in which cancer cells invade passive nerves to metastasize. Tumor innervation is the process where tumor cells attract nearby nerves to the tumor microenvironment (TME) for tumor growth and metastasis.
