Identification Of Holocarboxylase Synthetase Chromatin Binding Sites In Human Mammary Cell Lines Using The Damid Technology, Dipika Singh

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Holocarboxylase synthetase (HCS) is a chromatin protein that is essential for mediating the covalent binding of biotin to histones. Biotinylation of histones plays crucial roles in the repression of genes and repeats in the human genome. The objective of this project was to determine genomic binding sites of holocarboxylase synthetase (HCS) in the human chromatin of breast cell lines using an antibody independent technology. We tested the feasibility of DNA adenine methyltransferase identification (DamID) technology to map HCS binding sites in human mammary cell lines. Full-length HCS was fused to Dam for subsequent transfection into breast cancer (MCF-7) and normal …

Hydraulic Conductivity, Infiltration, And Runoff From No-Till And Tilled Cropland, Jessica H. Deck

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Infiltration and runoff are important processes that affect the efficiency of center pivot irrigation systems. No-till planting systems potentially influence the hydraulic properties of soils and the soil surface conditions. The result of long-term use of no-till could be higher infiltration and lower runoff from rainfall and irrigation.

This potential was investigated in Nebraska on two center pivot irrigated sites; Fillmore County and Phelps County, one furrow irrigated site; South Central Agriculture Laboratory (SCAL), and one dryland site; Rogers Farm. Paired treatments were used at each location, one that was no-till planted and one that used two to three operations …

Ultrafast Time Domain Optics Of Single-Cycle Laser Pulse Interaction With Materials, Ufuk Parali Dec 2010

Ultrafast Time Domain Optics Of Single-Cycle Laser Pulse Interaction With Materials, Ufuk Parali

Department of Electrical and Computer Engineering: Dissertations, Theses, and Student Research

In this thesis, interaction of an ultrashort single-cycle pulse (USCP) with a bound electron without ionization is studied for the first time. For a more realistic mathematical description of USCPs, Hermitian polynomials and combination of Laguerre functions are used for two different single-cycle excitation cases. These single-cycle pulse models are used as driving functions for the classical approach to model the interaction of a bound electron with an applied field. Two different new novel time domain modification techniques are developed for modifying the classical Lorentz damped oscillator model in order to make it compatible with the USCP excitation. In the …

Modeling And Simulation Of Tunable Photonic Crystals, Weiqing Yang Dec 2010

Modeling And Simulation Of Tunable Photonic Crystals, Weiqing Yang

Department of Electrical and Computer Engineering: Dissertations, Theses, and Student Research

Photonic crystals (PhCs) have wavelength scale periodically alternating refractive indexes. Photon in such structures is subject to strong scattering, experiencing distinctive redistribution of energy, yielding interesting properties such as photonic band gaps, field enhancement, strong nonlinear optic effects and photon confinement. The modified fields also alter the propagation of light beams. By proper setup, super collimation could be realized in PhCs where beams can travel long distance without spreading, while no waveguide structure is used. Redirection of light can extend the refraction to negative range, without violating physics rules. This distinguished phenomenon has been envisioned as the core mechanism for …

Improved Mobile Wireless In Vivo Surgical Robots: Modular Design, Experimental Results, And Analysis, Jeff Hawks Dec 2010

Improved Mobile Wireless In Vivo Surgical Robots: Modular Design, Experimental Results, And Analysis, Jeff Hawks

Department of Mechanical and Materials Engineering: Dissertations, Theses, and Student Research

Laparoscopic surgery results in superior patient outcomes as measured by less painful recovery and an earlier return to functional health compared to conventional open surgery. However, the difficulty of manipulating laparoscopic tools from outside the patient’s body generally limits these benefits to patients undergoing relatively simple procedures. The use of miniature in vivo robots that fit entirely inside the peritoneal cavity represents a novel approach to laparoscopic surgery. These robots enable more complex laparoscopic procedures, increasing the number of patients that benefit from laparoscopic surgery.

This thesis describes recent work focused on developing a modular wireless mobile platform that can …

Nanomanufacturing And Analysis Of Novel Integrated Continuous Nanofibers, John E. Hannappel

Department of Engineering Mechanics: Dissertations, Theses, and Student Research

Complex nanostructured materials have great potential for applications in many areas of nanotechnology. This potential is being unlocked by precise control of their nanoscale architecture and properties. Most current methods of creating these nanostructures are expensive and difficult to control, with the majority of techniques resulting in non-continuous nanostructures and nanoparticles. Electrospinning is an economic nanomanufacturing method resulting in continuous nanofibers. The method consists of spinning fiber-forming liquids in high electric fields. In this work, a modified electrospinning process was analyzed. The process utilized two concentric liquids that resulted in integrated continuous hollow or composite nanofibers. A new adjustable co-axial …

The objectives of this research were: characterize the pollutants in roadway runoff and determine the effectiveness of the existing stormwater BMPs at the study site. To accomplish these objectives, eleven rainfall events were sampled from November 2008 through November 2010.

First flush and composite highway runoff samples were analyzed for heavy metals, anions, nutrients, particulates, BOD, COD, VOCs, and …

Removal Of Low Density Sediments By Vegetative Filters, Brent M. Hall Nov 2010

Removal Of Low Density Sediments By Vegetative Filters, Brent M. Hall

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

The use of vegetative filter strips (VFS) is a longstanding best management practice for the removal of sediment and other pollutants from overland flow. Many attempts have been made to model the effectiveness of a VFS based upon soil, vegetation, and sediment properties, and also upon flow conditions, but little work has been done to investigate the reliability of the existing models when considering sediments, such as microbial pathogens, that have a lower density than the mineral sediments used for development of the models. The objectives of this study were to: 1) quantify the ability of a VFS to remove …

Changes In The Structure And Function Of Microbial Communities In Drinking Water Treatment Bioreactors Upon Addition Of Phosphorus, Xu Li, Giridhar Upadhyaya, Wangki Yuen, Jess Brown, Eberhard Morgenroth, Lutgarde Raskin

Department of Civil and Environmental Engineering: Faculty Publications

Phosphorus was added as a nutrient to bench-scale and pilot-scale biologically active carbon (BAC) reactors operated for perchlorate and nitrate removal from contaminated groundwater. The two bioreactors responded similarly to phosphorus addition in terms of microbial community function (i.e., reactor performance), while drastically different responses in microbial community structure were detected. Improvement in reactor performance with respect to perchlorate and nitrate removal started within a few days after phosphorus addition for both reactors. Microbial community structures were evaluated using molecular techniques targeting 16S rRNA genes. Clone library results showed that the relative abundance of perchlorate-reducing bacteria (PRB) Dechloromonas and Azospira …

Department of Agricultural and Biological Systems Engineering: Faculty Publications

Feedlot operators may increase profitability by including distillers grains in finishing diets. However, the nutrients remaining in the by‐product are concentrated by about a factor of three during the distillation process. Manure can be applied to meet single‐ or multiple‐year crop nutrient requirements. The water quality effects of the use of distillers grains in feedlot diets and multiple‐year manure application have not been well quantified. The objectives of this study were to (1) compare the runoff water quality effects resulting from the application of manure derived from corn and distillers grain diets, (2) examine the effects of till and no‐till …

Comparison Of Subsurface And Surface Runoff Phosphorus Transport Capacities In Alluvial Floodplains, Aaron R. Mittelstet, Derek M. Heeren, Daniel E. Storm, Garey A. Fox, M. J. White, R. B. Miller

Department of Agricultural and Biological Systems Engineering: Faculty Publications

There have been numerous studies on phosphorus (P) contributions from surface runoff, but studies comparing the contribution of surface versus subsurface P are limited, as subsurface transport is often considered negligible. Previous work has shown that the transport of P in the gravelly subsurface at two sites in northeast Oklahoma can be significant, especially in preferential flow paths (PFPs), hypothesized to be buried gravel bars. The objective of this project was to quantify subsurface P losses based on field data, and compare with surface runoff P losses derived from Pasture Phosphorus Management Calculator (PPM Plus) simulations. Ozark ecoregion study sites …

Electromagnetic Field Calculations For Microlens Optical Systems, Jian Wang Oct 2010

Electromagnetic Field Calculations For Microlens Optical Systems, Jian Wang

Department of Mechanical and Materials Engineering: Dissertations, Theses, and Student Research

Microlenses are becoming more widely used in modern optical equipment. When the microlens diameter is comparable with the incident electromagnetic illumination wavelength, diffraction effects through the microlens aperture dominate and significantly affect the microlens optical properties leading to differences from that predicted by ordinary geometrical optics theory. In this work, the continuous-profile symmetrical biconvex microlens is selected for investigation. Its optical properties, with both monochromatic plane wave and TEM00 mode Gaussian beam illumination, are studied using the full-field Separation-of-Variables method (SVM) in the oblate spheroidal coordinate system by calculating the electromagnetic field distributions inside of and adjacent to the microlens. …
