Retention Of Cellular Viability And Growth Potential In Cryopreserved Bovine Arterial Tissue: Implications For Clinical Use Of Cryopreserved Vascular Allografts, Kip G. Gardner

Segments of bovine calf aortas and pulmonary arteries were cryopreserved at -196° C for two weeks to 18 months and subsequently thawed. Cellular morphology, viability, and growth potential in these segments were then compared with the same attributes in segments from fresh (control) arteries. Scanning electron microscopy revealed no disruption of intimal surfaces attributable to cryopreservation. Endothelial cells were successfully cultured from all cryopreserved arterial segments. Endothelial monolayers grown from cryopreserved arteries had a cobblestone appearance and expressed Factor VIII antigen but not smooth muscle alpha-actin. Plating efficiency in primary cultures of endothelial cells was lower for cryopreserved arteries than …

Fucoidan is a polysaccharide commonly found in brown algae. It is a sulfated polymer of the monosaccharide L-fucose. Fucoidan is thought to function in water retention by algae. Commercially available fucoidan is extracted from a brown algae called Eurus vesiculosus.

This polysaccharide has immense potential as a biologically active agent: (i) it exhibits anticoagulant action, (ii) it has also been reported to block retroviral replication and (iii) it is capable of blocking sperm-egg binding in sea urchin, mouse and rabbit. A transient model to predict temperature within a tunnel ventilated broiler house during misting is developed. The model is calibrated with field data to obtain steady-state constants; transient predictions are compared to measured temperatures during cyclic misting for two different size birds. Measured temperatures during cyclic misting are shown to swing between steady-state asymptotes predicted from the model. Transient response of the model was faster than measured temperature data, in part due to temperature sensor dynamic response. The model predicts the length-wise temperature profile within the building during misting, and can be used to investigate alternate misting strategies and designs. …

A series of mechanical backup systems for electronic environmental controllers is presented for a typical finishing swine barn and a typical tunnel ventilated broiler house. The systems consist of mechanical thermostats and timers used in parallel with the electronic controller, designed to ensure animal survival in the event of controller or related hardware failure. For swine housing, three distinct mechanical backup functions are identified; for broiler housing, four distinct mechanical backup functions are identified. Schematic diagrams of the mechanical backup functions are provided and their implementation is described.

The variables considerd to be affecting the card sliver quality are as follow: blowroom lines/chute feed arrangement, carding machines were used throughout, days of production and cotton fibers. A statistical analysis were made of the sliver parameters, its size, count variations neps per gram and sliver irregularity, using three factor analysis of variance. Two new techniques for quantification of forces generated by an expansion screw apparatus placed between pairs of cylindrical endosseous titanium implants are described, along with preliminary data gained from their use in Macaca rhesus monkeys. Strain gages were bonded to the expansion apparatus and were calibrated either directly by micro load cell, or indirectly by Instron Machine to reflect the applied force.

Immediate loads of up to 6 kilograms have been applied to the implants with no increase in mobility as verified by a Periotest device. No previous studies on the orthodontic uses of implants have evaluated in vivo forces …

Sixteen commercially available electronic environmental controllers were evaluated. The units were classified according to enclosure type, analog versus microprocessor based control, power supply, sensors, alarms, control relays and triac output, interval timers, outside temperature feedback, and retail price. An assessment of these controllers indicated several critical limitations in the application of this technology.

The use of integrated controllers for animal production has the potential for substantial improvements in production efficiencies. If the limitations observed in the present controller technology, as represented by this sample, are addressed, industry acceptance of the technology can be accelerated. The integrated electronic control system will provide a new method for the day-to-day management of environmental control of animal production systems. No standards are currently accepted for transient overvoltage protection of these controllers. To assess the adequacy of existing designs, a test circuit was designed and used for a transient open circuit over-voltage waveform (ANSI/IEEE C62.41-1980) of 16 environmental control units: a maximum spike of 770 V was applied to the power supplies, and a spike up to 100 V was applied to temperature sensor lines. Farming in Western Australia, and in Australia, has undergone a revolution over the past 15 to 20 years. Two significant changes have been the cessation of ploughing to kill weeds, that is, the change from discs to tines, and the reduction in tillage through the use of agricultural chemicals. These have been important changes that have had conservation benefits. However, to handle stubble to best effect, further changes in machinery, newer machinery and combinations of machines will be needed. In this article, the author discusses current and possible future practces.

The strength and height of the fence kept domestic animals confined. For many years, conventional fences have served the farmer well. However because returns from livestock over the past ten years have been variable and because of the current cost/ price squeeze, electric fencing, as a cheaper form of fencing, has gained popularity.

A murine monoclonal antibody (MAb), 7E11-C5, recognizes a prostate organ-specific antigen. Under reducing conditions, the 7E11-C5 antigen exhibited major 100-Kd and minor 70-Kd bands, suggesting that the antigen comprises two peptide chains. The antigen was detected in pooled normal human seminal plasma, xenograft tissue extract of a prostate adenocarcinoma cell line, LNCaP, and tissue extracts of benign prostatic hyperplasia (BPH) and prostate carcinomas, but was not identified in any of the non-prostate tissue extracts tested. Antibodies of the three major prostate-associated antigens (prostatic acid phosphatase (PAP), prostate specific antigen (PSA) and prostate secretory protein (PSP)) were unable to block the …

This study describes morphological changes in the lung at several time intervals following a single injection of 1-Nitronaphthalene (100 mg/kg intraperitoneal) in male Sprague-Dawley rats using transmission and scanning electron microscopy. Dilation of the smooth endoplasmic reticulum and mitochondrial distension in the nonciliated bronchiolar (Clara) cells was apparent as early as 1 hour after injection, while adjacent ciliated cell showed minimal changes. At 6 hours, when respiratory distress was apparent in the animals, Clara and ciliated cells exhibited chromatin clumping, loss of mitochondrial cristae and cytoplasmic vacuolization. Also at 6 hours, the interstitial area around the terminal bronchioles was infiltrated …

Glycerophospholipids of mammalian cells exist as chemically diverse structures with various fatty acids at the sn-1 and sn-2 positions. Arachidonic acid, a polyunsaturated fatty acid, which may be converted to biologically active eicosanoids such as prostaglandins, thromboxanes, and leukotrienes, is found predominantly in the sn-2 position of glycerophospholipids. The purpose of this study was to examine, at the level of the individual molecular species, the incorporation of arachidonate into phospholipids and its release from phospholipids during stimulation. There is an expressed need to develop a greater variety of safe contraceptive methods which would find acceptance worldwide, particularly in developing countries. Immunization against spermatozoa might be such a method as judged by the accumulated evidence from studies on human females and in a number of female animal species.

Two extraction techniques were used for sperm membrane antigen isolation. The first technique involved NP-40 detergent for antigen extraction from human motile sperm and the second technique employed homogenized human testis for antigen extraction. Whole-Farm Planning : Success At Wilgi Creek, Kevin Shanhun

Ian and Bev Lynch own Wilgi Creek, a 376 ha mixed farming property at West Mount Barker in the 700 mm rainfall zone. In 1983, they started a whole-farm plan to overcome the problems of declining production caused by waterlogging (their biggest problem), salinity and deterioration of the remnant native vegetation. Today, their property is an example of a successful, wholefarm land conservation plan based on agroforestry, timber production, water harvesting and improved pastures.

Narrow-Winged Seeder Points Reduce Water Erosion And Maintain Crop Yields, Kevin Bligh

Sowing crops without loosening the topsoil by tillage reduces water erosion. It can increase infiltration of rainfall into loamy soils, thereby reducing runoff and increasing potential crop yields. Crop yields were maintained after I1 seasons of seeding an Avon Valley loam near Beverley with minimum and no-tillage seeding operations. Infiltration increased significantly from 80 per cent of the 1983 growing-season rainfall under the traditional three tillage operations, to 87per cent under a single tillage operation using a combine seed drill. Infiltration increased further to 96 per cent under a no-tillage system using a triple^lisc drill. At Gnowangerup, 80 per cent …

The Lake Mead Fertilization Project is a research program designed to investigate the potential for using large-scale artificial fertilization to enhance the game fisheries of this reservoir through an increase in the population of threadfin shad, the system's primary forage species. A substantial decline in the population of largemouth bass, together with poor condition of adult striped bass, are the two major issues affecting the Lake Mead game fisheries. Both issues have been hypothesized to be a result of an inadequate amount of forage in the reservoir. Previous studies have in turn suggested that a major factor limiting the shad …

A new concept for mechanically harvesting bell peppers, intended specifically for multiple-pass harvesting, utilized spaced horizontal fingers combing vertically upward through plant foliage in an elliptical path alternating from both sides of the row. An apparatus based on this concept was designed and built, and harvest tests were conducted to determine the effect of two main machine operating parameters, vertical picking speed and disk angle. Harvesting performance was evaluated in terms of harvest efficiency, fruit damage, and plant damage as it related to multiple-pass harvesting. Fruit removal, both harvestable size and immature, increased significantly as vertical picking speed increased. Branch …
