Immune checkpoint inhibitors (ICIs) are an effective therapy for various cancers; however, they can induce immune-related adverse events (irAEs) as a side effect. Myocarditis is an uncommon, but fatal, irAE caused after ICI treatments. Currently, the mechanism of ICI-associated myocarditis is unclear. Here, we show the development of myocarditis in A/J mice induced by anti-PD-1 monoclonal antibody (mAb) administration alone without tumor cell inoculation, immunization, or viral infection. Mice with myocarditis have increased cardiac infiltration, elevated cardiac troponin levels, and arrhythmia. Anti-PD-1 mAb treatment also causes irAEs in other organs. Subclinical mastitis (SCM) is an inflammatory state of the lactating mammary gland, which is asymptomatic and may have negative consequences for child growth. The objectives of this study were to: (1) test the association between the dietary inflammatory index (DII®) and SCM and (2) assess the differences in nutrient intakes between women without SCM and those with SCM. One hundred and seventy-seven women with available data on human milk (HM) sodium potassium ratio (Na:K) and dietary intake data were included for analysis. Animal bodies maintain themselves with the help of networks of physiological processes operating over a wide range of timescales. Many physiological signals are characterized by 1/f scaling where the amplitude is inversely proportional to frequency, presumably reflecting the multi-scale nature of the underlying network. Although there are many general theories of such scaling, it is less clear how they are grounded on the specific constraints faced by biological systems. To help understand the nature of this phenomenon, we propose to pay attention not only to the geometry of scaling processes but also to their energy. The first key assumption …

Our current understanding of the spectrum of TB and COVID-19 lesions in the human lung is limited by a reliance on low-resolution imaging platforms that cannot provide accurate 3D representations of lesion types within the context of the whole lung. To characterize TB and COVID-19 lesions in 3D, we applied micro/nanocomputed tomography to surgically resected, postmortem, and paraffin-embedded human lung tissue. We define a spectrum of TB pathologies, including cavitary lesions, calcium deposits outside and inside necrotic granulomas and mycetomas, and vascular rearrangement. Primary immune regulatory disorders (PIRD) represent a group of disorders characterized by immune dysregulation, presenting with a wide range of clinical disease, including autoimmunity, autoinflammation, or lymphoproliferation. Autosomal dominant germline gain-of-function (GOF) variants in STAT3 result in a PIRD with a broad clinical spectrum. Studies in patients have documented a decreased frequency of FOXP3+ Tregs and an increased frequency of Th17 cells in some patients with active disease. However, the mechanisms of disease pathogenesis in STAT3 GOF syndrome remain largely unknown, and treatment is challenging. Naturally produced peptides (<100 amino acids) are important regulators of physiology, development, and metabolism. Recent studies have predicted that thousands of peptides may be translated from transcripts containing small open-reading frames (smORFs). Here, we describe two peptides in Drosophila encoded by conserved smORFs, Sloth1 and Sloth2. These peptides are translated from the same bicistronic transcript and share sequence similarities, suggesting that they encode paralogs. Yet, Sloth1 and Sloth2 are not functionally redundant, and loss of either peptide causes animal lethality, reduced neuronal function, impaired mitochondrial function, and neurodegeneration. We provide evidence that Sloth1/2 are highly expressed in neurons, imported to mitochondria, and regulate mitochondrial complex III assembly. These results suggest that phenotypic analysis of smORF genes in Drosophila can provide a wealth of information on the biological functions of this poorly characterized class of …100>

Mechanisms that underlie adipose tissue remodeling to enhance metabolic health in response to exercise training remain inadequately understood. PURPOSE: We utilized mouse genetics and human GWAS to determine the impact of exercise training on mitochondrial DNA copy number, and interrogate the relationship between Esr1 and adipose tissue health. METHODS: We performed RNAseq on adipose tissue from 100 strains of inbred mice following exercise training and determined mitochondrial content by qPCR. Soil samples and compaction readings were taken from a corn maze site at Murray State University Pullen Farm. The scope of this project was compare the years of overlap with the different designs in the corn maze. This foot traffic compaction can be relayed back to the affect of livestock in fields.

It is 1977 in a Black neighborhood of Pittsburgh known as the Hill District. Drivers of unlicensed cabs (jitneys) fight for love, survival, and respect as the powers-that-be threaten to close down their garage in the name of neighborhood improvement.
Though Jitney is one of August Wilson’s first plays, it was the last of his 10-play American Century Cycle to premiere on Broadway. Using Wilson’s characteristically rich language, Jitney explores what happens when tempers flare, potent secrets are revealed, and the fragile threads that bind people together threaten to come undone.

OBJECTIVE To evaluate efficacy of a novel vaccine against rabbit hemorrhagic disease virus 2 (RHDV2) in domestic rabbits.
ANIMALS 40 New Zealand White rabbits obtained from a commercial breeder.
PROCEDURES Rabbits were vaccinated and held at the production facility for the duration of the vaccination phase and transferred to Colorado State University for challenge with RHDV2. Rabbits were challenged with oral suspensions containing infectious virus and monitored for clinical disease for up to 10 days. Rabbits that died or were euthanized following infection were necropsied, and livers were evaluated for viral RNA via RT-PCR.

This paper argues for the necessity of formalizing a new, practically-oriented sub-discipline of AI ethics by outlining the needs, highlighting shortcomings in current approaches, and providing a framework for ethical analytics, which is concerned with the study of the ethical issues surrounding the development, deployment, and/or dissemination of ML/AI systems and data science research, as well as the development of tools and procedures to mitigate ethical harms. Androgen deprivation therapies aimed to target prostate cancer (PrCa) are only partially successful given the occurrence of neuroendocrine PrCa (NEPrCa), a highly aggressive and highly metastatic form of PrCa, for which there is no effective therapeutic approach. Our group has demonstrated that while absent in prostate adenocarcinoma, the αVβ3 integrin expression is increased during PrCa progression toward NEPrCa. Here, we show a novel pathway activated by αVβ3 that promotes NE differentiation (NED).
