Midazolam (MDZ), a benzodiazepine derivative, is metabolized to 1′- and 4-hydroxylated metabolites (1′-OH-MDZ and 4-OH-MDZ, respectively) by cytochrome P450 3A (CYP3A). The purpose of this study was to investigate the CYP3A-mediated hydroxylation of MDZ in the rat brain mitochondria (MT). Brain microsomes (MC) and MT fractions were prepared from rats (n = 8) using differential and density gradient centrifugations, and the purity of the fractions was evaluated using VDAC1 and calreticulin as markers of MT and MC, respectively. Non-O157 Shiga toxin-producing Escherichia coli (STEC) are emerging serogroups that often result in diseases ranging from diarrhea to severe hemorrhagic colitis in humans. The most common non-O157 STEC are O26, O45, O103, O111, O121, and O145. These serogroups are known by the name “big six” because they cause severe illness and death in humans and the United States Department of Agriculture declared these serogroups as food contaminants. The lack of fast and efficient diagnostic methods exacerbates the public impact of the disease caused by these serogroups. Síi Túupentak (CA-SCA-565/H) is a late precontact ancestral Ohlone village and cemetery site in central California (ca. 540-145 cal. BP). Integration of proteomic, genomic, and osteological analyses provided highly confident biological sex estimates for remains of most individuals at this site (65 of 76) spanning all age groups—from perinatal infants to aged adults. The comprehensive nature of the data allowed us to generate sex-specific Kaplan-Meyer survivorship curves for this burial population of sedentary hunter-gatherers. As was common among societies pre-dating modern western medicine and antibiotics, infant mortality was high for both males and females. However, male infants (from birth to …

Shape memory polymers (SMPs) are a class of smart materials that can be temporarily stored in a deformed shape and can actively recover their original shape upon exposure to an external stimulus, such as heat, magnetic field, moisture, pH, light, or electric field. The ability of SMPs to change shape when required can be used for a wide range of applications, especially in the case of minimally invasive biomedical applications. In this work, polyurethane-based SMPs were explored for their use in tissue engineering, drug delivery, and wound healing applications.

"Analytical," "Angry." And "(Too) Honest"?! Learners' Stereotypes About Teachers Of German., Lucian Rothe

This study investigated stereotypes about teachers of German that 67 novice learners of German at a U.S. university had reportedly encountered. Consequently, the investigation explored the perceived accuracy of clichés associated with native-speaker and non-native-speaker instructors. Students' stereotypical notions were sorted into categories using principles of grounded theory. Statistical analyses showed that respondents were able to list various stereotypes about instructors of German. Responses also reflected societal narratives concerning native-speaker and non-native-speaker teachers' strengths and shortcomings. This dissertation charts the significant, if understudied, history of American horses during the era of World War I, from roughly 1914 to 1919. Its chapters trace how the US Army acquired, used, cared for, and ultimately demobilized horses over the course of that conflict. Beginning with their acquisition, via either an Army Horse Breeding Program or a complicated buying process, horses faced a complex introduction into military service. Life for these animals did not get any easier once they reached the European front. Dye design can influence the ability of fluorescently labeled imaging agents to generate tumor contrast and has become an area of significant interest in the field of fluorescence-guided surgery (FGS). Here, we show that the charge-balanced near-infrared fluorescent (NIRF) dye FNIR-Tag enhances the imaging properties of a fluorescently labeled somatostatin analogue.

Blank Spaces explores the way childhood nostalgia manifests in the physical world through the creation of mixed media pieces. The experimentation with different mediums in my work, such as painting, ceramics, and animation, is emblematic of childhood creative play. These mixed media artworks showcase how memories recalled from the past can influence our interpretation of the present. The familial connection to family and culture, represented through manipulated old family photographs and imaginary renderings of stories, blends representational imagery with fantasy; my art examines the movement of those memories in the “blank spaces” of the mind. Modern competitive video games are complex systems that require players to learn techniques and develop strategies in response to computer- or human-controlled opponents. Building on the work of both game studies scholars and game designers, this thesis analyzes various learning spaces within and surrounding competitive video games, including in-game tutorials, arcades, and online communities. It focuses on “fighting games,” those competitive video games in which, typically, two players control characters to face off in single combat. The ability to accumulate rewards while minimizing negative consequences is a valuable survival skill. Importantly, many psychiatric diseases such as substance use disorder (SUD; Bechara, 2005; Gowin et al., 2013), attention deficit hyperactivity (Magnus et al., 2021), anxiety (Hartley and Phelps, 2012), major depressive, bipolar, and schizophrenia disorders (Whitton et al., 2015) involve impaired decision-making that can lead to detrimental outcomes. Similar to previous pandemics, COVID-19 has been succeeded by well-documented postinfectious sequelae, including chronic fatigue, cough, shortness of breath, myalgia, and concentration difficulties, which may last 5 to 12 weeks or longer after the acute phase of illness. Both the psychological stress of SARS-CoV-2 infection and being diagnosed with COVID-19 can upregulate cortisol, a stress hormone that disrupts the efferocytosis effectors, macrophages, and natural killer cells, leading to the excessive accumulation of senescent cells and disruption of biological barriers. Prostate cancer is one of the most common types of cancer, affecting over 191,930 men, with an estimated 34,500 deaths for 2022 in the United States (NIH, 2022). As men age, the risk for prostate cancer increases; about 34% of all men diagnosed with prostate cancer are above the age of 50, and a staggering 70% are over the age of 80. To ensure proper wound healing it is important to elucidate the signaling cues that coordinate leader and follower cell behavior to promote collective migration and proliferation for wound healing in response to injury. Using an ex vivo post-cataract surgery wound healing model we investigated the role of class I phosphatidylinositol-3-kinase (PI3K) isoforms in this process. Our findings revealed a specific role for p110α signaling independent of Akt for promoting the collective migration and proliferation of the epithelium for wound closure. To identify novel insecticide target sites that can address the evolution of insecticide resistance, our group has tested the insecticidal potential of two potassium flux pathways: potassium-chloride (K+/Cl−) cotransporter (KCC) and inward rectifier potassium (Kir) channels. GABAergic signaling is the cornerstone of fast synaptic inhibition for neural signaling in arthropods; thus, the γ-aminobutyric acid (GABA)-receptor-chloride-channel (GRCC) complex has been a longstanding target for insecticides. Despite the importance to neuronal homeostasis, the upstream mechanisms enabling GRCC function are poorly defined. Populists and Progressives around the turn of the 20th century had similar reform goals and many of the same foes. Both wanted to expand the power of the federal government to counter corporate monopolies which were dominating the nation’s economy and political system. In the early 1900s, Progressives enacted numerous reforms that were championed by Populists, including regulating railroads and other large corporations, breaking-up monopolies, implementing a graduated income tax, among other reforms. With so much agreement on policy, why did Populists and Progressives actively work against one another in the 1890s?

Childhood obesity is a pandemic affecting almost 40 million individuals aged 5-18 globally. Many current interventions are relatively successful at treating childhood obesity, yet the rate of childhood obesity continues to increase from year to year, and current intervention methods are struggling to keep up. Nanotechnology is a multifaceted and a promising strategy in agricultural pest management for the development of insecticides or insecticide carriers. Research has shown that nanoparticles increase stability and efficacy of entrapped chemical pesticides to prevent active ingredient degradation while improving chemical stability and solubility. The following work evaluated nanoparticles synthesized from biopolymers, materials that originate from renewable sources also have an increased biodegradability.

The Florida manatee (Trichechus manatus latirostris) mainly resides within the state's coastal waters, but sharing this habitat with thousands of watercrafts, either registered, unregistered, or visiting, has proven challenging for its survival. The future of manatee research among scientists and citizen scientists has strong potential. By taking advantage of venues of opportunity such as boat shows, not only is local population data gathered, but a stronger positive relationship with the public is forged. When people feel they have a stake in the conservation of a species or habitat, they will likely become and remain engaged.

The emerging infectious disease (EID) crisis represents an immediate existential threat to modern humanity. Current policies aimed at coping with the EID crisis are ineffective and unsustainably expensive. They have failed because they are based on a scientific paradigm that produced the parasite paradox. The Stockholm paradigm (SP) resolves the paradox by integrating four elements of evolutionary biology: ecological fitting, sloppy fitness space, coevolution, and responses to environmental perturbations. It explains why and how the EID crisis occurs and is expanding and what happens after an EID emerges that sets the stage for future EIDs. The SP provides a number …
