BACKGROUND: The prolyl-hydroxylase domain family of enzymes (PHD1-3) plays an important role in the cellular response to hypoxia by negatively regulating HIF-α proteins. Disruption of this process can lead to up-regulation of factors that promote tumorigenesis. We observed decreased basal expression of PHD3 in prostate cancer tissue and tumor cell lines representing diverse tissues of origin. Furthermore, some cancer lines displayed a failure of PHD3 mRNA induction when introduced to a hypoxic environment. This study explores the mechanism by which malignancies neither basally express PHD3 nor induce PHD3 under hypoxic conditions.
METHODOLOGY/PRINCIPAL FINDINGS: Using bisulfite sequencing and methylated DNA enrichment …

INTRODUCTION: Tumors that express estrogen receptor alpha (ERα+) comprise 75% of breast cancers in women. While treatments directed against this receptor have successfully lowered mortality rates, many primary tumors initially or later exhibit resistance. The paucity of murine models of this "luminal" tumor subtype has hindered studies of factors that promote their pathogenesis and modulate responsiveness to estrogen-directed therapeutics. Since epidemiologic studies closely link prolactin and the development of ERα+ tumors in women, we examined characteristics of the aggressive ERα+ and ERα- carcinomas which develop in response to mammary prolactin in a murine transgenic model (neu-related lipocalin- prolactin (NRL-PRL)). To …

On The Effect Of Criticality And Topology On Learning In Random Boolean Networks, Alireza Goudarzi
On The Effect Of Criticality And Topology On Learning In Random Boolean Networks, Alireza Goudarzi
Systems Science Friday Noon Seminar Series
Random Boolean networks (RBN) are discrete dynamical systems composed of N automata with a binary state, each of which interacts with other automata in the network. RBNs were originally introduced as simplified models of gene regulation. In this presentation, I will present recent work done conjointly with Natali Gulbahce (UCSF), Thimo Rohlf (MPI, CNRS), and Christof Teuscher (PSU). We extend the study of learning in feedforward Boolean networks to random Boolean networks (RBNs) and systematically explore the relationship between the learning capability, the network topology, the system size N, the training sample T, and the complexity of the computational task. …

Journal of Tourism Insights
In this study, the investigators use scores from the Tourist Ecological Orientation (TEO) Scale (Uriely, Reichel & Shani, 2007) of Costa Rica visitors to understand how their ecological orientation and interest in ecological practices influence accommodation and destination site choices. Results from this study were intended to be used to inform professionals in the tourism industry on how they might best develop their tourism sites, policies and programs according to the ecological orientation of the visitors. Based on the findings from this study, however, it seems that the Tourist Ecological Orientation Score derived by Uriely, Reichel and Shani (2007) is …

Nature-Based Tourism Businesses In Colorado: Interpreting Environmental Ethics And Responsible Behavior, Diane Gaede, Drew Strickert, Richard R. Jurin
Journal of Tourism Insights
Tourism businesses operate from a primarily economic-centric point of view, but nature-based tourism businesses are also acutely aware of the need to sustain the natural resource that attracts the client to their outdoor recreation service. A preliminary qualitative inquiry reveals how nature-based tourism organizations in Colorado view themselves as operating from environmentally ethical positions, what specific actions they take to minimize negative environmental impacts, and how they educate their clients about resource conservation. Findings indicate that too often companies fail to realize opportunities in which they can encourage meaningful bonds between people and nature through the use of education and …

Orang-utan- is derived from a Malay term meaning man of the forest- and aptly describes the southeast Asian great apes native to Sumatra and Borneo. The orang-utan species, Pongo abelii (Sumatran) and Pongo pygmaeus (Bornean), are the most phylogenetically distant great apes from humans, thereby providing an informative perspective on hominid evolution. Here we present a Sumatran orang-utan draft genome assembly and short read sequence data from five Sumatran and five Bornean orang-utan genomes. Our analyses reveal that, compared to other primates, the orang-utan genome has many unique features. Structural evolution of the orang-utan genome has proceeded much more slowly …

The relationship between biological research and mathematical modeling is complex, critical, and vital. In this review, we summarize the results of the collaboration between two laboratories, exploring the interaction between mathematical modeling and wet-lab immunology. During this collaboration several aspects of the immune defence against viral infections were investigated, focusing primarily on the subject of heterologous immunity. In this manuscript, we emphasize the topics where computational simulations were applied in conjunction with experiments, such as immune attrition, the growing and shrinking of cross-reactive T cell repertoires following repeated infections, the short and long-term effects of cross-reactive immunological memory, and the …

The PI3K/AKT signaling pathway is aberrant in a wide variety of cancers. Downstream effectors of AKT are involved in survival, growth and metabolic-related pathways. In contrast, contradictory data relating to AKT effects on cell motility and invasion, crucial prometastatic processes, have been reported pointing to a potential cell type and isoform type-specific AKT-driven function. By implication, study of AKT signaling should optimally be conducted in an appropriate intracellular environment. Prognosis in soft-tissue sarcoma (STS), the aggressive malignancies of mesenchymal origin, is poor, reflecting our modest ability to control metastasis, an effort hampered by lack of insight into molecular mechanisms driving …

Cancún Summit On Global Climate Change A Success Or Failure, Depending On Whom You Ask, Carlos Navarro
NotiEn: An Analytical Digest About Energy Issues in Latin America
Depending on whom you ask, the UN conference on climate change in Cancún, Mexico, on Nov. 29-Dec. 11, 2010, was a total failure or a step in the right direction to reduce greenhouse-gas emissions, generally thought to be responsible for the warming of the Earth's atmosphere. The Cancún gathering was also known as COP16—-an abbreviation for the sixteenth edition of the Conference of the Parties of the UN Framework Convention on Climate Change (UNFCCC). Those who viewed the results of COP16 with the "glass-is-half-full" premise included the host of the gathering, Mexican President Felipe Calderón, who said the agreements reached …

Examination of 268 specimens of Mesobuthus eupeus eupeus (C. L. Koch, 1839) and 1765 specimens of M. eupeus phillipsii (Pocock, 1889) from 163 localities, 92 of them in Turkey, has enabled us to establish distribution of the two subspecies and limits of their ranges in southeastern Turkey. A map of distribution of these subspecies in Turkey and a key to Turkish Mesobuthus species and subspecies are provided. Buthus eupeus philippovitschi Birula, 1905 is synonymized with Mesobuthus eupeus eupeus, and Buthus eupaeus mesopotamicus Penther, 1912 is synonymized with Mesobuthus eupeus phillipsii.

Background
Genetic markers are pivotal to modern genomics research; however, discovery and genotyping of molecular markers in oat has been hindered by the size and complexity of the genome, and by a scarcity of sequence data. The purpose of this study was to generate oat expressed sequence tag (EST) information, develop a bioinformatics pipeline for SNP discovery, and establish a method for rapid, cost-effective, and straightforward genotyping of SNP markers in complex polyploid genomes such as oat.
Results
Based on cDNA libraries of four cultivated oat genotypes, approximately 127,000 contigs were assembled from approximately one million Roche 454 sequence reads. …

Organic Dairying: Role Of Grazing, Jake Schmitz
Organic Dairying: Role Of Grazing, Jake Schmitz
Kentucky Grazing Conference
Certified organic and many conventional dairy farmers are using grazing techniques for a number of reasons, including building better soil, reducing feed costs, improving milk quality, and enhancing herd health. Additionally, certified organic farmers must utilize their pastures because the United States Department of Agriculture’s (USDA) organic rule requires organic cows to graze. Both the farmer and the USDA are working hard to ensure that “organic dairy” is synonymous with “grazing cows,” because the organic consumer vehemently demands dairy products from pasture-grazed livestock.

Afforestation And Timber Management Compliance Strategies In Climate Policy. A Computable General Equilibrium Analysis, Melania Michetti, Renato Nunes Rosa
Fondazione Eni Enrico Mattei Working Papers
This paper analyzes the role of afforestation-reforestation and timber management activities, and their major and secondary economic effects in stabilizing climate during the first commitment period of the Kyoto Protocol. In particular, with a Computable General Equilibrium framework, the ICES model, it is inferred how forest carbon sequestration fits within the European domestic portfolio of a 2020-20 and 2020-30 climate stabilization policy. Afforestation and land use are accounted for by introducing their effects in the model. This is done by relying on carbon sequestration curves provided by Sohngen (2005), which describe the average annual cost of sequestration for selected world …
