The bone marrow microenvironment composed in part of stromal cells provides the niche in which normal hematopoiesis occurs. Hematopoiesis generates cellular components of the immune system and blood through proliferation and differentiation of multipotent hematopoietic stem cells (HSC). In preparation for bone marrow transplantation, radiation or chemotherapy treatment temporarily disrupts the balance of hematopoiesis as many HSC and progenitor cells are destroyed. Following treatment cessation, transplanted HSC and progenitor cells migrate, or "home", back to the bone marrow microenvironment and initiate productive hematopoiesis. While the mechanism of bone marrow homing is not completely understood, several soluble factors and adhesion molecules …

The Myogenic Regulatory Factor family (MRFs) is a family of transcription factors that plays a key role in the development of skeletal muscle. MRFs include Myo D, myf-5, myogenin, and MRF-4. Myo D and myf-5 are genes expressed during myoblast proliferation, whereas myogenin and MRF-4 are expressed during myotube formation and differentiation. Insulin-Like Growth Factor 1 (IGF-1) controls muscle development both embryonically and during postnatal growth and repair by inducing myoblast proliferation and muscle hypertrophy. Abandoned mine lands are areas affected by past coal mining activities that often pose significant water quality problems from acid mine drainage (AMD). Weather, a primary determinant of AMD, is inherently stochastic. The economic costs of treating AMD problems are an issue of importance to policymakers and citizens. This study develops a spatial model to devise cost effective plans for treating AMD in a watershed when pollution loads are stochastic and the level of certainty of treating all of the AMD is allowed to vary. The UROtsa cell line is a unique model of human urothelium with considerable utility in toxicology-based research. UROtsa cells maintained in serum-free medium have several morphological features of transitional epithelium of the urinary bladder including a multilayered appearance and apically located umbrella-like cells that exhibit tight junctions. These cells also have a basal expression pattern of heat shock protein (hsp) and metallothionein (MT) genes similar to that observed in in situ urothelium. Integrated biomarkers of physiologic and genetic damage in sperm are needed to identify factors that affect male fertility and influence a father's risk of producing abnormal progeny. Offspring studies show a preferential paternal origin for heritable structural chromosomal abnormalities, presenting the need for methods to efficiently detect structural chromosomal abnormalities in human sperm. This dissertation breaks new conceptual ground toward the understanding of paternally transmitted genetic disease by adapting FISH technology to detect human sperm that carry chromosomal breaks and partial chromosomal duplications and deletions and by integrating state-of-the-art biomarkers of semen quality including computer-assisted sperm analysis (CASA). The Ca-ATPase of sarcoplasmic reticulum is a 110 kDa integral membrane protein responsible for calcium transport from the cytosol into the sarcoplasmic reticulum lumen to promote muscle relaxation. In the heart, the Ca-ATPase is regulated by phospholamban, a 52 residue integral membrane protein, which inhibits the Ca-ATPase by decreasing the apparent calcium affinity of the enzyme. Phospholamban phosphorylation relieves Ca-ATPase inhibition. At present, the physical mechanism by which phospholamban regulates the Ca-ATPase is not known. Birds have few reliable indicators of aging. Pentosidine (Ps), a biomarker of aging, is a product of non-enzymatic glycation, which accumulates in the tissues of an animal over its lifespan. The intent of this study was to determine if comparable change in Ps concentrations could be established in the skin of wild birds of known ages. Skin samples were obtained from the breast of 44 birds of various species. Foot webbing samples were obtained from 24 California Gulls. Mountaintop surface mining for coal has been practiced in West Virginia for over three decades, but its environmental impacts have become increasingly scrutinized in recent years. Therefore, this study evaluated soil development on different aged mined lands. Four different ages (2, 7, 11, and 23 years) of minesoils and two different slope classes, nearly level to gently sloping and steep to very steep, along with three contiguous native soils on a reclaimed mountaintop removal mine in Logan County, West Virginia were described and sampled. Extensive biochemical and genetic analyses suggest that majority of retroviruses employ similar mechanisms for specific RNA packaging. Better understanding of mechanisms of this process may help to develop new anti-retroviral drugs against such serious pathogens as human immunodeficiency virus types 1 and 2.;This study concerns with specific packaging of RNA by two simple retroviruses, murine leukemia virus (MLV) and spleen necrosis virus (SNV). This study examined gender and age-related differences in reasons for living, using a sample of 50 young adults (M = 20.2), 53 middle-aged adults (M = 49.3), and 55 older adults ( M = 78.5). Participants completed a combined version of the Reasons for Living Inventory, the Reasons for Living Inventory for Younger Adults, and the Reasons for Living Inventory for Older Adults. Although no gender differences emerged in reasons for living, younger adults placed more importance on fear of social disapproval, moral objections, survival and coping skills, peer acceptance, survival and coping beliefs, fear of suicide, and future optimism …

The purpose of this study was to determine the effects of concept mapping on academic achievement in anatomy and transfer of anatomy knowledge to kinesiology and to describe the participants' experience with this new study strategy. Methods. Thirty-eight junior physical therapy and occupational therapy students volunteered to participate in this study in the fall of 2000. Thirty students matriculated into the spring semester and completed the study. All participants signed informed consents. This research focuses on diffusion of an education innovation in a large, traditional University. In March 1999, the Blackboard digital course management system was installed for enterprise-wide availability at Temple University, the 39th largest university in the United States. The web-enabled database of Temple Blackboard logs the adoption date, course ID, and course title for every Blackboard course, unobtrusively, twenty-four hours a day. Temple Blackboard serves as 4 digital approximation of the cumulative recorder pioneered by B. F. Skinner, recording more than 2800 course adoptions across 30 months, in real time. Ohio River electrofishing surveys identified embayments as preferred macrohabitat of subadult largemouth bass. Catch rates within macrohabitats were low ranging from 0.00--2.73 subadult bass/hour. Embayment electrofishing CPUE was higher than tributary or main channel except in spring, when tributary CPUE was highest.;Core use areas were calculated from biotelemetry data and ranged from 88--59,400 m2, representing 0.02--89.00% of available study location macrohabitat. Subadult bass movement was limited to nursery embayments or tributaries with infrequent movement to nearby main channel areas. Bass preferred a mixed microhabitat consisting of a combination of vegetation and woody debris. This mixed microhabitat was less common in …

The Deckers Creek drainage basin covers approximately 16,600 hectares (62.9 square miles) in Monongalia and Preston Counties of West Virginia. Throughout the twentieth century, this watershed has received high levels of pollution in several forms. A 1974 study examined the water chemistry in Deckers Creek over a six-month period at 29 sample points. Water samples from the same sites were collected and analyzed monthly from March 1999 through October 2000 for acidity, alkalinity, total iron, aluminum, manganese, and other metals, as well as for fecal coliform and fecal streptococcus concentrations. The purpose of this study was to provide information that might be useful to researchers and extension personnel at West Virginia University in developing and designing educational programs concerning pest management for corn farmers. This research identified current trends in management practices, insect, fungal, and weed problems, and educational needs so that instructional materials can be designed to meet clientele needs. Data were collected via 100 questionnaires mailed to farmers in the top ten corn producing counties in West Virginia. In porcine ovaries, endothelin-1 (ET-1) may have an autocrine/paracrine role in follicular development. Endothelin-converting enzyme-1 (ECE-1) converts big endothelin-1 to ET-1. In situ hybridization was used to determine the ovarian compartment that synthesizes ECE-1. ECE-1 mRNA was detected in granulosa cells of large (10 mm) and medium (3 mm) antral follicles. This study, in conjunction with previous studies suggests a novel autocrine role of ET-1 as a possible local ovarian factor of follicular development. ET-1 may play a role during prostaglandin (PG) F2 alpha-induced luteal regression in the cow. Effects of PGF2 alpha treatment were examined during early and mid-luteal …

Raptor abundance and diversity were compared between 4 treatments (grassland, shrub/pole, fragmented forest, and intact forest) over 3 seasons (migration, summer, and winter) on 3 mountaintop/valley fill mines in southern West Virginia. Habitat characteristics of Red-shouldered Hawks (Buteo lineatus) were examined at landscape and microhabitat scales. Forty-eight points were surveyed monthly (Feb. 2000--Jan. 2001) using broadcast calls. I quantified microhabitat characteristics within an 11.3 m radius plot and landscape characteristics within 1000 and 564 m buffer zones at each point. Abundance was highest in grasslands; richness did not differ among treatments. Overall, I detected a shift from a forest to …

This dendrochronological study used dated and measured tree-ring data to examine relationships between radial growth and climate for four hard wood species, yellow-poplar, northern red oak, chestnut oak and red maple, growing on contrasting topographic aspects of a watershed in north-central West Virginia.;The study showed that all species except northern red oak showed significant differences in growth between the mesic and xeric aspects. Where significant differences were found all except chestnut oak exhibited higher growth rates at the mesic aspect.;Yellow-poplar showed a sharp decline in growth from late 1950's to late 1960's, which was evidently caused by several years of …

Chronic, inescapable stress during pregnancy (i.e. prenatal stress) modifies the behavior of the adult offspring. Several behaviors were evaluated in adult prenatally stressed (PS) rats, most notably the acoustic startle response and behavioral inhibition (i.e. freezing). PS rats do not differ in baseline peak startle responding, or in the habituation of the peak startle response when compared to the non-prenatally stressed (CON) rats. However, PS rats have greater peak responses and longer latencies to reach the peak response following systemic doses of the 5-HT 1A agonist, 8-OH-DPAT, compared to CON rats. Previous studies demonstrated that quinidine causes G1/G0 cell cycle arrest and inhibition of proliferation in MCF-7 human breast cancer cell line (Woodfork, K. et al., 1995). The goal of studies reported here was to understand the molecular mechanisms of c-myc gene regulation by quinidine. C-myc is one of the most common oncogene aberrations in breast cancers (Deming, S. L. et al., 2000). C-myc functions include regulation of cell cycle, proliferation, differentiation, and apoptosis. Stand dynamics and disturbance histories of five eastern, oak-dominated old-growth stands were investigated using ecological field sampling techniques, dendrochronologogical methods, and a review of historical resources of original forest composition and patterns of human and natural disturbance. Witness tree tallies from 18th century land surveys show that presettlement forests in the study areas were oak-dominated forests. Quercus alba was, at minimum, twice as abundant as Q. velutina, the second ranked species. Acer saccharum and Fagus grandifolia were among the top ten ranked species at each site; however their importance value was consistently less than one-third the value for Q. alba. …

The development of a general route to the N-[2-(1-alkynyl)phenyl]ketenimines (enyne-ketenimines) and N-[2-(1-alkynyl)phenyl]carbodiimides (enyne-carbodiimides) and their subsequent thermal behavior are described. Like enyne-allene and enyne-ketene systems, these enyne-hetereocumulenes undergo cycloaromatization through two competing biradical mechanisms under thermal conditions and therefore could serve as potential DNA cleaving agents. The resultant nitrogen-containing hetereocyclic compounds are also biologically interesting. The research described herein provides a easy access to the synthesis of benzo[b]carbazole, indoloquinoline and indolonaphthyridine alkaloids. In addition, the new synthetic strategies for these compounds are very versatile, providing access to a diverse array of analogs of these alkaloids.

This dissertation examines the interaction between the various forces of Polish "nationalism" and "feminism" in the construction of women's physical culture in modern Poland from the late nineteenth century until the end of the twentieth century. The basic premise of this work is that unlike most western countries where sportswomen have typically been marginalized in their athletic endeavors, Polish female athletes have earned far greater respect and recognition in the development of their physical culture. This present study was designed to examine the relation between cardiovascular reactivity to stress and the behaviors of individuals classified as either "extraverts" or "introverts." Although experimental psychophysiological studies have been conducted to examine the relation between extraversion and physiological arousal, little is known about the ways in which extraversion and cardiovascular reactivity to stress are related. According to the optimal arousal theory, both extraverts and introverts would be expected to be more reactive to a social challenge than to a mundane non-social task, with introverts exhibiting greater reactivity to both tasks than extraverts. A mild and efficient route to substituted carbazolones has been developed. This novel procedure consists of two consecutive palladium-catalyzed reactions, an intermolecular Stille coupling followed by an intramolecular reductive N-heteroannulation. For example, 1,2-dihydro-4(3H)-carbazolone was prepared in good isolated yield (74%) by the reductive cyclization of 2-(2-nitrophenyl)-2-cyclohexen-1-one using Pd(dba)2 (6 mol%), 1,3-bis(diphenylphosphino)propane (6 mol%), 1,10-phenanthroline monohydrate (12 mol%), and carbon monoxide (90 psi) in DMF at 80°C. 2-(2-Nitrophenyl)-2-cyclohexen-1-one was prepared via a Stille coupling of 2-iodo-2-cyclohexen-1-one and 2-(tri-n-butylstannyl)-1-nitrobenzene. This study addresses the genotoxicity of oxidative stress. We induced oxidative stress in cultured rat oligodendrocytes by exposure to hydrogen peroxide (H2O2). Intact genomic DNA was isolated by agarose embedding and its fragmentation was assessed by field inversion gel electrophoresis (FIGE). To detect single strand fragmentations, the DNA was digested with S1 nuclease before FIGE. We found that the exposure of oligodendrocytes to pathological concentrations of H2O2, induces a rapid digestion of nuclear chromatin in a pattern consistent with higher order chromatin degradation (HOCD). The responses of college freshmen placed on academic probation to a survey examining the factors leading to their poor academic performance were studied to ascertain the perceived problem areas. One hundred and seventy-two college freshmen selected factors concerning motivation, time management, social, and financial components, which they believed contributed to their poor academic performance during their freshman year. For males, inadequate balance between time and other activities was perceived as the number one factor contributing to their demise. A study was conducted to determine brake-specific emissions from diesel-powered off-highway heavy equipment that were exercised on steady-state and transient cycles. Two vehicles were evaluated in this study, a streetsweeper (114 hp) and a rubber-tired loader (110 hp). Due to the lack of appropriate transient cycles available for these vehicles, methods were developed to document infield operation for the creation of test cycles and strategies.
