This study focuses on the application evaluation of a prototype backscatter imaging LDV system (BILS). This non-intrusive instrument has the ability to record particle images and two-dimensional velocity data simultaneously. A series of validation tests were conducted to verify the operation of the system. These included comparing the velocity of a wire, attached to a rotating disc, with known angular frequency (measured by digistrobe), with that measured by the instrument. The results were found to be in good agreement. For applications in civil infrastructure rehabilitation and new construction, wood is being reinforced with externally bonded laminates and fabrics of fiber-reinforced polymer (FRP) composites. The potential benefits of external reinforcement with bonded FRP composites have been demonstrated through several studies, and the field implementation of this technology has been successfully proven in numerous projects. This study investigated the relationship between faculty-student communication, student time management, student expectations, and quality of instruction and the level of student success in distance courses. The population of the study consisted of undergraduate students enrolled in Independent Study courses at Mountain State University (N = 1007) during the summer and fall 2001 terms. The sample consisted of 500 students randomly selected from the population. Participants completed the Distance Education Satisfaction Survey developed by the researcher. The objective of study was to design, develop, and evaluate techniques to measure exhaust emissions from diesel-powered off road equipment. Emissions data was collected from a Komatsu hydraulic excavator and a Caterpillar track-type tractor.;For the excavator, data was logged during in-field operation, and test cycles were created on a dynamometer test bed using this data. For the tractor, on-board emissions measurement equipment collected mass emissions rate data in field.;Vast differences were found between transient and 8-mode brake-specific test results. Transient cycle HC, CO, CO2, NO, and PM emissions were 50%, 65%, 10%, 35%, and 50% lower than 8-Mode results, respectively. …

The target population consisted of all alumni that graduated between the years 1991 and 2001. The objective of the study was to characterize the mRNA and protein expression of the MT gene in response to Cd2+ exposure in the 267-B1 immortalized normal human neonatal prostate cell line. The cells were exposed to 18muM Cd2+ for 4hr and rescued for 48hr in P4-8F normal medium. Samples were obtained at 0, 1, 2, 4hr exposure and 4, 8, 12, 16, 24, 36, and 48hr recovery. The risk based sport fish consumption advisory protocol, West Virginia Sport Fish Consumption Advisory Guide (the Guide), was drafted for the state. Fish were collected from ten West Virginia sites (5 previously identified as contaminated, 5 previously identified as uncontaminated). Fish samples were collected, processed and analyzed for total mercury concentration and Aroclors (PCBs). Recommended meal frequencies were calculated from contaminant levels and based on the Guide. Mean fillet sample contaminant levels were compared to composite sample levels and found to require the same level or in one case a more protective of consumption advisory. Neurons using gamma-aminobutyric acid (GABA) as a neurotransmitter are critically important as inhibitory neurons in the CNS. A major hindrance to further studies of this important and highly diverse class of neurons is their relative scarcity, which makes it difficult to identify and target them for morphological and physiological studies. To overcome this difficulty, we developed a novel protocol for visualizing detailed morphology of GABAergic neurons in living mouse brain slice cultures, using biolistic transfection with GAD67-GFP. The morphological, immunocytochemical and physiological characteristics of GAD67-GFP expressing neurons indicated that they were functional GABAergic neurons. Uric acid is an important antioxidant due to its role as a free radical scavenger, and methods to elevate its plasma concentration may be important in animal health. In a preliminary study, the effect of dietary protein on plasma uric acid (PUA) was determined in 3-wk-old chicks. Twenty-four broiler chicks were randomly assigned to 4 diets: low protein (LP) containing 10% casein, medium protein (MP) containing 20% casein, high protein (HP) containing 45% casein and a commercial diet (COD) for 2-wks. The objective of this research was to determine if disease on susceptible tomato cultivars is reduced by intercropping with resistant cultivars. A second objective was to investigate the mechanism of the disease reduction observed. Early blight (caused by Alternaria solani) rate of disease increase and lesion expansion were lower on susceptible tomato cv. 'Brandywine' when intercropped with resistant cv. 'Juliet' than with 'Brandywine' monoculture in both the field and greenhouse. Yield from 'Brandywine' plants was 17.3% greater when intercropped with 'Juliet' than when grown in monoculture. Reduction in lesion expansion on 'Brandywine' when intercropped with 'Juliet' suggests an interaction initiating …

Since the advent of nitroxide based spin-probes the electron paramagnetic resonance (EPR) study of spin-labeled macromolecules has been able to provide insight into structural and dynamics properties of DNAs, proteins, and related systems. Spin labels have been extensively used to study the dynamics of oligonucleotides. The heat shock proteins (hsps) are believed to provide protection against a variety of insults such as elevated temperature, heavy metals and chemical agents. The goal of this study was to determine if the immortalized HK-2 human proximal tubule cell line could provide a model system to study the stress response of the proximal tubule cell. Heat stress, elevated temperature at 42.5°C for 1 hr, caused a marked increase only in hsp 70 mRNA and protein, but not that of hsp 27 or hsp 60 mRNA and protein. Two first to second order streams (Kent Run and Munnell Hollow) at Ryerson Station State Park, Greene County, Pennsylvania, were compared as to water parameters (dissolved oxygen, hardness, temperature, pH, and current velocity), species diversity, and abundance of both immature and adult insects. Samples were taken biweekly for one year from May, 1999 through April, 2000 for a total of 26 sampling periods. Data were analyzed using a repeated measures ANOVA. Kent Run had lower dissolved oxygen, higher pH and hardness, lower temperatures, and higher current velocity. A total of 137 taxa were collected at both streams. Of the total …

Vanadate is potent toxic and carcinogenic agent. The mechanisms of its toxic and carcinogenic actions are still under investigation. Cell cycle arrest is an important mechanism in maintaining genomic stability and integrity in response to environmental stress. The present study investigated the possible mechanisms involved in vanadate-induced cell cycle arrest and its signal transduction pathway. Cemented total hip arthroplasty (THA) has enjoyed widespread success since its introduction over thirty years ago. Despite the successes, there are threats to the long term stability of THA, chief among them aseptic loosening of the femoral stem. Femoral component loosening is a significant problem because it prevents the efficient transfer of hip joint forces through the cement to the proximal femur. During 1998--2000, I examined the relationship between the reproductive success of a declining Neotropical migrant songbird, the wood thrush ( Hylocichla mustelina), and surrounding habitat within the highly forested region encompassing the northwestern portion of the Monongahela National Forest, West Virginia, USA. I measured nest fate, food delivery rates, and nest attendance rates of 56 videotaped nests and then related these variables to habitat measured at three concentric scales: nest site (11.3 m radius), territory (100 m), and landscape (1000 m). The study was conducted to investigate and model phosphorus sorption in soils and to study the effectiveness of acid mine drainage (AMD) floc in controlling phosphorus (P) loss to surface runoff from soils. Phosphorus sorption capacity of surface and subsurface horizons of four benchmark soil series - Berks, Huntington, Lindside, and Monongahela - of West Virginia was measured. The soils of these four series were loaded with increasing amounts of P up to 60% of their P retention capacity. As there are over twenty potential PKC phosphorylation sites in AFAP-110 structure, deletional mutagenesis was employed to determine which regions of AFAP-110 are most likely to contain crucial sites for PKC phosphorylation. Our data indicate that functional PKC phosphorylation sites must be localized to the amino acid stretch 266--294 within STK region of AFAP-110. There are four PKC candidate phosphorylation sites in this region; they are being substituted by alanine residues in order to determine their significance in serine/threonine phosphorylation regulation of AFAP-110 function.;Analysis of AFAP-110 amino acid sequence has also revealed that AFAP-110 contains twelve potential 14-3-3 binding sequences. …

Bacterial infections associated with mastitis reduce pregnancies in cattle. Effects on pregnancy and incidence of mastitis in sheep were investigated after immunization with peptidoglycan-polysaccharide (PG-PS) and killed cells from Strep. pyogenes. Rambouillet (n = 100) and mixed breed (n = 18) ewe lambs were immunized (∼d42 and 22 before breeding) with PG-PS (30mugPG/kg/bw) or killed cells or were not immunized (Control, n = 117). IgG antibodies were detected by ELISA. Ewes were bred at synchronized estrus. All immunized and half of non-immunized ewes were challenged with PG-PS (60mugPG/kg/bw) d5 after breeding. Soil series occurring above 1020 m in elevation in the Monongahela National Forest in West Virginia were sampled and analyzed for soil organic and soil microbial biomass carbon as well as various fertility parameters. Additionally, vegetative biomass estimates were made at the sampling sites. The soil series sampled (established and tentative) are frigid, well and moderately-well drained inceptisols and spodosols, covered by northern hardwood or spuce-dominated forests. A longtitudinal study was performed in prepubertal heifer calves to determine when the individual components of the hypothalamic-pituitary-ovarian axis were capable of functioning. In study 1, the hypothalamus and anterior pituitary of prepubertal dairy heifers were challenged with exogenous hormones (estradiol and GnRH, respectively) to determine at what age gonadotropin surges could be induced. Two-, four-, six-, and eight-month-old heifer calves were challenged with GnRH or estradiol. The superior paraolivary nucleus (SPON) is a group of neurons located within the superior olivary complex, a constellation of brainstem nuclei involved in auditory processing. The major inputs to the SPON arise from the contralateral ear and SPON axons target primarily the ipsilateral inferior colliculus. However, little is known regarding the neurochemical phenotypes present in the SPON and how these neurons respond to auditory stimuli. Understanding the neurochemical and physiological properties of the constituent neurons will provide insight into the functional role of the SPON and will contribute to our understanding of the neural circuitry involved in hearing. Although psychologists are beginning to be employed in medical settings, little is known about medical patients' preferences when encountering mental health problems. One population that could be better served by this type of research is diabetes mellitus because of the poor adherence rates and high depression associated with the disease. The current study aimed to identify diabetes patients' preferences for treatments and helpers when encountering emotional difficulties. Data were collected on 80 patients from an Appalachian region of the United States who had previously been diagnosed with either type 1 or type 2 diabetes. The advantages and limitations of current web GIS software for forest fragmentation information and analysis functionality were investigated using Landsat Thematic Mapper data of 1987 to 1999 for a test site in northern West Virginia. ESRI's ArcIMS technology was used to build a Web-based forest fragmentation analysis system to query, represent, and analyze the status of forest fragmentation using landscape metrics. Both ArcIMS HTML and Java fragmentation analysis tools were constructed. The web GIS was evaluated with respect to accessibility, navigation, interactive cartographic functionality, and spatial analysis functionality.
