Hypoxia inducible factor-1 is a heterodimeric basic-loop-helix transcription factor composed of two subunits, HIF-1alpha and HIF-1beta. HIF-1beta is constitutively expressed in cells, while HIF-1alpha is upregulated by hypoxia as well as growth factors and oncogenes. In this study we focus on the regulation of HIF-1alpha expression by phosphatidylinositol-3 kinase (PI3K), and novel functions of HIF-1alpha. Chapter 1 of this dissertation, consists of background and a brief review of the literature regarding PI3K signaling and HIF-1alpha. Chapter 2 focuses on the observation that basal HIF-1alpha expression is upregulated in ovarian cancer and the mechanism by which PI3K regulates this effect. Chapter …

The organization of eukaryotic genomes requires a harmony between efficient compaction and accessibility. This is achieved through the packaging of chromatin and the influence of chromosomal proteins, a topic not well understood. A chromosomal protein that remains a mystery at present is the D1 protein. Identified in 1974 by Alfageme et al., D1 has been highly characterized but its function remains unknown. The goal of this study was to elucidate the function of D1 by overexpression analysis using the GAL4/UAS system in Drosophila melanogaster. Specialized epithelial cells, called hair cells, detect stimuli perceived by the organs of the inner ear. Such stimuli deflect the sensing organelle of the hair cell, the hair bundle. This force is proposed to create tension in the tip link, an extracellular filamentous structure, and open mechanically-gated ion channels. This process, called mechanotransduction, allows ions to enter the hair cell and results in its depolarization. During prolonged stimuli, the sensitivity of the hair cell is maintained by adaptation. Currently, it is perceived that brook trout (Salvelinus fontinalis ) are experiencing reductions in both range and abundance across their east coast distribution. Data identifying key habitat features influencing growth rates of these fish will be valuable to managers conducting remediation efforts as growth at age has important consequences to survival and fecundity. Knowledge of the interaction between proteins is necessary for understanding how the cellular machinery operates. Multiple methods have been developed to study protein-protein interaction, but only two have been able to provide large amount of information for S. cerevisae protein-protein network. Due to the multiple spurious data coming from these methods, there is a need of new complementary methods to improve confidence of the interactions. The enantioselective syntheses of digitoxigen 2,6-dideoxy-beta-L-ribo-hexopyranoside, its analogues and digitoxin monosaccharide analogues have been achieved. Key to this approach is the iterative application of the palladium-catalyzed glycosylation reaction, reductive 1,3-transposition and diastereoselective dihydroxylation. Anticancer tests showed that these analogues have even better anticancer activities.

Fictional depictions of feminine reading and writing practices reveal transformations in expectations of women's roles due to changes in economic, political, social, and familial structures during the eighteenth century. Luce Irigaray's concern with the relationship between cultural influences and individual development offers a new perspective for considering female authors' representations of literary women and femininity. This dissertation utilizes Irigaray's cultural and psychoanalytic critiques of masculine discourse to develop a methodology for analyzing women's reactions, as they are revealed through fictional characters, against prohibitions that limited their engagement in literary activities. Suicide remains stable as the third leading cause of death of adolescents in the United States (CDC, 2004). The rate of adolescent suicide is 10 per 100,000 or approximately one suicide every two hours (AAS, 2006). Adolescent substance abusers are a unique population with a high risk for suicide. The relation between suicide and substance abuse has been examined yet the nature of this relation is unclear (Goldston, 2004). Significant evidence indicates the existence of cross-talk between PKCα, cSrc and PI3-kinase signaling pathways. Activation of each of these kinases results in substantial changes in actin filament concomitant with increased cell motility and invasive potential. The ability of PKCα to induce cSrc activation requires the adaptor protein, AFAP-110. This results in the disruption of the actin cytoskeleton and the formation of motility structures. Activation of PI3-kinase has been shown to direct up-regulation of PKCα activity. However the ability of PKCα to activate PI3-kinase remains controversial. Environmental pollutants and toxicants expose cells to oxidative and electrophilic challenges. Oxidative and xenobiotic stresses caused by environmental toxicants are known to cause human diseases including cancer and diabetes. To protect against the insults, cells have developed detection and response mechanisms. One such sensor-defense cellular mechanism involves the Nrf2 pathway. Decision aids for treatment selection have improved the decision-making process by increasing patient knowledge, decreasing decisional conflict (or uncertainty), increasing strength of preference for an option, and increasing decision satisfaction. Decision aids provide information and assist patients in making informed medical decisions. Guided decision aids help patients weigh pros and cons of options. This study involved the creation and evaluation of a guided decision aid for patients with follicular lymphoma, a disease with multiple complex treatment options. Olfactory systems must detect and discriminate odors while maintaining a degree of perceptual invariance of identity in a highly variable environment. Studies have suggested that olfactory systems of both vertebrates and invertebrates detect odors based on the interaction of specific molecular features of odorant molecules with olfactory receptor neurons. It has also been shown that changing stimulus concentration can elicit substantive changes in spatial representations of odorants. The current study was a chart review that examined 31 female sex offenders (FSO), 31 male sex offenders (MSO), 31 female violent offenders (FO), and 31 male violent offenders (MO) using a 2 (female/male) by 2 (sex/violent offender) design. This is the first study to include three comparison groups when researching female sex offenders. Several variables appeared to relate to gender. Having a co-perpetrator during the crime and being a passive participant in the crime was related to being a female. The critical shortage of clinical laboratory scientists indicates an urgency for planning for the future. A three-round Delphi study explored the future of the clinical laboratory science profession as predicted by a panel of experts. Thirty-two nationally known authorities in the field were invited and 24 agreed to participate as an expert panel. The first round asked the panelists to respond to the statement, “As I consider the next 20 years of clinical laboratory science as a profession, I believe that the following events might occur.†Round One generated more than 200 events which the investigator synthesized into a 147-event …

Previous studies have shown that chronic treatment of broilers with hemin decreases plasma concentrations of uric acid and increases leukocyte oxidative activity (LOA) whereas the inclusion of inosine produces an inverse effect. The objective of these studies was to determine whether or not inosine could serve as a potential therapy for oxidative stress. Oxidative stress was induced in broilers by the administration of hemin. Dietary inclusion of inosine was then begun either before the onset of oxidative stress induced by hemin, or as a post treatment once oxidative stress was established. De novo asymmetric synthesis is a strategy that uses asymmetric catalysis to construct chiral molecules from achiral starting materials. De novo approach to carbohydrate builds saccharides from commercially available achiral starting material which will enable the medicinal chemist to install a much broader range of carbohydrates on natural products and in a SAR-amenable fashion. The palladium-catalyzed glycosylation reaction stereoselectively converts 2-substituted 6-tert-butoxycarboxy-2H-pyran-3(6H)-ones into 2-substituted 6-alkoxy-2H-pyran-3(6H)-ones. By using this reaction, a divergent library of the natural and unnatural 1,4-manno-trisaccharides as well as branched 1,4- and 1,6-linked trisacchardes were synthesized. The 1,4-α-manno-disaccharides were achieved in eight total steps starting from achiral acylfuran. …

A 3-step method for the preparation of 2-methyl-l,3-syn diols was developed and demonstrated on several examples. A previous incarnation of this method was used in the formal synthesis of two polyoxygenated natural products. RK-397 is a 32-membered oxopolyene macrolide antibiotic which also exhibits anti-cancer activity. Virginiamycin M2 (also known as flavofungin I) is a member of the streptogramin antibiotic family which are active against vancomycin-resistant s. aureus. The methodology employed for setting many of the stereocenters in these two products is asymmetric catalysis. This allows for either enantiomer to be created and also is efficient by means of chirality multiplication.

Childhood obesity is an edpidemic in the United States, and is directly associated with risk factors for both cardiovascular disease (CVD) and type 2 diabetes. Endothelial dysfunction (EDF) is thought to be an initiating factor in both these diseases, and recent studies have established the presence of EDF in obese children. An integral approach to estimating stem green and dry weight for yellow-poplar in West Virginia was compared to traditional ratio equation methods. The data were based on stem analysis of 26 trees from the Central Appalachian Broadleaf Forest province in northern West Virginia and 18 trees from the Eastern Broadleaf Forest province in west central West Virginia. The proposed equation generally performed better for the whole tree, as well as for sections within the tree, based on the 9 relative height classes examined. The focus of this project was on the development of a software tool for analysis of crime data at the West Virginia State Police Forensics Laboratory (WVSPFL). The tool enables users to conduct statistical analysis on criminal activity; in addition, data mining methods are implemented to identify possible crime patterns. A Crime database was implemented; it contains information on incidents submitted to WVSPFL over the last fifteen years. The WVSPFL database was integrated with a hypothetical conviction database. Recent evidence supports a role for luteal endothelial cells in mediating PGF2αinduced luteolysis via production of endothelin-1 (ET-1). Three experiments were designed to examine how the endothelin system mediates luteolytic actions of PGF2α. The first objective was to examine in vitro the anti-steroidogenic actions of PGF2α and ET-1 in the presence of a type-A or B receptor antagonist in day-8 luteal minces. Welfare recipients have always been stigmatized. The welfare reform legislation of 1996 reflects widespread attitudes and stereotypes of welfare recipients, which are translated into the structure of state welfare programs and are carried down through the administration to the welfare caseworker—the “face” of welfare for recipients. Literature on welfare stigma has focused on clients, but not their caseworkers. Using qualitative methods, this paper presents a Goffmanian analysis of the effects of welfare stigma on these caseworkers, who not only interact with clients in their jobs everyday, but who also implement a stigmatizing social program. This thesis includes two chapters. The first (Chapter 1) is a literature review focusing on the ecological consequences of introduced fishes on host communities, and specifically on native fishes of similar morphology and phylogenetic origins. The second (Chapter 2) is a manuscript synthesizing the aquaria based study of the interspecific relationships, with respect to habitat utilization, between two syntopically occurring congeneric species pairs in the Greenbrier River. A Selective Catalytic Reduction (SCR) System was designed and built to reduce NOx emissions of the 2003 West Virginia University (WVU) Diesel Hybrid-Electric FutureTruck. FutureTruck was a student competition that challenged students to redesign a new Sport Utility Vehicle (SUV) to improve fuel economy and reduce emissions. A comprehensive literature review discussing all aspects of SCR was completed. The review focuses on harmful effects of NOx emissions, future emission standards, NOx reduction technologies and last presents an in-depth review of SCR. Research has consistently shown a relation between attention and the experience of pain. Pain disrupts and demands attention, interfering with the ability to direct attention to other stimuli in the environment. Yet, various studies of pain management strategies, including studies which have yielded mixed results for the strategy of distraction, often have not specifically examined the relation of pain management, attention, and pain behavior. The iris is a popular biometric that has been demonstrated to have high matching accuracy and permanence under ideal conditions. However, there are several limiting factors that have yet to be comprehensively addressed.
