The last half-century has seen momentous and accelerating changes in humankind's economic activities, political relations, and social and demographic profile. A prominent feature of this change is the increasing scale of human impact on Earth's natural biophysical systems: the climate system, stratospheric ozone, biodiversity, terrestrial and marine food-producing ecosystems, and the great cycles of water, nitrogen, and sulfur (Meyer 1996, Vitousek et al. 1997). These systems sustain the conditions on which life depends, and their weakening may therefore have profound long-term implications for human population health (McMichael 1993, Last 1997).

An epizootic causing mortality among cultured summer flounder Paralichthys dentatus occurred in summer of 1998 at a land-based facility on Narragansett Bay, Rhode Island, USA. The disease, flounder infectious necrotizing enteritis (FINE), was characterized by reddening around the anal area, distended abdomens filled with opaque serosanguineous fluid, enteritis and necrosis of the posterior intestine. In extreme cases of the disease, the posterior intestine was detached from the anus and was observed coming out the vent. The intestine of individuals that recovered from the dsease ended in a blind-sac; the abdomens of these fish were distended, due to food and water …

The Ganges had been the world's 8th largest river in terms of the volume of water discharge of 490 km3/yr to the ocean 4 prior to 1975. Since that time, over a period of two decades, India has reduced the Ganges discharge through Bangladesh to 40% from its original annual average discharge of 1,932+7-223 m3 /s by diverting water for irrigation in her upper states. The , resulting consequences have been disastrous due to the depletion of the surface water resources. One of the devastating consequences has been the generation of extreme climate. Portions of penis and prepuce were collected from 24 bulls with current or recent Tritrichomonas foetus infection. Epididymides were collected from seven of the bulls, and seminal vesicles and prostate were collected from four. Following immunohistochemical staining with two monoclonal antibodies (34.7C4.4 and TF1.15) prepared against T. foetus surface antigens, trichomonads were identified in sections from 15 of the bulls. Organisms were most often located in penile crypts in the midshaft and caudal regions and less often in preputial crypts. Trichomonads were not observed in sections from other genitalia or in subepithelial tissue. T. foetus antigen, however, was present in …

The aim of this project was to investigate the novel use of the atomic force microscope (AFM) as a technique in the elucidation of leukocyte abnormality. To this end, comparative AFM imaging and elasticity studies were performed on normal and myeloid leukaemic leukocytes.

Initially, an evaluation of techniques for the isolation of specific leukocyte (e.g. lymphocytes, granulocytes, etc.) populations from whole blood or leukocyte concentrates, was carried out. It was found that a silanised glass surface failed to select and immobilise a leukocyte population. However, centrifugation of whole blood in an Isopaque-ficoll gradient solution yielded a viable mixed leukocyte population …

Avoparcin is a macrocyclic glycopeptide antibiotic structurally related to vancomycin, teicoplanin, and ristocetin A. When attached to 5-μm spherical silica gel, the avoparcin proved to be an effective chiral stationary phase (CSP) that could be used in the reversed-phase, normal- phase, and polar-organic modes. The avoparcin CSP was complimentary to the other macrocyclic glycopeptide CSPs in that it could resolve some racemates that the others could not, and vice versa. Some important compounds resolved on the avoparcin CSP include verapamil, thyroxine, mephenytoin, and 2- imidazolidone-4-carboxylic acid. The use of this CSP and the optimization of separations on it are discussed. …

Assessing The Accuracy Of A New Diagnostic Test When A Gold Standard Does Not Exist, Todd A. Alonzo, Margaret S. Pepe

Often the accuracy of a new diagnostic test must be assessed when a perfect gold standard does not exist. Use of an imperfect test biases the accuracy estimates of the new test. This paper reviews existing approaches to this problem including discrepant resolution and latent class analysis. Deficiencies with these approaches are identified. A new approach is proposed that combines the results of several imperfect reference tests to define a better reference standard. We call this the composite reference standard (CRS). Immunological Relationship Between The Class I Epitope Of Streptococcal M Protein And Myosin, Anthony Quinn, Kent Ward, Vincent A. Fischetti, Mark E. Hemric, Medeleine W. Cunningham

The class I epitope of streptococcal M protein is an epidemiological marker for acute rheumatic fever (ARF)-associated serotypes of group A streptococci and is recognized by anti-M protein monoclonal antibody (MAb) 10B6. Using MAb 10B6, we determined the relationship between the class I epitope of M protein and the a-helical coiled-coil protein myosin. MAb 10B6 reacted by enzyme-linked immunosorbent assay and Western blotting with human cardiac myosin and rabbit skeletal myosin and its heavy meromyosin (HMM) subfragment. Overlapping synthetic peptides of M5 protein were used to identify the region of M5 protein recognized by MAb 10B6. Two C repeat peptides …

In October 1995, epidemic “hemorrhagic fever,” without jaundice or renal manifestations, was reported in rural Nicaragua following heavy flooding; 2259 residents were evaluated for nonmalarial febrile illnesses (cumulative incidence, 6.1%) and 15 (0.7%) died with pulmonary hemorrhage. A case-control study found that case-patients were more likely than controls to have ever walked in creeks (matched odds ratio [MOR], 15.0; 95% confidence interval [CI], 1.7–132.3), have household rodents (MOR, 10.4; 95% CI, 1.1–97.1), or own dogs with titers ≥ 400 to Leptospira species (MOR, 23.4; 95% CI, 3.6–`). Sites on antibodies having affinity for photoaffinity compounds, in particular purine or azidopurine containing compounds are taught. These sites provide for the site-specific attachment of nucleotide photoaffinity compounds to antibodies, e.g., ATP- or GTP-analog photoaffinity compounds by photochemical means. These nucleotide photoaffinity compounds may additionally be attached to molecules having a desired therapeutic or diagnostic activity, and the resultant conjugates used as diagnostics or therapeutics.

This software agent will be integrated into wearable technology which is embedded in the protective clothing worn by the humans. The Canadian Cooperative Wildlife Health Centre (CCWHC) was established in 1992 as an organization among Canada's 4 veterinary colleges, with a mandate to apply veterinary medicine to wildlife management and conservation in Canada. A major function of the CCWHC is nation-wide surveillance of wild animal diseases. Disease surveillance is conceived as consisting of 4 different activities: detection, diagnosis, information management, and use of information. Circular covariance is important in modelling phenomena in epidemiological, communications and numerous physical contexts. We introduce and develop a variety of methods which make it a more versatile tool. First, we present two classes of estimators for use in the presence of missing observations. Using simulations, we show that the mean squared errors of the estimators of one of these classes are smaller than those of the Maximum Likelihood (ML) estimators under certain conditions. Next, we propose and discuss a parsimonious, autoregressive type of circular covariance structure which involves only two parameters. We specify ML and other types of estimators …

Pursuit Of A Chiral Amino Aldehyde Intermediate In The Synthesis Of (+)-Obafluorin, A B-Lactone Antibiotic, Jim Cwik '96

This research is focused on synthesizing a chiral amino aldehyde from L-Serine. The proposed synthesis of the amino aldehyde would yield a stereochemically pure product working through an L-Serine beta-lactone intermediate. The amino aldehyde is a proposed intermediate in the synthesis of (+)-obafluorin, a beta-lactone antibiotic of interest. The proposed synthesis might provide a simpler, more versatile way to synthesize obafluorin.
