Turkish Journal of Medical Sciences

Under conditions that may risk the success of flap surgery, free skin grafting may be an option for the reconstruction of the irradiated area. Clinical observations reveal that skin grafts applied to irradiated area undergo ulceration, resulting in chronic open wounds. The role of hyperbaric oxygen therapy (HBO) in problem wounds is to increase the tissue oxygen tension to optimize fibroblast proliferation and collagen synthesis, to enhance the oxidative killing capacity of the white blood cells and to stimulate angiogenesis. HBO has also been shown to stimulate angiogenesis in radiation-damaged tissue. In view of this data, an animal model is …

Effect Of Vitamin E On Kidney Preservation Using Isolated Perfused Dog Kidney, Adnan Şahi̇n, Cem Algin, Çağatay Sezgi̇n, Enver İhti̇yar

Turkish Journal of Medical Sciences

Unsuccessful cadaveric kidney transplantation still remains as an important problem in organ transplantation. Cold storage solutions are not very efficient for kidney preservation. Vitamin E is an important dietary antioxidant, which may play a vital role in preventing free radical perfusion injury. We investigated the effect of vitamin E on kidney preservation with Euro-Collins (EC) solution using the isolated perfused dog kidney model. Recipients were randomly divided into 3 groups: group 1 (n = 6), immediately reperfused with EC solution, given laboratory pelleted diet and drinking water for 4 weeks; and group 2 (n = 6), flushed, and stored with …

Soft Tissue Profile Changes In Anatolian Turkish Girls And Boys Following Orthodontic Treatment With And Without Extractions, Serpi̇l Hazar, Sercan Akyalçin, Hayal Boyacioğlu

Turkish Journal of Medical Sciences

The choice of extraction is critical in treatment planning, and its impact on the final facial appearance can be quite dramatic. Evaluation of facial profile esthetics both before and after treatment should be made cautiously. An orthodontist should be conscious of changes brought about by treatment and by late adolescent growth as well as differences in the dentofacial structures of various ethnic groups. The purpose of this cephalometric investigation was to determine the changes in the soft tissue profile of Turkish children treated with and without extractions. An assessment of pre- and post-treatment faces, chosen from our clinical records by …

Comparative Cytotoxicity Of Fludarabine In Human Peripheral Blood Mononuclear Cells And In The Promyelocytic Leukemia Cell Line (Hl60), A. Lale Doğan

Turkish Journal of Medical Sciences

Fludarabine is an inhibitor of DNA/RNA synthesis with antitumor activity on lymphoproliferative malignancies. We investigated the dose-dependent cytotoxic effects of the drug on human peripheral blood mononuclear cells, the HL60 cell line and the differentiated HL60 cells. Dimethylformamide was used for the neutrophilic differentiation of HL60 cells and the MTT assay was used to estimate drug cytotoxicity. Undifferentiated tumor cells had higher proliferation rates compared to the differentiated subline. There was 57% cell death in HL60 cells at a dose of 5 µg/ml of the drug. On the other hand, in differentiated HL60 cells and nonmalignant cells, cell death did …

Turkish Journal of Medical Sciences

Genetic biomonitoring of human populations exposed to potential mutagens/carcinogens can be performed using different genetic markers. Sister chromatid exchange (SCE) is one of the most extensively used markers of the early biological effects of DNA damaging agents. In order to assess the genotoxicity associated with exposure to cotton dust, we determined SCE frequency in peripheral blood lymphocytes cultured from 20 cotton gin workers and 20 controls. Student’s-t test indicated an increased frequency of SCE in lymphocytes of the workers (14.66 ± 4.18) compared to the controls (10.40 ± 1.37) (P < 0.0001). The Mann-Whitney U test indicated that the duration of exposure to cotton dust (10 years) and the work area were not related to the frequency of SCE (P > 0.05). The textile industry is rapidly expanding in Turkey, and …

Genotoxic Evaluation Of The Antibacterial Drug, Ciprofloxacin, In Cultured Lymphocytes Of Patients With Urinary Tract Infection, Mevli̇t İkbal, Hasan Doğan, Hati̇ce Odabaş, İbrahi̇m Pi̇ri̇m

Turkish Journal of Medical Sciences

Ciprofloxacin is a quinolone carboxylic acid derivative and is commonly used in medicine. The genotoxicity of ciprofloxacin was evaluated in cultured human peripheral blood lymphocytes in patients with urinary tract infection. Sister chromatid exchange (SCE), mitotic index (MI) and replicative index (RI) were measured before and after ciprofloxacin therapy. Our results showed that SCE frequency significantly increased after ciprofloxacin therapy (P < 0.001), but MI and RI decreased (P < 0.001). The results of this study suggest that clinical therapy using ciprofloxacin may have a moderate genotoxic potential.
