Evidence That Clusterin Has Discrete Chaperone And Ligand Binding Sites, Johnathon Lakins, Stephen Poon, Simon Easterbrook-Smith, John Carver, Martin Tenniswood, Mark Wilson

Mark R Wilson

Clusterin is the first identified extracellular mammalian chaperone and binds to a wide variety of partly unfolded, stressed proteins. Clusterin also binds to many different unstressed ligands including the cell surface receptor low density lipoprotein receptor-related protein-2 (LRP-2). It is unknown whether clusterin binds to all of these many ligands via one or more binding sites. Furthermore, the region(s) of clusterin involved in these many binding interactions remain to be identified. As part of an investigation of these issues, we expressed recombinant human clusterin in the yeast Pichia pastoris. The resultant protein had variable proteolytic truncations of the C-terminal region …

Cooper, Michael T. Ferdig, Xin-Zhuan Su, Lyann M. B. Ursos, Jiabing Mu, Takashi Nomura, David A. Fidock, Paul D. Roepe, Thomas E. Wellems

Roland A. Cooper

Chloroquine resistance (CQR) in Plasmodium falciparum is associated with multiple mutations in the digestive vacuole membrane protein PfCRT. The chloroquine-sensitive (CQS) 106/1 line of P. falciparum has six of seven PfCRT mutations consistently found in CQR parasites from Asia and Africa. The missing mutation at position 76 (K76T in reported population surveys) may therefore be critical to CQR. To test this hypothesis, we exposed 106/1 populations (10(9)-10(10) parasites) to a chloroquine (CQ) concentration lethal to CQS parasites. In multiple independent experiments, surviving CQR parasites were detected in the cultures after 28 to 42 days. These parasites showed novel K76N or …

Mutations in SCN1A, the gene encoding the brain voltage-gated sodium channel alpha1 subunit (NaV1.1), are associated with at least two forms of epilepsy, generalized epilepsy with febrile seizures plus (GEFS+) and severe myoclonic epilepsy of infancy (SMEI). We examined the functional properties of four GEFS+ alleles and one SMEI allele using whole-cell patch-clamp analysis of heterologously expressed recombinant human SCN1A. One previously reported GEFS+ mutation (I1656M) and an additional novel allele (R1657C), both affecting residues in a voltage-sensing S4 segment, exhibited a similar depolarizing shift in the voltage dependence of activation. Additionally, R1657C showed a 50% reduction in current density …

Fish Production Of A Temperate Artificial Reef Based On The Density Of Embiotocids (Teleostei: Perciformes), Daniel J. Pondella Ii, John S. Stephens Jr., Matthew T. Craig

Daniel Pondella

The abundance of two embiotocids (Embiotoca jacksoni and Rhacochilus vacca) was surveyed continually from 1974 to 1998 on the breakwaters comprising King Harbor in Redondo Beach, California, and a nearby natural rocky-reef as a reference. Embiotocids provide an interesting model for reef-fish production because they are viviparous and their entire ontogeny can be observed on the reefs. We deduce that the two reef populations of both species were stable at their carrying capacity. This carrying capacity was greater in King Harbor throughout the study. We also report a linear decline in survival of juveniles of both species over the 25-year …

New Species Of Tricolor Leptotyphlops (Reptilia: Squamata: Leptotyphlopidae) From Central Peru, Edgar Lehr, Van Wallach, Gunther Kohler, Cesar Aguilar

Edgar Lehr

A new species of slender blind snake, Leptotyphlops alfredschmidti, is described from two localities (2940 and 3090 m) from the western Andean slopes of central Pern (Departamento de Ancash, Provincia de Huarmey). Leptotyphlops alfredschmidti is referred to the Leptotyphlops tesselatus group. The new species shares with Leptotyphlops teaguei and Leptotyphlops tricolor a tricolor body pattern of red, black, and yellowish longitudinal stripes but differs from all members of the group in several features of coloration pattern and pholidosis.

Factors Related To Success And Satisfaction In Online Learning, Patricia A. Beffa-Negrini, Brian Miller, Dr. Nancy L. Cohen

We investigated factors that may relate to positive outcomes in a web-based introductory nutrition course: age, gender, prior nutrition knowledge, nutrition attitude, attitude toward technology, satisfaction with the instructor, and satisfaction with peer interaction. Fifty-four students completed pre-post surveys of knowledge, altitudes, and course satisfaction. When adjusted for multiple variables, satisfaction with the instructor and prior nutrition knowledge related to achievement in nutrition knowledge from pre- to posttest. Age, satisfaction with the instructor, and prior nutrition knowledge predicted course grade. Satisfaction with the instructor and satisfaction with peer interaction related to self-reported learning. Satisfaction with peer interaction, satisfaction with the …

A Long Way From Home: Transatlantic Sea Star Migration, William Jaeckle, Paul Kehle Dec 2001

A Long Way From Home: Transatlantic Sea Star Migration, William Jaeckle, Paul Kehle

William Jaeckle

For a marine biologist strolling along the eastern coast of South America, finding some sea stars (popularly called starfish, and technically members of the class Asteroidea of the phylum Echinodermata) is not all that unusual. However, when closer inspection reveals the sea stars to be ones that are also found on the shores of the western coast of Africa, the marine biologist now faces an interesting question: How did these sea stars come to be so far from home? Larval Productivity Of A Mature Artificial Reef: The Ichthyoplankton Of King Harbor, California, 1974–1997, John Stephens Jr, Daniel Pondella Ii Dec 2001

Larval Productivity Of A Mature Artificial Reef: The Ichthyoplankton Of King Harbor, California, 1974–1997, John Stephens Jr, Daniel Pondella Ii

Daniel Pondella

Do artificial reefs serve as productive marine fish habitats (sources) or do fish assemblages of such reefs contribute little to the gene pool of succeeding generations (sinks)? Using data from a 24-year study of the breakwater at King Harbor (California, USA), annual densities of reef fish larvae were compared with densities observed elsewhere in the Southern California Bight. Larval production at King Harbor has decreased over the study period, as has the size of the fish assemblage. Both declines may be related to recorded increases in water temperature. Larval densities from similar water depths (0–15 m) throughout the bight vary …
