The Allegheny woodrat (Neotoma magister) is found throughout much of the central and southern Appalachians and adjacent portions of the Interior Highlands. Allegheny woodrats have declined in the northern portions of their range and are state-listed as threatened, endangered or sensitive species of concern in every state where they occur. Until recently, biologists have had to rely on biological data collected from the closely related eastern woodrat (N. floridana) because of limited research on the Allegheny woodrat. We have been studying the ecology and natural history of woodrats in Virginia and West Virginia since 1990. On …

In vivo electroporation is an efficient means of increasing plasmid DNA delivery to normal tissues, such as skin and muscle, as well as directly to tumors. In the experiments described here, plasmid DNA was delivered by in vivo electroporation to B16 mouse melanomas using two very different pulsing protocols. Reporter expression increased 21- or 42-fold, respectively with electroporation over injection alone. The growth of experimental melanomas with an approximate diameter of 4 mm on the day of treatment was monitored after electroporation delivery of reporter plasmid DNA. Electric field enhanced molecular delivery for cancer research and treatment is a new technology that has demonstrated its effectiveness in clinical trials using bleomycin or cisplatin (Heller, R., Gilbert, R., Jaroszeski, M. J. Clinical applications of electrochemotherapy, Advanced Drug Delivery Reviews 35,119-129, 1999), as chemotherapeutic agents. The technology is being investigated in research applications for applicability as a method to enhance gene expression in a target tumor. Antibiotic Resistance Patterns (ARPs) of Enterococcus spp. were used as a phenotypic fingerprint to compare and categorize unknown-source isolates in an impaired segment of the Roanoke River, Roanoke County, Virginia. Antibiotic resistance analysis (ARA) of enterococci has been effectively used to differentiate among sources of fecal contamination in many geographic regions in the United States. Enterococcus spp. were used as a fecal indicator in a library consisting of 1,562 known-source isolates. Two-way analysis indicated that approximately 95% of the unknown-source isolates collected were of animal origin. A 3-way analysis indicated that 61% of the unknowns were of livestock origin while …

I examined entrance patterns of Atlantic croaker, Micropogonias undulatus by comparing the abundance, length, age and growth rates, of larvae as they entered Oregon and Ocracoke Inlets, NC. These biological factors were then compared against environmental factors to assess the influence the physical environment has on recruitment variability. Entrance abundance into Oregon Inlet was an order of magnitude higher than abundance into Ocracoke Inlet. However, the difference was influenced by three peak events in Oregon Inlet and non-parametric tests found no difference in median abundance between the two inlets. Patterns of larval amphibian distribution and abundance have historically been ascribed to direct predation. However, adult oviposition site selection is an alternative mechanism that can produce identical patterns and significantly influence community assembly. Although such behavioral habitat selection has been documented in several amphibian species whose larvae lack particular anti-predator defenses, the sensitivity of adult amphibians in detecting predator cues and avoiding these habitats has not yet been determined. If larval survivorship is greatly reduced in sites containing predatory fish, selection to detect and avoid habitats with low fish densities should be strong. Elevated blood pressure is associated with varying degrees of arterial remodeling. The mechanisms by which extracellular mechanical stress is converted into intracellular alterations in signal transduction and gene expression have yet to be fully elucidated. Our goal was to investigate the early events in the vascular smooth muscle response to acute hypertension and to identify mediators involved in long-term hypertensive remodeling.

Minirhizotron images are assigned color codes to approximate the age of individual roots in situ. Triphenyl-tetrazolium chloride (TTC) has been used to measure the vitality of plant tissue by detecting dehydrogenase activity. TTC was used to assess the vitality of root tissue by color in a scrub oak ecosystem at Kennedy Space Center in central Florida. Roots were excavated, sorted into four color classes and stained with TTC. The product of the reduction of TTC by dehydrogenase enzyme, formazan, was extracted and its absorbance recorded using a spectrophotometer. Interferon α may be used as a single agent therapy for metastatic malignant melanoma or as an adjuvant to chemotherapy. Delivery of interferon α by gene therapy offers an alternative to recombinant protein therapy. Electrically mediated delivery enhances plasmid expression in a number of tissues, for instance skin, liver, muscle and tumors including melanomas. Here we compare the effect of delivery of a plasmid encoding mouse interferon α on growth of visible B16 mouse melanomas following electrically mediated delivery to muscle or directly to the tumor. Ovaries of black drum, Pogonias cromis, collected from the Chesapeake Bay region in 1992, were used to describe reproductive strategy and fecundity. Histological examination showed that black drum spawn in the Chesapeake Bay region from April through early June. Distributions of oocyte diameter showed distinct oocyte-developmental groups indicating that Chesapeake Bay black drum are group-synchronous batch spawners. Female black drum are extremely fecund ranging from 414,000 to 3,736,000 hydrated oocytes (mean = 1,389,000) per batch with a spawning periodicity of 3.8 days. Populations of female grass shrimps (Palaemonetes pugio and P. vulgaris) were sampled from five coastal embayments in Delaware, Maryland, and Virginia (Delmarva) and compared with respect to reproductive and life history attributes. We observed interspecific differences in timing of reproduction, carapace length, ratio of carapace length to total body length, body weight, clutch weight, clutch size, and egg volume. Onset of reproduction in P. vulgaris lagged behind P. pugio. Although there was no difference in the relationship between clutch size and carapace length for the two species, carapace length/total body length in P. pugio was greater than …

Autotrophic picoplankton and phytoplankton composite samples were collected from the surface, pycnocline, and bottom water layers at three stations in the lower Chesapeake Bay between June and September 1993. Using light microscopy a total of 170 phytoplankton taxa were identified. Utilizing epifluorescent microscopy, the picoplankton taxa were distinguished by size, representing species with cells less than 2 microns. Diatoms were the dominant taxa during this period, with lesser representation by the other phytoplankton components. Within the picoplankton category, cyanobacteria were dominant. This study investigated the physical factors influencing the vertical distribution of zooplankton at the mouth of the Chesapeake Bay and attempted to derive biological data from acoustic Doppler current profiler (ADCP) backscatter. A single site was occupied in the North Channel at the mouth of Chesapeake Bay (37° 04' N, 75° 58' W) from 17.0–19.7 November 1999. Temperature, salinity, fluorescence, light transmission, current velocity and backscatter were measured throughout the sampling period. Plankton samples were collected every hour from 3.5, 7.5, and 11.5 m.
We found little association between ADCP backscatter and plankton concentrations, but stronger correlations between backscatter and …

Active anting behavior occurs when songbirds apply ants to their feathers. During anting displays, a bird draws its wings forward in front of the head and swings its tail around to the far right or left. Both wing and tail feathers are then rubbed with ants held in the bird's beak. The potential associations among ants, feather ectoparasites and birds make anting behavior an important community-level interaction. Although this behavior is cosmopolitan in distribution and occurs in a wide variety of passerine birds, it remains poorly understood. Benzo[α]pyrene is a ubiquitous pollutant produced from the incomplete combustion of organic material such as fossil fuels. It is found in the workplace, urban air, drinking water, and the food supply. Recently, it has been proposed that benzo[α]pyrene may be the causative agent in the formation of lung adenocarcinomas among some Taiwanese women exposed to cooking oil fumes without adequate ventilation. In this study, calf thymus DNA was modified in vitro with benzo[α]pyrene-diol epoxide (BPDE) to a level consistent with that found in biological samples. Nuclear factor-kappa B (NF-kB) is an important transcription factor that participates in the genetic regulation of inflammation in many tissues. Cocaine and heroin are drugs of abuse associated with renal and cardiac pathology. The purpose of this study was to test the hypothesis that exposure to cocaine or morphine (a metabolite of heroin) would activate NF-kB in renal epithelial cells (COS-7) and cardiac myocytes (H9c2) grown in culture. COS-7 and H9c2 cells were co-transfected with an experimental reporter specific for NF-kB activation and a control reporter with constitutive activity. A colonial cyanoprokaryote, Aphanocapsa holsatica and autotrophic picoplankton abundance were monitored weekly over a two year period in Lake Joyce, Virginia. Significant differences were observed in both the cyanoprokaryote and picoplankton abundance over the study period and an inverse relationship was observed between these two plankton groups. Disaggregation of colonies was shown to contribute to picoplankton populations where water temperature and precipitation input apparently trigger colony dispersion. This relationship is suggested to occur in other aquatic habitats. Results of this work and its implications for ecosystem dynamics are discussed.

Fluorescence in situ hybridization (FISH) permits the detection of unique DNA sequences of target DNA within single cells. FISH allows the identification of DNA sequences within individual cells and readily identifies genetic changes within a mixed population of cells. However, problems arise in maintaining nuclear morphology, particularly following harsh denaturation procedures where cells are heated to 80°C, causing the nuclear boundary to become blurred or the cell to completely burst. In an attempt to preserve cellular morphology and high hybridization efficiency, particularly in fresh tissue samples, we evaluated a FISH protocol using alkaline denaturation in place of heat.

This study examined the ability of ticks to maintain multiple species of spotted fever group rickettsiae via transovarial transmission. Using a capillary feeding method, previously established Rickettsia montana- and Rickettsia rhipicephali-infected cohorts of Dermacentor variabilis (Say) were exposed to R. rhipicephali and R. montana, respectively, in two reciprocal challenge experiments. Eggs collected from individual females, for two successive generations, of each cohort were assessed for rickettsial infection by polymerase chain reaction for each challenge experiment. We evaluated an artificial capillary feeding method to infect nymphal Ixodes scapularis (Say) ticks with Borrelia burgdorferi, the causative agent of Lyme disease. Thirty to 70% of the nymphs were infected after feeding for 2.5 h from glass capillary tubes filled with a solution of spirochetes. Capillary infection was stable and persisted in the nymphs for at least 10 d after feeding. Capillary feeding also maintained natural vector competence patterns because I. scapularis ticks acquired infection unlike Dermacentor variablis (Say), which did not become infected. Leukemia inhibitory factor (LIF) enhances in vitro murine preimplantation development in a time- and dose-dependent fashion. Knockout experiments have demonstrated that endometrial LIF is essential for in vivo murine implantation. We assessed the impact of LIF and an anti-LIF polyclonal antibody (pab) on in vivo development and developed a novel and successful nonsurgical method of embryo transfer for this species, a transcervical blastocyst transfer technique. Trichodesmium spp. fix atmospheric N2 and so an element other than N limits production by these species in the oligotrophic ocean. Because dissolved inorganic phosphorus (DIP) is in short supply in many marine systems, it has been hypothesized that P is a growth-limiting nutrient for these species in nature. However, Trichodesmium is capable of hydrolyzing dissolved organic P (DOP) compounds and the inorganic products from hydrolysis may provide an additional source of P for growth. In addition to a soluble response, many invertebrates control bacterial infections by means of phagocytosis or melanotic encapsulation. In some insects, Escherichia coli growth is reported to be inhibited by aggregation/encapsulation. Soluble and phagocytic responses to bacterial challenge have been reported in ticks, but evidence of an aggregation/encapsulation response was reported only for inanimate (araldite) implants. Ticks were challenged by direct inoculation of bacteria into the hemocoel cavity. By plating, no viable E. coli were detected 6 h postinoculation. Six species of phytoplankton, representing 6 major phylogenetic groups (2 oceanic species: a cyanobacteria, Synechococcus sp., and a coccolithophorid, Emiliania huxleyi; and 4 coastal species: a prasinophyte, Tetraselmis sp., the green algae Dunaliella tertiolecta, the diatom Skeletonema costatum and a dinoflagellate Amphidinium carterae) were tested for their ability to reduce iodate to iodide in batch cultures. They all did so to varying degrees. Thus, the reduction of iodate to iodide by phytoplankton may be a general phenomenon in the marine environment. We used the otoliths from a sample of Dissostichus eleginoides pre-recruits, whose length density was distinctly polymodal, to see whether ages estimated by reading otoliths were congruent with the length modes observed. Length densities by age were compared graphically with the overall length density observed. Additionally, ages were predicted for each fish based on length, and compared with ages estimated from reading otoliths in a goodness-of-fit test. The majority of the otoliths (83.6%) were estimated to be from fish 1+ or 2+ years old, with mean total lengths of 32.5 cm and 41.3 cm respectively. No difference was found between …

Interleukin-12 (IL-12) has been used in numerous immunotherapy protocols against melanoma. However, delivery of IL-12 in the form of recombinant protein can result in severe toxicity, and gene therapy has had limited success against B16.F10 murine melanoma. The purpose of this study was to examine the effectiveness of in vivo electroporation for the delivery of plasmid DNA encoding IL-12 as an antitumor agent against B16.F10 melanoma. We treated mice bearing established B16.F10 melanoma tumors with intratumoral (i.t.) or intramuscular (i.m.) injections of a plasmid encoding IL-12, followed by in vivo electroporation. A self-consistent model analysis of electroporation in biological cells has been carried out based on an improved energy model. The simple energy model used in the literature is somewhat incorrect and unphysical for a variety of reasons. Our model for the pore formation energy E(r) includes a dependence on pore population and density. It also allows for variable surface tension, incorporates the effects of finite conductivity on the electrostatic correction term, and is dynamic in nature. An improved electroporation model is used to address membrane irreversibility under ultrashort electric pulse conditions. It is shown that membranes can survive a strong electric pulse and recover provided the pore distribution has a relatively large spread. If, however, the population consists predominantly of larger radii pores, then irreversibility can result. Physically, such a distribution could arise if pores at adjacent sites coalesce. The requirement of close proximity among the pore sites is more easily satisfied in smaller organelles than in outer cell membranes.
