The plainfin midshipman fish, Porichthys notatus, provides an excellent opportunity for delimiting the influence of neurochemical content on vertebrate vocal behavior, in part because the production and recognition of social-acoustic signals is vital to their reproductive behavior. There are two distinct reproductive male morphs that follow divergent developmental trajectories with corresponding alternative reproductive tactics: type I males are the territorial/nesting morph that vocally court females during the summer breeding season while type II males do not court females, but instead sneak spawn in competition with type I males. The interaction of two RNA molecules involves a complex interplay between folding and binding that warranted recent developments in RNA-RNA interaction algorithms. However, biological mechanisms in which more than two RNAs take part in an interaction also exist.

A typical algorithmic approach to such problems is to find the minimum energy structure. Often the computationally optimal solution does not represent the biologically correct structure of the interaction. Trypanosome Lytic Factor (TLF) is an innate immunity complex that was originally discovered to protect against African Trypanosomes. The major components of TLF are Apolipoprotein A1 (APOA1), Apolipoprotein L1 (APOL1) and HPR (Haptoglobin Related Protein), where APOL1 is necessary and sufficient for trypanolysis. Recently we have shown that TLF ameliorates infections by cutaneous Leishmania species. Here we investigated the effect of different primate and human TLF against different Leishmania sp. Central nervous system injuries usually produce motor impairments that are exacerbated by pathologically altered muscle tone. Abnormal muscle tone interferes with voluntary movement and is associated with loss of dexterity. Prior work in our laboratory demonstrated that 30-second trans-spinal direct current (DC) stimulation can temporarily modify muscle tone in anesthetized spastic mice after spinal cord injury (SCI). These experiments described DC-induced muscle tone responses to be polarity-dependent. That is, anodal stimulation (current passed from the lumbar spine to sciatic nerve) decreased muscle tone, and cathodal stimulation (current passed from the sciatic nerve to the lumbar spine) increased it. Previous studies have shown that haplorhine diet is affected by phylogeny; however, until now studies in Malagasy strepsirrhines were lacking. The evolution of differences in Malagasy primates’ diets appears to differ from the pattern shown in haplorhines. My results indicate that niche differentiation may be a stronger predictor of diet.

Ecological niche modeling investigates how climatic variables have influenced taxonomic diversity in Papio. Models performed well suggesting climatic variables influence the distribution of baboon species. Niche overlap among all possible pairs of taxa determined that species exhibited significantly different niches. The results of these models support a parapatric speciation scenario.

DNA testing on touched objects is a valuable tool in forensic investigations, but DNA is usually present in low amounts, causing poor STR typing results. For touch DNA evidence, there is a clear need for additional individualization, especially for highly probative samples. This could be achieved by testing genetically variable proteins. Introduced species provide rare opportunities to test evolutionary hypotheses in situ by creating so-called natural experiments. Natural experiments are situations in nature that resemble laboratory studies by allowing for comparisons of a “control” group (i.e., a species’ native range) with “experimental” groups (i.e., a species’ introduced range). Conservation biology is a scientific discipline that draws on methods from diverse fields to address specific conservation concerns and inform conservation actions. This field is overwhelmingly focused on charismatic animals and vascular plants, often ignoring other diverse and ecologically important groups. Estrogen receptor positive (ER+) breast cancers often have MDM2 overexpression indicating a critical role for MDM2 in breast cancer tumorigenesis. The cancer genome atlas (TCGA) found that increased MDM2 expression is one of the four pathways that correlate with all breast cancer subtypes. MDM2 is mainly known as the negative regulator of wild type p53. However, aggressive breast cancers often have MDM2 overexpression and mutant p53 (mtp53). The series of investigations described in this thesis were designed to assess the effect of diet on mortality through decreasing temperature in two genetic varieties of M. mercenaria important to the natural environment and the aquaculture industry: “wild” from natural stocks and “notata” selected for fast growth. Data from these novel investigations demonstrate a temperature specific sensitivity to each dietary component designating 20:5n-3 as homeoviscously favorable in the upper thermal range above 12°C, and 22:6n-3 as favorable in the lower. An original method to extract and detect barbiturates, specifically secobarbital, in hair by enzyme-linked immunosorbent assay (ELISA), has been developed. Drug-free hair specimens were fortified with an exaggerated amount of secobarbital to produce a homogeneous, true positive sample to be utilized to test the drug extraction efficiency from hair. Drug extraction using hair extraction buffer (HEB) from Immunalysis was compared to a base digestion using 1M NaOH. After LCMS/MS analysis, HEB extraction was deemed a successful method for extracting barbiturates from hair. Using an undergraduate sample, the present study examined how psychopathic traits (assessed by the Psychopathic Personality Inventory-Revised) and empathic concern (assessed by the Interpersonal Reactivity Index) affect emotional-information processing using event-related potentials (ERPs) and a sentential priming paradigm. Participants had their electroencephalogram (EEG) recorded while they silently read sentences with three types of endings: congruent, incongruent, and emotionally negative. We hypothesized that participants with high levels of psychopathic traits and participants with low levels of empathic concern would find the emotionally negative sentence endings less unexpected and disturbing (yielding a smaller N400), compared to participants with low levels of psychopathic …

Wastewater-based epidemiology is an innovative approach that uses the analysis of human excretion products in wastewater to obtain information about exposure to drugs in defined population groups. We developed and validated an analytical method for the detection and quantification of opioids (morphine, oxycodone, hydrocodone, oxymorphone and hydromorphone), and cannabinoids (9-tetrahydrocannabinol, 11-nor 9-carboxy- tetrahydrocannabinol (THCCOOH) and THCCOOH-glucuronide) in raw influent wastewater samples by ultra-high performance liquid chromatography-tandem mass spectrometry. Method validation included linearity (5–1 000 ng/L for opioids, 10–1 000 ng/L for cannabinoids), imprecision (<21.2%), accuracy (83%–131%), matrix effect (from –35.1% to –14.7%) and extraction efficiency (25%–84%), limit of detection (1–5 ng/L) and quantification (5–10 ng/L) and auto-sampler stability (no loss detected). River, sewage overflow and wastewater samples were collected in triplicate from different locations in New York City and stored at -20 C until analysis. River water samples were negative for all the compounds. Water from sewage overflow location tested positive for morphine (10.7 ng/L), oxycodone (4.2–23.5 ng/L), oxymorphone (4.8 ng/L) and hydromorphone (4.2 ng/L). Wastewater samples tested positive for morphine (133.0– 258.3 ng/L), oxycodone (31.1– 63.6 ng/L), oxymorphone (16.0–56.8 ng/L), hydromorphone (6.8–18.0 ng/L), hydrocodone (4.0– 12.8 ng/L) and THCCOOH (168.2– 772.0 ng/L). This method is sensitive and specific for opioids and marijuana determination in wastewater samples.

Packs of African wild dogs, Lycaon pictus, were analyzed for coloration patterns and social aggregation tendencies. Mapped locations determined if the coat patterns followed a geographic distribution that corresponded to Southern or Eastern phenotypic forms. Social aggregation tendencies were observed to determine grouping behavior presumably related to individuals’ roles.

Animal personality has a significant impact on an animal’s physiology and welfare. Eight captive red pandas were observed to determine if personality can be assessed in this species using an ethological approach. Analysis revealed two behavioral dimensions: 1) Active/Exploratory and 2) Maintenance. Age differences were found in the Active/Exploratory dimension.

Ruffed lemur (Varecia spp.) color vision research was conducted using a multidisciplinary approach: psychophysics, genetic analysis, technology, and animal training. The behavioral manifestation of Varecia spp. trichromacy was shown using a touchscreen apparatus (SMARTA). Trichromats performed better than dichromats when discriminating red from green (G2 = 78.10, p < 0.001).

Purpose: FHFs are cytosolic proteins that bind to voltage-gated sodium channels (NaVs) and modulate their functions to control membrane excitability. FHFs raise the voltage dependence of NaV fast inactivation to promote excitability, while A isoforms FHF (A-FHFs) also capture open sodium channels into a long-term inactivated (LTI) state to limit excitability. My research has addressed how FHFs balance membrane excitability as it relates to normal and pathological brain functions.
