In the past decade, layer-by-layer (LbL) nanoassembly has been used as a tool for immobilization and surface modification of materials with applications in biology and physical sciences. Often, in such applications, LbL assembly is integrated with various techniques to form functional surface coatings and immobilized matrices. In this work, integration of LbL with microfabrication and microfluidics, and tissue engineering are explored. In an effort to integrate microfabrication with LbL nanoassembly, microchannels were fabricated using soft-lithography and the surface of these channels was used for the immobilization of materials using LbL and laminar flow patterning. In this study, we applied the traditional Electrostatic layer-by-layer (ELBL) assembly procedure to fabricate nanothin films over flat surfaces, and modify particle surfaces to influence the drug particle size, and drug release. The ELBL assembly has previously been applied to fabricate multilayer nano-scale thin films, but its ability to instantaneously influencing particle size is unique. Other unique observations such as influence on drug release as a result of polymer complexation, and thermal changes occurring during layer fabrication are recorded.

In this dissertation, the modeling, design, and function of fluorescent spherical enzymatic microsensors for minimally-invasive diabetic monitoring are described. The devices reported herein are novel and their experimental construction and theoretical analysis have not been previously reported, thus laying the foundation for an intensive set of studies. These sensors are based on the encapsulation of an enzymatic fluorescent assay for glucose within hydrogel alginate microspheres with diameters on the order of tens of microns, which are of the appropriate size for intradermal implantation. This dissertation addresses the research for the development of spinal cord-computer interface (SCCI). The main objective of SCCI is to generate voluntary motor control signals for individuals with spinal cord injury (SCI).

In the neuroscience aspect, organization of the fibers in the descending tracts of the dorsolateral funiculus of the cervical spinal cord was investigated in cats. The spinal cord was penetrated with silicon substrate microelectrodes at 400 μm intervals in the medio-lateral direction at the C5/C6 and C6/C7 segmental borders. An enzyme-catalyzed synthesis of novel polymers within layer-by-layer (LbL) constructed polyelectrolyte microcapsules has been developed. This approach is based on the selective permeability of polyelectrolyte-capsule walls to monomer molecules. Conversely biocatalysts and forming polymeric chains cannot exit the micro-capsule interior because of their characteristic high molecular weight. Horseradish Peroxidase (HRP) was encapsulated into four bilayer PSS (poly-styrenesulfonate)/PAH (poly-allylamine hydrochloride) capsules with an average diameter of 5 μm using pH-driven pore opening. The polymerization of 4-(2-Aminoethyl) phenol hydrochloride (tyramine) catalyzed by HRP produces easily detectable fluorescent polymeric products after the addition of hydrogen peroxide to the system. Site-specific irrigation enables maximizing yields and water use efficiency for fields with variation in soil water availability. Distributed control for fixed irrigated systems, with controllers close to the sensors and actuators in the field, is easier to install and maintain, and less susceptible to damage by lightning strikes compared to centralized control, but is only economically viable with affordable, low-power controllers. A low cost, solar-powered, feedback irrigation controller for distributed control of fixed irrigation systems was developed and tested. Consequently, a decision must be made for irrigation management about which areas in the field should be treated differently or grouped. Research objectives. One source of falls in the elderly may be an inability to sufficiently adjust to transient postural perturbations or slips. Identifying useful predictors of fall potential, as well as factors that affect the ability of an individual to detect a movement of the standing support surface may provide insight into postural stability and methods to increase stability in elders. The t(4;9)(B3;E3.2)12Gso reciprocal translocation is an autosomal recessive mouse mutation involving chromosome 4, band B3 (Mmu4B3) and chromosome 9, band E3.2 (Mmu9E3.2). The most striking phenotype of the 12Gso homozygote involves axial skeleton deformities, resulting in significant shortening of body length, scoliosis, displaced hips and kinky tails. Homozygous animals are significantly smaller than normal littermates and frequently exhibit early lethality, as many of the mutants perish by 1 day postnatal, and many others do not survive the three weeks to weaning age. The purpose of this study was to improve breast cancer diagnosis by reducing the number of benign biopsies performed. To this end, we investigated modular and ensemble systems of machine learning methods for computer-aided diagnosis (CAD) of breast cancer. A modular system partitions the input space into smaller domains, each of which is handled by a local model. An ensemble system uses multiple models for the same cases and combines the models' predictions.

Cardiovascular disease is the leading cause of death in North America. Cardiac infarction caused by thrombus plaque rupture can often lead to sudden death. Arterial stenosis caused by atherosclerosis is an important precursor leading to thrombus formation. Shear stress, abnormal lipid metabolism, and subendothelial layer exposure are the major contributors to arterial thrombus formation. Platelet activation by the above factors, followed by adhesion is the basic sequence in thrombus formation.

In the present dissertation, an automated neural network-based ECG diagnosing system was designed to detect the presence of myocardial infarction based on the hypothesis that an artificial neural network-based ECG interpretation system may improve the clinical myocardial infarction. 137 patients were included. Among them 122 had myocardial infarction, but the remaining 15 were normal. The sensitivity and the specificity of present system were 92.2% and 50.7% respectively. The sensitivity was consistent with relevant research. The relatively low specificity results from the rippling of the low pass filtering. Nitric oxide (NO) has been shown to suppress platelet activation, induce vasodilation, inhibit smooth muscle cell proliferation, and act against infection. Activated platelets generally have opposite effects from these. They secrete platelet-derived growth factor (PDGF), which stimulates smooth muscle growth, serotonin, which is a platelet agonist, and a number of agents that promote further platelet aggregation. However, activated platelets also produce NO through the enzymatic functions of the constitutive form of nitric oxide synthase (NOS). An alternative way of blood oxygenation is proposed by introducing small bubbles directly into the blood stream. To be used for blood oxygenation, the bubble should be small enough to pass through the capillary beds. To explore the feasibility of producing small bubbles, bubble formation phenomena in a crosscurrent liquid flow in a silicon microchannel were studied. Sliding hip screws are routinely used to repair Garden III femoral neck factures. This research seeks to better understand the influence of the hip screw angle on the performance of the fixation. The mechanics of fractured femurs repaired with 135- and 150-degree sliding fixation devices are explored using experimental, finite element, and analytical modeling.
