Ganciclovir (GCV) has been shown to be an effective antiviral for the treatment of congenital cytomegalovirus (cCMV) induced sensorineural hearing loss. However, dose-dependent adverse effects (e.g. neutropenia) require strict, burdensome monitoring, limiting GCV use in some patients. Quercetin is a flavonoid with antioxidant properties that has demonstrated antiviral therapy against CMV. Although more benign than GCV, quercetin has limited therapeutic value due to its poor solubility. We previously reported that GCV improves hearing thresholds in a murine model of cCMV. We also noted that solubilizing quercetin with poloxamer 188 (P188) and combining with GCV therapy amplified CMV inhibition in murine …

Aquatic therapy for rehabilitation can be performed in a variety of environments, which can vary from a traditional swimming pool to a self-contained underwater treadmill. While kinematic analysis has been performed in large volume swimming pools using specific underwater motion capture systems, researchers may only have access to a land-based motion-capture system, which is not waterproof. Additionally, underwater motion capture systems may not fit within the confines of a smaller underwater treadmill. Thus, the purpose of this study was to design and analyze methodology to quantify lower limb kinematics during an aquatic treadmill session, using a land-based motion capture system. …

Highly Branched Polymers Based On Poly(Amino Acid)S For Biomedical Application, Marisa Thompson, Carmen Scholz

PRC-Affiliated Research

Polymers consisting of amino acid building blocks continue to receive consideration for biomedical applications. Since poly(amino acid)s are built from natural amino acids, the same building blocks proteins are made of, they are biocompatible, biodegradable and their degradation products are metabolizable. Some amino acids display a unique asymmetrical AB2 structure, which facilitates their ability to form branched structures. This review compares the three forms of highly branched polymeric structures: structurally highly organized dendrimers, dendrigrafts and the less organized, but readily synthesizable hyperbranched polymers. Their syntheses are reviewed and compared, methods of synthesis modulations are considered and variations on their traditional …

Effects Of Flight Altitude On The Lift Generation Of Monarch Butterflies: From Sea Level To Overwintering Mountain, Madhu K. Sridhar, Chang-Kwon Kang, Brian D. Landrum, Hikaru Aono, Shannon L. Mathis, Taeyoung Lee Mar 2021

Effects Of Flight Altitude On The Lift Generation Of Monarch Butterflies: From Sea Level To Overwintering Mountain, Madhu K. Sridhar, Chang-Kwon Kang, Brian D. Landrum, Hikaru Aono, Shannon L. Mathis, Taeyoung Lee

PRC-Affiliated Research

Aerodynamic efficiency behind the annual migration of monarch butterflies, the longest among insects, is an unsolved mystery. Monarchs migrate 4000 km at high-altitudes to their overwintering mountains in Central Mexico. The air is thinner at higher altitudes, yielding reduced aerodynamic drag and enhanced range. However, the lift is also expected to reduce in lower density conditions. To investigate the ability of monarchs to produce sufficient lift to fly in thinner air, we measured the climbing motion of freely flying monarchs in high-altitude conditions. An optical method was used to track the flapping wing and body motions inside a large pressure …

A major challenge for astronauts in long-duration space travel is combatting the hazardous spaceflight environment caused by microgravity and increased levels of ionizing radiation. Microgravity damages cellular DNA by increasing the production of harmful reactive oxygen species, while ionizing radiation damages DNA by creating double-stranded DNA (dsDNA) breaks. Cellular damage due to microgravity and radiation has been investigated using ground-based models, but most models consider microgravity and ionizing radiation alone, or asynchronously. Synchronous modeling better mimics spaceflight conditions and can be used to understand the combined effects of microgravity and ionizing radiation. However, commercially available devices to model microgravity and …

A green and simple process for preparing the polyethylene glycol passivated fluorescent carbon dots (CDs-PEG) have been studied by a microwave pyrolysis method, using gelatin and PEG as starting materials. This method is very effective for development of carbon-based quantum dots from gelatin with high quantum yield (QY). The synthesized CDs-PEG were found to emit blue photoluminescence (PL) with a maximum QY of 34%. At the following research, we investigated the effect of the presence of PEG on PL intensity, and the result showed that CDs-PEG becomes stronger PL properties than pure CDs from gelatin. The synthesized CDs-PEG were characterized …

The treatment of seeds and plants by electrically generated cold atmospheric pressure plasma can accelerate seed germination and radicle growing rates. The plasma generated reactive oxygen and nitrogen species, UV photons, and high frequency electromagnetic fields can penetrate into seed coats and modify their surface properties. Atomic force microscope data shows that cold helium or argon plasma induces strong corrugation of pumpkin seed coats, produces pores and surface defects. These structural deformations and poration enhance water uptake by seeds during the imbibing process, accelerate seeds germination, and increase seed growth. The cold atmospheric pressure plasmas treatment of pumpkin seeds also …

Despite the recent interest in flapping wing aerodynamics, the aerodynamic sound generation mechanism of a flapping wing is inadequately understood. In this paper, the interplay between the wing motion, resulting unsteady aerodynamics, and aeroacoustics of a flapping wing flyer is investigated. The wing motion is varied in terms of the flapping amplitude, pitching amplitude, and the phase difference between flap and pitch. The unsteady flow around the flapping wing is numerically calculated using a well-validated Navier-Stokes equation solver. Acoustic pressures are computed using the Ffowcs-Williams-Hawkings equation in a three-dimensional space at varying distances from the wing. Two main sound generation …

Biological systems often generate unique and useful structures, which can have industrial relevance either as direct components or as an inspiration for biomimetic materials. For fabrication of nanoscale silica structures, we explored the use of the silaffin R5 peptide from Cylindrotheca fusiformis expressed on the surface of the fd bacteriophage. By utilizing the biomineralizing peptide component displayed on the bacteriophage surface, we found that low concentrations (0.09 mg/mL of the R5 bacteriophage, below the concentration range used in other studies) could be used to create silica nanofibers. An additional benefit of this approach is the ability of our R5-displaying phage …

Late-stage diagnosis of lung cancer occurs ~95% of the time due to late manifestation of its symptoms, necessitating rigorous treatment following diagnosis. Existing treatment methods are limited by lack of specificity, systemic toxicity, temporary remission, and radio-resistance in lung cancer cells. In this research, we have developed a folate receptor-targeting multifunctional dual drug-loaded nanoparticle (MDNP) containing a poly(N-isopropylacrylamide)-carboxymethyl chitosan shell and poly lactic-coglycolic acid (PLGA) core for enhancing localized chemo-radiotherapy to effectively treat lung cancers. The formulation provided controlled releases of the encapsulated therapeutic compounds, NU7441 - a potent radiosensitizer, and gemcitabine - an FDA approved chemotherapeutic drug for lung …

Cartilage injury induced by acute excessive contact stress is common and mostly affects young adult. Although early detection of cartilage injury may prevent serious and lifelong arthritic complications, early detection and treatment is not possible due to the lack of a reliable detection method. Since chondrocyte injury and subsequent cell death are the early signs of cartilage injury, it is likely that cartilage cell apoptosis can be used to predict the extent of injury. To test this hypothesis, a near infrared probe was fabricated to have high affinity to apoptotic cells. In vitro tests show that this apoptosis probe has …

Highly environment-sensitive fluorophores have been desired for many biomedical applications. Because of the noninvasive operation, high sensitivity, and high specificity to the microenvironment change, they can be used as excellent probes for fluorescence sensing/imaging, cell tracking/imaging, molecular imaging for cancer, and so on (i.e., polarity, viscosity, temperature, or pH measurement). In this work, investigations of the switching mechanism of a recently reported near-infrared environment-sensitive fluorophore, ADP(CA)2, were conducted. Besides, multiple potential biomedical applications of this switchable fluorescent probe have been demonstrated, including wash-free live-cell fluorescence imaging, in vivo tissue fluorescence imaging, temperature sensing, and ultrasound-switchable fluorescence (USF) imaging. The fluorescence …

Simultaneous imaging of multiple targets (SIMT) in opaque biological tissues is an important goal for molecular imaging in the future. Multi-color fluorescence imaging in deep tissues is a promising technology to reach this goal. In this work, we developed a dual-modality imaging system by combining our recently developed ultrasound-switchable fluorescence (USF) imaging technology with the conventional ultrasound (US) B-mode imaging. This dual-modality system can simultaneously image tissue acoustic structure information and multi-color fluorophores in centimeter-deep tissue with comparable spatial resolutions. To conduct USF imaging on the same plane (i.e., x-z plane) as US imaging, we adopted two 90 -crossed ultrasound …

Decellularized extracellular matrix (ECM) contains complex tissue-specific components that work in concert to promote tissue repair and constructive remodeling and has been used experimentally and clinically to accelerate epithelial wound repair, leading us to hypothesize that lung-derived ECM could mitigate acute lung injury. To explore the therapeutic potential of ECM for noninvasive delivery to the lung, we decellularized and solubilized porcine lung ECM, then characterized the composition, concentration, particle size and stability of the preparation. The ECM preparation at 3.2 mg/mL with average particle size <3 μm was tested in vitro on human A549 lung epithelial cells exposed to 95% O2 for 24 hours, and in vivo by tracheal instillation or nebulization into the lungs of rats exposed intermittently or continuously to 90% O2 for a cumulative 72 hours. Our results showed that the preparation was enriched in collagen, reduced in glycosaminoglycans, and contained various bioactive molecules. Particle size was concentration-dependent. Compared to the respective controls treated with cell culture medium in vitro or saline in vivo, ECM inhalation normalized cell survival and alveolar morphology, and reduced hyperoxia-induced apoptosis and oxidative damage. This proof-of-concept study established the methodology, feasibility and therapeutic potential of exogenous solubilized ECM for pulmonary cytoprotection, possibly as an adjunct or potentiator of conventional therapy.
