The small Mr Rab4-like GTPase, RabD, localizes to the endosomal pathway and the contractile vacuole membrane system in Dictyostelium discoideum. Stably transformed cell lines overexpressing a dominant negative functioning RabD internalized fluid phase marker at 50% of the rate of wild-type cells. Mutant cells were also slower at recycling internalized fluid. Microscopic and biochemical approaches indicated that the transport of fluid to large postlysosome vacuoles was delayed in mutant cells, resulting in an accumulation in acidic smaller vesicles, probably lysosomes. Also, RabD N121I-expressing cell lines missorted a small but significant percentage of newly synthesized lysosomal alpha-mannosidase precursor polypeptides. However, the …

We have investigated the effects of Concanamycin A (CMA), a specific inhibitor of vacuolar type H(+)-ATPases, on acidification and function of the endo-lysosomal and contractile vacuole (CV) systems of D. discoideum. This drug inhibited acidification and increased the pH of endo-lysosomal vesicles both in vivo and in vitro in a dose dependent manner. Treatment also inhibited endocytosis and exocytosis of fluid phase, and phagocytosis of latex beads. This report also confirms our previous conclusions (Cardelli et al. (1989) J. Biol. The role of myosin Is in endosomal trafficking and the lysosomal system was investigated in a Dictyostelium discoideum myosin I double mutant myoB-/C-, that has been previously shown to exhibit defects in fluid-phase endocytosis during growth in suspension culture (Novak et al., 1995). Various properties of the endosomal pathway in the myoB-/C- double mutant as well as in the myoB- and myoC- single mutants, including intravesicular pH, and intracellular retention time and exocytosis of a fluid phase marker, were found to be indistinguishable from wild-type parental cells. Highly purified lysosomes, prepared by magnetic fractionation of homogenates from Dictyostelium discoideum cells fed colloidal iron, were lysed under hypoosmotic conditions, and the membrane-associated proteins were subjected to gel electrophoresis. Thirteen major membrane polypeptides, ranging in molecular weight from 25,000 to 100,000 were identified. The isoelectric points of these proteins ranged from below 3.8 to greater than 7.0. Most of these proteins were stripped from membranes exposed to a chaotropic agent, 3,5-diodo-2-hydroxybenzoic acid lithium salt, and were therefore classified as peripheral membrane proteins. In the course of screening a cDNA library for ras-related Dictyostelium discoideum genes, we cloned a 0.7 kb cDNA (rabD) encoding a putative protein that was 70% identical at the amino acid level to human Rab4. Rab4 is a small M(r) GTPase, which belongs to the Ras superfamily and functions to regulate endocytosis in mammalian cells. Southern blot analysis indicated that the rabD cDNA was encoded by a single copy gene while Northern blot analysis revealed that the rabD gene was expressed at relatively constant levels during growth and differentiation. A new species of Trichoptera, Ceraclea (C.) enodis, of the Ceraclea (C.) senilis Group, was discovered in freshwater sponge in the Little River in Montgomery County, North Carolina. Illustrations and descriptions of the male, female, larva, pupa, and larval case are given. Adults resemble those C. cancellata (Betten), the mature larva looks like that of C. maculata (Banks), the younger larva may be confused with C. transverasa (Hagen), and the pupa has anal rods rather like those of C. spngillovorax (Resh); but all life history forms can be distinguished reliably from these species by consistent morphological characters and the sponge …

Modifications were made in the configurations of the unitizing, nonselective wire cutters used by Alper et al.(1992) for mass cuttings of Stage II Citrullus lanatus cv. Charlee (watermelon) plant tissue cultures to further enhance productivity. Mounting the cutter in an inverted position over the receiving vessel eliminated time required for filling. This cut-and-dump technique became 4.8 times more productive for the total transfer process than the conventional scalpel and forceps technique when both time and yield of cut segments with visible buds were considered. Unitized, nonselective mass cutting of Stage II Citrullus lanatus cv. Charlee (watermelon) plant tissue cultures has been achieved with various configurations and sizes of wire cutter devices. Time studies revealed that the wire cutters increased the productivity of the cutting function over conventional scalpel and forceps by a factor of 14, Total transfer productivity, including the manual functions of removing tissue from an initial culture vessel and filling (sorting and placing cut tissue segments) new vessels, was increased by a factor of 1,8, The square grid-type cutting devices yielded from 48 to 59% as many viable bud clusters per culture …

In North Carolina, Precis coenia that emerge during the Summer months exhibit a ventral hindwing (VHW) with well-defined reddish-brown and brown pattern elements on a light tan background. During late Summer and early Fall, however, individuals begin to appear with poorly defined or obscured pattern elements on a dark reddish-brown background. The present study shows that the Fall (rosa) color morph can be induced by either low rearing temperatures or short daylengths. The effect of such conditions seems to be cumulative throughout the larval life, although animals are much more sensitive during the last 24 hours of larval …

Ninety-three species of caddisflies, representing 14 families, were identified from collections obtained from two localities on Upper Three Runs Creek, Savannah River Site, Aiken County, South Carolina. Collections of adult caddisflies were made biweekly over a 1-yr period with ultraviolet light traps. The similarity index, Jaccard's Coefficient of Communities, showed that the two sites were over 78% similar with 73 species in common. Three species, Oxyethira setosa Denning, Triaenodes smithi Ross, and Nyctiophylax serratus Lago & Harris, are new distributional records for South Carolina. Two species of Triaenodes are new to science. Other species, which were considered to be endemic …

Investigations of materials and methods for growing plant tissue in a continual-flow, liquid nutrient medium as an alternative to semisolid agar medium have been made. Enhanced growth of plant tissue on microporous
polypropylene membranes floating on a liquid nutrient has been demonstrated. Moreover, in vitro plantlets on the microporous membrane are free from entanglement with the support matrix and readily available to mechanized
handling. Trained growth of plantlets through polypropylene netting shows potential for mechanization by mass handling (separation, singulation, and transfer) of plant tissue cultures.

Logging of baldcypress (Taxodium distichum) in the swamps of the southeastern United States is once again becoming common and an area of particular concern in Louisiana is the regeneration of cypress in its natural environment. One way to ensure the proper stocking of cypress is to plant seedlings, but nutria usually damage or destroy newly planted seedlings and are a deterrent to cypress regeneration in flooded areas. In 1985 cypress seedlings were planted in a flooded logged area and in an area where flooding was preventing the establishment of natural seedlings. Nutria destroyed 86% of the seedlings in …

Vexar seedling protectors were tested for preventing nutria damage to baldcypress (Taxodium distichum (L.) Rich) seedlings planted in Louisiana
swamp forests. Five areas were planted with 1-year-old baldcypress seedlings. Half the seedlings in each area were protected with Vexar seedling protectors. The protectors slowed down the rate of destruction in some areas, but after 3 months, 85% of the guarded seedlings and 87% of the unguarded seedlings were destroyed.

A phylogeny of the families of Trichoptera is reviewed to provide a basis for understanding the probable evolution of feeding tactics and case or retreat constructions by larvae. At lease 48 hierarchically inclusive homologues are known, mostly from larval, pupal, and adult morphology. Their resulting phylogeny indicates that Rhyacophilidae, Hydrobiosidae, Glossosomatidae, and Hydroptilidae are more closely related to Philopotamidae, Hydropsychidae, and their allies than to Limnephilidae, Leptoceridae, and their allies. This phylogeny implies that the ancestral caddisfly larva was probably a tube-dwelling detritivore, inhabiting humus and detrial mats near the shores of lentic or lotic-depositional habitats. As discussed in detail in Part I of this series (Morse et al. 1980), aquatic insects were collected biweekly from six locations on Upper Three Runs Creek, Savannah River Plant near Aiken, South Carolina, from September 1976 through August 1977, using light traps and semi-quantitative benthic sampling methods. See part I (Morse et al. 1980) for site and habitat descriptions and for a discussion of the methods used. In that publication, faunistic results were provided for all insect taxa except Diptera and relative abundance data for all but Diptera and non-dryopoid Coleoptera.

Upper Three Runs Creek and its tributary sources, located mostly on property of the Savannah River Plant near Aiken, South Carolina (Fig. 1), is well-known as an unpolluted, blackwater drainage system which is probably typical of smaller, Sandhills waterways of primeval southeastern North America. Probably no other stream at the Plant's Flowing Stream Laboratory (or Thermal Effects Laboratory) has been documented by a research team from the Academy of Natural Sciences of Philadelphia (A.N.S.P.).
