This study was developed to use aquatic environmental DNA (eDNA) as a tool to detect the presence of the Santa Ana sucker (Catostomus santaanae), a federally threatened fish species, across the Santa Ana River in Southern California. Environmental DNA is any genetic material that an organism sheds into the environment. We can get an idea of what aquatic species are present when sampling water at a given site. We sequenced the complete mitochondrial genome (mitogenome) of the Santa Ana sucker and designed custom DNA primers to improve eDNA detection accuracy. Understanding the evolution of infectious diseases within a dynamic population is essential for formulating effective public health strategies. This work introduces an enhanced SEIRS epidemic model that incorporates demographic variation and a single-dose vaccination strategy, supporting that immunity can be reduced over time. The model reflects diseases in which individuals may return to the susceptible state after recovery. Extending prior reduced three-dimensional models, this study develops a complete four-dimensional SEIRS framework, thereby increasing the model’s applicability to real-world scenarios. Biogeographical ancestry (BGA) inference is an important tool in forensic science, providing supplemental information when comparison/reference DNA is not available for traditional DNA analyses. Forensic taphonomy examines the postmortem processes that affect human remains within specific environmental contexts and plays a critical role in medicolegal death investigation. Despite significant advances in decomposition research, experimental studies remain geographically and environmentally uneven, limiting the transferability of postmortem interval (PMI) estimation methods across regions. In 1985, India banned the export of human remains. This ended the major trade route for primarily US and European anatomical supply companies. Until this point, companies like Carolina Biological Supply exploited and erased vulnerable people's identities, molding them into anonymous anatomical specimens. This study investigates the life and postmortem history of twenty-five individuals, previously used as anatomical specimens, housed at the University of Nebraska–Lincoln. Using an osteobiographical approach, this research reconstructs aspects of these individuals' lived experiences and examines how anatomical preparation practices reflect structural violence and apathy.

This study explores the development and validation of an automated, deep learning system designed to differentiate human from large mammal long bone fragments using micro-CT imagery. A comprehensive dataset of cross-sectional µCT images was assembled from human and animal skeletal material processed at the DPAA, Nebraska. A convolutional neural network (CNN) based on a ResNet-18 architecture, utilizing transfer learning from ImageNet weights, was trained to classify µCT images as human or animal. The model achieved a mean classification accuracy of 89.85% (± 8.27%) across five sample-level cross-validation folds, with a sensitivity of 96.21% and a 95% bootstrap confidence interval of …

Over the last thirty years the mortality rate of the undocumented population at the US-Mexico border has risen, requiring methods distinguishing US-born individuals from immigrants to improve the likelihood of identification and repatriation through foreign consulates. Forensic anthropologists with the Pima County Office of the Medical Examiner (PCOME) have advocated for the use of the biocultural profile, combining the biological profile with factors including oral health, osteological stress, and personal effects. Pepper spray is a common self defense tool used in both law enforcement and civilian settings, with the goal of incapacitating the sprayed individual. In real casework where pepper spray was used, a red-brown stain which tested positive presumptively for blood provided no DNA profile after further testing six months later. A previous graduate student, Emma Crawn, attempted to emulate these casework conditions and instead achieved a DNA profile with slight indications of inhibition or degradation. While DNA profiling of sexual assault evidence can identify biological contributors, determining the tissue source of recovered profiles remains a significant challenge, particularly in cases of suspected oral assault involving mixtures of epithelial cells. The initial goal of this study was to identify non-genetic signatures for differentiating two types of epithelial cells relevant to oral assaults, saliva epithelial cells and epidermal skin cells. To develop tissue-specific signatures, reference cell populations from saliva and epidermal skin surfaces were collected from 20 donors and analyzed using full spectrum flow cytometry.

Massively Parallel Sequencing (MPS) is a new Forensic Science field allowing multiple samples to be sequenced simultaneously. In contrast to CE, multiple types of genetic markers including those from different nucleic acids can be used in the same assay. Additionally, MPS has opened the doors to certain fields of Forensic DNA Analysis like Forensic Investigative Genetic Genealogy and Forensic Phenotyping assays containing upwards of 100 SNPs. While quite expensive and requiring a high cost for initiating, MPS has the potential to save time and greatly increase the throughput of Forensic DNA Analysis in crime labs.

This study compared the efficiency of two carrier nucleic acid methods, specifically using salmon sperm DNA (spDNA), in robotic solid-phase extractions of cotton swab samples. Robotic extractions reduce processing time and workload but result in substantially lower DNA recovery than manual organic extraction. This research tested 10 µg of spDNA being added to the lysis buffer or pre-treating the swabs with it before DNA was added. Both methods showed a significant increase in recovery compared to extractions with no carrier. In the buffer extractions, recovery ranged from 69.4% to 117.5% and the swabs being pre-treated ranged from 84.2% to 125.8% …

Dental cementum is characterized by lifelong deposition of alternating opaque annuli in fall/winter and translucent bands in spring/summer. This study aims to test the applicability of cementochronology to estimate season-of-death, by observing the properties of the last layer, with direct implications for forensic anthropology and bioarchaeology. The sample includes canine teeth from 146 adults (84 males, 62 females), age-at-death ranging from 20-97 years (mean=61.7 years, SD=18.6 years), from two contemporary skeletal collections. Of these individuals, 78 (53.4%) died in spring/summer and 68 (46.6%) died in fall/winter. Overall accuracy was low at 64.4% (94/146). Accuracy was higher for spring/summer deaths 100.0% …

This study investigated how tree species diversity influences the variety of organisms living on or around trees across different habitat types. Field data were collected through random tree selection, measurement of tree diameter, identification of species, and estimation of surrounding ground cover types such as grass, herbaceous plants, woody plants, and leaf litter. Biodiversity associated with each tree was analyzed using the Shannon Diversity Index across several ecosystems, including grassland, woodland, and pond environments.
